A Study on the Evaluation Methods of Indoor Thermal Comfort Index in Building (실내(室內) 온열환경지표(溫熱環境指標)의 평가방법에 관한 연구)

  • This objective of this paper is to investigate the evaluation and indiction of human thermal comfort in building environment. The issue of defining the boundaries of acceptable thermal comfort conditions in buildings and urban may have significant implication for building design and also may have urban design by climate considerations. And then it is to apply the thermal comfort condition to environmental design by using passive methods in Korea. Since 1920. architects have conducted studies to measure thermal comfort in houses under hot and humid conditions, while industrial hygienists have studied the effects of temperature and humidity on the performance of factory workers. Thermal comfort can be influenced by many variables. This paper conducted to review the previous researches and the human heat balance equation, and to analyse in order to reveal the meaning and usage of the thermal comfort index in two traditional essays, Fanger's PMV and Gagge's ET* Their comfort indexes compared with each other. They were based on human heat balance equation and psychological and physiological responses in the laboratory tests. The researchers and the architectural engineers using thermal comfort index shall be careful in decided the use of indexes and be necessary to recognize the value concept of the design criteria for thermal comfort. Therefore, The opinion of the authors is that different comfort standards have to apply for each building and urban with different climatic conditions.

  • Unripe indigenous persimmons which contain rich tannins have been used as natural dye materials traditionally and have been using continuously for dyed clothes named Gal-ot in Jeju. Those persimmons were cheap and easy to use as dyes because of inedible and widely cultured in Korea. Persimmon juice dyes not only make fabrics brown-color but also give functional and hygienic properities such as stiffness, air ventilation of clothes, antibacterial activity, protectivity against ultraviolet light. However there are several serious problems which are ristriction of dyeing periods, longtime irradiation, uneven color and low color fastness etc. in persimmon juice dyeing. This study purpose to improve dye effect and method in order to enlarge useability of persimmon juice dyeing. Cotton fabrics were pad-dyed to 100% pick-up using padding machine after dipping in persimmon juice extracted from unripe persimmons indigenous from Jeju. It was possible and available to control pick-up rate. The color of dyed cotton fabrics by padding method was more even and repeatable than which by traditional hand method. Persimmon juice concentrations were 4 types of 10, 25, 50 and 100%. The more concentration increased, the more color deepened. UV Irradiation instead of sunlight was applied to color developing. Irradiation times were shortened till 1∼8 hrs. Same color values could be taken without water wetting which were required in sunlight irradiation. Tensile strengths of cotton fabrics pad-dyed with low concentration of persimmon juice decreased but recovered at high concentrations. Elongations(%) of cotton fabrics pad-dyed with persimmon juice were increased 1% more than undyed cotton in sunlight irradiation. Drape stiffness increased upto double times as much as.

Research on University Students' Recognition of Recreation (대학생들의 레크리에이션에 관한 인식 조사)

  • This study speculates on the Korea's current recreation activity via surveys on students' recognition of recreation activity. For the research, the number of 200 university students (subjects) in D metropolitan city who had attended a recreation class or program was collected as a population, among which the data of 188 subjects were used for the real analysis. The collected data were analyzed through 'Frequency Anaalysis' and 'Chi-square Test' using SPSS Ver. 12.0. First, 'gender' was not significant in the area of students' recognitional difference according to their personal data (gender, department); on the other hand, a slight difference was found in 'department'; but in general, the recreation activity was regarded as a sort of entertainment in which people gather and have fun together, and it was thought to be involved in leisure. Second, the recreational activity in students' minds was presented into school MT, orientation, athletic meeting, camping, etc. The results demonstrate that the concept of recreation in our country became considerably reduced compared to its original meaning, and a recreation leader's authority came to be also low-valued.

  • To elucidate the mechanism underlying the embryotropic effect of extracellular matrix(ECM) on the preimplantation development of mammalian embryos, the involvement of mitogen-activated protein kinase(MAPK) downstream the integrin signaling was examined in mouse blastocysts. Blastocysts were cultured in the presence of growth factor-reduced(GFR) Matrigel(0.5%, v/v). MAPK activity was measured by in vitro phosphorylation of myelin basic protein by the Erk1/2 antibody immunoprecipitates of embryonic extract following the Matrigei treatment. MAPK activity of the early blastocysts rapidly increased within 10 min fo1lowing the Matrigel treatment. When the embryos were cultured for 12 h in the presence of Matrigel, the MAPK activity was significantly higher than that ot the control embryos. PD098059, a MAPK kinase(MEK) inhibitor, attenuated the effect of Matrigel on the change in MAPK activity. Taken together, it suggested that the embryotropic effect of ECM proteins might be mediated by the activation of MAPK cascade.

  • The purpose of research is to complement the circulating pump of gas boiler for the domestic and to develop a pump housing with internal three way valve: Housing and three way valve have been apart in the existing pump system. Based on the experimental result on can-typed circulating pump with existing housing, a new housing with internal three way valve was designed and manufactured. The performance of can-typed circulating pump with the new housing, and the performance of circulating pump of boiler system were tested. As a result ot the test, the new housing with internal three way valve has been excellent in respect of pump performance, weight and manufacturing cost. So It is expected to have an effect of import substitution.

  • The effect of media feeding rate on cell growth and monoclonal antibody production in the fed-batch culture ot hybridoma A4W was studied. In the batch culture, the highest specific antibody production rate was observed at the begining of the culture period but its value tended to decrease rapidly with the culture time. The final antibody concentration and volumetric productivity was 65 $\mu g$/ml and 13 mg Mab/l/day, respectively. In the fed-batch culture, the specific antibody production rate, $q_p$ rebounded sharply within a few hours after the media feeding was started and it remained high until the end of culture if the media feeding was continued. The final antibody concentration was 220 $\mu g$/ml and the volumetric productivity was 45.1 mg/l/day. Further increase in final antibody concentration was achieved by applying a modified media of which component was fortified with glucose and glutamine, hence the final antibody concentration in this case was 270 $\mu g$/ml and the volumetric productivity was 51.8 mg/lday, which is as four tinlcs as high cuixparinf! to that of batch culture.

  • Isolation of the toxic substance which was produced by Streptentyces sp. and it's biorhemical characterestics and toxicity to fishes were reported in the previous paper. The present report includes antibiotic activity of the substance, inhibitory activity of the substance on the succinic dehydrogenase of fishes, and its effect on the blood corpuscles of a rabbit. An evident antibiotic activity of this substance was observed on Candida yeasts, but n$ot^1$on molds or bacteria. The substance inhibited the growth of Candida japonica and C. utilis to the 50% at the concentrations of 7.5 and 10.2ug per ml, respectively. The activity of succinic dehydrogenase obtained from various organs of Cyprinous carpio L. was also found to be inhibited by this substance. Original activities of the enzymes from tht brain, kidney, and liver, were inhibited by 75.4%, 38.2%, and 26.2%, respectively, but not the enzymes from the heart and spleen. Neither leukopenia nor leukocytosis was detected after the intravenous administration of the substince to the rabbit at the level of 6 mg per 2 kg body weight.

  • As the areas of computer application are expanded, the real-time applicition enviroments that must process as many transactions as possible within their deadlines have been increased recently. Conventional disk based databaes system is not appropriate in real-time transaction processing due to delying time for disk I/O processing. When the system is overloaede, the performance of transaction scheduling technique using earliest deadline first deteriorates rapidly because it can assign the highest priority ot a transaction that has already missed or is about to miss tis deadline. Therfore, the performance of suggested transaction secheduling technique is made to improved by propos-ing the doule locking mechanism based on priority. Finally, in order to evaluate the performance of the proposed priority-based double locking techniques under single proessor and main memory database system environments, the simulation model was developed using the SLAM II language.

  • During the period of the Cochlodinium polykrikoides red tide occurrence in the southern coast of Korea in 1997, we obseved the vertical migration of red tide organisms in situ, along with environmental factors such as water temperature, dissolved oxygen, pH, chlorophyll $\alpha$ and nutrients for practical application of loess suspension techniques ot diminish fisheries damage. To cope with serious outbreaks of red tide of C. polykrikoides, the finfish culturists resorted with loess suspension to disperse the bloom watermass by turning around it with fishing boats in full speed. Peak in afternoon the cell numbers were diminished in the surface layer after sunset red tide organisms settled down in the middle and bottom layer progressively again after sunrise. Chlorophyll $\alpha$ and dissolved oxygen showed the same trends of variation with the cell number. In the course of controlling the red tide with loess suspension we could not find any severs oxygen deficiency to kill the culture species.

Induction of Leptin cDNA Expression in Esherichia coli Cells (대장균 세포에서 Leptin 유전자의 발현 유도)

  • Leptin gene, an obesity gene, has been known to involve in the regulation of food intake and body weight. It is also thought to be related to the glucose metabolism, insulin secretion and type II diabetes mellitus. Recently, the production of recombinant leptin protein has been attempted for the application in the treatment of obesity and the correction of hereditary obesity and type II diabetes. In the present study, leptin cDNA was cloned from mouse fat cells by RT-PCR and prokaryotic expression of leptin was attempted in order ot prepare a leptin-specific antigen. Immunization of a rabbit with the leptin-specific antigen into a rabbit resulted in the generation of leptin-specific antiserum that could be useful in the detection of leption expressed in various tissues. The sequence of leptin cDNA prepared in the present study wa identical to the previously reported one. Transformation of E. coli(DH5a) cells with the leptin cDNA-inserted translation vector, pGEX-4T-3-leptin followed by treatment with IPTG (0.1mM) resulted in the expression of a large amount of GST-leptin fusion protein with a molecular weight of 44 KDa as an inclusion body. Denaturation of the insoluble fusion protein by 8M urea, 6M guanidium-HCI or 0.1% 2-mercaptoethanol followed by a slow oxidation could not solubilize the inclusion body. The cell extract was subjected to SDS-PAGE and GST-leptin protein electroeluted from the gel was then injected into a rabbit subcutaneously for the immunization. Anti-GST-leptin rabbit antiserum which had a cross reactivity to the GST-leptin protein was generated. Leptin protein expressed in mouse brain and fat tissues was detected by Western blot immunodetection system using the antiserum generated in the present study.
