This study was conducted to investigate the physiological characteristics of water and ethanol extracts from leaves and stems of Achyranthes japonica. The highest contents of total polyphenol and flavonoid compounds were 58.27 and 42.22 mg/g in water extract from leaves, respectively. The protein content was the highest at 16.42 mg/g in water extract from leaves. Ethanol extract from stems showed the highest content of reducing sugars at 11.35 mg/g. In the measurement of electron donating ability (EDA), ethanol extract from stems showed the highest EDA at 93.41% at a concentration of 1.0 mg/mL. Superoxide dismutase-like activity of ethanol extract from leaves was the highest at 8.13% at a concentration of 1.0 mg/mL. In the analysis of nitrate scavenging activity, water extract from leaves showed the highest activity at 94.90% at pH 1.2, and the activity increased as concentration increased and pH decreased. In the measurement of xanthine oxidase inhibition, ethanol extract from stems showed the highest inhibitory activity at 66.67% at a concentration of 1.0 mg/mL. Especially, nitrate scavenging activities of water extract from leaves were the highest under all pH conditions. These results verify that extracts from leaves of A. japonica have strong antioxidant activity and can be used as an effective antioxidant source for nutraceutical foods, medicines, and cosmetic stuffs.

In order to use nutraceutical components in Yulmoo (Coix lachryma-jobi L. var. ma-yuen Stapf), the acceptability of Yulmoo for Takju (Korean rice wine) was examined in terms of the production of volatile components. When Yulmoo was added to the Takju made by commercial koji as a substitute of starch at 0 (rice 100%), 15, 30, 60, and 100%, the ethanol content slightly increased to 13.6, 15.2, 15.2, 14.1, and 13.8%, the Production of isobutanol and iso-imyl alcohol increased as the amount of Yulmoo increased. While the use of Yulmoo to the Takju as the nuruk (Korean traditional koji), inoculated with Aspergillus kawachii (AK) and Rhizopu japonicus (RJ), decreased the contents of n-propanol, iso-butanol, and iso-amyl alcohol as well as the contents of ethanol from 14.3% to 11.2 [AK], 7.5 [RJ], and 10.4% [AK + RJ]. The Yulmoo nuruk in Takju increased the production of acetaldehyde and ethyl acetate indicating the negative effect in the organoleptic evaluation. It was determined that the addition of Yulmoo as a starch source or as nuruk has a critical effect on the production of fusel oils and other volatile compounds in Takju fermentation.

Tyrosinase catalyzes melanin synthesis in skin melanocytes. The effects of Paeonia sufficinalis water extract (MDP) on antioxidant and tyrosinase activities have been studied in a cell-free system and mouse melanoma B16 cell. Radical scavenging activity of MDP was tested by DPPH assay and it showed high DPPH radical scavenging activity. The cellular tyrosinase activity was measured in mouse melanoma B16 cell by RT-PCR and enzyme activity. Treatment with MDP for 24 hr resulted in decreased tyrosinase mRNA level. Tyrosinase activity was decreased, compared with control, in cells exposed to MDP. Thus, Paeonia sufficinalis water extracts may be a candidate for cosmetic use.

This study was carried out to estimate beneficial effects of medicinal plant [Oenanthe javanica (MNR), Coicis lachryma-jobi L. var. (YM), Plantaginis asiatica L. (CJJ)] water extracts on activities of key enzymes such as lipoprotein lipase (LPL), acyl-CoA synthetase (ACS) and carnitine acetyltransferase (CAT) on lipid metabolism. LPL and ACS were extracted from the epididymal adipose tissue and liver of Zucker lean rats (lean) and Zucker fatty rats (fa/fa). MNR or YM water extract treatment significantly reduced the activity of lean and fa/fa LPL. When 10000 ppm of MNR, YM, and CJJ water extracts were tested, they decreased fa/fa LPL activity by 32.5%, 30.1% and 22.8%, respectively. The lean ACS activity was significantly higher in YM water extract treatment compared to the control and the MNR water extract treatment significantly increased the activity of fa/fa ACS, compared to the activity in the control (p<0.05). MNR water extract activated fa/fa ACS activity by 12-fold compared with control at 10000 ppm concentration. CAT activity was significantly higher in 10000 ppm and 20000 ppm CJJ water extract treatment than in the control. Thus, the MNR, YM and CJJ water extracts may have beneficial effects due to activities of enzymes related with fat metabolism in obese humans.

We investigated the effects of feeding diet containing medicinal plant water extracts (MPWEs) on body weight, epididymal adipose tissue weight, adipocyte size of epididymal adipose tissue and plasma lipid levels in high fat (HF) diet-induced obese mice. To test antiobese effects of diet containing the MPWEs, C57BL/6J mice were fed with HF diet for 11 weeks. In the last 6 weeks, the HF diet was supplemented with 0 (HFD) or MPWEs (5 g/kg, HFD+MPWEs) or orlistat [0.5 g/kg, HFD+orlistat (antiobesity drug)]. The HF-free diet group was fed normal chow for 11 weeks. Eleven-weeks feeding with HFD resulted in significant increase in lipid levels, body weight, liver and epididymal adipose tissue weights, compared with the HF-free group. Diet containing MPWEs significantly reduced plasma total cholesterol, LDL-cholesterol, triglyceride and glucose concentrations as well as body weight, liver weight and epididymal adipose tissue weight. Plasma triglyceride levels were significantly lower in the HFD+Forlistat group after 6 weeks and a similar effect was found with HFD+MPWEs group. The adipocyte size of epididymal adipose tissue in HFD group was significantly larger than those of HF-free group. MPWEs and orlistat (positive control) significantly decreased the size of epididymal adipocytes but orlistat was slightly more effective than MPWEs. These results suggest that oral feeding of the MPWEs may have antiobesity effects by suppressing body weight gain, adipose tissue formation and adipocyte size increase.
This study was performed to observe the effect of hot-water extracts from garlic and 13 kinds of medicinal plants composites (GMP) on hyperlipidemia and hepatoprotective activity in rats administered with alcohol. Male Sprague-Dawly rats were fed an AIN-93 diet (Normal), a normal diet plus ethanol (control, 10 ml of 40% ethanoljkgjday), a control diet plus 0.5% garlic and 1.0% medicinal plants composites extracts (GMP-I), and a control diet plus 1.0% garlic and medicinal plants composites extracts (GMP-II) for 7 days. Blood glucose was higher than the control, but it was markedly decreased in the GMP-II group. Elevation total lipids, cholesterol, triglyceride and phospholipids in serum were markedly decreased in rats fed with GMP-I. GMP-II also inhibited the increase of lipid content in serum. Activities of GOT, GPT, $\gamma$-GTP and ALP in serum elevated by alcohol were significantly inhibited in the GMP group. TBARS content of serum was significantly decreased in GMP groups administered with garlic and medicinal plant extracts. Extracts of garlic and medicinal plants play an important role in recovering liver function in rats from alcohol induced damage.

In this study, anti-inflammatory activity of hot water extract of Aronia fruits (AF-H) was examined. Pre-treatment with AF-H significantly inhibited production of nitric oxide (NO) and prostaglandin E-2 in a dose-dependent manner in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The inhibitory effect of AF-H on LPS-induced inflammation was also confirmed by down-regulation of inducible NO synthase as well as cyclooxygenase-2 protein expression. Furthermore, treatment with AF-H significantly inhibited secretion of inflammatory cytokines such as tumor-necrosis $factor-{\alpha}$ and interleukin-6. Signal transduction pathway studies further indicated that AF-H inhibited LPS-induced activation of nuclear $factor-{\kappa}B$, but not mitogen-activated protein kinase. Treatment with AF-H also partially protected against LPS-induced lethal shock in C57BL/6 mice, although its effect was not statistically significant. These results suggest that AF-H is a more promising nutraceutical or medicinal agent for inhibition of LPS-induced inflammation or inflammation-related diseases.
This study evaluated the change in anti-allergy activity of ${\beta}-glucan$ by electron beam irradiation. ${\beta}-Glucan$ was irradiated at dose of 50 kGy and then orally pre-treated with electron beam irradiated and non irradiated ${\beta}-Glucan$ for 7 days. After pre-treatment, allergy was induced by injection of ovalbumin (OVA). Serum total immunoglobulin E (IgE) and OVA-specific IgE levels in the allergic mice was significantly increased but the mice pre-treated 50 kGy electron beam irradiated ${\beta}-glucan$ was significantly decreased the levels of total IgE and OVA-specific IgE, respectively. Moreover, cytokine production (interleukin-4) was also decreased in the 50 kGy electron beam irradiated ${\beta}-Glucan$ pre-treated mice. These results indicate that pre-treatment of 50 kGy electron beam irradiated ${\beta}-glucan$ may elevate the anti-allergy activity. Therefore, electron beam-irradiated ${\beta}-glucan$ could be used for nutraceutical foods in food industry.

This study was carried out to investigate the mutagenic, antimutagenic, cytotoxicity and antitumor effect of Codonopsis lanceolata (CL). CL was extracted with 70% ethanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water. Antimutagenic, cytotoxicity and antitumor effects of CL extracts were measured by using Ames test, SRB method, and the tumor growth inhibition test. CL extracts did not show any mutagenicity in the Ames test; however, 70% ethanol extracts and its fractions had strong antimutagenic effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and 4-nitroquinoline-1-oxide (4NQO). The ethyl acetate fraction of CL (200 ${\mu}g$/plate) showed approximately 72.1% inhibitory effect on the mutagenesis induced by 4NQO against TA98 strain, whereas 69.6% and 67.0% inhibitions were observed on the mutagenesis induced by MNNG and 4NQO against TA100 strain. In anticancer effects, the cytotoxicity of CL extract and its fractions against cancer cell lines including human cervical adenocarcinoma (HeLa), human hepatocellular carcinoma (HepG2), human breast adenocarcinoma (MCF-7), human lung carcinoma (A549) and transformed primary human embryo kidney (293) were investigated. The treatment of 1 mg/mL CL ethyl acetate fraction had the highest cytotoxicity of 74.5%, 70.7% and 80.3% against HeLa, MCF-7 and A549 cells, respectively. In contrast, the extract and its fractions showed only 2$\sim$31% cytotoxicity for a normal human kidney cell line (293). In vivo anticancer effect of CL extract was tested using Balb/c mice transplanted sarcoma-180 cells. CL ethyl acetate fraction showed the highest inhibition rate of 56.4% at the 50 mg/kg concentration.

The root of Adenophora triphylla is widely used as traditional herbal medicine in Korea. We studied its effects on sodium nitroprusside (SNP) cytotoxicity and antioxidant genes expression in HepG2 cells. To study whether Adenophora triphylla ethylacetate extract (ATea) inhibited NO-induced cell death, HepG2 cells were preincubated for 24 hr with 50 and 100 $\mu$g/mL ATea followed by 24-hr exposure to 0.5 mM SNP (exogenous NO donor). No-induced cytotoxicity was inhibited by pretreatment of ATea, as assessed by mitochondrial dehydrogenase activity (MTT assay). We further investigated the effects of ATea on mRNA levels of various enzymes of the antioxidant system such as Cu, Zn superoxide dismutase (SOD 1), Mn SOD (SOD 2), glutathione peroxidase (GPx), catalase and several enzymes of the glutathione metabolism [glutathione reductase (GR), $\gamma$-glutamyl-cystein synthetase (GCS), glutathione-S-transferase (GST), $\gamma$-glutamyltranspeptidase ($\gamma$-GT), glucose-6-phosphate dehydrogenase (G6PD)] by RT-PCR. CAT, GCS, GR and G6PD mRNA levels were increased after treatment with ATea. The SOD 1, SOD 2, GPx, GST and $\gamma$-GT mRNA levels were not affected in ATea-treated HepG2 cells. We concluded that ATea have an indirect antioxidant effects, perhaps via induction of CAT, GCS, GR and G6PD.
