Antioxidant Properties of Erigeron annuus Extract and Its Three Phenolic Constituents

  • The antioxidant activity of the extract of Erigeron annuus was assessed by means of two different in vitro tests: bleaching of the stable 1,1-diphenyl-2-picrylhydrazyl radical (DPPH test) and the scavenging of authentic peroxynitrite in company with peroxynitrite generation from 3-morpholinosydnonimine (SIN-1). In both tests, the 85% aq. MeOH and n-BuOH soluble fractions of the crude extract showed a significant scavenging effect on peroxynitrite and DPPH radical in comparison to L-ascorbic acid. And bioassay-guided fractionation of the n-BuOH soluble fraction led to the isolation of three compounds: Apigenin (1), quercetin-3-O-glucoside (2), and caffeic acid (3). The structures of the isolated compounds were elucidated on the basis of their spectroscopic data and their antioxidant activities were measured by determining their capacity to scavenge peroxynitrite and the DPPH radical.

  • A new phenolic glucoside (1), pursargentoside, was isolated from the leaves of Prunus serrulata var. spontanea, along with three other known compounds, orobol 7-O-glucoside (2), $1{\beta}$, $2{\alpha}$, $3{\alpha}$, 24-tetrahydroxy-urs-12-en-28-oic acid (3), and chlorogenic acid (4). The structure of pursargentoside (1) was identified by spectroscopic data analysis including 1D and 2D NMR spectroscopy, as 2-O-${\beta}$-(6'-benzoyl)-glucopyranosyl o-(Z)-coumaric acid. Compounds 1, 2, and 4 exhibited ONOO- scavenging activity, whereas compound 3 was determined to be virtually inactive.

호두 추출물의 항산화 활성과 신피질에서 세포 손상과 지질과산화 방지효과 (Antioxidant Activity of Extract from Walnut Uuglans sinensis Dode) and Its Protective Effect on Cell Injury and Lipid Peroxidation in Renal Cortical Slices)

  • 본 연구에서는 호두박 분획층의 항산화 활성을 검색하기 위해 MeOH, $CH_2Cl_2$, EtOAc, BuOH 그리고 $H_2O$ 분획으로 추출하여 수행하였고, 각 분획층의 산화적 세포손상과 지질 과산화의 방지 효과를 in vitro에서 확인하였다. 그 결과 호두박의 free radical (DPP보 radical) 소거 활성은 EtOAc층에서 가장높게 나타났고, 활성산소종 억제율과 peroxynitrite $(ONOO^-)$ 소거 활성은 $CH_2Cl_2$층을 제외한 모든 분획층에서 높게 나타났다. 신피질 절편에 t-BHP의 처리 시 LDH의 방출과 지질과 산화를 증가시켰고 이러한 변화는 호두의 MeOH, EtOAc, n-BuOH 분획에 의해서 완전하게 방지되었다. 이러한 결과는 호두 추출물이 t-BHP에 의한 신장 세포 손상에 효과적이고 이러한 효과는 항산화력에 의한 것으로 추측된다. 또한 이러한 결과는 호두 추출물에서 많은 질병의 원인이 되고 있는 산화적 스트레스를 방지할 수 있는 효과적인 약물 개발의 가능성을 시사한다

고지방식사로 유도된 신장 산화스트레스를 개선하는 가압볶음 무말랭이 열수추출물 효과 (Effects of hot water extracts of roasted radish against renal oxidative stress induced by high-fat diet)

  • 고지방식사를 섭취한 쥐의 신장에서 가압볶음 무말랭이 추출물의 산화방지, 항염증 효과를 확인하였다. 실험군은 chow diet와 증류수를 경구 투여하는 NOR group, 고지방식사와 증류수를 경구 투여하는 CON group, 고지방식사와 237 mg/kg bw/day농도의 DR과 RDR를 각각 경구 투여하는 DR group, RDR group으로 나누어 12주간 사육하였다. 신장의 산화스트레스 지표인 ROS, $ONOO^-$, 그리고 지방질과산화물의 농도를 확인한 결과 CON group이 NOR group에 비해 유의적으로 증가하였다. 반면 DR group과 RDR group에서 유의적으로 감소하였다. 체내 산화방지 지표인 글루타싸이온의 농도와 산화방지단백질의 발현은 DR group과 RDR group 모두 CON group에 비해 증가하였다. NF-${\kappa}B$ 발현은 CON group이 NOR group에 비해 증가하였으나 DR group과 RDR group은 모두 감소하였다. RDR group은 DR group에 비해 $ONOO^-$ 및 지방질과산화물 농도가 감소하였고 글루타싸이온 농도와 산화방지 관련 단백질 발현 중 Nrf2, HO-1, 카탈레이스, 그리고 GPx가 증가하였고 염증반응 전사인자인 NF-${\kappa}B$의 발현이 낮아졌다. RDR group의 산화스트레스 개선효과는 유의적으로 높은 것으로 확인되었다. 따라서 가압볶음무말랭이를 차로 제조하여 섭취할 때 고지방식사로 유도된 신장의 산화스트레스를 억제할 수 있을 것으로 사료되며, 그 효과는 무말랭이보다 높은 것으로 확인되었다.

천년초 줄기의 항균, 항산화 및 항암효과에 관한 연구 (The Effects of Antimicrobial, Antioxidant, and Anticancer Properties of Opuntia humifusa Stems)

  • 본 연구는 천년초 줄기를 이용하여 항균, 항산화, 항암 등 천년초의 생리활성 효과를 연구하였으며 그 결과는 다음과 같다. 천년초 줄기의 추출물중 gram 양성균인 4가지 균주(Rhodococcus equi, Staphylococcus aureus, Clostridium perfringens, Bacillus cereus)에서 항균 효과가 나타난 것은 메탄올과 핵산층이었으며, 4가지 균주 중 Bacillus cereus를 제외한 3종에 대해서는 우수한 항균효과를 나타냈다. 천년 초 줄기의 peroxynitrite($ONOO^-$) 제거능 측정결과 핵산층을 제외한 메탄올층, 부탄올층, 수층의 항산화 활성은 천년초 줄기 추출물의 농도가 증가할수록 항산화 효과가 유의적으로(p<0.05) 높게 나타났다. 천년초 줄기 추출물의 각 용매별 DPPH 유리기 소거능은 농도가 증가함에 따라 소거능도 유의적으로(p<0.05) 증가하였으며, 부탄올>메탄올>수층>핵산층 순으로 높게 나타났다. 천년초 줄기의 암세포 증식 억제 효과에서 피부암 세포의 경우 $50\sim100{\mu}g/mL$의 농도에서 메탄올층과 핵산층이 수층과 부탄올층에 비하여 유의적으로(p<0.05) 높게 나타났다. 간암세포인 HepG2의 경우 추출물 $100{\mu}g/mL$의 농도를 기준으로 메탄올층이 유의적으로(p<0.05) 높았고, 핵산층>부탄올층>수층의 순으로 나타났다. 대장암세포인 HT-29 역시 메탄올층이 유의적으로 가장 높았고, 핵산층>수층>부탄올층 순으로 나타났다. 유방암세포인 MCF-7은 메탄올층>핵산층>수층>부탄올층 순으로 효과가 높게 나타났다. 이와 같은 결과로 볼 때 천년초줄기의 항균, 항산화, 항암 효과가 높게 나타나 줄기를 활용한 항균제품이나 여러 가지 가공제품을 개발하는데 기초 자료로 활용할 수 있다.

MIA로 유발된 골관절염 동물모델에서 산수유 추출물의 골관절염 개선 효과 (Protective Effect of Corni Fructus Extracts on MIA-induced Animal Model of Osteoarthritis: Effect of Corni Fructus Extracts on OA)

  • Objectives: Osteoarthritis (OA) is a chronic and degenerative joint disease characterized by progressive degeneration of articular cartilage. Inflammation is a recognized and important factor of OA progression. The present study was designed to investigate the protective effect of Corni Fructus water extract (CFW) on a monosodium iodoacetate (MIA)-induced rat model of OA. Methods: Osteoarthritis was induced by injection of MIA (50 µL; 80 mg/mL) into the knee joint cavity of rats. After an adaptation period for seven days, the rats were divided into 4 groups (n=8/group): normal, control, indomethacin-treated (5 mg/kg), and CFW-treated (200 mg/kg) groups. The rats were treated orally for 14 days. Pain was evaluated by determining hind paw weight distribution. For biochemical analyses, we measured the changes in reactive oxygen species (ROS) and peroxynitrite (ONOO-) in the knee joint. The presence of anti-oxidant proteins and inflammatory proteins was determined by western blotting. Results: The administration of CFW significantly improved the hind paw weight distribution. The ROS and ONOO- levels of knee joint were significantly decreased in the CFW group. CFW inhibited the production of inflammatory mediators, such as COX-2, and inflammatory cytokines, including IL-6 and IL-1β, via the NF-κB signaling pathway. The expression of anti-oxidant enzymes, such as catalase and GPx-1/2 also increased significantly. Conclusions: The findings indicate that CFW has a therapeutic and protective effect on OA by suppression of inflammation. Therefore, CFW could represent a potential and effective candidate for OA treatment.

울릉 마가목의 클로로겐산 이성체의 최종당화산물의 생성 저해 및 라디칼 소거 활성 (Chlorogenic Acid Isomers from Sorbus commixta of Ulleung Island Origin and Their Inhibitory Effects against Advanced Glycation End Product (AGE) Formation and Radical Scavenging Activity)

  • 천연물로부터 당뇨합병증에 효과적인 소재를 개발하기 위하여 본 연구를 수행하였으며, 울릉도산 마가목 열매의 80% methyl alcohol 추출물의 ethyl acetate 가용부로부터 최종 당화산물 생성 억제능($IC_{50}$; $181.2{\pm}2.8{\mu}g/mL$)을 확인하였다. 효능성분의 동정을 위하여 $C_{18}$ 겔 등을 활용한 column chromatography를 수행하여 3종의 페놀성 화합물을 분리하였고, 각 화합물의 화학구조는 NMR 스펙트럼 데이터 해석 및 표품과의 HPLC 직접 비교를 통하여 neochlorogenic acid(1), cryptochlorogenic acid(2) 및 chlorogenic acid (3)로 동정하였다. 이들 화합물에 대해 최종당화산물 생성억제능을 평가한 결과 neochlorogenic acid(1)가 가장 강한 $167.5{\pm}3.5{\mu}M$$IC_{50}$ 값을 나타내었고, cryptochlorogenic acid(2)는 $185.9{\pm}3.7{\mu}M$$IC_{50}$ 값을 나타내었다. 또한, 이들 물질에 대해 $ONOO^-$ 소거 활성을 평가한 결과 cryptochlorogenic acid(2)가 가장 강한 라디칼 소거 활성인 $4.0{\pm}0.2{\mu}M$$IC_{50}$ 값을 나타내었고, 구조이성질체인 neochlorogenic acid(1)가 $4.5{\pm}0.3{\mu}M$$IC_{50}$ 값을 나타내었으며, 이들 활성은 caffeic acid가 결합 양상에 따른 화합물의 구조에 따라 다름이 시사되었다. 향후 이들 활성 물질의 활성 기작에 대한 연구가 필요하며 본 연구 결과는 더욱 우수한 최종당화산물 생성 억제능을 가지는 새로운 선도화합물 발굴을 위한 기초자료로 이용될 수 있을 뿐만 아니라 당뇨합병증 예방 및 치료에 효과적인 천연물질의 상업화를 위한 기초자료로 이용될 수 있을 것으로 생각한다.

태충·삼음교의 침 자극이 Streptozotocin으로 유발된 당뇨쥐의 신장 손상에 미치는 영향 (The Effects of LR3 and SP6 Acupuncture on Renal Damage in Streptozotocin-induced Diabetic Mice)

  • Objectives : This study was performed to investigate the effects of $LR_3$ and $SP_6$ acupuncture on renal damage in streptozotocin(STZ)-induced diabetic mice. Methods : ICR male mice were stabilized for a week and divided into four groups: a normal mice group(N), no-acupuncture diabetic mice group(Control), $LR_3$ acupuncture diabetic mice group($LR_3$), and $SP_6$ acupuncture diabetic mice group($SP_6$). Diabetes was experimentally induced by intraperitoneal injection of STZ(150 mg/kg) in citrate buffer(pH 4.5). For two weeks, $LR_3$ and $SP_6$ acupunctures were administered bilaterally at each point once a day. After two weeks, the animals' weight was measured and they underwent a laparotomy. Serum glucose and blood urea nitrogen(BUN) were measured from the blood taken from the heart. We measured glucose, reactive oxygen species(ROS), peroxynitrite($ONOO^-$) and thiobarbituric acid reactive substances(TBARS) in the kidney and compared expression levels of superoxide dismutases(SOD), glutathione peroxidase(GPx), nuclear factor-kappa B(NF-${\kappa}B$), cyclooxygenase-2 (COX-2), inducible nitric oxide synthase(iNOS) and Interleukin-1 beta(IL-$1{\beta}$). Results : BUN significantly decreased in $LR_3$, $SP_6$ compared to the control group. $LR_3$ showed significantly decreased glucose compared to the control group. $LR_3$, $SP_6$ significantly decreased in ROS and $ONOO^-$ compared to the control group. $LR_3$ significantly decreased in TBARS compared to the control group. $SP_6$ significantly increased in expressions of SOD-1, catalase, and GPx compared to the control group. $LR_3$, $SP_6$ significantly decreased in COX-2 compared to the control group. $SP_6$ significantly decreased in IL-$1{\beta}$ compared to the control group. Conclusions : This study suggests that $LR_3$ acupuncture may be effective in controlling glucose and lipid peroxidation and that $SP_6$ acupuncture may have anti-oxidative and anti-inflammatory effects on renal damage in STZ-induced diabetic mice.

고지방식이 비만모델에서 방기황기탕(防己黃芪湯) 합(合) 영계출감탕(苓桂朮甘湯)의 항비만 효과 (Anti-obesity Effects of Banggihwnggi-tang-hap-yeonggyechulgam-tang in High Fat Diet Induced Obese Mice Model)

  • Objectives This study is to investigate anti-obesity effects of Banggihwanggi-tang-hap-yeonggyechulgam-tang (BY), an herbal formula, in high fat diet induced obese mice model. Methods Fourty five male C57Bl/6J mice were randomly assigned to normal group fed with normal research diet (Nor, n=9), high fat diet control group treated with water (Veh, n=9), high fat diet group treated with orlistat (Oris; n=9, Orlistat 40 mg/kg), high fat diet group treated with low concentraion BY (BYL; n=6, BY 0.87 g/kg) and high fat diet group treated with high concentration BY (BYH; n=6, BY 1.74 g/kg). Results Seven weeks later, antioxidative capacity, body weight, epididymal fat pad and liver weight, reactive oxygen species (ROS), peroxynitrite ($ONOO^-$), alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol, triglyceride, high density lipoprotein (HDL), low density lipoprotein (LDL), superoxide dismutase (SOD), catalase, glutathione peroxidase (Gpx), heme oxygenase (HO)-1 and histology of liver were evaluated. In the BYH group, 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis (3 ethybenzothiazoline-6-sulfonic acid) radical scavenging activity were more than L-ascorbic acid. Body weight gain were significantly less than Veh group. Epididymal fat pad and liver weight gain were significantly less than Veh group. ROS and $ONOO^-$ were significantly less than with Veh group. ALT and AST were significantly less than with Veh group. Total cholesterol, triglyceride and LDL were significantly less, HDL were significantly more than Veh group. SOD, catalase, Gpx, HO-1 significantly increased compared with Veh group. Injury on liver was lesser than Veh group. Conclusions It can be suggested that BY has anti-obesity effects in high fat diet induced obese mice model.

MIA로 유발된 골관절염 동물모델에서 산약 추출물의 골관절염 개선 효과 (Protective Effect of Dioscoreae Rhizoma Extracts in MIA-induced Rat.)

  • Objectives : Osteoarthritis is characterized by degeneration of articular cartilage, which is characterized by chronic pain, stiffness and decrease range of motion. The present study was designed to compare the therapeutic effect of Dioscoreae Rhizoma water extract (DRW) and Dioscoreae Rhizoma 30% ethanol extract (DRE) on the monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods : Osteoarthritis was induced by injection of MIA ($50{\mu}{\ell}$ with $80mg/m{\ell}$) into the knee joint cavity of rats. After adaptation period for seven days, rats were divided by 5 groups (n=10/group): normal group, control group, positive control (indomethacin 5 mg/kg), DRW 200 mg/kg treated group, DRE 200 mg/kg treated group (n=10/group). The hind paw weight distribution was measured with the changes of reactive oxygen species (ROS), peroxynitrite ($ONOO^-$) in articulation tissue. Also, the cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), tumor necrosis factoralpha ($TNF{\alpha}$), interleukin-6 (IL-6), matrix metalloproteinase-1 (MMP-1), and tissue inhibitor of metalloproteinases-1 (TIMP-1) were investigated by western blot analysis. Results : The administration of DRW and DRE significantly decreased the hind paw weight distribution. The ROS and $ONOO^-$ levels of cartilaginous tissue were significantly decreased in DRW and DRE compared to control group. The results showed that DRE decreased inflammatory cytokines such as iNOS and $TNF{\alpha}$. Also DRE decreased MMP-1 and increased TIMP-1. Conclusions : Based on the above results, Dioscoreae Rhizoma extract seems to have the therapeutic effect on osteoarthritis via suppression of inflammation.
