In the present study, the anti-oxidative properties of 80% ethanol extracts from 140 kinds of natural plants were investigated. Persicaria perfoliata had the highest anti-oxidative relative potency(VitaminE, $ORAC_{PE}$=1.0), followed by Sorbaria sorbifolia var. stellipila, Rosa multiflora, Lysimachia vulgaris var. davurica, Quercus aliena, Quercus mongolica and Lespedeza bicolor ($ORAC_{PE}$${\geq}$ 1.50). Among them, the anti-oxidative activity values of the n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH, and water fractions of the 80% EtOH extracts of P. perfoliata and S. sorbifolia in the ORAC assay system were $1.13{\pm}0.002$, $1.10{\pm}0.004$, $3.67{\pm}0.232$, $1.62{\pm}0.049$ and $1.05{\pm}0.003$; $1.02{\pm}0.007$, $1.00{\pm}0.016$, $2.94{\pm}0.130$, $1.80{\pm}0.019$ and $1.04{\pm}0.012$, respectively, with marked anti-oxidative activity in the ethyl acetate fraction from selected the 2 plant extracts.

Rice is widely grown in Asia and is one of the major dietary staples in the world. Also, rice contains antioxidants which can prevent from oxidative stress related diseases, including cancer, atherosclerosis, and diabetes. Because the rice is consumed cooked, the effect of the cooking process on the antioxidative and antigenotoxic properties of rice is lacking. The aim of this study was to determine the effects of cooking on the antioxidant and antigenotoxic effects of white rice (WR), brown rice (BR), and germinated brown rice (GBR). The antioxidant activities were measured for total phenolic content (TPC), DPPH radical scavenging activity (DPPH RSA), total antioxidant capacity (TRAP), and oxygen radical absorbance capacity (ORAC). The highest TPC was found in uncooked BR (18.4 mg gallic acid equivalent/100 g). After cooking, the TPC of WR significantly increased, while the TPC of BR and GBR were reduced by 47.7% and 36.7%, respectively. The $IC_{50}$ for DPPH RSA was not significantly different in uncooked rice, while the DPPH RSA of WR and GBR decreased after cooking and the DPPH RSA of BR significantly increased. TRAP values in BR and GBR increased after cooking, while the value of WR decreased. The ORAC values of uncooked WR, BR, and GBR were 5.3, 4.3, and $3.9{\mu}M$ trolox equivalent at the concentration of $50{\mu}g/mL$. After cooking, the ORAC value of BR remained unchanged, while the value of GBR increased and the value of WR decreased. The antigenotoxic activities of WR, BR, and GBR were determined by measuring the inhibitory effects of $H_2O_2$-induced DNA damage on human leukocytes using the comet assay. The results showed that all rice tested showed a significant antigenotoxic effect against oxidative stress, except for the cooked white rice. Overall, our results indicate the addition of brown rice and/or germinated brown rice to cooked white rice is a good option for improving the benefits of rice.

The antioxidant activities of 29 components isolated from the aerial parts of Artemisia iwayomogi were evaluated in vitro and in cell culture. Among the tested compounds, 2, 6, 8, 10, 13, and 14 exhibited the greatest peroxyl radical-scavenging activities in the oxygen radical absorbance capacity (ORAC) assay, and 2, 10, and 14 also showed significant reducing capacities. However, all compounds showed weak metal chelating activities. Their cellular antioxidant activities were evaluated in HepG2 cells. At $10{\mu}M$, compounds 6, 8, and 14 exhibited stronger protection against 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress than compounds 2, 10, and 13. Moreover, Compounds 2 and 8 were more effective in protecting against $Cu^{2+}$-induced oxidative stress than compounds 6, 10, 13, and 14 at $10{\mu}M$. These results suggest that the phenolic compounds in A. iwayomogi have the potential to be developed as natural antioxidants for the treatment of oxidative stress-related diseases.
Arginyl-fructose (AF) and arginyl-fructosyl-glucose (AFG) were chemically synthesized and purified. Their in vitro and cellular antioxidant activity was investigated using oxygen radical absorbance capacity (ORAC) and cellular antioxidant activity assay, respectively. The peroxyl radical scavenging activity of AF was much higher than that of AFG, which was in good agreement with their reduction capacity to donate electrons or hydrogen atoms. On the other hand, the hydroxyl radical scavenging activity of AF was weaker than that of AFG, which was consistent with their metal chelating activity, suggesting that AFG-$Cu^{2+}$ complex may be less redox-active than AF-$Cu^{2+}$ complex due to 1 glucose molecule attached. The cellular antioxidant activity of AF and AFG appeared to depend on both their permeability into cell membrane and the scavenging activity on peroxyl or hydroxyl radicals. These results indicate that AF and AFG, Maillard reaction products, may have a high potential as a material for the development of nutraceutical food with antioxidant activity.
The present study aimed to characterise anti-oxidant peptides from water-soluble protein extracts of Hanwoo beef and evaluate their anti-proliferative effect on human colorectal carcinoma cells (HCT116). Antioxidant peptides were purified from the low-molecular-weight fraction (<3 kDa) of Hanwoo beef extract. Antioxidant activity of peptide fractions was determined using the oxygen radical absorbance capacity (ORAC) assay. Purified peptide (P3) displayed higher ORAC activity than the low-molecular-weight fraction ($202.66{\mu}M\;TE/g$ vs $167.38{\mu}M\;TE/g$ of dry matter, respectively) (p<0.05). The peptide sequence of P3 was Cys-Cys-Cys-Cys-Ser-Val-Gln-Lys (888.30 Da). The novel peptide P3, at $250{\mu}g/mL$, also significantly inhibited HCT116 cell proliferation up to 25.24% through phosphorylation of ERK, JNK, and p38 kinase (p<0.05). Hence, antioxidant peptide P3 from Hanwoo beef extract can be used as an antioxidative and anticancer agent in the functional food industry.
This study examined the antioxidative and lipid accumulation inhibitory effects of 14 plants from Mongolia and Myanmar on 3T3-L1 and HepG2 cells. The total phenolic and flavonoid contents (TPC and TFC) of 14 plant extracts were measured, and the antioxidative activities were analyzed using DPPH, ABTS, FRAP, and ORAC. After measuring the pancreatic lipase levels and performing the thiobarbituric acid assay, the degree of lipid accumulation was determined by lipid (Oil Red O) staining and triglyceride assay in 3T3-L1 and HepG2 cells. M. paniculate (259.43 mgGAE/g) and C. benghalensis (130.78 mgNAE/g) had the highest TPC and TFC, respectively, among the 14 plants. R. acicularis Lindl. had the highest antioxidant activity in DPPH. The ABTS, FRAP, and ORAC results showed that the antioxidant activity of 11 species was higher than that of the positive control. The pancreatic lipase inhibitory effect of C. angustifolium Scop. was reduced to 23.65% at 0.1 mg/mL, and the level of lipid peroxidation of C. abrorescens Lam. was 0.63 nmol/mg. Five selected plants inhibited the lipid accumulation and triglyceride content, respectively, in 3T3-L1 and HepG2 cells. These results provide scientific evidence for developing functional foods using 14 plants from Mongolia and Myanmar, which have antioxidant activities and lipid accumulation reduction effects.
This study was conducted in order to evaluate the antioxidant activity of extruded ginseng in different extracted fractions. Each of the fractions obtained from extruded ginseng and ginseng (control) were extracted with 80% ethanol, and then the lipophilic components were removed with ether while the hydrophilic components were separated with water-saturated butanol. Each of the 80% ethanol/butanol/water layers were collected and evaporated to acquire samples for tests of saponin content and antioxidant activity. The antioxidant activity of extruded ginseng fractions and ginseng fractions were determined via the oxygen radical absorbance capacity (ORAC) assay. Overall, the extruded ginseng samples harbored saponin contents of 2.2 (Rg1), 2.3 (Re), 1.2 (Rc), 1.3 (Rb2), and 2.2 (Rd) times that measured in the ginseng prior to extrusion. Antioxidant capacity was also higher, not only in the 80% ethanol/butanol which harbor a significant quantity of saponin, but also in the water fractions, which harbor relatively low quantities of saponin as compared to the control samples. Hypsizygus marmoreus (white cultivar), also called white beech mushrooms, are edible mushrooms commercially cultivated in Korea and Japan. This study was carried out to evaluate the antioxidant properties of H. marmoreus. H. marmoreus fruit bodies were divided into pileus and stipe. The pileus and stipe were extracted into water and 80% ethanol and their antioxidant activities were analyzed. The total polyphenol content was highest in the water extract (pileus 1137.39±0.38 mg of GAE (gallic acid equivalents)/100 g, stipe 700.86±0.06 mg of GAE/100 g) compared to the ethanol extract (pileus 923.47±0.18 mg of GAE/ 100 g, stipe 324.05±0.03 mg of GAE/100 g). Ethanol extracts from pileus showed better scavenging ability on DPPH (47.32±0.23% at 10 mg/ml) and ABTS (57.33±0.10% at 10 mg/ml) than the stipe and water extract groups. Water extract from pileus were more effective in reducing power and ORAC (oxygen radical absorbance capacity) value than stipe and ethanol extract. The toxicity of water and ethanol extracts was investigated using WST-1 (Water Soluble Tetrazolium salt) assay on the mouse macrophage cell line RAW 264.7. Overall, total polyphenol content and antioxidant activities of extracts from H. marmoreus increased in a dose dependent manner while pileus was showed better total polyphenol content and antioxidant activities than stipe.
This study was carried out to evaluate potential of Hypsizygus marmoreus (brown cultivar) as a functional food and drug materials. H. marmoreus were divided into pileus and stipe and extracted in hot water and 80% ethanol. The total polyphenol content was highest in the hot water extracts (pileus 17.15±0.19 mg of GAE g, stipe 7.37±0.16 mg of GAE/g) and pileus compared to the ethanol extracts (pileus 10.23±0.14 mg of GAE/g, stipe 3.76.±0.19 mg of GAE/g) and stipe. Also, hot water extracts of pileus from H. marmoreus (brown cultivar) was more effective DPPH, ABTS, ORAC value, reducing power than ethanol extracts and stipe extracts. The pileus and stipe extracts were confirmed to be non-cytotoxic in the mouse macrophage cell line RAW 264.7 determined by WST-1 (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) assay. Overall, extracts of H. marmoreus (brown cultivar) was higher antioxidant activity than other mushrooms, and no cytotoxicity. Therefore, H. marmoreus (brown cultivar) showed potential as a functional food and drug materials. The brown cultivar of H. marmoreus have higher antioxidant activity than white cultivar, H. marmoreus seem to have different antioxidant activity depending on the cultivar.

We aimed to provide the basic data for the development of a beverage using the juices from bioresources from Jeju. Our results show that pH and $^{\circ}Bx$ of the bioresources ranged 2.0-6.5 and 3.3-16.8, respectively. Rubus coreanus Miquel juice had the highest total phenol content (47.3 mg gallic acid equivalent (GAE)/100 mL). Citrus sphaerocarpa juice showed higher rates of 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging (86.8%) than those of other juices. However, the oxygen radical absorbance capacity (ORAC) value ($2,409.5{\mu}M$ TE/mL) of Citrus sudachi Hort. ex Shirai juice was higher than those of other juices. A high correlation (R=0.7343) was observed between the pH and ORAC values for the 20 bioresources. Furthermore, a high correlation (R=0.8752) was found between the phenolic contents and DPPH radical scavenging for the 5 citrus fruits. These results suggest that the bioresources in Jeju could be used as natural antioxidants for the development of functional foods, including healthy beverages.
