Development of Multi-residue Analysis and Monitoring of Persistent Organic Pollutants (POPs) - Used Organochlorine Pesticides in Korea (국내에 사용되어 금지된 유기염소계 농약류의 다중분석법과 모니터링)

  • This study was conducted to investigate residual organochlorine pesticides in green house soil and oriental melon, green pepper, and lettuce. The majority of them were designated as persistent organic pollutants (POPs) by the international community at the Stockholm Convention on Persistent Organic Pollutant. Extraction and clean-up method were developed using the QuEChERS method for residual organochlorine pesticides (OCPs) in soil and oriental melon, green pepper and lettuce. Recovery of OCPs in greenhouse soil and oriental melon, green pepper, and lettuce ranged from 73.3-110.6%. Limit of detection (LOD) of OCPs in soil and 3 crops were 0.01-0.08 and $0.11-0.17{\mu}g/kg$. The residues of OCPs in oriental melon, green pepper and lettuce greenhouse soil were analyzed by the developed method, and dieldrin, ${\beta}-endosulfan$ and endosulfan sulfate were detected at 1.4-72.5, 0.1-78.7 and $0.0-214.1{\mu}g/kg$, respectively. The detection frequency of 3 compounds in soils were 52 (29.7%), 34 (19.4%) and 57 (32.6%) among 175 samples, respectively. However, these compounds were not detected in all crop samples. The residue level in 3 crops were lower than 1/58.8 of maximum residue level of them. These results showed that the OCPs residue in oriental melon, green pepper, and lettuce greenhouse soil were not as high as crop safety threatening.

  • The analytical method of extracting compounds from human blood to examine accumulated organochlorine pesticides (OCPs) has been widely used the traditional liquid-liquid extraction (LLE) method and solid-phase extraction (SPE) method, yet these methods have certain limitations in purification and usafe of a large amount of sample. In order to overcome the se problems reside in these, solid-phase microextraction (SPME), known as a highly efficient extration method with less samples and relatively simple, was employed to collect 18 different kinds of OCPs in blood as extraction method in this study. To optimize extraction method, we examine various experimental SPME-parameters such as adsorption (fiber type, adsorption time, adsorption temperature, salting out effect), and desorption (desorption time, desorption temperature etc.). From the experimental results, the optimal conditions are as follows: fiber was polyacrylate with $85{\mu}m$, adsorption time was for 5 min, adsorption optimum temperature was at $280^{\circ}C$, and salting out effect was NaCl with 0.1 g. MDL, precision and accuracy was in the ranges of 0.05~0.20 ng/mL, 5.59~13.39%, respedively, and accuracy was -0.5% ~24.5% for all OCPs.

Oral Contraceptives, Abortion and Breast Cancer Risk: a Case Control Study in Saudi Arabia

  • Background: Several studies have examined the relationship between oral contraceptive pill (OCP) use, abortions and breast cancer, with mixed results. Hormonal changes associated with OCP use and abortion may increase risk of breast cancer over time, but there is a lack of studies studying this association in Saudi Arabian women. Materials and Methods: We thererfore conducted a case control study in 192 women (92 as cases and 100 as controls), aged 30 to 65, and collected information on variables including examples related to study objectives and those which may confound findings. The Chi square test was used to detect associations between various factors and risk of breast cancer. Results: We found no evidence of interaction between history of abortion or frequency of abortion and breast cancer risk (Chi square=0.422, p =0.420 and 1, p =0.169) respectively. Oral contraceptives did not confer risk for breast cancer overall (OR=0.276, 95%CI 0.092-0.829, p=0.524), while long term use of OCP was associated with increased risk of breast cancer (OR=0.297, 95%CI 0.158-0.557, p=0.001), with higher association for those who used 10 years or more of OCPs (OR=0.282, 95%CI 0.095-0.835, p=0.02). Age at first use of OCPs had no effect on breast cancer risk (p=0.452) or age at diagnosis (p=0.074). Conclusions: Prolonged use of OC (more than 10 years) may be associated with increased risk of breast cancer in Saudi women. Larger population based studies are needed to confirm this finding in this population.

Suppressive Effect of Acanthopanax sessiliflorus Extract on the DNA and Cell Damage by Dieldrin (Dieldrin에 의한 DNA와 세포 손상에 대한 오가피 추출물의 억제효과)

  • Dieldrin, one of the organochlorine pesticides (OCPs), induced the damages in neuroblastoma cells and DNA damages in lymphocytes. The ethanol extracts of A. sessiliflorus leaves were examined for the suppressive effects on the dieldrin-induced cell damages. Moreover, the extract was used to test whether it might inhibit the oxidative DNA damage of lymphocytes using Comet assay. The cell and DNA damage by dieldrin were suppressed in vitro upon treating A. sessiliflorus extract. This result suggests that A. sessiliflorus extract might be useful to reduce dieldrin toxicity.

Analytical method of New POPs in environmental samples (환경 중 신규 POPs 분석방법 고찰)

  • The purpose of this study is to set up the analytical method of new persistent organic pollutants (POPs) such as chlordecone, endosulfan, ${\alpha}$-HCH, ${\beta}$-HCH, ${\gamma}$-HCH. The analytical methods for these compounds listed as new POPs by the Stockholm Convention need to be newly established. Therefore, we proposed the analytical method for 5 organic chlorinated pesticides (OCPs) and then applied the analytical method to environmental samples. To do this, the pre-treatment such as florisil and activated carbon cleanup process in the Korean official method for classic POPs had been reviewed. All of compounds except chlordecone were pre-treated simultaneously with reviewed cleanup process and detected by GC/MS and HRGC/HRMS respectively. There is a problem that chlordecone could not get a high sensitivity by GC analysis, but in this study GC/MS method was proposed.

Investigation of Residual Organochlorine Pesticides in Apple and Pear Orchard Soil and Fruit (사과 및 배 과수원의 토양 및 과실 중 잔류성유기염소계 농약류 잔류량 조사)

  • BACKGROUND: Residual organochlorine pesticides (OCPs) are designated as persistent organic pollutants (POPs) by Stockholm Convention because they bioaccumulate through the food web, and pose a risk of causing adverse effect to human health and the environment. Apple and pear is economic crop in agriculture, and its cultivation area and yield has been increased. Therefore, we tried to investigate the OCPs residue in apple and pear orchard soils and fruits. METHODS AND RESULTS: Extraction and clean-up method were developed using the modified QuEChERS method for residual organochlorine pesticides (ROCPs) in apple and pear orchard soil and fruits. Recovery and limit of quantitation (LOQ) of ROCPs in soil and fruits were 75.4-101.4 and 76.9-93.4%, 0.03-0.21 and $0.6-1.2{\mu}g/kg$, respectively. Detected ROCPs in apple and pear orchard soil was 2,4-DDT, 4,4-DDD, 4,4-DDE, 4,4-DDT, and endosulfan sulfate, the residues were 2.2, 1.9-48.0, 1.3-84.1, 90.6-863.1, and $11.3-239.0{\mu}g/kg$, respectively. But five pesticides in all fruit samples were not detected. CONCLUSION: These results showed that ROCPs residues in apple and pear orchard soil had no effect on safety of agricultural products.

Analytical Head-space Supercritical Fluid Extraction Methodology for the Determination of Organochlorine Compounds in Aqueous Matrix

  • The proposed head-space supercritical fluid extraction (SFE) methodology as an alternative to an existing conventional procedure was explored for the determination of organochlorine compounds in aqueous matrix. In this study, polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) were utilized as target analytes. To enhance the recovery efficiency, the factors such as the $CO _2$ density, the extraction time, and the extraction mode were investigated. Furthermore, the analytical procedures and the results obtained were compared with those provided by the conventional method (the U.S. EPA method 8080). Under the optimized conditions, i.e., a combination of static with dynamic SFE mode at 2,000 psi and 40 ${^{\circ}C}$, the head-space SFE methodology gave equivalent or better to the conventional method in recovery efficiencies with clear advantages such as simple sample treatment and fast analysis time as well as reduced solvent and reagent consumption.

Benthic Pollution Assessment Based on Macrobenthic Community Structure in Gamak Bay, Southern Coast of Korea

  • Benthic pollution assessment based on macrobenthic community structure with environmental variables was carried out at twelve stations during two periods on a presumed pollution gradient in Gamak Bay. Univariate and multivariate methods were applied to investigate structural changes in the benthic communities. A clear gradient of pollution effects on the macrobenthic community was observed from the interior to the exterior of the bay. The community on the northwestern basin was severely disturbed due to a low level of hydrodynamics and a large amount of pollutant input from nearby cities. Exterior regions on the southern basin appeared to have the best benthic environmental characteristics among all stations according to most methods of analysis. Central ridge regions and two stations around the islets in the mouth of the bay exhibited intermediate levels of perturbation when compared to the more disturbed interior and undisturbed exterior regions. Pollution effects on the communities were attenuated at the southern area of the central ridge during spring compared to those of summer, where aquacultural farming was densely distributed. The environmental variables primarily correlated to the macrobenthic community structure were total organic carbon (C), organochlorine pesticides (OCPs), and tributyltins (TBTs), contents found on the surface sediment, as anthropogenic variables indicating organic materials.

A.c. Impedance Measurement of CP-Ti in 0.1 M NaOH Solution

  • A.c. impedances of mechanically polished CP-Ti specimens were measured at open-circuit potential (OCP) with immersion time and under applied anodic potentials between -0.2 and 1 $V_{Ag/AgCl}$ in 0.1 M NaOH solution. Capacitances of native oxide films ($C_{ox,na}$) grown naturally and capacitances of anodic oxide films ($C_{ox,an}$) formed under applied anodic potentials were obtained to examine the growth of native and anodic oxide films in 0.1 M NaOH solution and how to use $C_{ox,na}$ for the surface area measurement of Ti specimen. $1/C_{ox,na}$ and $1/C_{ox,an}$ appeared to be linearly proportional to OCP and applied potential ($E_{app}$), with proportional constants of 0.086 and 0.051 $uF^{-1}\;V^{-1}$, respectively. The $C_{ox,na}$ also appeared to be linearly proportional to geometric surface area of the mechanically polished CP-Ti fixture specimen, with proportional constants of 11.3 and $8.5{\mu}F\;cm^{-2}$ at -0.45 $V_{Ag/AgCl}$ and -0.25 $V_{Ag/AgCl}$ of OCPs, respectively, in 0.1 M NaOH solution. This linear relationship between $C_{ox,na}$ and surface area is suggested to be applicable for the measurement of real surface area of Ti specimen.
