  • 1. 종자형성 계통군의 출현은 흑색 PE 피복, 녹색 PE 피복 및 투명 PE 피복이 무피복보다 각각 1일, 2일 및 3일 빨랐으며, 유식물체형성 계통군의 출현은 흑색 PE 피복, 녹색 PE피복 및 투명 PE피복이 무피복보다 각각 2일, 2일 및 3일 빨랐다. 2. 생근경중은 종자형성계통 군에서 투명 PE 피복이 무피복보다 16.7% 증수하였으며, 유식물체형성 계통 군에서는 녹색 PE 피복 및 투명 PE 피복이 무피복보다 각각29.4%, 26.5% 증수하였다. 3. 메탄올엑스함량은 종자형성 계통 군에서는 투명 PE피복이 42.60%로서 가장 많았으며, 흑색 PE 피복이37.50%로서 무피복보다 많았으나, 유식물체형성 계통군에서는 각처리 모두 무피복과 차이가 없었다. 4.조사포닌 함량은 종자형성 계통군에서는 투명 PE피복이 8.25%로서 무피복, 흑백 PE 피복 및 녹색 PE피복보다는 높았으나, 흑색 PE 피복과는 차이가 없었으며, 유식물체형성 계통군에서는 투명 PE피복이 7.75%로서 흑색 PE 피복이나, 녹색 PE 피복보다 높은 함량을 나타냈으나, 무피복 및 흑백 PE 피복과는 차이가 없었다.

Effects of Non-saponin Red Ginseng Components on the Function of Brain Cells

  • Non-saponin gingseng fraction components (NSRG) have been known to have a variety of biological activity. However, the effects of these components on the function of brain cell have not been characterized in detail. In this study, we investigated the preventive effect of non-saponin red ginseng components on acrylamide (ACR)-induced suppression of neural cell adhesion molecule (NCAM), which is highly expressed in neuronal cells. The data showed that NSRG blocked the suppression of NCAM expression by ACR in neuroblastoma cells (SK-N-SH). In addition, NSRG significantly increased NCAM expression in ACR-nontreated neuroblastoma cells. NSRG treatment resulted in the increase of cell proliferation in a concentration-dependent manner. We also examined whether NSRG could modulate the NO production of astrocytes. When glioma cells (C6) were treated with various concentrations of NSRG (100-300 ug/ml) in the presence or absence of $IFN-{\gamma}$ for 24 hours, NO production was suppressed in $IFN-{\gamma}-$stimulated C6 cells. Taken together, these results demonstrate that treatment of brain cells with NSRG results in the enhancement of proliferation, the suppression of NO production and the protective effect on NCAM expression impaired by ACR. Thus, the present data suggest that NSRG has proliferative and neuroprotective effects and these effects could be useful in neuronal diseases.

초임계 유체로 추출된 산삼 부정 배양근 오일의 효모균 처리에 의한 비사포닌계 지방산 함량 변화 및 Collagenase 및 Elastase 저해 활성 증대 (Changes in Non-saponin Fatty Acid Content and Increases in Inhibitory Activities of Collagenase and Elastase by Treatment with Saccharomyces cerevisiae of the Supercritical Fluid Extracted Oil of the Adventitious Roots Culture of Wild Mountain Ginseng)

  • Background: To obtain useful cosmetic resources, this study aimed to determine the non-saponin fatty acid and inhibitory activities of collagenase and elastase by treatment of Saccharomyces cerevisiae in supercritical fluid extracted oil of the adventitious root culture of wild mountain ginseng. Methods and Results: We performed supercritical fluid extraction at various conditions such as pressure, temperature, time, and use of co-solvents, unlike the n-hexane extraction for the adventitious roots culture of wild mountain ginseng. The non-saponin-fatty acid obtained from the oil of the adventitious roots culture was incresed by treatment with S. cerevisiae. The supercritical fluid extraction was conducted using gas chromatography. Non-saponin-fatty acid content, in the oil of adventitious roots culture of wild mountain ginseng treated with S. cerevisiae for 2 days were three times higher than that in the control. In addition, the oil of the adventitious roots culture treated with S. cerevisiae was investigated for the anti-wrinkle effect by using collagenase and elastase. The oil of adventitious roots culture treated with S. cerevisiae exhibited higher collagenase and elastase inhibitory activities than those in the control. Conclusions: Supercritical fluid extracted oil of the adventitious roots culture of wild mountain ginseng treated with S. cerevisiae was found to have decreased ratio of saturated fatty acids and incresed ratio and content of unsaturated fatty acids increased. Furthermore, it showed anti-wrinkle effects in vitro.

  • The ameliorating effect of the MeOH extract of the root of Platycodon grandiflorum on the ethanol-induced cognitive dysfunction in mice was investigated. The total MeOH extract was devided into two fractions, saponin rich fraction and non-saponin fraction. The mice with repeated administration of the total MeOH extract and those with saponin rich fraction both showed a markedly prolongεd latency time upon the step-through test (passive avoidance performance) performed after acute ethanol intoxication, while those with non-saponin rich fraction did not. The results suggest that the extracts from the root of Platycodon grandiflorum exert a beneficial effect on ethanol-induced memory impairment in mice and also suggest that these effects of the extracts might be ascribed to the saponin constituents.

Long-term administration of red ginseng non-saponin fraction rescues the loss of skeletal muscle mass and strength associated with aging in mice

  • Background: Sarcopenia is a new and emerging risk factor aggravating the quality of life of elderly population. Because Korean Red Ginseng (RG) is known to have a great effect on relieving fatigue and enhancing physical performance, it is invaluable to examine its potential as an anti-sarcopenic drug. Methods: Anti-sarcopenic effect of non-saponin fraction of Korean Red Ginseng (RGNS) was evaluated in C2C12 myoblasts treated with C2-ceramide to induce senescence phenotypes, and 22-month-old mice fed with chow diet containing 2% RGNS (w/w) for 4 further months. Results: The RGNS treatment significantly alleviated cellular senescence indicated by intracellular lipid accumulation, increased amount of lysosomal β-galactosidase, and reduced proliferative capacity in C2C12 myoblasts. This effect was not observed with saponin fraction. In an aged mouse, the 4-month-RGNS diet significantly improved aging-associated loss of muscle mass and strength, assessed by the weights of hindlimb skeletal muscles such as tibialis anterior (TA), extensor digitorum longus (EDL), gastrocnemius (GN) and soleus (SOL), and the cross-sectional area (CSA) of SOL muscle, and the behaviors in grip strength and hanging wire tests, respectively. During the same period, an aging-associated shift of fast-to slow-twitch muscle in SOL muscle was also retarded by the RGNS treatment. Conclusions: These findings suggested that the long-term diet of RGNS significantly prevented aging-associated muscle atrophy and reduced physical performance, and thus RGNS has a strong potential to be developed as a drug that prevents or improves sarcopenia.

The non-saponin fraction of Korean Red Ginseng ameliorates sarcopenia by regulating immune homeostasis in 22-26-month-old C57BL/6J mice

  • Background: The non-saponin fraction (NSF) of Korean Red Ginseng is a powder in which saponin is eliminated from red ginseng concentrate by fractionation. In this study, we examined the effect of NSF on age-associated sarcopenia in old mice. Methods: NSF (50 or 200 mg/kg/day) was administered orally daily to young (3-6-month-old) and old (20-24-month-old) C57BL/6 J mice for 6 weeks. Body weight and grip strength were assessed once a week during the oral administration period. The gastrocnemius and quadriceps muscle were excised, and the muscle fiber size was compared through hematoxylin and eosin staining. In addition, the effect of NSF on sarcopenia and inflammation/oxidative stress-related factors in hindlimb muscles was investigated by western blotting. Flow cytometry analysis was conducted to investigate the effect of NSF on immune homeostasis. Blood samples were collected by cardiac puncture, and the serum levels of insulin-like growth factor 1, pro-inflammatory cytokines, and glutathione were evaluated. Results: NSF significantly alleviated muscle strength, mass, and also fiber size in old mice. Age-associated impairment of immune homeostasis was recovered by NSF through retaining CD11b+F4/80+ macrophages and regulating inflammatory biomarkers. NSF also decreased the age-induced expression of oxidative stress factors. Taken together, NSF showed the effect of improving sarcopenia by inhibiting low-grade chronic inflammatory/oxidative stress factors. Conclusion: NSF exhibited anti-sarcopenia effects by regulating chronic inflammation and oxidative stress in old mice. Thus, we suggest that NSF is a promising restorative agent that can be used to improve sarcopenia in the elderly as well as maintain immune homeostasis.

대두 사포닌의 활성에 관한 기전 연구

  • Saponins are glycosidic compounds present in many plant foods. They are characterized by their ability to lyse cell membranes due to their surface-active properties. Saponins are believed to interact primarily with cholesterol in the cell membrane. In this study, the interaction of soybean(SS) with cell membrane was investigated using erythrocytes as a model. Mechanisms of interaction was also investigated by measuring their binding capacity with different membrane lipid fractions. Throughout the study, gypsophilla saponin(GS) and quillaja saponin(QS) were used to evaluate the membranolytic activity of soybean saponins. All saponins released hemoglobin in a concentration-dependent manner. SS induced 40% hemolysis at the concentration of 400 ppm, however there was no increase in hemoglobin release above 400ppm concentration. 5ppm of GS and 8 ppm of QS hemolyzed 100% of erythrocytes. Isolation of SS fractions by thin layer chromatography revealed that only one non-polar saponin possesses strong hemolytic activity. When saponins were incubated decreased the release of cholesterol. When the hemolytic activity of saponins was measured in the presence of other major membrane lipid components, sphingomyelin significantly reduced the hemolytic activity of SS, while cholesterol reduced the activity of QS. GS showed high affinity to other component(s) in the incubation media as well as lipids. These results suggest that the membranolytic activity of saponins are related to their specific chemical structure, which determines the interaction behavior between saponins and different membrane components, and thereby influence the biological activity.

  • This studies were conducted to investigate the changes of saponin pattern, pH and organic acid contents of malt wort added ginseng components during alcoholic fermentation by Sacch. uvarum. The results are as follows. Saponin patterns of fermented wort were same as that of the non- fermented wort, but the weight of former was decreased comparing to that of the latter. pH value of fermented wort contained 0.1∼0.5% of ginseng extract were almost same as that of control(PH 4.23). Lactate, pyruvate, succinate and fumarate, pyroglutarate and citrate contents of the fermented wort were increased by the addition of ginseng extract and pyruvate content, particularly, was increased from 28.4 to 214mg/100 ml while that of control was 33.2mg/100m1. Citrate content of fermented wort contained ginseng saponin was almost same as control (37. 5mg/100m1) . But pyruvate content was tower 4-8.6mg/100m1 than that of control(33.2mg/100m1) .

홍삼 분획물이 감마선을 비사한 생쥐 간에서 항산화물질과 지질과산화에 미치는 방사선 보호효과 (Radioprotective Effect of Ginseng Components on Antioxidant Enzymes, Glutathione and Lipid Peroxidation of Liver in ${\gamma}$-Irradiated Mice)

  • In the present study, to determine whether the antioxidative components of Korean red ginseng protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of total saponin (200 mg/kg, ip) and lipophilic fraction (200 mg/kg, oral) preferment of mice on the survival ratio, major antioxidant enzymes (SOD, catalase and glutathione peroxidase) activities, glutathione levels and lipid peroxidation in the liver were exiled for 2 weeks after whole ${\gamma}$-body ${\gamma}$-irradiation (6.5 Gy). The 30-day survival ratio increased from 10% to 57% and 40% for mice treated with total saponin and lipophilic fraction, respectively. On day 14 after ${\gamma}$-irradiation, the ginseng total saponin pretreatment produced a slight increase of antioxidant enzymes activities and significantly Increased reduced glutathione (GSH) contents (p<0.05) in the liver compared with non-treated group. Pretreatment with ginseng total saponin significantly deceased GSSG/total GSH ratio (p<0.05) without change of GSSG in the liver and inhibited the radiation-induced incense in the hepatic malondialdehyde levels. (p<0.05) In these results, GSH plays an important role in the liver in several detoxifications and the reduction of lipid peroxides. Thus, it appears that total saponin of red ginseng exerts its radioprotective effect by accelerating the production of endogenous antioxidants, such as glutathione from radiation induced damages and thereby oxygen free radicals.

Colloidal Gas Aphrons의 제조와 안정성에 대한 연구 (A Study on Preparation of Colloidal Gas Aphrons and Stability)

  • 비이온성 및 이온성 계면활성제를 이용하여 제조한 CGA(cllodial gas aphron)의 안정성을 고찰하였다. 실험에서 이용한 계면활성제는 SDS(Sigma-Aldrich, Co.), Triton X-100, Tween 80, Quillaja Saponin이다. CGA발생장치를 이용하여 단일 성분의 계면활성제 및 혼합 계면활성제에 의한 CGA를 제조하였고, 농도, 온도, 시간에 따른 CGA의 안정성을 조사하였다. 단일 성분 CGA의 경우, Saponin이 143분의 지속시간을 보여 가장 높은 안정성을 나타내었고 Triton X-100, SDS, Tween 80순으로 안정성이 높았다(상온). CGA제조시 계면활성제 용액을 $70^{\circ}C$로 가열한 경우, SDS가 가장 높은 안정성을 보였으며(116분) Saponin은 상온에 비해 안정성이 낮아졌다. (105분). 혼합 계면활성제일 경우, 두 계면활성제의 중간 수준의 안정성을 보였으며, SDS+Saponin 조합이 139분으로 가장 안정성이 컸다. 가열한 혼합 계면활성제에서는 Saponin+Triton X-100 조합이 가장 안정성이 낮아 53분에 불과했다. CGA는 초기 평균 입경이 약 140 ${\mu}m$ 였다가 융합이 일어나면 그 크기가 약 190 ${\mu}m$로 상승하고 이후 붕괴하는 양상을 띠었다. 가열하여 제조한 CGA에서는, Saponin이 포함된 경우 상온에 비해 그 크기가 감소하였으며, 타 계면활성제에서는 일반적으로 그 크기가 증가하였다. 요약하면 CGA는 가열 했을 때 보다는 상온일때, 분자간 상호작용이 크게 작용하는 혼합 계면활성제보다 단일 계면활성제일 경우, CGA 지속시간이 길어져 안정성이 높아졌다.
