석창포에 의한 발기부전 개선 효과 (Effect of Acorus Gramineus on the Relaxation of Corpus Cavernosum Smooth Muscle)

  • The aim of the present study is to investigate whether an ethanol extract of Acorus gramineus Soland (EAG) augments penile erection in vitro and in vivo experiment. Preconstructed with phenylephrine (PE) in isolated endothelium-intact rabbit corpus cavernosum, EAG relaxed penile smooth muscle in a dose-dependent manner, which was inhibited by pretreatment with NG-nitro-L-argininemethylester (L-NAME), a nitricoxide synthase inhibitor, and 1H-[1,2,4]-oxadiazole-[4,3-${\alpha}$]-quinoxalin-1-one (ODQ), a soluble guanylylcyclase (sGC) inhibitor, respectively. EAG-induced relaxation was significantly attenuated by pretreatment with tetraethylammonium (TEA), a nonselective $K^+$ channel blocker. EAG increased cGMP levels of the rabbit corpus cavernosum in a concentration-dependent manner without changes in cAMP levels. In addition, EAG caused increase of peak intracavernous pressure (ICP), ICP/MAP ratio and area under the carve (AUC) in SD rats. Taken together, these results suggest that EAG augments penile erection via NO-cGMP system and $K^+$ channels in corpus cavernosum.

해수산 클로렐라(Chlorella ellipsoidea) 유기용매 추출물의 항염증 효과 (Anti-Inflammatory Effect of Chlorella ellipsoidea Extracted from Seawater by Organic Solvents)

  • Chlorella has been reported to have certain beneficial physiological effects, including hypocholesterolemic, antihypertensive, antioxidative, and anti-tumor activities in animal and human studies. The aim of this study was to determine the anti-inflammatory activities of an 80% methanol extract(CE-Met), hexane fraction (CE-Hex), and ethyl acetate fraction (CE-EA) of Chlorella ellipsoidea in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage. Treatment with various concentrations of the C. ellipsoidea extract resulted m a significant, dose-dependent reduction in nitric oxide (NO) production by LPS-induced macrophages. The C. ellipsoidea extract significantly inhibited LPS-induced NO production accompanied by an attenuation of IL-6 and TNF-$\alpha$ formation in macrophages. These results suggest potent inhibit이y effects on the production of inflammatory mediators by a C. ellipsoidea extract. Thus, C. ellipsoidea extract may be a potent anti-inflammatory agent for troubled skin.

Neuroprotective Effects of a Butanol Fraction of Rosa hybrida Petals in a Middle Cerebral Artery Occlusion Model

  • The neuroprotective effects of a butanol fraction of white rose petal extract (WRPE-BF) were investigated in a middle cerebral artery occlusion (MCAO) model. Seven week-old male rats were orally administered WRPE-BF for 2 weeks and subjected to MCAO for 2 h, followed by reperfusion. Twenty-four h later, MCAO-induced behavioral dysfunctions were markedly improved in a dose-dependent manner by pretreatment with WRPE-BF. Moreover, higher dose of WRPE-BF not only decreased infarction area but also effectively reduced astrogliosis. The expression of inducible nitric oxide synthase, cyclooxygenase-2, and glial fibrillary acidic protein in MCAO model were markedly inhibited by WRPE-BF treatment. Notably, WRPE-BF decreased nitricoxide and malondialdehyde levels in the striatum and subventricular zone of stroke-challenged brains. These data suggested that WRPE-BF may exert its neuroprotective effects via anti-oxidative and anti-inflammatory activities against ischemia-reperfusion brain injury and could be a good candidate as a therapeutic target for ischemic stroke.

  • Objectives: Three yin-tonifying formulae (Ssanghwa-tang, Yukmijihwang-tang and Jaeumganghwa-tang) were applied to investigate their immunological activities on antigen (Ag)-specific or Ag-non-specific immune responses in the murine macrophage cell line (RAW 264.7) and in ovalbumin (OVA)-immunized mice. Methods: This study was carried out in nitricoxide (NO) synthesis in RAW 264.7 cells and cellular proliferation in mouse splenocytes in association with three herbal formulas. C57BL/6 mice were immunized intraperitoneally with OVA/aluminum ($100\;{\mu}g/200\;{\mu}g$/mouse) on days 1, 8, and 15. Three herbal formulas were administrated to mice orally for 3 weeks from day 1. On day 22, OVA-, lipopolysaccharide (LPS)-, and concanavalin A (Con A)-stimulated splenocyte proliferation and antibodies (OVA-specific antibodies of the IgG, IgG1, and total IgM classes) in plasma were measured. Results: All three yin-tonifying formulas significantly enhanced cellular proliferation by LPS and Con A in splenocytes from OVA-immunized mice (p<0.001). Also, these herbal formulas all significantly enhanced plasma OVA-specific IgG, IgG1, and total IgM levels compared with the OVA/Alum group. Conclusion: These results suggested that the three yin-tonifying formulae could be used as stimulators of immune response.

  • 12주간의 저강도 트레드밀 운동은 본태성 고혈압 쥐의 안정시 심박수와 혈압, LPOA와 호모시스테인 수준의 개선과 함께 심장근의 eNOS mRNA 및 골격근의 sNOS 단백질 발현량을 증가시킨 반면 심근의 ET-1 mRNA 수준을 감소시키는 것으로 나타났다. 이러한 결과들은 결국 운동이 혈압조절 뿐만 아니라 고혈압에 의한 심근비대현상 관련 유전자들의 기능개선을 가져와 고혈압을 개선시키는 작용을 한다는 것을 확인 할 수 있었다.

감마선 조사에 의한 LPS의 면역 독성 저감화 (Reduction of Immune Toxicity of LPS by Gamma Irradiation)

  • The purpose of this study is to reduce the immune toxicity of lipopolysaccharide (LPS) by gamma irradiation. LPS was gamma-irradiated at the various doses of 20, 100 and 200 kGy and then evaluated on the immune toxicity through the cell proliferation, nitricoxide production and cytokine release. Cell proliferation significantly decreased in the intact LPS treated groups, whereas gamma-irradiated LPS treated group were not reduced the cell proliferation. Similarly, the production of nitric oxide and cytokine showed the high levels in the intact LPS treated group. However, gamma-irradiated LPS treated group remarkably decreased the production of nitric oxide and cytokine in dose-dependent manner. Therefore, gamma irradiation may effective method to reduce the immune toxicity of LPS.

Anti-inflammatory Activity of Complex Saponin Separated from Pueraria flos and Adzuki Beans Mixture Extract

  • Pueraria flos and Adzuki Beans contains several bioactive compounds, such as saponin, oleanolic acid, and flavone. Pueraria flos and Adzuki Beans has traditionally been used to treat disorders of antioxidant activity, diabetes and liver detoxication, and it has antinociceptive and anti-inflammatory properties. However, complex saponin were validity of the anti-inflammatory activity has not been scientifically investigated. In this study, to determine anti-inflammatory activity of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract on nitric oxide and prostaglandinE2 assay. The anti-inflammatory activities of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract were evaluated for inhibitory activities against lipopolysacchride induced nitric oxide and prostaglandinE2 production protein expressions in RAW264.7 cell lines. The complex saponin separated from Pueraria flos and Adzuki Beans mixture extract inhibitory activity for both tests with protein high depressions(%) values showed in the ranges of 50~100 ㎍/ml. Overall, prostaglandinE2 tests had a higher inhibitory effect on inflammation than nitricoxide tests. Theseis result suggest a potential role of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract as source of anti-inflammation agent.

생쥐 시각피질에서 melanopsin을 가지는 신경세포의 면역조직화학적 위치 (Immunocytochemical Localization of Melanopsin-immunoreactive Neurons in the Mouse Visual Cortex)

  • Melanopsin은 옵신과 비슷한 광색소로 망막 광민감성 신경절 세포에서 적은 비율로 발견 된다. Melanopsin은 일주기 리듬조절에 관여 하고 동공반사를 조절한다. 이번 연구의 목적은 melanopsin이 망막 이외의 중추 시각계 에서도 발견 되는지를 알아 보는 것이다. 우리는 생쥐 시각 피질에 존재하는 melanopsin을 가지는 신경세포를 면역조직화학법을 통해 염색하여 melanopsin의 분포와 형태를 분석하였다. Melanopsin 면역반응은 핵을 제외한 신경세포의 세포체에서 일어났다. 우리는 melanopsin이 성체 생쥐의 시각 피질의 2,3층에서 높은 밀도로 모여서 존재하는 것을 확인했다. 시각 피질 1층에서 melanopsin 면역반응을 보인 신경세포는 드물게 발견 되었다. 생쥐 시각 피질에서 발견된 melanopsin의 세포종류는 주로 round/oval 세포였으나, vertical fusiform 그리고 pyramidal 세포 등도 드물게 발견 되었다. 염색된 세포들의 분포는 특이적이지 않았다. 우리의 실험 데이터로 melanopsin이 시각피질에도 분포한다는 것을 알 수 있었다. 또한, melanopsin을 가지는 신경세포는 nitric oxide synthase, calbindin and parvalbumin과도 같이 발현 되는 것을 관찰했다. 이러한 실험결과는 망막 이외의 부분에서 발견되는 melanopsin의 기능에 대한 연구가 필요함을 야기한다.

감태 효소 추출물 및 폴리페놀 추출물의 생리활성에 관한 연구 (Biological Potential of Enzymatic and Polyphenol Extracts from Ecklonia cava)

  • 본 연구에서는 감태 효소 추출물과 그것의 폴리페놀 추출물의 화장품 원료로서의 효능을 알아보기 위하여 항산화, 항당화, 미백, 항염 효과와 관련된 실험을 실시하였다. 감태 효소 추출물과 폴리페놀 추출물은 강력한 라디컬 소거능을 가지고 있으며 BSA/Glucose 시스템에서 최종당화생성물의 형성을 저해하는 항당화 활성과 타이로시네이즈 저해를 통한 우수한 미백력을 가지고 있음을 확인하였다. 또한 두 추출물 모두 세포 내에서 $PGE_2$와 NO 생성 저해를 통한 항염 효과를 나타내었다. 이러한 결과를 종합해 볼 때, 감태 효소 추출물과 그 폴리페놀 추출물은 화장품 원료로서의 응용 가능성이 있을 것으로 사료된다.
