각 돈피추출물의 수율과 단백질함량은 고분자 PS 처리구에서 높은 함량을 나타내었으며, 특히 단백질 함량은 저분자 LPS 처리구에 비해 유의적으로 높았고, 약 10배 정도의 높은 단백질 함량을 나타내었다(p<0.05). 항산화활성 측정결과 처리 농도가 증가할수록 높은 항산화 활성을 나타내었으며(p<0.05), 특히 고분자 PS 처리구에 비해 저분자 LPS 처리구에서 유의적으로 높은 효과를 나타내었다. ORAC 활성은 LPS 농도 1 mg/mL일 때, $141.39{\mu}M$ TE/g의 높은 활성을 나타내었다. 각 돈피추출물을 신경모세포종 SH-SY5Y 세포에 고농도로 처리한 결과 세포에 독성을 나타내지 않았다. 신경세포에 과산화수소를 처리하여 유발시킨 산화적 스트레스에 대한 PS와 LPS의 신경세포 보호효과를 확인한 결과, 모든 처리구에서 농도 의존적으로 세포 보호효과를 나타내었다. 특히 저분자인 LPS 처리구 농도 $100{\mu}g/mL$일 때, 86.45%의 세포생존율을 보였으며 $H_2O_2$ 대비 29.98%의 신경세포보호효과를 나타내었다. 독성 단백질인 $A{\beta}_{1-42}$를 처리하여 신경세포 보호효과를 확인한 결과, 고분자 PS 처리구보다는 저분자인 LPS 처리구 농도 $100{\mu}g/mL$일 때, 82.01%의 생존율을 보였으며 $A{\beta}_{1-42}$ 대비 14.38%의 신경세포보호효과를 나타내었다(p<0.05). AChE 저해효과를 확인해 본 결과, 고분자 PS 처리구에서는 거의 효과가 나타나지 않았으나, 저분자인 LPS 처리구에서는 농도 의존적으로 증가하는 경향을 나타내었으며, 농도 100 mg/mL일 때 33.62%의 높은 저해 효과를 나타내었다. 따라서 본 연구결과 돈피에서 분리한 3 kDa 이하의 저분자 효소분해물인 LPS는 높은 항산화 활성을 나타내었고, $H_2O_2$와 $A{\beta}_{1-42}$로 유발시킨 산화스트레스에 대한 신경세포 보호효과 및 AChE 저해 효과를 나타내어 향후 항산화 활성 및 신경세포보호를 위한 축산식품 소재로 이용 가능성이 있을 것으로 판단된다.
The present study was aimed to explore the neuroprotective role of imatinib in global ischemia-reperfusion-induced cerebral injury along with possible mechanisms. Global ischemia was induced in mice by bilateral carotid artery occlusion for 20 min, which was followed by reperfusion for 24 h by restoring the blood flow to the brain. The extent of cerebral injury was assessed after 24 h of global ischemia by measuring the locomotor activity (actophotometer test), motor coordination (inclined beam walking test), neurological severity score, learning and memory (object recognition test) and cerebral infarction (triphenyl tetrazolium chloride stain). Ischemia-reperfusion injury produced significant cerebral infarction, impaired the behavioral parameters and decreased the expression of connexin 43 and phosphorylated signal transducer and activator of transcription 3 (p-STAT3) in the brain. A single dose administration of imatinib (20 and 40 mg/kg) attenuated ischemia-reperfusion-induced behavioral deficits and the extent of cerebral infarction along with the restoration of connexin 43 and p-STAT3 levels. However, administration of AG490, a selective Janus-activated kinase 2 (JAK2)/STAT3 inhibitor, abolished the neuroprotective actions of imatinib and decreased the expression of connexin 43 and p-STAT3. It is concluded that imatinib has the potential of attenuating global ischemia-reperfusion-induced cerebral injury, which may be possibly attributed to activation of JAK2/STAT3 signaling pathway along with the increase in the expression of connexin 43.
Formation of ${\beta}$-amyloid $(A{\beta})$ fibrils has been identified as one of the major characteristics of Alzheimer's disease (AD). Inhibition of $A{\beta}$ fibril formation in the CNS would be attractive therapeutic targets for the treatment of AD. Several small compounds that inhibit amyloid formation or amyloid neurotoxicity in vitro have been known. Citrate has surfactant function effect because of its molecular structure having high anionic charge density, in addition to the well-known antibacterial and antioxidant properties. Therefore, we hypothesized that citrate might have the inhibitory effect against $A{\beta}$ fibril formation in vitro and have the protective effect against $A{\beta}$-induced neurotoxicity in PC12 cells. We examined the effect of citrate against the formation of $A{\beta}$ fibrils by measuring the intensity of fluorescence in thioflavin-T (Th-T) assay of between $A{\beta}_{25-35}$ groups treated with citrate and the control with $A{\beta}_{25-35}$ alone. The neuroprotective effect of citrate against $A{\beta}$-induced toxicity in PC12 cells was investigated using the WST-1 assay. Fluorescence spectroscopy showed that citrate inhibited dose-dependently the formation of $A{\beta}$ fibrils from ${\beta}$-amyloid peptides. The inhibition percentages of $A{\beta}$ fibril formation by citrate (1, 2.5, and 5 mM) were 31%, 60%, and 68% at 7 days, respectively in thioflavin-T (Th-T) assay. WST-1 assay revealed that the toxic effect of $A{\beta}_{25-35}$ was reduced, in a dose-dependent manner to citrate. The percentages of neuroprotection by citrate (1, 2.5, and 5 mM) against $A{\beta}-induced$ toxicity were 19%, 31 %, and 34%, respectively. We report that citrate inhibits the formation of $A{\beta}$ fibrils in vitro and has neuroprotective effect against $A{\beta}$-induced toxicity in PC12 cells. Neuroprotective effects of citrate against $A{\beta}$ might be, to some extent, attributable to its inhibition of $A{\beta}$ fibril formation. Although the mechanism of anti-amyloidogenic activity is not clear, the possible mechanism is that citrate might have two effects, salting-in and surfactant effects. These results suggest that citrate could be of potential therapeutic value in Alzheimer's disease.
본 연구는 꾸지뽕 열매 조다당류 분획물(CTP)의 산화방지 활성 및 신경세포 보호 효과에 관하여 알아보기 위하여, 항산화 활성인 라디칼 제거능 및 환원력을 평가한 결과, 꾸지뽕 열매 조다당류의 농도가 증가할수록 산화방지 활성이 유의적으로 증가하는 것으로 나타났다. 또한, 이러한 꾸지뽕 열매 조다당류의 뇌신경세포 보호 효과에 관하여 알아보기 위하여 생쥐의 해마 유래 뇌신경세포에 꾸지뽕 열매 조다당류를 처리한 후 과산화수소로 산화적인 스트레스를 유도하여 세포 독성에 관하여 알아본 결과, 꾸지뽕 열매 조다당류의 처리는 농도 의존적으로 뇌신경 세포의 생존율을 증가시켰으며, 이에 따라 산화방지 효소인 SOD 활성이 증가하고 지방질 과산화 생성물인 MDA level이 감소한 것을 알 수 있었다. 이상의 결과들로 꾸지뽕 열매 조다당류의 산화방지 활성 및 뇌신경세포 보호 효과에 관하여 확인할 수 있었으며, 추후 어떤 메커니즘으로 신경세포를 보호하는지 추가적인 연구가 필요할 것으로 생각한다. 꾸지뽕 열매 조다당류 분획물은 산화방지 및 산화적 스트레스로부터 신경세포 보호효과를 나타내어 퇴행성 신경질환 예방에 유용한 건강기능성 식품 소재가 될 것으로 사료된다.
Objectives : The purpose of the present study is to observe the neuroprotective effect of the $NeuBo153^{\circledR}$ on transient focal cerebral ischemia in rats. Methods : $NeuBo153^{\circledR}$ was made by mixing the herbs, mainly the root of Panax ginseng, the root of Rehmannia glutinosa and Poria cocos, the stem bark of Acanthopanax senticosus, the root of Scutellaria baicalensis and Mel, and heating for 96 hours. Transient Focal cerebral ischemia (2 h of ischemia, 22 h of reperfusion) was induced by intraluminal suture method with SD rats. Sensory motor function was tested by rotarod test, prehensile traction test, beam balance test and foot fault test at 24 h after ischemia. The brain slices were stained by 2% 2, 3, 5-triphenyltetrazolium chloride and the infarct volume was measured by graphic analyzer at 24 h after ischemia. Results : $NeuBo153^{\circledR}$ treated group did not show significant differences compared with vehicle treated group in body temperature. Oral administration of $NeuBo153^{\circledR}$ reduced brain infarct volume by 29.7% compared with vehicle treated group. $NeuBo153^{\circledR}$ also showed protective effects on sensory motor functional deficits. Conclusion : $NeuBo153^{\circledR}$ treatment reduced brain damage and improved functional deficits induced by MCAo. It showed neuroprotective effects even when treatment was relayed 2 h after injury. Further research is required to evaluating long term functional recovery am accurate therapeutic range and mechanisms.
Objectives: This experiment was designed to investigate the effect of the InSamYangYoung-tang(Renshenyangrongtang) extract on $A{\beta}$-induced AD model. Methods: The effects of the InSamYangYoung-tang(Renshenyangrongtang) extract on neural damages of cultured PC12 cells induced by $A{\beta}$ were investigated. The effects of the InSamYangYoung-tang(Renshenyangrongtang) extract on neural damages of hippocampal and cortical neurons in the mouse induced by $\beta$-amyloid were investigated. Results: 1. $A{\beta}$ treatment into neuronal cells activated cell death pathway when analyzed by MTT assay and by histological analysis. Then InSamYangYoung-tang(Renshenyangrongtang) treatment improved cell survival to a similar level as in normal group. 2. $A{\beta}$ treatment increased caspase 3 protein levels but decreased phospho-Erk1/2 in neuronal cells. InSamYangYoung-tang(Renshenyangrongtang) treatment reversed the production levels of two proteins close to those in normal group. 3. $A{\beta}$ treatment induced the atrophy of neuronal cells in terms of neuronal processes and cell body shrinkage, but InSamYangYoung-tang(Renshenyangrongtang) greatly improved their morphology. 4. Neuroprotective activity, as observed in InSamYangYoung-tang(Renshenyangrongtang)-treated groups, was similarly observed in cells treated with galantamine which was used as a positive control. Moreover, overall recovery pattern by InSamYangYoung-tang(Renshenyangrongtang) was similar between cultured PC12 cells and in vivo hippocampal and cerebral cortical neurons in the mouse brain. Conclusions: This experiment shows that the InSamYangYoung-tang(Renshenyangrongtang) may play a protective role in neural tissues damaged by cytotoxic substances. Since neuronal damage seen in degenerative brains such as AD are largely unknown, the current data may provide possible insight into therapeutic strategies for AD treatments. InSamYangYoung-tang(Renshenyangrongtang) might be effective for the treatment of AD. Investigation into the clinical use of the InSamYangYoung-tang(Renshenyangrongtang) for AD is suggested for future research.
신경흥분독성과 간질발작발현은 glutamate 수용체활성과 연관이 있다고 알려져 있다. a-amino-3-hydroxy-5-methyl-4-isoxazole-propionate (AMPA), kainate 수용체에 대한 glutamate 활성을 포함하는 다양한 기전을 가진 항전간제인 Topiramate는 신경보호작용을 가진다는 증거가 제시되어 Topiramate가 간질발작 후 해마의 glutamate 수용체 발현에 미치는 효과를 관찰하였다. 흰쥐에 kainate를 복강 내 주사하여 간질중첩증을 유발시킨 후 Topiramate를 1주일 주사하였다 Apop tag in situ detection kit를 이용하여 세포손상을 관찰한 결과 kainate 유발 간질중첩증 1주일 후 해마의 CA1, CA3에서 심각한 세포사를 보였으나, Topiramte 처리 군에서는 세포사가 현저히 감소하였다. 간질중첩증 이후 NMDA 수용체 아형 1,2a, 2b 발현이 현저히 증가했으나 Topiramate 처치에 의해 NMDA수용체의 발현에는 뚜렷한 변화가 없었다. AMPA수용체에서는 GluR1이 간질중첩증 이후 현저히 상향 조정되었고 GluR2는 현저히 하향조정 되었다 Topiramate 1주일 처리 시 간질중첩증으로 인해 변화된 CluR1과 GluR2의 발현이 역전되었다. 결론적으로 Topiramate는 간질중침증에 의한 CluR1/CluR2 발현비의 증가로 유발되는 흥분성 신경세포사를 억제시킴으로써 신경보호작용이 있는 것으로 보인다.
Background: Ginseng, the root of Panax ginseng (PG), is used widely as a herbal medicine to prevent and treat various diseases. Panax ginseng has pharmacological effects on neurodegenerative diseases such as Alzheimer's disease (AD). The present study evaluated the neuroprotective effects of PG and its possible neuroprotective mechanisms in advanced glycation end product (AGE)-induced AD in a rat model. Methods: Advanced glycation end products were injected bilaterally into the CA3 region of the rats' brains. The Morris water maze test and step-down type passive avoidance test were performed to evaluate their memory and cognitive abilities. The oxidation indexes in the hippocampus were detected. Immunohistochemistry was conducted to visualize the receptors for advanced glycation end products (RAGEs) and nuclear factor-kappa-light-chain-enhancer of activated B cell (NF-${\kappa}B$). Results: Behavioral results showed that PG (1 g/kg, 0.5 g/kg, and 0.25 g/kg) significantly shortened the escape latency, remarkably increased the number of crossing times, significantly decreased the number of errors, and prolonged the latency in rats with AGE-induced AD. Panax ginseng also significantly reduced the malondialdehyde level, increased the glutathione content, and increased superoxide dismutase activity in the hippocampus. Panax ginseng significantly decreased the expression of RAGE and NF-${\kappa}B$. The blockade of anti-RAGE antibody could significantly reduce AGE-induced impairments and regulate these expressions. Conclusion: Our results demonstrated that PG significantly inhibits AGE-induced memory impairment and attenuates Alzheimer-like pathophysiological changes. These neuroprotective effects of PG may be associated with the RAGE/NF-${\kappa}B$ pathway. Our results provided the experimental basis for applying PG in preventing and treating AD.
본 연구에서는 석곡(Dendrobium nobile Lindl.) 추출물에 대한 신경세포 보호 효과를 확인하기 위하여 $H_{2}O_{2}$에 의해 유도된 스트레스 상태의 PC12 세포주에서 MTT-dye reduction assay와 LDH release assay를 이용하였다. MTT reduction assay 결과, 스트레스 상태에서는 47%의 생존율을 보인데 반하여 석곡 추출물 50${\mu}$g/ml 농도로 처리하였을 때 99.5%의 높은 세포 생존율을 확인할 수 있었다. 이 결과는 LDH release assay 에서도 일치하는 결과를 확인하였다. 그리고 광학 현미경을 이용한 형태학적 변화를 관찰한 결과에서도 신경돌기의 출현 유도를 통한 신경세포 생존을 확인 할 수 있었다. 또한 핵의 변화에 미치는 영향을 관찰한 결과 정상 세포의 핵은 타원형의 온전한 핵 모양을 나타낸 반면 산화적 손상을 입은 세포는 apoptotic body가 핵 주변에 나타나는 전형적인 apoptosis를 나타내었고 석곡 추출물을 처리한 결과 핵의 condersation현상과 fragmentation이 현저히 감소함을 확인 할 수 있었다. 그리고 $H_{2}O_{2}$에 의한 capase-3의 활성은 석곡 추출물을 50${\mu}$g/ml 처리 후 약 1.1배 이하로 감소함을 보였다.
알츠하이머 질환은 대표적인 신경퇴행성 질환으로, 뇌 내에서 $A{\beta}$ 단백질 축적은 알츠하이머 질환의 원인으로 알려져 있다. 본 연구에서는 amyloid beta ($A{\beta}$)로 손상을 유도한 SH-SY5Y 신경세포에서 kaempferol, kaempferol-3-O-glucoside, quercetin, quercetin-3-${\beta}$-D-glucoside의 신경세포 보호 효과에 대해 검토하였다. SH-SY5Y 신경세포에 $A{\beta}_{25-35}$ ($25{\mu}M$)를 처리하였을 때, 처리하지 않은 normal군에 비해 세포생존율이 유의적으로 감소하였다. 반면, kaempferol, quercetin 및 그 배당체들을 각각 처리하였을 때 $A{\beta}_{25-35}$만을 처리한 control군에 비해 유의적으로 세포생존율의 증가를 나타내었다. 또한, apoptosis에 관여하는 cleaved caspase9, Bcl-2-associated X protein (Bax) 단백질 발현을 측정한 결과, normal군에 비해 control군에서 유의적으로 cleaved caspase9 및 Bax 단백질 발현의 증가를 나타내어 $A{\beta}$ 유도 신경세포 손상으로 인한 apoptosis가 유발됨을 확인 하였으며, kaempferol, quercetin 및 그 배당체들의 처리 시 apoptosis 관련 단백질 발현이 감소함으로써 신경세포 보호 효능이 나타냄을 확인하였다. 이러한 결과는 kaempferol, quercetin 및 그 배당체들이 apoptosis 조절을 통해 신경세포 보호 효과를 나타내며, 신경세포 손상으로 인한 알츠하이머 질환을 예방하는 유용한 소재로써 사용 가능성이 있음을 보여준다.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
