Effect of Kyejakjimo-tangkami (Guishaozhimu-tangjiawei) on Osteoarthritis (계작지모양가미방(桂芍知母湯加味方)이 골관절염에 미치는 영향)

  • Objectives The purpose of this study is to prove the effect of Kyejakjimotangkami(KMK) on osteoarthritis. Methods We checked antioxidant activity and measured production of $IL-1{\beta}$, IL-6, TNF-${\alpha}$ in RAW 264.7 cell after treat by KMK. Then we measured hind paw weight of Wister Rat with arthritis induced by MIA after KMK oral administration, checked Prostaglandin E2, IL-$1{\beta}$, IL-6, TNF-${\alpha}$, Osteocalcin, TIMP-1, MMP-9, LTB-4 in serum, ran histopathological test and ${\mu}CT$-arthrography. Results 1. DPPH radical Scavenging was increased depend on concentration of KMK ethanol extract in RAW 264.7 cell. 2. Production of NO was significantly decreased by KMK ethanol extract on concentration of $200{\mu}g/ml$ in RAW 264.7 cell. 3. Production of IL-$1{\beta}$ was significantly decreased by KMK ethanol extract on concentration of $200{\mu}g/ml$. And Production of IL-6, TNF-${\alpha}$ were significantly decreased KMK ethanol extract of every concentration in RAW 264.7 cell. 4. Result of checking hind paw weight when administered KMK ethanol extract to Wister Rat with arthritis induced by MIA was significantly higher than control group and similar to normal group. 5. Production of Prostaglandin E2, IL-$1{\beta}$, Osteocalcin, TIMP-1, MMP-9 and LTB-4 in serum was significantly decreased by KMK ethanol extract after administerd to Wister Rat with arthritis induced by MIA. 6. In Hematoxylin & Eosin staining and Safranin-O staining, we could find inflammation of synovial cell, infiltration of macrophage and granulocyte and degeneration of cartilage and bone were decreased in comparison with control group. 7. When checked cartilage volume to examine degree of cartilage degeneration using ${\mu}CT$-arthrography, volume of cartilage was increased in comparison with control group. Conclusions Comparison of the results for this study showed that KMK ethanol extract have anti-inflammatory effectiveness and can protect cartilage and bone. So we expect that KMK can be used as a effective drugs for osteoarthritis.

Antioxidative and Antigenotoxic Activity of Extracts from Rhododendron mucromulatum Turcz. Flowers (진달래 꽃 추출물의 항산화 및 항유전독성 활성)

  • The present study describes the preliminary evaluation of antioxidant activities and antigenotoxic effect of Rhododendron mucromulatum Turcz. flowers (RMF). The samples were prepared by extracting RMF with four different solvents (methanol, ethanol, acetone, and water), and antioxidant properties were evaluated by determining total phenolic contents (TPC), DPPH radical scavenging activity (RSA), and reducing power (RP). Water extract showed the highest total phenol content (328.1 mg/g gallic acid equivalents). Acetone extract showed the most potent RSA and RP. The $IC_{50}$ for RSA and RP in the acetone extract were 78 mg/mL and 454 mg/mL, respectively. The 200 mM $H_2O_2$ induced DNA damage, measured by Comet assay, was inhibited with water, methanol and acetone extract in dose dependent manner in human leukocytes. The inhibition rates were 42, 62, and 52% at the concentrations of 50 mg/mL of water, methanol and acetone extracts, respectively. These results suggest that R. mucromulatum Turcz. has significant antioxidative activity and protective effect against oxidative DNA damage.

Effects of Pumpkin (Cucurbita moschata Duch.) Leaf Powder on Quality Characteristics, Antioxidant Activities, and Retarding Retrogradation by Shelf-life of Sponge Cake (호박잎 분말을 첨가한 스폰지 케이크의 품질 특성, 항산화 활성 및 저장 기간에 따른 노화 억제 효과)

  • This study investigated the quality characteristics, antioxidant activities, and retarding retrogradation of sponge cakes made with 0%, 1%, 3%, 5% and 7% pumpkin (Cucurbita moschata Duch.) leaf powder. Specific capacity significantly increased with the addition (0.33~0.38), but baking loss and dough yield were not significantly different (p<0.05). In color, there was a decrease in the L-value, a-value and b-value, but ${\Delta}E$ increased in proportion to the amount of pumpkin leaf powder. Moisture content increased in the 0~3% additions, from 27.90~31.68%, but decreased in 5% and 7% (22.37% and 28.15%, respectively). pH tended to decrease significantly according to the amount of pumpkin leaf powder (p<0.05). Hardness increased with the addition of pumpkin leaf powder, and pumpkin leaf groups presented higher springiness and cohesiveness than the control. Chewiness was not significantly differ (p<0.05). In retarding retrogradation, Avrami exponent (n) showed that addition of 5% (0.1329) had more retarding retrogradation effect than the control (0.4319), whereas time constant (1/k) showed both 3% (100.00) and 5% (70.42) addition had more effect than control (18.45). Total phenols and flavonoids content increased proportionate to the addition levels. ABTS radical scavenging activity tended to increase according to the level of pumpkin leaf powder. In sensory properties, 5% addition showed maximum color, and 3% addition had the highest scores in flavor, moistness, sweetness, chewiness and overall acceptability. These results suggested that pumpkin leaf was a good addition to improve the quality characteristics, health and sensory preferences. The most appropriate proportion to have the effect of retarding retrogradation in sponge cake is 3%.

Brewing and Properties of Low-Malt Beer with a Sweet Potato Paste (고구마를 첨가한 저맥아 맥주의 양조와 품질 특성)

  • Sweet potato-malt worts were prepared by using sweet potato paste of Shinyulmi and Shinjami as the main adjunct, enzymes, malt, hop, and water. We brewed low-malt beers of the lager- or ale-type by using these worts and inoculating bottom and top fermenting yeast, respectively. Moreover, the componential and functional characteristics of the resulting beers were evaluated. During saccharification of sweet potato, the addition of an enzyme agent containing ${\alpha}-amylase$ caused an improvement in filterability and an increase of total sugar. The sugar content of sweet potato-malt wort which was prepared by the addition of 0.1% enzyme agent containing ${\alpha}-amylase$ and a three-step infusion procedure was $13.5^{\circ}Brix$ adequate for beer brewing. The polyphenol and anthocyanin contents of Shinjami beer were increased with increasing content of the paste, and also increased DPPH and ABTS radical scavenging activities. But in Shinyulmi beer it were decreased. A strong correlation was obserbed between antioxidave activities and polyphenol and anthocyanin contents of sweet potato beers. In all lager- and ale-type low-malt beers using 41.6% of Shinjami pastes, sensory attributes very similar to those of 100% malt beer were obtained and they were very good as they had unique red color.

Interface Traps Analysis as Bonding of The Silicon/Nitrogen/Hydrogen in MONOS Capacitors (실리콘/수소/질소의 결합에 따른 MONOS 커패시터의 계면 특성 연구)

  • The effect of hydrogen-nitrogen annealing on the interface trap properties of Metal-Oxide-Nitride-Oxide-Silicon (MONOS) capacitors is investigated by analyzing the capacitors' gate leakage current and the interface trap density between the Si and $SiO_2$ layer. MONOS samples annealed at $850^{\circ}C$ for 30 s by rapid thermal annealing (RTA) are treated by additional annealing in a furnace, using annealing eases $N_2$ and 2% hydrogen and 98% nitrogen gas mixture $(N_2-H_2)$ at $450^{\circ}C$ for 30 mins. Among the three samples as-deposited, annealed in $N_2$ and $N_2-H_2$, MONOS sample annealed in an $N_2-H_2$ environment is found to have the lowest increase of interface-trap density from the capacitance-voltage experiments. The leakage current of sample annealed in $N_2-H_2$ is also lower than that of sample annealed in $N_2$.

The effect of Cultivated Environments on the Antioxidant Enzyme Activities of Codonopsis lanceolata (생육환경이 더덕(Codonopsis lanceolata)의 향산화효소 활성에 미치는 영향)

  • The activities of the antioxidative enzymes in the roots of Codonopsis lanceolata have been compared depending on the cultivated environments - wildness, cultivate paddy fields and cultivate dry fields - and the parts of the root. In the Codonopsis lanceolata raised in cultivate paddy fields, the activity of SOD was higher in 2 yrs old than 1 yr old, but the activity in 1 yr old was higher than in 2 yrs old for the plants raised in the cultivate dry fields. The specific activity of SOD in wildness plants 86.069unit/mg protein was the highest among plants studied. The tissue distribution of the SOD activity showed differences depending on the enviroment. The highest activity of SOD was shown in the upper part of the root for the cultivate paddy fields, the loewr parts for the cultivate dry fields and middle parts for the wildness. The specific activity of POD was increased with ages of the plants, and that in the wildness was the highest 68 unit/mg protein among the plants studied. The activity of POD in the parts of the roots was shown as middle>lower>upper. The activity of POD in the middle part of the root, rasied in Soebick province was 85 unit/mg protein. The specific activity of CAT was decreased with ages of the plants. The activities of wildness and cultivate paddy fields was similar, but that in cultivate dry fields was lower than others. The tissue distribution in the parts of the roots was upper>lower>middle. The activity of CAT middle part of rasied in the Sebuck area was 5.359 unit/mg protein. The activities antioxidative in the cells cultured in MSID(Murashige and Skoog +2.4-D 1mg/$\iota$) was followings: 1564 for CAT. 30 for POD and 22200 unit/mg protein for SOD. These figures were lower than that in in vivo.

Black ginseng extract ameliorates hypercholesterolemia in rats

  • Background: Ginseng (Panax ginseng Meyer) is a well-characterized medicinal herb listed in the classic oriental herbal dictionary as "Shin-nong-bon-cho-kyung." Ginseng has diverse pharmacologic and therapeutic properties. Black ginseng (BG, Ginseng Radix nigra) is produced by repeatedly steaming fresh ginseng nine times. Studies of BG have shown that prolonged heat treatment enhances the antioxidant activity with increased radical scavenging activity. Several recent studies have showed the effects of BG on increased lipid profiles in mice. In this study report the effects of water and ethanol extracts of BG on hypercholesterolemia in rats. To our knowledge, this is the first time such an effect has been reported. Methods: Experiments were conducted on male Sprague Dawley rats fed with a high-cholesterol diet supplemented with the water and ethanol extracts of BG (200 mg/kg). Their blood cholesterol levels, serum white blood cell levels, and cholesterol-metabolizing marker genes messenger RNA (mRNA) expression were determined. Liver and adipose tissues were histologically analyzed. Results: We found that BG extracts efficiently reduced the total serum cholesterol levels, low-density lipoprotein (LDL) levels with increased food efficiency ratio and increased number of neutrophil cells. It also attenuated the key genes responsible for lipogenesis, that is, acetyl-coenzyme A (CoA) acetyltransferase 2, 3-hydroxy-3-methyl-glutaryl-CoA reductase, and sterol regulatory element-binding protein 2, at the mRNA level inside liver cells. Furthermore, the BG extract also reduced the accumulation of fat in adipose tissues, and inhibited the neutral fat content in liver cells stained with hematoxylin and eosin and oil red O. Conclusion: Administration of BG extracts to Sprague Dawley rats fed with high-cholesterol diet ameliorated hypercholesterolemia, which was mediated via modulation of cholesterol-metabolizing marker genes. This data throw a light on BG's cardioprotective effects.

Removal Characteristics of Dichloroacetic Acid at Different Catalyst Media with Advanced Oxidation Process Using Ozone/Catalyst (담지체를 달리한 오존/촉매 AOP공정에서 디클로로아세트산의 제거 특성)

  • Pd/activated carbon (Pd/AC) and Pd/alumina (Pd/AO) catalysts were prepared by the impregnation of palladium into activated carbon and alumina. The catalytic characteristics according to the kinds of support materials were compared. The decomposition efficiencies of ozone according to kinds of support materials are about the same when these were compared by adding 10 g of catalysts into the water saturated with ozone. The decomposition efficiencies and the oxidation characteristics (TOC, $COD_{Cr}$) of dichloroacetic acid were compared with the ozone only process and the catalytic ozonations using Pd/activated carbon and Pd/alumina catalysts. The decomposition efficiencies of dichloroacetic acid by catalytic ozonations were better than the one by ozone only process, but there was slight difference of the one between Pd/activated carbon and Pd/alumina catalyst. The decomposition efficiency of dichloroacetic acid was increased with increasing ozone dose at a constant concentration of dichloroacetic acid, but the one was little increased with increasing ozone dose at more than 1.0 L/min of ozone dose. It was seemed that the bicarbonate and the chloric ion formed throughout the decomposition of dichloroacetic acid acted as the scavenger of hydroxyl radical.

Carcinoma of the Tonsillar Region -Results of External Irradiation- (편도선 암의 방사선 치료)

  • Thirty-six patients with carcinoma of the tonsillar region treated with radical radiotherapy at Yonsei Cancer Center between Jan. 1971 and Dec. 1980 were retrospectively reviewed. The purpose of this study is to evaluate the clinical characteristics and the treatment results. Seventy-five per cent of patients were in advanced stage when they were first seen. Incidence of nodal involvement at the time of initial presentation was $64\%$. Bilateral nodal involvement was found in $19\%$ of cases. Three-year local control rates for T1, T2, T3, and T4 were $88\%,\;46\%,\;25\%,\;and\;11\%$, respectively. Advanced neck nodes were poorly controlled $(N1\;70\%,\;N2\;20\%,\;N3\;12\%)$. The prognosis is mainly affected by the status of the primary and neck nodes Overall three-year local control rate was $42\%$.

Study of Apoptotic Effect on Hydrogen Peroxide-induced Rat PC-12 Cells by Aster tataricus Water Extract (자완 수추출물(水抽出物)이 $H_2O_2$에 의해 유도(誘導)된 PC-12 세포주(細胞株)의 세포사(細胞死)에 미치는 영향(影響))

  • Objective : Alzheimer's disease (AD) is a geriatric dementia that is widespread in old age. With an aging populace, AD is a looming problem in public health service. Alzheimer's disease is characterized by specific neuronal degeneration in certain areas of the brain. Mutations and abnormal expression of several genes are associated with ${\beta}-amyloid$ deposits and Alzheimer's disease; among them APP, PS1, and PS2, SOD, free radical, ROS. Methods:We studied herbal medicines that have a relationship to brain degeneration. From pre-modern times, although a variety of oriental prescriptions of Aster tataricus have been traditionally utilized for the treatment of AD, their pharmacological effects and action mechanisms have not yet been fully elucidated. Result : Based on morphological observations by phase-contrast microscope, TUNEL assay and MTT in the culture media, $H_20_2-induced$ cell death was significantly inhibited by Aticus. We examined by ROS formation, catalase activity and GSH activity. We studied the protective effect and inhibitory effects of neurotoxicity in $H_20_2-induced$ PC-12 cells by Aticus. Findings from our experiments show that Aticus inhibits apoptosis, which has neurotoxicities and cell damage in PC-12 cells. In addition, treatment with Aticus ($>25{\mu}g/ml$ for 6hrs) partially prevented $H_20_2-induced$ cytotoxicity in PC-12 cells, and induced a protective effect. Conclusion : As the result of this study, in the Aticus group, the apoptosis in the nervous system was inhibited, protected against the degeneration of PC-12 cells by $H_20_2$. Taken together, Aticus exhibited inhibition of $H_20_2-induced$ apoptotic cell death. Aticus was found to induce protective effect on GSH and catalase in PC-12 cells. Based on these findings, Aticus may be beneficial for the treatment of AD.
