An Analysis of Vegetation-Environment Relationships of Quercus mongolica Communities by TWINSPAN and DCCA (TWINSPAN과 DCCA에 의한 신갈나무군집(群集)과 환경(環境)의 상관관계(相關關係) 분석(分析))

  • Vegetational data from 81 quadrats of Quercus mongolica communities in Mt. Odae and Mt. Jumbong were analysed by applying two multivariate methods : two - way indicator species analysis(TWNSPAN) for classification and detrended canonical correspondence analysis(DCCA) for ordination. The dominant tree species of Quercus mongolica communities were found in the order of Quercus mongolica, Acer pseudosieboldianum. Tilia amurensis, Carpinus cordata, Acer mono, and Fraxinus rhynchophylla. The forest vegetation of Quercus mongolica community was classified into Quercus mongolica, Quercus mongolica - Carpinus cordata, and Quercus mongolica - Abies nephrolepis groups according to the TWNSPAN. The relationships between the distribution of dominant groups for forest vegetation and soil condition in Quercus mongolica communities were investigated by analysing elevation and soil nutrition gradients. Quercus mongolica group was distributed in the low elevation and poor nutrition area of total nitrogen, Quercus mongolica - Carpinus cordata group was distributed in the low elevation and good nutrition area of $Mg^{{+}{+}}$ and $Ca^{{+}{+}}$, while Quercus mongolica Abies nephrolepis group was distributed in the high elevation and poor nutrition area of $Mg^{{+}{+}}$ and $Ca^{{+}{+}}$. The two dominant factors influencing community distribution were elevation and total nitrogen.

Removal of Organic Matter and Nitrogen in a Model System of Riverbed Filtration (하상여과 모형에서 유기물과 질소의 제거)

  • A column experiment was performed to investigate the influence of the sludge cake development on the riverbed and the hydraulic gradient imposed by the drawdown at the well on the filtrate quality in order to offer a guideline in the design and operation of the riverbed filtration. Results show that the sludge cake on the riverbed plays an important role in the removal of the organic matter. Under the conditions of this study the COD removal rate increased from 17% to 50% along with the sludge cake development, which was equivalent to the BCOD removal of 22% and 67%, respectively. The active removal of the organic matter took place in the sludge cake and the upper 40 cm of the riverbed. As the flow rate increased owing to the increase in the head difference imposed on the column, the slope of the COD profile near the column inlet decreased, however, the profiles converged in about 40 cm from the inlet. In 10 days of sludge cake formation the dissolved oxygen was depleted at the depth of 70 cm, which suggests the denitrification can take place beyond the depth. This depth was further reduced to $20{\sim}40\; cm$ as the sludge cake developed. From this study the removal of organic matter can be expected through the riverbed filtration even with the depth of as shallow as 3 m, which is frequently met in Korea, while the removal of nitrogen through denitrification is not expected to be active under the condition.

Temporal Variations of Submarine Groundwater Discharge (SGD) and SGD-driven Nutrient Inputs in the Coastal Ocean of Jeju Island (제주도 연안에서 해저 지하수 및 지하수 기원 영양염류 유입량의 시간적 변화)

  • To determine the temporal variations of submarine groundwater discharge (SGD) and SGD-driven nutrients inputs, we measured the seepage rate and the nutrient concentrations of pore water/groundwater in Bangdu Bay of Jeju Island at two and three month intervals from September 2009 to September 2010. The seepage rate of groundwater ranged from 0 to 330 cm/day (average ~170 cm/day) during the five sampling periods, which increased sharply from high tide to low tide due to changes in hydraulic pressure gradient between water table in land and water sea level in the coastal ocean by the tidal cycles. The submarine inputs of groundwater were also relatively higher in summer than in winter. The nutrient fluxes from SGD were about 90~100%, 70~95%, and 65~100% of the total input (except from open ocean waters) for dissolved inorganic nitrogen (DIN), phosphorus (DIP), and silicate (DSi), respectively, potentially supporting about 0.9~33 g $carbon/m^2/day$ of new primary production in Baugdu Bay. Thus, our study suggests that SGD-driven nutrients may play an important role in the eutrophication and biological production in the coastal ocean of Jeju Island.

Evaluating Effect of Density Flow from Upstream on Vertical Distribution of Water Quality at the Paldang Reservoir (팔당호 수질의 연직분포에 대한 밀도류 영향 평가)

  • Paldang is a river reservoir in the Midwest of Korea, which is a drinking water source for the metropolitan area. Since the Paldang Reservoir is shallow, and has a short hydraulic residence time, its water quality is directly impacted by two incoming rivers, the north Han River (NHR) and the south Han River (SHR). The NHR has different seasonal patterns of water temperature from the SHR because the NHR is greatly impacted by the discharge water from upstream dams. The electrical conductivity (EC) and other material concentrations of the SHR are usually higher than those of the NHR because its basin is limestone-based. The difference in water temperature in the two rivers causes density flow, and the distribution of the EC within the reservoir can be an indicator for monitoring density flow. From the vertical gradient of the EC at the dam site, from spring to fall, it was confirmed that the SHR flowed into the upper layer, and the NHR flowed into the lower layer, and vice versa at other times. The relative difference (RD) of the EC between the upper layer and the lower layer at the dam site was used as an indicator for density flow. The RD of the EC showed a very significant correlation with the RD of total organic carbon (r = 0.70, p < 0.001) and the RD of total nitrogen (r = 0.58, p < 0.01). This relationship is based on the assumption that the difference in electrical conductivity and water quality between the SHR and the NHR is constant. However, in many cases this assumption is inconsistent. Thus, further study is needed on more suitable indicators to evaluate the impact of density flow on water quality.

Analysis on the Growth Environment of Disporum smilacinum A. GRAY for Development of Shade-tolerance Groundcover Plant (내음성 지표식물 개발을 위한 애기나리의 생육환경분석)

  • The purpose of this study was to find the basic application of Disporum genus in landscape-arc-hitectual space. Environment of native site (Daeducksan, Piseulsan) and growth characteristics of experiment plot were investigated and analyzed in this study. The results of this study were as follows; 1. In the case of environment of native site, the relative light intensity was about 10% and distribution of species were primarily north slope. Gradient of Daeducksan and Piseulsan were 10$^{\circ}$, 18$^{\circ}$ Soil textures was Lic both and field capacity of Daeducksan and Piseulsan were 69%, 73.5%. The soil pH of Daeducksan and Piseulsan were 6.1 and 5.8. The both content of organic matter and C.E.C. were high markedly. The P2O5 content of Daeducksan showed high but total nitrogen showed nearly the same level when comparing with the common dry field condition. 2. In tree layer, Daeducksan composed of broadleaved forest of 80% involving Quercus variabilis, and Piseulsan composed of coniferous forest of 80% involving Pinus densiflora on the other hand. Daeducksan, therefore, was more appearance species than Piseulsan in herbaceous layer. 3. Result in ANOVA Test, significance followed with light intensity was recognized in plant height, leaf width, leaf length, leaf thick and No. of leaf, while in the case of soil was not. Plant height was longest in 1% and leaf width, leaf length was largest in 25%. Therefore ornamental value of leaf was the very best in 25%. 4. Result in correlation coefficient analysis, plant height was correlated leaf width, leaf length. The longer plant height, the smaller leaf width, leaf length. In relative light intensity of 1%, flower was hardly expected because of the more shade, the less flower. Disporum smilacinum is expected great use as ground covers in the shades of tall-building or hdavy woods.

  • Recovery component for cell division in UV-irradiated E. coli B was detected with use of the cell extract of E. coli B/r which is a resistant mutant of E. coli B against UV-irradiation. The active substance was non-dialyzable and increased the activity by adding B-NAD remarkably. One more factor for increasing or promoting the restoration recognized was magnesium. Magnesium was effective to stabilze the substance in procedure of isolation. Two active substances were obtained from sucrose gradient centrifugation. One of them was recovred from the botton area and the other from top area just below below surface. the former was not stabilized by magnesium, while the latter stabilized the activity by it remarkably. The former which did not require magnesium was insensitive to protease and the latter which required magnesium was sensitive to it. Both were insensitive to RNase and DNase. Recovery ratio was doubled by using nitrogen gas than aeration in purification process. DNA-ligase less mutant was revealed same activity on it's recovery ratio with the parent strain of E. coli K-12. The active substance stimulating the filament cell may exist as a complex which is inactivated easily in the dissociated state ana requrie B-NAD or magnesium.

Estimation of Nutrients Released from Sediments of Deukryang Bay (득량만 퇴적물로부터 영양염 용출 평가)

  • The purpose of the present study is to estimate the release of dissolved Inorganic nitrogen and photophorous from sediments of Deukryang Bay. One method used In this study is to calculate nutrients released from a concentration gradient between sediment porewaters and the overlying water based on periments. The calculated and measured ammonium released from the sediments were 8.93mg-atN/$m^2$ hr and 60.4mg/atN/$m^2$ hr, respectively In July. 8.57mg-atN/$m^2$. hr and 32.9mg/atN/$m^2$ hr, respectively in October. The ammonium was released more highly in truly than in October, and the measured ammonium flux was higher than the calculated one. The calculated nitrate plus nitrite released from the sediments were 0.31mg/atN/$m^2$. hr in July and 0.84mg/atN/$m^2$. hr in October. The measured nitrate plus nitrite released from the sediments was 282mg/atN/$m^2$. hr in October. The calcuated was lower than the measured because the content of the nutrients in the sediments was always much more than in the overlying waters, and it has shown a differently seasonal pattern compared to the ammonium flux. The calculated phosphorous released from the sediments were 0.97mP/atN/$m^2$ hr and measured negative fluxed -6.50mP/atN/$m^3$ hr In truly, and alculated 0.18mg/atP/$m^2$. hr and measured 24.6mg/atP/$m^2$. hr, respectively in October.

Analysis and Quantification of Ammonia-Oxidizing Bacteria Community with amoA Gene in Sewage Treatment Plants

  • The analysis and quantification of ammonia-oxidizing bacteria (AOB) is crucial, as they initiate the biological removal of ammonia-nitrogen from sewage. Previous methods for analyzing the microbial community structure, which involve the plating of samples or culture media over agar plates, have been inadequate because many microorganisms found in a sewage plant are unculturable. In this study, to exclusively detect AOB, the analysis was carried out via denaturing gradient gel electrophoresis using a primer specific to the amoA gene, which is one of the functional genes known as ammonia monooxygenase. An AOB consortium (S1 sample) that could oxidize an unprecedented 100% of ammonia in 24 h was obtained from sewage sludge. In addition, real-time PCR was used to quantify the AOB. Results of the microbial community analysis in terms of carbon utilization ability of samples showed that the aeration tank water sample (S2), influent water sample (S3), and effluent water sample (S4) used all the 31 substrates considered, whereas the AOB consortium (S1) used only Tween 80, D-galacturonic acid, itaconic acid, D-malic acid, and $_L$-serine after 192 h. The largest concentration of AOB was detected in S1 ($7.6{\times}10^6copies/{\mu}l$), followed by S2 ($3.2{\times}10^6copies/{\mu}l$), S4 ($2.8{\times}10^6copies/{\mu}l$), and S3 ($2.4{\times}10^6copies/{\mu}l$).

Distribution of Epilithic Diatom Communities in Relation to Land-Use and Water Quality in the Geum River System, South Korea (금강수계의 토지이용 및 수질구배에 따른 돌부착 돌말류의 분포특성)

  • To study the distribution characteristics of diatom communities in relation to the gradient of water quality and land-use, water samples and epilithic diatoms were collected from 90 sites including mainstream and tributaries, in the Geum River System (GRS) during no rainfall, May 2011. Of 239 taxa identified, Achnanthes convergens and Cymbella minuta were dominated over all the sites. Cluster analysis on the GRS divided into three groups. Group 1 (G1) is the mountainous upstream, a relatively good water quality, and mainly consisted of saproxenous species. Group 2 (G2) is the urban sites, polluted (high in electric conductivity, total phosphate and soluble reactive phosphorus), and saproxenous and common species. Group 3 (G3) is the agriculture sites, and polluted (turbidity, total nitrogen, nitrate), and saprophilous species. There were some discrepancies between abiotic and biotic variables in GRS; biologically similar between G1 and G2, but abiotically similar between G2 and G3. These differences may attribute to not only physiological characteristics of diatom cells, but complicate relationships between microhabitat and water quality. Thus, an urgent development of generalized or standardized methods to diminish the differences between epilithic diatom community and environments is advent.

Separation of Calcium-binding Protein Derived from Enzymatic Hydrolysates of Cheese Whey Protein

  • This study was carried out to separate the calcium-binding protein derived from enzymatic hydrolysates of cheese whey protein. CWPs (cheese whey protein) heated for 10 min at $100^{\circ}C$ were hydrolyzed by trypsin, papain W-40, protease S, neutrase 1.5 and pepsin, and then properties of hydrolysates, separation of calcium-binding protein and analysis of calcium-binding ability were investigated. The DH (degree of hydrolysis) and NPN (non protein nitrogen) of heated-CWP hydrolysates by commercial enzymes were higher in trypsin than those of other commercial enzymes. In the result of SDS-PAGE (sodium dodecyl sulphate polyacrylamide gel electrophoresis), $\beta$-LG and $\alpha$-LA in trypsin hydrolysates were almost eliminated and the molecular weight of peptides derived from trypsin hydrolysates were smaller than 7 kDa. In the RP-HPLC (reverse phase HPLC) analysis, $\alpha$-LA was mostly eliminated, but $\beta$-LG was not affected by heat treatment and the RP-HPLC patterns of trypsin hydrolysates were similar to those of SDS-PAGE. In ion exchange chromatography, trypsin hydrolysates were shown to peak from 0.25 M NaCl and 0.5 M NaCl, and calcium-binding ability is associated with the large peak, which was eluted at a 0.25 M NaCl gradient concentration. Based on the results of this experiment, heated-CWP hydrolysates by trypsin were shown to have calcium-binding ability.
