Determination of Aloesin in Plasma by High-Performance Liquid Chromatography as Fluorescent 9-Anthroyl Derivative

  • A sensitive high-performance liquid chromatographic (HPLC) method for the determination of aloesin in plasma was developed. After solid-phase extraction from plasma and derivatization of aloesin and compound AD-1, which was prepared from aloesin as a internal standard, with 9-anthroylnitrile in the presence of quinuclidine, the derivatives were separated on a Inertsil ODS-3 column using acetonitrile/methanol/water (3:1:6) as a mobile phase, and detected fluorimetrically at 460nm with excitation at 360nm. The detection limit of aloesin was 3.2ng/ml in plasma (S/N=3).

  • A stereospecific HPLC method has been developed for the resolution of the enantiomers of salbutamol in human urine. After solid-phase extraction and derivatization with (S)-(-)-${\alpha}$-methyl-benzyl isocyanate, the diastereomeric derivatives were resolved $(R_s=1.59)$ on $5{\mu}M$ octadecylsilan column using 47% methanol as a mobile phase with fluorescence detection. The detection limit of each enantiomer was 10 ng/ml (S/N=3).

  • A study was undertaken to determine the incidence of short oestrus cycles and associated serum progesterone levels in Nili-Ravi buffaloes. Forty lactating buffaloes were kept under observation for oestrus activity for a period of sixty days. Blood samples from jugular vein from twenty buffaloes were used to determine associated serum progesterone levels during short oestrus cycles. Incidence of short oestrus cycles was 15.5 percent (Nine out of fifty eight oestrus cycles) and most of the short oestrus cycles occurred between first and second oestrus after calving. Lengths of short oestrus cycles ranged from 6 to 14 days. The most frequent length was 11 days. Serum progesterone levels were lower at oestrus and increased gradually attaining peak levels of 1.3 and 1.8 ng/ml on 7th day of 9 and 11 day short oestrus cycles. One buffalos exhibiting a short oestrus cycle of 12 days conceived which indicated that ovulation took place at the oestrus of short cycle.

  • In this work energy and entransy characteristics of heat exchangers are analyzed for 12 different flow arrangements of heat exchangers. The dimensionless parameters are number of entransy dissipation (Ng), number of entransy dissipation-based thermal resistance (Nr), and entransy dissipation-based effectiveness of heat-exchanger (εg). The dimensionless parameters are expressed analytically in terms of the effectiveness of heat exchanger (ε), heat capacity ratio (c), and number of transfer unit (N) for optimal performance of heat exchangers. Results showed that the dimensionless parameters based on the entransy dissipation can be useful concepts for optimal design of heat exchangers.

Sewage disposal system management policy in Korea (우리나라의 하수도사업현황 및 시책방향)

  • We had passed through a serious water pollution with a rapid urbanization and industrialization in the 1960's. Seoul Chunggyechun Sewage Disposal System set uP in 1966 and finished in 1976, as the first sewage disposal system in Korea, had covered only 4 percentage of the sewage disposal system per capita for 10 years. Through holding the Asian Games in 1986 and the Olympic Games in 1988, we expanded the sewage disposal system so that it became increased 18 percentage of the population in 1986. Finally, we have installed about 60 sewage disposal system by 1994 for a large environmental investment which was critically caused by "Phenol Accident in Nakdong River" . Now, the sewage disposal system per capita covers 42 percentage and the activity for water quality improvement is going on rapidly. The method of sewage disposal is mainly "activated sludge process" . However, the technical ability for the sewage disposal has largely developed since 1991 so the "extended aeration process" is used in Po$\v{u}$n, Tangyang, Mun$\v{u}$i, "rotating biological contact process" in Onch$\v{u}$ng, Pukok, and "oxidation ditch process" in K$\v{o}$ch'ana.

Determination of DL-Carnitine Hydrochloride in Pharmaceutical Preparation by HPLC using UV Absorption Derivatives (자외부 흡광 유도체를 이용한 염산 DL-카르니틴의 분석)

  • A reverse-phase HPLC method to determine DL-Carnitine Hydrochloride in pharmaceutical preparation is described. UV absorption derivatives of DL-Carnitine Hydrochloride were formed with p-Bromophenacyl Bromide in an essentially quantitative manner using crown ether as catalyst. The DL-Carnitine-bromophenacyl ester absorbed UV radiation strongly at 254nm, allowing the detection of as small a quantity as 12.5ng of DL-Carnitine Hydrochloride. A linear defection range was $5\;{\times}\;10-8 \;{\sim}\;5\;{\times}\;10-7M$ of DL-Carnitine Hydrochloride. And the linear regression at various drug concentration was =0.999 (n=10). The DL-Carnitine Hydrochloride in pharmaceutical preparation was successfully derivatized and separated from other constituents by reverse phase HPLC with detection at 254 nm.

  • Analytical method of benzophenone (BP) in sediment and soil was developed by gas chromatography/mass selective detector/selected ion monitoring (GC/MSD/SIM). The ultrasonic extraction of US EPA (method 3550B) method and liquid-liquid extraction for sediment and soil samples were used for the analysis of BP from sediment and soil. BP was extracted with n-hexane. Organic layer was washed with 5% sodium chloride solution. 1∼2 l of the concentrated solution of organic layer was applied to GC/MSD. The retention time of BP peak was 11.10 min. Recovery (%) of BP by ultrasonication from sediment and soil samples was 96.0∼100.6% and 40.0∼83.0%, respectively. Recovery of BP by liquid-liquid extraction was 51∼59% in soil samples. The detection limit of BP in sediment and soil samples were determined to 0.1 ng/g.

  • This report presents a voltammetric assay of dinitrotoluene using a DNA immobilized onto a carbon nanotube paste electrode (PE). The cyclic voltammetry (CV) and square wave (SW) stripping voltammetry parameters of the optimized conditions were obtained. An anodic peak current appeared at 0.3 V (versus Ag/AgCl) in a 0.1-M $NH_4H_2PO_4$ electrolyte solution. The detection limit was found to be $0.6ngL^{-1}$(S/N = 10), within a deposition time of 100 sec.

Electrochemical Detection of Trace Level Copper in in vivo Cell (생체 세포내 미량 구리의 전기화학적 검출에 관한 연구)

  • In order to measure the minute amount of Cu(II) in our environment, cyclic voltammetry (CV) and square-wave stripping voltammetry (SWSV) were performed for a trace copper assay using bismuth immobilized on a carbon nanotube paste electrode. An analytical working range of 30 to $240{\mu}g/L$ Cu(II) was obtained for CV and SWSV. The SWSV precision obtained was 0.47 % (n = 15) RSD in $30.0{\mu}g/L$ Cu(II). The detection limit obtained was 3.1 ng/L Cu(II) using SWSV, while the CV yielded the nano-range detection limit through the pre-concentration step. By using this research method, Cu(II) value could be determined in the urine of human sample and in the brain of fish sample. This research can be effectively applied to other cases of measuring minute amount of Cu(II) in living organisms.

  • Ocher block has started to be studied recently as a new environment friendly alternative building material. This article investigates the use of Rice Husk Ask (RHA) as an additive to improve properties of the ocher block. Ocher mixtures incorporating various proportions of RHA were compared. Compressive strength at various ages of these ocher blocks was evaluated ad the resistance to wet environment was also examined. These test result help to determine the effect of RHA on such properties of the ocher block as compaction, mix proportion, compressive strength and water absorption indexes. Based on these results, the new alternative building material and low cost construction techniques might be developed through more intensive research efforts.
