Transgenic Rice Expressing Snowdrop Lectin (Galanthus nivalis agglutinin; GNA) Shows Resistance to Rice Brown Planthopper (Nilaparvata lugens Stål) (Snowdrop lectin (Galanthus nivalis agglutinin: GNA) 유전자 도입에 의한 벼멸구 저항성 형질전환 벼 개발)

  • Transgenic rice plants with increased resistance to rice brown planthopper (Nilaparvata lugens St${\aa}$l) were generated by particle bombardment-mediated transformation of plants with a gene encoding snowdrop lectin (Galanthus nivalis agglutinin; GNA) under control of the rice Rubisco small subunit (rbcS) promoter.. A large number of transgenic rice plants containing the GNA gene were generated. The integration, expression, and inheritance of this gene in the $R_1$ and $R_2$ generations were demonstrated by Southern and western blot analyses. The plants contained one to five copies of the transgene. The GNA protein comprised approximately 0.01-2.0% of total soluble protein in the $R_1$ and $R_2$ transgenic plants. Insect bioassays and feeding studies showed that the GNA protein expressed in the $R_2$ transgenic rice plants reduced the survival of brown planthoppers. The introduction of GNA into rice plants therefore can help to control insect pests.

  • The brown planthopper, Nilaparvata lugens, is among the most serious insect pests of rice. It is widely distributed in Asia, Australia and Pacific islands. An earlier mitochondrial DNA study revealed that there exist significant genetic differences between populations north and south of the Red River Delta region in Vietnam. However the mitochondrial DNA was not sufficiently variable to examine the sources of immigration. For a more detailed analysis of geographic population structure of N. lugens, we developed microsatellite markers. Thirty-seven putative microsatellite loci were isolated using a magnetic biotin method, and five primer pairs designed from the flanking regions of sequenced microsatellite clones were labeled with fluorescent. Of these five primer sets, two have proven to be useful across all the samples we used in this study. We used variation at these two microsatellite loci to test the hypothesis that N. lugens biotypes (1, 2, and 3) sampled from laboratory selection constituted distinct genetic units. Allele frequency differences among the three major biotype categories were not significantly different at one locus (27035). However, the other (7314) did show differences among the major three biotypes. The methods we describe here will be useful for studying population structure of crop pest and for tracking the patterns of migratory pest like the rice planthoppers.

Studies on the Distribution of the Brown Planthopper(Nilaparvata lugens St${\aa}$l)Biotypes Migrated in the Southern Regions of Korea (경남, 전남 남부지방에 비래된 벼멸구 생태형 분포에 관한 연구)

  • Biotypes of the brown planthopper(BPH), Nilaparvata lugens St l, population migrated from maninand China in to southern coastal regions of Korea were investigated in 1985-1987 by honeydew excretion test using Korean cultvars,, 'Milyang 23'(no resistant gene), 'Cheongcheo-ngboy'(bph 1 gene), 'Milyang 63'(bph 2 gene) and 'Gayabyeo'(bph 1 and bph 2 gene). There was a correlation between the amount of honeydew excreted and the ability of each biotype to infest resistant cultivars: 'Milyang 23' with no resistant gene was highiy susceptible to bioty-pe-1, 2 and 3 ; 'cheongcheongbyeo' with resistant bph 1 was susceptible to biotype-2: and 'Milyang 63' with resistant bph 2 gene was highiy resistant to biotype-1, 2 and 3. Percent biotype dis-tribution of the female BPH collected in the southern coastal regions of Korea showed that in 1985, 1986 and 1987, the BPH biotype-1 was predominant accounting for 64.7%, 61.2% and 57.9%, respectivley, whereas bitype-2 was 22.0%, 21.9% and 29.7%, and biotype-3 was 13.3%, 17.0% and 12.6%, repestively.

  • These experiments were performed to investigate the effects of sublethal doses of fungicides (validamycin A 3 Sp and neoasozin 6.5 Lq) and insecticides (diazinon 34Ec and decamethrin 1Ec) on the possible ways to the resurgence of brown planthoppers (BPH) (Nilaparvata lugens $St{\aa}l$). The possible factors tested in this experiment were fecundity of BPH, longevity, egg-hatchability, amount of honeydew excreted, population build-up following the pesticide treatments in the next generation, and hopperburn rate of potted rice plants. Among the insecticides tested, the applications of diazinon and decamethrin in rice fields may induce the BPH resurgence and accelerate the hopperburn of rice plants through stimulating the fecundity, increasing the adult longevity, promoting the feeding activity and causing the positve population build-up of the BPH

Impact of Pesticide Treatment on an Arthropod Community in the Korean Rice Ecosystem

  • An arthropod community in a rice ecosystem was surveyed to determine the impact of two insecticides frequently used in Korean rice ecosystems: carbofuran 3GR, which targets the rice water weevil, Lissorhoptrus oryzophilus Kuschel (Coleoptera: Curculionidae) in the early season and fenobucarb EC, which targets the brown planthopper, Nilaparvata lugens ($St{\aa}l$) (Hemiptera: Delphacidae) in the mid- and late seasons, respectively. Overall, the application of the insecticides reduced density of total arthropods by 48.4% compared to the untreated field, but their impact on each functional group were different. Carbofuran GR treatment on 1 June reduced the L. oryzophilus population significantly until mid-season. The population of filterfeeding chironomids was also reduced by 50%, whereas the spider population was less disturbed. Fenobucarb EC treatment on 16 August significantly reduced N. lugens and detrivorous entomobryid populations until the late season. Both web-building and wandering spiders were also significantly disturbed by fenobucarb EC although the impact differed according to their behavioral differences. While the population of web-building spiders significantly decreased over time, that of wandering spiders recovered from the disturbance a few weeks later.

Life Table Analysis of the Brown Planthopper, Nilaparvata lugens Stål (Hemiptera: Delphacidae) on Rice of Resistant Cultivars (벼멸구 저항성 품종벼에서 벼멸구의 생명표 분석)

  • Development, survival, and reproduction of brown planthopper (BPH) Nilaparvata lugens $St{\aa}l$ (Hemiptera: Delphacidae), were studied in laboratory at $25{\pm}2^{\circ}C$, $65{\pm}5%$ RH and a 16L : 8D hours photoperiodism on five rice cultivars of: Dongjin 1ho, Chungchungbyeo, Jangseongbyeo, Chinnongbyeo and Jungmo 1045. BPH nymphs successfully survived on all rice cultivars, although survival rate was lowest on Jangseongbyeo (36.0%). Developmental time of immature stages ranged from $11.7{\pm}0.59d$ on Jungmo 1045 to $12.8{\pm}0.59d$ on Chinnongbyeo. Reproductive period and female longevity were longest on Dongjin 1ho, Chinnongbyeo and Jungmo 1045 while highest fecundity of N. lugens being observed on these three rice cultivars. Highest and lowest net reproductive rates were calculated on rice cultivars, Jungmo 1045 and Jangseongbyeo, respectively. Mean generation time was the longest on rice cultivar Dongjin 1ho. Respective descending order of intrinsic rates of population increase were on Jungmo 1045, Chinnongbyeo, Dongjin 1ho, Chungchungbyeo and Jangseongbyeo. These population parameters showed that N. lugens can successfully survive and reproduce on Chinnongbyeo and Jungmo 1045.

Effects of Sublethal Doses of Imidacloprid on the Brown Planthopper, Nilaparvata lugens $St{\aa}l$ (Homoptera: Delphacidae) (Imidacloprid의 아치사량이 벼멸구(Nilaparvata lugens $St{\aa}l$)의 생물적 특성에 미치는 영향)

  • Effects of sublethal dose of imidacloprid on biological characteristics, such as longevity, fecundity and excretions from brown planthopper (BPH) and its repellency to imidacloprid were examined. Topical application of imidacloprid to BPH at the dose of $LD_{10}$ and $LD_{30}$ (These values were determined at 48 hours after topical application of imidacloprid) was remarkably increased the mortality from 3 days after application and showed over 90% mortality on 4 days after application. Topical application of imidacloprid at dose of $LD_{10}$ (16 days) showed no difference compared to the control (16.9 days) in the longevity, while application of $LD_{30}$ was reduced the longevity of BPH to 13.5 days. Application of sublethal dose of imidacloprid to BPH adults reduced the number of eggs laid per day and for adult stage, and the hatchability of their eggs. By root zone application at the dose of $LC_{30}$, 50% longevity and 40% fecundity was reduced respectively, but showed no effect on number of eggs laid per day. The day on which BPH laid 50 % of eggs in number were 7.6 days at the $LC_{10}$ and 5.9 days the of $LC_{30}$ respectively. Hatchabilities of the groups treated with imidacloprid were lower than that of the control. Topical application to BPH at the dose of $LD_{10}$ did not reduce the amount of excretes, but application at the dose of $LD_{30}$ was reduced 26.5% of excretion compared to the control. Leaf dipping into the solution of concentration of $LC_{10}$ and $LC_{30}$ was reduced 96.4% and 98.0% of excretion, respectively. And root zone treatment at the concentration of $LC_{10}$ and $LC_{30}$ reduced 79.5% and 96.6% of excretion, respectively. Repellency rates of BPH to imidacloprid were 20% regardless of the dose of application.

Studies on the Mechanism of Varietal Resistance of Rice to the Brown Planthopper(Nilaparvata lugens $ST{\AA}L$ (벼멸구에 대한 수도품종(水稻品種)의 저항성기작(抵抗性機作)에 관(關)한 연구(硏究))

  • This paper was performed to study the nature of varietal resistance of some Korean-new rice cultivars to the brown planthopper (BPH), Nilaparvata lugens ($ST{\AA}L$). The rice cultivars tested were Cheongcheong, Gaya, Hangangchal, Samgang, Nampoong and Yeongpoong which have been reported as having resistant genes for the BPH. The check varieties were Jinheung, Sangpoong and Chucheong for susceptible and IR-36 for resistant. The factors studied were referred to the seedling responses, preference in feeding and oviposition of BPH, antibiosis (nymphal development, adult emergence and sex ratio, adult body weight, population build-up, feeding amount and amylase activity), and chemical composition (inorganic components, chlorophyll contents, cell wall components, amino acids and esterase isozymes) of leaf- sheath and/or roots of rice plants. In conclusion, the natures of varietal resistance of rice cultivars to the BPH were not only correlated with the resistant reaction of rice plant, but also they were related with the non preference in feeding and oviposition and those resistant cultivars had the antibiotic effects to the insects. Their antibiotic effects of rice cultivars to the BPH would be related with some of the chemical components of rice plants, such as the contents of magnesium oxide (MgO), and chlorophyll and the different esterase isozymes.

Preference, Development and Fecundity of the Brown Planthopper(Nilaparvata lugens St${\aa}$l) Biotypes Fed on Different Cultivars of Rice with Various Resistance Gence (벼의 품종저항성이 벼멸구 각 생태형의 선호성, 발육 및 증식능력에 미치는 영향)

  • Some attempts were made to investigate the biological characteristics of the brown planthopper(BPH), Nilaparvata lugens St${\aa}$l, biotypes in terms of perference in feeding oviposition on rice field, egg and nymphal periods, egg hatchability, emergemce ratio, growth index, adult lingevity and gecundity fed on 60-day-old rece cultivaes with different resistance genes. Feeding and oviposition perference of the three BPH biotypes on Dongjinbyeo with no resistance gene were shown very higher than on rice cultivars with resistance genes. Those of biotype-2 on Cheongcheongbyeo with bph 1 gene and bitype-3 on Milyang 63 with bph 2 gene were relatively high, however, they were still remarkably lower than those of Dongjibyeo. The egg and nymphal periods of the three BPH biotyes on Milyang 23 were shorter than on the other rice cultivars. The periods of biotype-2 on Cheongcheongbyeo and biotype-3 on Milyang 63 were as short as those of the three BPH biotypes on Milyang 23. The egg hachability, emergence ratio, and growth index of the three BPH bioyoes on Milyang 23 were higher than on the other rice cultivars and those of biotype-2 on Cheongcheongbyeo and biotyoe-3 on Milyang 63 were as high as on Milyang 23. The female adult longevity of the BPH biotype-1 on Milyang 23 was longer than on the other cultivaes, and that of biotype-2 on Cheongcheongbyeo and bioty-3 an Milyang 63 were as ling as on Milyang 23. The number of egg laid by a female of the three BPH biotypes were greatly increased on Milyang 23. Those of biotype-2 on Cheongcheongbyeo and biotype-3 an Milyang 63 were also greatly increased.
