Whitening Effect with Fruit and Whitening Toothpaste (과일 및 미백치약으로 처리한 치아의 미백효과)

  • The purpose of this study is to confirm the effect of natural tooth whitening using fruits that can be easily accessed in real life by increasing interest in tooth whitening in modern society. Twenty premolar teeth were divided into five groups (n=4). Strawberry (A), lemon (B), banana skin (C), baking soda (D) and whitening toothpaste (E) were treated with a toothbrush on the enamel surface. The tooth surface was treated with a toothbrush at the same time for 3 minutes for breakfast, lunch, and dinner, then washed with sterile distilled water, and stored in saline solution. This procedure was repeated for 4 weeks, and the color change of teeth was observed at intervals of one week. After 4 weeks, there was a significant difference in tooth color change after 3 weeks(p<0.05). Among them, strawberry and lemon showed higer effects to whitening toothpaste. It is believed that strawberries and lemons have a higher whitening effect than whitening toothpaste. Whitening toothpaste may be replaced with a safe natural whitening effect using strawberries and lemons instead of chemical ingredients that may be harmful to human body.

Cudrania tricuspidata Root Extract as Whitening and Antiwrinkle Cosmetic Agent (꾸지뽕나무 뿌리 열수추출물로부터 미백 및 주름개선 화장품 소재의 개발)

  • Water extracts of root, leaf, branch and fruit of Cudrania tricuspidata were tested to use as natural antiwrinkle and whitening cosmetic ingredient. Root extract indicated 86.5 mg/g polyphenol concentration and 55.5 mg/g flavonoid concentration. From MTT test, root, branch and fruit extract did not show cell toxicity up to $1,000{\mu}g/mL$, however, leaf extract displayed significant cell toxicity in concentrations higher than $200{\mu}g/mL$. Cudrania tricuspidata root extracts displayed excellent 81% tyrosinase inhibition (whitening effect), 58% elastase inhibition (antiwrinkle effect) at $500{\mu}g/mL$ and showed minor UVB absorbance. When solution and lotion formulation were made including Cudrania tricuspidata root extract, they are stable at $25^{\circ}C$ for 21 days, but showed significant deviation in viscosity and particle size at $47^{\circ}C$. From the study, Cudrania tricuspidata root extract demonstrated good potential for natural whitening and antiwrinkle cosmetic agent.

The Effect of Inhibition of Uncaria rhynchophylla as an Inhibitor of Melanogenesis and an Antioxidant in B16F10 Melanoma Cells (B16F10세포에서 멜라닌 생성 억제제 및 항산화제로서 조구등의 억제 효과)

  • Many people of all ages wish to have lighter skin for cosmetic reasons, and natural products attract more attention than chemically synthesized compounds. Uncaria rhynchophylla is widely used in Asia as a traditional herbal medicine. In order to find novel skin whitening agents, the present study evaluated the antioxidant activity and potential tyrosinase-inhibiting properties of U. rhynchophylla. Specifically, this study analyzed the antioxidant capacity of a 70% ethanolic extract of U. rhynchophylla as well as its effects on tyrosinase activity and melanin synthesis. Total mRNA levels were examined using reverse transcription polymerase chain reaction. The results revealed that U. rhynchophylla extracts exhibit great antioxidant capacity and significant levels of polyphenol and flavonoid compounds. U. rhynchophylla extracts can also powerfully inhibit tyrosinase activity. This same capacity was observed in melanoma B16F10 cells; that is, U. rhynchophylla extracts suppressed intracellular tyrosinase activity and reduced the amount of melanin in treated cells. In addition, a 1 mg/ml concentration of U. rhynchophylla extract significantly reduced the mRNA expression levels of tyrosinase. U. rhynchophylla extracts decrease tyrosinase and inhibit melanogenesis in B16F10 cells. This finding suggests that U. rhynchophylla has great potential as a natural whitening agent in skincare products.

Isolation of Bioactive Compounds from the Ethylacetate Fraction of Fermented Garlic Complex and their Tyrosinase Inhibition Activities (대산(大蒜)을 포함하는 복합발효물의 에틸아세테이트 분획으로부터 Tyrosinase 저해활성 성분의 분리 및 동정)

  • Fermented complex from garlic and nine medicinal plants were developed as a natural whitening material. Tyrosinase inhibition activity was determined and four active compounds were isolated. The nutritional components of fermented garlic complex (FGC) were analyzed to confirm the applicability as a functional food material. Tyrosinase inhibitory effect of FGC was 88.6%. Methanol extract was partitioned with EtOAc, n-BuOH and H2O. From the EtOAc fraction (47 g), which showed the highest yield, active fractions were separated by repeated TLC, silica gel and ODS column chromatography to isolate active compounds. The chemical structures of the isolated compounds were analyzed by NMR and MS spectra. Phenylpropanoid compounds of 2,4,5-trihydroxy-benzenepropanoic acid (1) (1.9 mg) and 2,3,5-trihydroxy-benzenepropanoic acid (2) were confirmed. In addition, 2,4-dihydroxy-hydrocinnamic acid (3) (3.3 mg) and (+)sesamin (4) (6.1 mg) were isolated. These compounds will be useful as index compounds or functional compounds in FGC.

The Inhibitory Effects of Acanthopanax sessiliflorum Seeman on Melanogenesis (오가피추출물의 멜라닌 생성 저해 효과)

  • To develop a new natural whitening agent for cosmetics, we investigated effects of Acanthopanax sessiliflorum Seeman on melanogenesis. We prepared phenolic acid-rich extract including two phenolic acids, chlorogenic acid and caffeic acid, as predominant constituents from Acanthopanax sessiliflorum Seeman. Phenolic acid-rich extract showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.43{\pm}0.35{\mu}g/mL$ and $158.91{\pm}1.57{\mu}g/mL$, respectively. Phenolic acid-rich fraction reduced melanin contents of B16 melanoma cells dose-dependantly and the decrease was $27.27{\pm}2.66%$ at a concentration of $100{\mu}g/mL$. And the phenolic acid-rich fraction reduced intracellular tyrosinase activity about $53.67{\pm}8.55%$ at a concentration of $100{\mu}g/mL$. Phenolic acid-rich extract inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that phenolic acid-rich fraction reduced melanin formation by the inhibitions of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that phenolic acid-rich extract could be used as a whitening ingredient in cosmetics.

Tyrosinase Inhibitory Compounds Isolated from Persicaria tinctoria Flower (쪽 꽃에서 분리한 타이로시네이즈 저해 화합물)

  • To develop a new natural whitening agent, we investigated the tyrosinase inhibitory effects of Persicaria tinctoria Flower extracts (PTFE). PTFE showed inhibitory activity on mushroom tyrosinase with the $IC_{50}$ values of $70.8{\pm}2.2{\mu}g/mL$. We purified two active compounds from PTFE by LH-20 column chromatography and prep-high performance liquid chromatography (HPLC) and identified as quercetin-3-O-rhamnoside (Q3R) and myricetin-3-O-rhamnoside (M3R) by $^1H$nuclear magnetic resonance (NMR) and liquid chromatography-mass spectrometry (LC-MS) analysis. Q3R and M3R showed tyrosinase inhibitory activities with the $IC_{50}$ values of $47.0{\pm}0.1{\mu}g/mL$ and $150.5{\pm}1.6{\mu}g/mL$, respectively. These results suggest that PTFE and its active compounds reduced melanin formation by the inhibition of tyrosinase activity. Thus, P. tinctoria flower extracts may be a candidate for cosmetic use.

Tyrosinase Inhibitory Activity and Melanin Production Inhibitory Activity of Taraxinic Acid from Taraxacum coreanum (흰민들레(Taraxacum coreanum)에서 분리한 taraxinic acid의 tyrosinase 활성저해 및 melanin 생성저해 효과)

  • This study was to investigate the Taraxinic acid from Taraxacum coreanum on tyrosinase activity and melanogenesis. In B16BL6 cell, Taraxinic acid did not show cytotoxicity even at concentrations of up to 100 ㎍/mL. In addition, tyrosinase inhibitory activity and melanogenesis inhibitory activity were confirmed by stimulation with α-melanocyte stimulating hormone (α-MSH) in the presence of taraxinic acid. Taraxinic acid was added to cells at concentrations of 10, 50 and 100 ㎍/mL and treated for 48 hours to confirm tyrosinase inhibitory activity and melanin production. The tyrosinase inhibitory activity increased in proportion to the amount of the sample, and showed an inhibitory activity of about 54.5% at a concentration of 50 ㎍/mL. Melanin production decreased in proportion to the sample amount, and it was about 62.2% at the concentration of 10 ㎍/mL. From the above results, it was found that Taraxinic acid had higher tyrosinase activity and melanin synthesis inhibitory activity in melanocyte than arbutin. The results suggest that Taraxinic acid can be utilized in natural whitening cosmetics.

The Inhibitory Effects of Pogostemon cablin Bentham Extract on Melanogenesis (광곽향 추출물의 멜라닌 생성 저해 효과)

  • To develop a new natural whitening agent for cosmetics, we investigated the inhibitory effects of Pogostemon cablin Bentham extracts (PCE) and its active component on melanogenesis. PCE showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $24.2{\pm}2.85{\mu}g/mL$ and $IC_{50}=118{\pm}0.43{\mu}g/mL$, respectively. PCE reduced melanin contents of B16 melanoma cells in a dose-dependant manner and decreased to about 23 % at a concentration of $20{\mu}g/mL$ without cell cytotoxicity (below $100{\mu}g/mL$). And the PCE reduced intracellular tyrosinase activity about 18 % at concentration of $50{\mu}g/mL$. We purified one active compound from PCE and identified its structure. It was identified as patchouli alcohol, sesquiterpene family, by 1H-NMR, $13_C$-NMR, and Mass analysis. Patchouli alcohol also inhibited ROS scavenging activities in DPPH radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.14{\pm}0.12{\mu}g/mL$ and $49{\pm}3.24{\mu}g/mL$, respectively. Patchouli alcohol inhibited melanin synthesis in a dose dependent manner ($IC_{50}=3.9{\mu}g/mL$). And the patchouli alcohol reduced intracellular tyrosinase activity about 40 % at concentration of $10{\mu}g/mL$. Patchouli alcohol inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that PCE and patchouli alcohol reduced melanin formation by the inhibited of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that PCE could be used as a useful whitening agent.

The Inhibitory Effects of Poria cocos Bark Extract on Melanogenesis (복령피 추출물의 멜라닌 생성 저해 효과)

  • To develop a new natural whitening agent for cosmetics, we investigated the inhibitory effects of Poria cocos Bark extracts (PCBE) and its active compound on melanogenesis. PCBE showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $19.4{\pm}2.21{\mu}g$/mL and $IC_{50}=103{\pm}3.33{\mu}g$/mL, respectively. PCBE reduced intracellular tyrosinase activity about 34 % at concentration of $50{\mu}g$/mL. And PCBE reduced melanin contents of B16 melanoma cells about 51 % at concentration of $50{\mu}g$/mL without cell cytotoxicity (below $100{\mu}g$/mL). We purified one active compound from PCBE and identified its structure. It was identified as 3-$\beta$-hydroxylanosta-7,9(11),24-trien-4-oic acid, triterpene family, by $^1H$-NMR, $^{13}C$-NMR and Mass analysis. 3-$\beta$-hydroxylanosta-7,9(11),24-trien-4-oic acid showed ROS scavenging activities in DPPH radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $4.3{\pm}0.15{\mu}g$/mL and $54{\pm}1.67{\mu}g$/mL, respectively. Also, it was shown that 3-$\beta$-hydroxylanosta-7,9(11),24-trien-4-oic acid reduced intracellular tyrosinase activity about 43 % at concentration of $10{\mu}g$/mL. And it inhibited melanin synthesis in a dose dependent manner ($IC_{50}=3.6{\mu}g$/mL) without cell cytotoxicity (below $100{\mu}g$/mL). 3-$\beta$-hydroxylanosta-7,9(11),24-trien-4-oic acid inhibited tyrosinase, TRP-1 and TRP-2 expression at protein level. These results suggest that PCBE and 3-$\beta$-hydroxylanosta-7,9(11),24-trien-4-oic acid reduced melanin formation by the inhibition of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that PCBE could be used as a useful whitening agent.
