제조방법을 달리한 경옥고의 고콜레스테롤혈증 흰쥐에 미치는 효능 비교 연구 (Lipid Improvement Effect of New manufacturing method produced Kyeongok-go on hyperlipidemia rats)

  • Objectives : Kyeongok-go has the effect of antioxidant, anti-cancer, improving fatigue, and hyperlipidemia, but it is not easy to use in clinical practice becausee of the complicated manufacturing process of heat control and moisture control. The aim of this study was to explore the lipid improvement effect of new manufacturing methods prodeced Kyeongok-go in hyperlipidemia rats. Methods : The SD rats were divided randomly into 8 groups of six animals each : Normal rats (Nor), hyperlipidemia rats (Con), hyperlipidemia rats treated with Spirulina 60 mg/kg(SP60), hyperlipidemia rats treated with Commercial Kyeongok-go 1 g/kg (K0), hyperlipidemia rats treated with Kyeongok-go manufactured by heating mantle 1 g/kg (K1), hyperlipidemia rats treated with ginseng fermentation Kyeongok-go 1 g/kg (KF), hyperlipidemia rats treated with Kyeongok-go with black ginseng 1 g/kg (BK), hyperlipidemia rats treated with black ginseng fermentation Kyeongok-go 1 g/kg (BKF). The experimental group induced hyperlipidemia by eating high cholesterol diet for 8 weeks. Also, we compared Kyeongok-go manufacturing method using the new method compared with traditional method. Results : As a result, the improvement effect of hyperlipemia using the new method showed the similar or better improvement effect compared with the conventional method, and it is considered that the Kyeongok-go manufacturing method using the new method can be substituted for the traditional Kyeongok-go manufacturing method. Conclusion : In conclusion, these results suggest that new manufacturing methods prodeced Kyeongok-go has the potential as a anti-hyperlipidemia material which possess lipid improvement activities.

Dietary corn resistant starch regulates intestinal morphology and barrier functions by activating the Notch signaling pathway of broilers

  • Objective: This study was conducted to investigate the effects of dietary corn resistant starch (RS) on the intestinal morphology and barrier functions of broilers. Methods: A total of 320 one-day-old broilers were randomly allocated to 5 dietary treatments: one normal corn-soybean (NC) diet, one corn-soybean-based diet supplementation with 20% corn starch (CS), and 3 corn-soybean-based diets supplementation with 4%, 8%, and 12% corn resistant starch (RS) (identified as 4% RS, 8% RS, and 12% RS, respectively). Each group had eight replicates with eight broilers per replicate. After 21 days feeding, one bird with a body weight (BW) close to the average BW of their replicate was selected and slaughtered. The samples of duodenum, jejunum, ileum, caecum digesta, and blood were collected. Results: Birds fed 4% RS, 8% RS and 12% RS diets showed lower feed intake, BW gain, jejunal villus height (VH), duodenal crypt depth (CD), jejunal VH/CD ratio, duodenal goblet cell density as well as mucin1 mRNA expressions compared to the NC group, but showed higher concentrations of cecal acetic acid and butyric acid, percentage of jejunal proliferating cell nuclear antigen-positive cells and delta like canonical Notch ligand 4 (Dll4), and hes family bHLH transcription factor 1 mRNA expressions. However, there were no differences on the plasma diamine oxidase activity and D-lactic acid concentration among all groups. Conclusion: These findings suggested that RS could suppress intestinal morphology and barrier functions by activating Notch pathway and inhibiting the development of goblet cells, resulting in decreased mucins and tight junction mRNA expression.

수도권 지역 직장인의 음식 관련 스마트폰 어플리케이션 이용 실태와 만족도 (Usage and Satisfaction of Food-related Smartphone Applications of Office Workers in Seoul Area)

  • This study was conducted to figure the usage status and satisfaction of food-related smartphone applications by generic characteristics and food purchase attributes of Seoul area workers. The results show that health vitality-oriented group, married compared to single, those with higher eating-out expenses possess more food-related applications (p<0.05). The primary reason for the usage of food-related applications was the need for food-related information (53.7%). The highest application subjects in use were restaurant-related information and recipe information. The real-life aid food-related applications utilized most were also restaurant information (60.7%) and recipe information (28.3%). Health vitality-oriented respondents especially turned out to use nutrition information and food functionality information often (p<0.01), and recipes or calories & diet information usage frequency was higher in women than in men (p<0.001). Restaurant-related information were more frequently used by singles, highly educated, and those with high income and eating-out expenses (p<0.05). Satisfaction of food-related applications was normal (3.06), showing that the satisfactory level is not yet high. Satisfaction regarding purchase attributes showed that the health vitality-oriented group (3.19) was more satisfied compared to other groups (p<0.05), and women (3.16) were more satisfied that men (2.89) were (p<0.05). Inconveniences of food-related applications were highest in usage fee (3.29), simplicity of information (3.28), lack of reliability of information and need for update (3.10). The results of this study implies that various subdivisions of food-related applications users should be implemented; at the same time, food-related applications covering diverse subjects that regard each group's characteristics should be developed in order to utilize food-related knowledge and information as a marketing tool in the food industry; this can efficiently be done by paying attention to the quality of information and updates within applications.

Effect of Genotype and Dietary Protein Level on Growth Performance and Carcass Characteristics of Fattening Pigs in Central Vietnam

  • This study aimed to determine the optimum dietary crude protein level in a typical diet for fattening pigs fed ad libitum under normal climate conditions in Central Vietnam. One hundred and ninety two gilts of Mong Cai local breed (MC), $F_1$ Large White${\times}$Mong Cai and $F_2$ crossbreds of (Landrace${\times}$Mong Cai)${\times}$Large White were used. At the start of the experiment, Mong Cai pigs weighed 12 kg at 11 weeks of age, $F_1$ pigs 12.1 kg at 8 weeks of age and $F_2$ pigs 12.2 kg at 8 weeks of age. Four diets differing in crude protein (CP) content (10.1, 13.1, 16.1 and 18.9% in DM) were formulated from rice bran, corn meal, cassava meal and fish meal. Calculated digestible energy content of the diets ranged from 13.5 to 13.8 MJ per kg DM. Pigs were housed individually in pens of $2.5\;m^2$ each and had ad libitum access to feed in a trough as well as water in bowls. The final weights after a growing period of 150 days were 66, 86 and 96 kg for MC, $F_1$ and $F_2$, respectively. Feed intake of MC pigs was highest at 13.1% CP while $F_1$ and $F_2$ had the highest feed intake at 16.1% CP. The results showed that for MC the maximum gain was obtained at levels between 13 to 16% CP. For the $F_1$ the maximum gain was at dietary protein levels of 16-17%. For $F_2$ the max gain was obtained at CP levels of 16 to 18%. Feed conversion was highest in MC pigs (~4.0) followed by $F_1$ (~3.3) and $F_2$ (~3.1), and within genotypes was lowest at the optimum CP level (p<0.05). Back fat thickness in MC (33.1 mm), $F_1$ (23.0 mm) and $F_2$ (20.5 mm) pigs was different and within genotypes was the lowest at intermediate CP levels. In conclusion, increasing the dietary crude protein contents in practical diets for pigs in Vietnam can increase production on small holder farms. Optimal performance for MC, $F_1$ and $F_2$ pigs is achieved at different dietary crude protein contents.

Effects of Supplementing with Single or Multiple Trace Minerals on Growth Performance, Fecal Mineral Excretion and Nutrient Utilization in Pullets from 1 to 18 Weeks of Age

  • This study investigated whether supplemental Cu, Fe, Zn, and Mn are needed in a practical diet for pullets. Four hundred and twenty females of an egg-laying strain (1-d-old, Lohmann Brown Layer) were randomly distributed into 4 groups, consisting of 7 replicates of 15 birds each. During the 18-week experimental period, chicks were given three basal diets in sequence, each with single or multiple Mn, Zn and Cu supplementation to improve the mineral balance gradually. In the Control, no Mn, Zn, and Cu were added; in the single Mn supplemented group (sMn) Mn was added to 120, 60, and 60 mg/kg for 1-6, 7-12, and 13-18 weeks of age, respectively; in the multiple Mn and Zn supplemented group (mMnZn), Mn was added to 180, 90, and 90 mg/kg and Zn was added to 120, 105, and 105 mg/kg for 1-6, 7-12, and 13-18 weeks of age, respectively; in the multiple Mn, Zn, Cu supplemented group (mMnZnCu), Mn, Zn, and Cu were added to the same multiple of basal Fe concentration relative to NRC (1994) recommendations. Energy and protein metabolizability were determined by subtracting energy/protein intake by energy/protein excretion (from both feces and urine) and dividing by energy/protein intake. There were no statistically significant differences between groups in terms of feed intake, final body weight or tibia length throughout the experiment. Optimal growth performance was observed in the Control, while adding trace minerals to basal diets tended to result in decreased productive performance. Protein metabolizability was increased by mMnZn and mMnZnCu treatments, but energy metabolizability was not affected. Concentrations of Mn, Zn, Cu in excreta varied greatly related to dietary content, and the retentions of Cu, Fe, Zn and Mn were all increased due to the improvement of mineral balance. Based on these results, it is suggested that the concentrations of Cu, Fe, Zn and Mn in typical basal diets used in this study were adequate for normal growth for pullets from 1 to 18 weeks of age.

절식이 마우스 장내미생물에 미치는 영향 (Comparison of mice gut microbiota before and after fasting for a day)

  • 본 연구에서는 절식이 일반사료 및 고지방사료를 섭취한 마우스의 장내미생물생태에 미치는 영향에 관하여 조사하였다. 일반 사료 및 고지방사료를 마우스에 16주 동안 무한급이 형태로 섭취하게 하여 실험종료 직전에 1일(24시간) 간 절식시켰으며, 절식 전 후의 분변 샘플을 채집하고 MiSeq을 이용하여 장내미생물생태 분석을 진행하였다. 본 연구의 결과는 고지방사료를 섭취한 마우스에서는 절식 전후 장내미생물생태 내의 종 풍부성과 균등성이 감소한 반면, 일반사료를 섭취한 마우스에서는 차이를 나타내지 않았다. 또한, 고지방사료를 섭취한 마우스의 장내미생물생태에서는 절식 후 변화하는 것을 확인하였으며, 일반 사료를 섭취한 마우스의 절식 전후 변화가 없었다. Difference abundance analysis는 일반사료를 섭취한 마우스에서 절식 이후 S24-7로 분류되는 OTU는 증가하고, Ruminococcaceae로 분류되는 OTU가 감소하는 것으로 확인되었다. 반면, 고지방사료를 섭취한 마우스에서는 절식 이후 10OTUs가 감소하고, 3OTUs가 증가하였는데, 대부분 Ruminococcaceae로 분류되는 것으로 확인되었다. 본 연구 결과는 마우스에서의 절식이 장내미생물생태 중 Ruminococcaceae의 abundance에 영향을 미치며, 일반사료를 섭취한 마우스에 비해 고지방사료를 섭취한 마우스에서 절식에 대한 효과가 더 명확하게 나타난다는 것을 시사한다.

인삼, 산양산삼 및 홍삼 추출물의 항비만 효과 (Anti-obesity effects of cultivated ginseng, -wild simulated ginseng and -red ginseng extracts)

  • This study was to investigate whether cultivated ginseng (CG), cultivated wild simulated ginseng (CWG) and cultivated red ginseng (CRG) extracts influences on the obesity. The saponin contents of 3 kinds of ginsengs were analysed by HPLC-ESI-TOF-MS. Total saponin contents were determined in CG on the most contents and since red ginseng has the highest PD (protopanaxadiol type) / PT (protopanaxatriol type) ratio, there may be differences between ginseng, wild ginseng, and red ginseng with respect to their pharmacological effects. Male C57BL/6J mice were fed a normal diet(N), HFD (60% Kcal fat, C), HFD with CG, CWG and CRG extracts (800 mg/kg) for 5 weeks. We observed change of total body weight, degree of hepatic lipid accumulation and immunohistochemical change of GLP-1 and insulin-secreting cells. Also this study attempts to use the physiological analysis method to analyze the changes of blood lipids, insulin and leptin concentration. The change of body weight and size of accumulated lipid droplets in liver lobules decreased in all of the experimental groups than the control(C) group. In the pancreas, the immunohistochemical density of insulin-secreting cells were significantly stronger in the CWG and CRG than C group. The levels of serum insulin and leptin significantly decreased 55.6%, 54.3% respectively in CWG and CRG. The changes of triglyceride, total cholesterol in serum decreased in CRG than the C group. Obesity related CG, CWG and CRG extracts might have contribute to improvement of obesity by regulating the levels of blood lipids and biochemical indicator of fat accumulation.

보중익기탕(補中益氣湯)이 streptozotocin 유발 당뇨병성 위부전마비 백서에 미치는 영향 (The Effects of Bojungikgi-tang on Streptozotocin-induced Diabetic Gastroparesis Rat Model)

  • Objective : Diabetic gastroparesis is a complication that is defined as delayed gastric emptying and upper gastrointestinal symptoms and often occurs in long-standing diabetic patients. Bojungikgi-tang (BJT) is a traditional oriental herbal formula that has long been used for the treatment of digestive disorders. The purpose of this study was to investigate the effects of BJT on streptozotocin (STZ)-induced diabetic gastroparesis rat model. Methods : Sprague-Dawley (SD) male rats (250-270g) were divided into 13 groups including normal group, STZ-induced diabetic control group, BJT diet (7 various concentrations), and insulin-, glibenclamide-, metformin-treated group were used for the experiments for the comparison. Diabetic gastroparesis was induced by intraperitoneal injection of STZ. The water intake, food intake, body weights and fasting blood glucose levels were measured. After 4 weeks the animals were sacrificed and gastrin, leptin, insulin, hemoglobin A1C (HbA1c), lactate, lactate dehydrogenase (LDH), bilirubin, creatinine, albumin and lipid levels were evaluated. Results : Intraperitoneal injection of BJT for 4 weeks resulted in increased levels of gastrin in blood and decreased leptin and lactate concentration in STZ-induced diabetic gastroparesis rat model. BJT did not affect insulin, fasting glucose, HbA1c, and lipid levels in STZ-induced diabetic gastroparesis rat model. Conclusion : These results indicated that BJT would have protect effect on diabetic gastroparesis through the improvement effect of gastric motility and fatigue syndrome in STZ-induced diabetic rats. This study shows that BJT might be effective for treatment of diabetes and its complications such as gastroparesis.

인구통계학적 특성에 따른 성인 대상별 산림치유 프로그램 선호도 분석 (The Preference Analysis of Adults on the Forest Therapy Program with regard to Demographic Characteristics)

  • 본 연구에서는 성인 대상별 산림치유 프로그램의 선호도를 분석함으로써 산림치유 프로그램의 효율적인 개발 및 운영을 위한 기초 자료를 얻고자 한다. 일반 성인 남 여 516명을 대상으로 산림치유 프로그램 37개에 관한 선호도 설문조사를 실시하였다. 자료분석은 SPSS 21.0 프로그램을 사용하여 각 프로그램의 기술통계분석 및 상관관계를 살펴본 결과 많은 공변량이 확인되어, 자료를 의미있게 축약하기 위해 요인분석을 실시하였으며, 신뢰도 분석, 빈도분석, T-검정, 분산분석을 통해 인구통계학적 특성(성별, 연령, 지역, 직업)에 따른 산림치유 프로그램 선호도의 차이를 살펴보았다. 37개의 산림치유 프로그램을 축약하기 위하여 베리멕스 직각회전법을 이용한 요인분석을 실시하였다. 이를 통해, 프로그램은 7개의 프로그램군(1: 심리치료중심 프로그램, 2: 강의 상담중심 프로그램, 3: 캠핑 산림욕중심 프로그램, 4군: 식이중심 프로그램, 5: 자연 식물중심 프로그램, 6: 명상중심 프로그램, 7: 호흡 요가중심 프로그램)과 7개의 독립프로그램(1: 숲속 잠자기, 2: 비전 퀘스트, 3: 물속에 손과 발 담그기 4: 신발 신고 숲속 걷기, 5: 숲 경관 보기, 6: 등산, 7: 숲속 운동회)으로 축약되었다. 이를 중심으로 성별, 연령, 지역, 직업에 따라 산림치유 프로그램 선호도의 차이를 검증하였다. 첫째, 성별에 따라서 심리치료중심 프로그램군, 식이중심 프로그램군, 명상중심 프로그램군, 호흡 요가중심 프로그램군, 등산 프로그램에 대해서 남 여 간에 유의한 차이가 있는 것으로 검증되었다. 둘째, 연령에 따라서 강의 상담중심 프로그램군, 캠핑 산림욕 중심 프로그램군, 자연 식물중심 프로그램군, 명상중심 프로그램군, 호흡 요가중심 프로그램군, 숲속 운동회 프로그램에 대하여 유의한 차이가 있는 것으로 나타났다. 셋째, 지역에 따라서 심리치료 중심 프로그램군, 강의 상담중심 프로그램군, 식이중심 프로그램군, 자연 식물중심 프로그램군, 명상중심 프로그램군, 비전 퀘스트, 등산, 숲속 운동회에 있어서 유의한 차이가 있는 것으로 나타났다. 넷째, 직업에 따라서 자연 식물중심 프로그램군, 명상중심 프로그램군, 비전 퀘스트, 신발 신고 숲속 걷기, 숲 경관 보기, 등산에 대하여 유의한 차이가 있는 것으로 나타났다. 이상에서 살펴본 바와 같이, 인구통계학적 특성에 따른 산림치유 프로그램의 선호도에 대한 다양성을 고려하여 산림치유 프로그램을 개발하여야 할 것으로 사료된다. 본 연구의 분석 결과가 성인을 대상으로 하는 산림치유 프로그램 개발을 위한 기초자료로 활용되기를 기대한다.

뽕잎추출물이 고콜레스테롤 식이 흰쥐의 지질대사에 미치는 영향 (Effects of Mulberry Leaf Extract Feeding on Lipid Status of Rats Fed High Cholesterol Diets)

  • 본 연구는 뽕잎추출물의 지질대사 개선효과를 검정하고자 하였다. 실험 동물은 체중 100$\pm$10 g 내외의 Sprague-Dawley 종 수컷을 구입하여 난괴법 (randomized complet block design)에 의해 정상식이군(N group), 정상식이에 0.16% 뽕잎추출물 공급군(NM group), 고콜레스테롤식이 공급군(HC group)과 고콜레스테롤식이에 뽕잎추출물 공급군으로 나눈 후 다시 뽕잎추출물의 급여 수준에 따라 뽕잎추출물을 0.08% 공급한 군(HCL group), 뽕잎추출물을 0.16% 공급한 군(HCM group), 뽕잎추출물을 0.32% 공급한 군(HCH group) 등 각 군 모두 10마리씩 하였다. 식이와 식수는 자유 공급시켰으며 4주간 사육한 후 희생시켰다. 체중증가량, 식이섭취량, 식수섭취량 및 식이효율은 실험군간의 유의적인 차이는 없었다. 혈중 중성지방, 총콜레스테롤, LDL-콜레스테롤, A.I 등은 고콜레스테롤군에 비해 뽕잎추출물 공급군이 유의적으로 감소되었으며 HDL-콜레스테롤은 고콜레스테롤군에 비해 뽕잎추출물 공급군이 유의적으로 증가되었다. UDP-glucuronyl transferase(UDP-GTase) 활성은 N군에 비해 HC군에서 유의적으로 증가되었으나 고콜레스테롤 식이군간의 유의적인 차이는 없었다. 간조직중의 총지방 및 콜레스테롤 함량은 고콜레스테롤군에 비해 뽕잎추출물 공급군에서 유의적으로 감소되었다. 콜레스테롤 함량은 고콜레스테롤 식이군 간의 유의적인 차이가 없었다. Lipoprotein lipase활성은 HC군에 비하여 HCM 및 HCH군에서 유의적으로 감소되었다. 부고환 지방 중의 총지질, 중성지방, 콜레스테롤 함량은 HC군에 비해 뽕잎추출물 공급군에서 유의적으로 감소되었다. 분변 중으로 배설되는 총지질, 중성지방, 콜레스테롤 함량은 HC군에 비해 뽕잎추출물 공급군에서 증가되었다. 이상의 결과에서 뽕잎추출물의 고콜레스테롤 식이 흰쥐에서 혈액, 간, 부고환 조직 및 분변중의 지질대사 개선효과가 규명되었다.
