Analysis of Diapause-Associated Changes in Silkworm Egg Proteins

  • The patterns of diapause-associated proteins of silkworm eggs were analyzed by two-dimensional (2-D) gel electrophoresis. Among the hundreds of spots on the 2-D gels, at least two proteins were considered to be associated with diapause. A protein, spot 4, with an approximate molecular weight of 38 kDa and pI 6.1 was observed in the HCI-treated, cold-treated, and diapause eggs, respectively. Spot 4 was undetectable in unfertilized eggs and non-diapause eggs at two days after oviposition, suggesting that this protein may be associated with the entrance to diapause. A protein, spot 11, with an approximate molecular weight of 21 kDa and pI of 61 was detected in the unfertilized, HCl-treated, and cold-treated eggs, respectively, after oviposition by normal moths. In diapausing eggs, a protein corresponding to spot 11 was observed in 3-, 5-, and 30-day-old eggs, while the protein was not detected one-day-old eggs. The protein corresponding to spot 11 was not detected in unfertilized and non-diapause eggs obtained from subesophabeal ganglion (SG)-extirpated moths either. Spot 11 was also considered to be a diapause specific protein, which occurred at only early embryonic stage under the control of diapause-downregulated gene.

Hormonal Control of Induction and Termination of Diapause in Pieris rapae L. (배추흰나비(Pieris rapae L.)의 휴면 유도 및 타파에 대한 호르몬의 조절)

  • Non-diapausing larvae and diapausing pupae of Pieris rapae L. were treated with 20-HE and JH, and the concentration of 20-HE and JH in body were also measured to determine mechanisms of diapause induction and termination. When early last instar larvae of non-diapausing stage were injected with 20-HE or JH-I, diapause was not induced in both cases. 20-HE concentration of pre-diapausing last instar larvae was very low, and 20-HE peak of non-diapausing two-day-old pupae was not present in diapausing pupae. There was no difference in concentration of JH-I between diapausing and non-diapausing stages. When early and late diapausing pupae were injected with 20-HE, JH-I, or JH-III. diapause was terminated by 0.1-0.5 $\mu$g of 20-HE, but not by JH-I, or JH-III. The above results indicate that diapause of Pieris rapae was induced by decrease of 20-HE concentration and also terminated by gradual accumulation of 20-HE during diapause process.

Diapause and Voltinism in Ostrinia furnacalis (Lepidoptera: Crambidae) in Suwon, and Larval Instar Sensitivity to Diapause Induction (수원 지방에서 조명나방 휴면과 발생 세대수 및 휴면유도에 대한 유충의 민감성)

  • The diapause induction season in Ostrinia furnacalis (Lepidoptera: Crambidae) was estimated in Suwon. Three batches of adult generations were observed, the first one from early May to early July, the second from early or mid-July to early or mid-August, and the third from mid-August to October. In outdoor larval rearing, colony rearing occurring from mid-July to mid-August produced both non-overwintering and overwintering larvae, whereas late-reared colonies produced only overwintering larvae. Larvae collected during July and August in maize fields produced both non-overwintering and overwintering larvae, whereas late-collected larvae produced only overwintering larvae. The results indicated that O. furnacalis has a bi- or trivoltine complex life cycle in this area. In the laboratory, when larvae of all instars within 9 h after molting were first treated to a diapause induction condition (11:13 h = light:dark photoperiod and 20℃), almost all larvae were induced to diapause. However, when similar treatments were conducted age-specifically for the 5th instar larvae, diapause induction rates in 3- and 4-day-old larvae of the 5th instar decreased. In contrast, when larvae were subjected to the diapause induction treatment only during the periods from the hatching stage to the 2nd, 3rd, and 4th instar, almost all larvae were not induced to diapause. The results suggest that the early age of the 5th larval instar is the last stage for sensitivity to diapause induction stimuli. In the diapause-induced larvae, hemolymph trehalose content increased and body supercooling points dropped, compared with those in non-diapause larvae.

Quantitative Changes of Free Sugars and Glycogen during Embryonic Devdlopment of Non-diapause Eggs of the Silkworm, Bombyx mori L. which were Stored at Low Temperature (저온 저장한 누에 비휴면란의 배발육기에 있어서 유리당류의 양적 변동)

  • It was carried out to find the carbohydrate metabolism for the glycogen utilization and the effects of low temperature(1$\circ$C, 5$\circ$C)on embryonic development of the silkworm, Bombyx mori L. The content of sorbitol and glycerol in the chillihg eggs stored at 5$\circ$C or 1$\circ$C on 24 hours after oviposition showed the highest level on the 50th day. After that, the corresponding value in the eggs stored at 5$\circ$C were decreased, and the value in the eggs stored at 1$\circ$C maintained the similar level. The trehalose level was rapidly increased on 50$\sim$60th day or thereabouts, and then, it was accumulated. When the eggs stored at low temperature were tranaferred and developed the embryo at 25$\circ$C, sorbitol and glycerol were respectively changed to glycogen. Futhemore, the content of trehalose and glucose showed specific of patterns with each embryonic stage at low temperature. With the aforementioned results, the intemediary metabolism of carbohydrate was discussed with regard to embryonic development of non-diapause eggs of silkworms.

Effects of Temperature on Survival, Development, and Reproduction of the Non-diapause Asian Corn Borer, Ostrinia furnacalis (Lepidoptera: Crambidae) (조명나방(나비목: 포충나방과) 비휴면태의 생존과 발육, 생식에 미치는 온도의 영향)

  • We analyzed the effects of temperature on the survival, development, and reproduction of the non-diapause Asian corn borer, Ostrinia furnacalis (Lepidoptera: Crambidae). A 16:8 h light:dark photoperiod was supplied to the non-diapause stages of O. furnacalis. The insects were raised on an artificial diet at seven constant temperatures between 15 and 35℃ for immature stages, and eight temperatures between 13 and 33℃ for the adult stage. The survival rate of eggs exceeded 70% at all temperatures, whereas survival of larvae was as low as 7.4% at 15℃. The developmental periods of the immature stages decreased with increasing temperature, but the larval period did not decrease further at 35℃. The pupal weight increased with increasing temperature, but the female weight decreased at 35℃. Variations with different last larval instars within the colonies at different temperatures were observed. Adults produced offspring at all tested temperatures. Adult longevity and the pre-oviposition and oviposition periods decreased with increasing temperature, but the pre-oviposition period increased at 33℃. Total fecundity exceeded 400 viable eggs at 22℃ and 31℃. The mean daily fecundity during the oviposition period and the mean daily fecundity on the day of oviposition increased with increasing temperature, but decreased at 33℃. Daily fecundity sharply increased at earlier adult ages and slowly decreased thereafter. The simulated oviposition frequency was greatest at 22℃. The lower developmental threshold temperatures were estimated to be the lowest at 9.7℃ for the first instar larva and the highest at 14.7℃ for the 5th to last instar larval stage, using a linear model.

Rate of Oxygen Uptake in Antheraea mylitta in Various Stages of Development and during Diapause

  • Respiration in Antheraea mylitta was studied using constant pressure respirometer. Oxygen uptake in different stages of life showed that moth has highest rate of oxygen uptake as compared to larva and pupa. At each stage significant sexual differentiation was observed. Depression in the rate of oxygen uptake in diapausing pupa was found to be higher in male ($65.75\%$) than female ($60.65\%$) as compared to non-diapausing counterparts. During diapause lowest oxygen uptake was recorded in the month of February, and were in the order of February < January < December < March < November < April < May < June in male, and, February < January < December < March < April < November < May < June in female. Significant sexual differentiation in oxygen uptake was recorded throughout the period of diapause (November to June) where female pupae registered lower rate of oxygen uptake than that of male. Oxygen uptake in female pupae declined upto $28.89\%$ in non-diapausing and $18.29\%$ in diapausing broods over male. Highest respiratory quotient value was recorded in the moth of November (0.68 in male and 0.70 in female) and, the lowest in the month of March in male (0.54) and May in female (0.55). Percentage loss in pupa weight always remained at a significantly higher level in male (except in February and March). The lowest oxygen uptake rate and weight loss in the month of February shows that the pupae were at peak of their dormancy during this month.

Attraction and Seasonal Occurrence of Piezodorus hybneri monitored with Aggregation Pheromone Traps of Riptortus clavatus (톱다리개미허리노린재의 집합페로몬 트랩에 의한 가로줄노린재의 유인과 발생양상)

  • Seasonal occurrence of the stink bug Piezodorus hybneri (Gmelin) (Heteroptera: Penta-tomidae) was monitored at a soybean field and the campus of Gyeongsang National University, Gyeongnam. Korea, in which host plants are less available for the bug with aggregation pheromone traps of Riptortus clavatus Thunberg (Heteroptera: Alydidae) in 2004. At soybean field, P. hybneri began to be attracted to traps from June 28, earlier than flowering stage of soybean plants. Number of P. hybneri attracted increased sharply after October 11. At the campus the bugs were not attracted to traps from March 21 to October 5, but began to be attracted since October 11. Difference in the occurrences in the two sites may suggest that the stink bug immigrates actively into soybean field to find host plant. P. hybneri that had been attracted to traps since October 11 was assumed to be diapausing adults emigrating to hibernation sites. There was no difference between sexes in trap catches. The fact that P. hybneri was attracted from June to late November may suggest that the aggregation pheromone of R. clavatus was attractive to both non-diapausing and diapausing adults of P. hybneri. The aggregation pheromone traps, when baited with 16.7+16.7+16.7 mg or 7+36+7 mg of (E)-2-hexenyl (Z)-3-hexenoate, (E)-2-hexenyl (E)-2-hexenoate, and myristyl isobutyrate, attracted significantly higher number of both sexes of P. hybneri adults than the live male traps baited with ten R. clavatus males and hexane control.
