Structure Prediction of KiSS1-derived Peptide Receptor Using Comparative Modelling

  • KiSS1-derived peptide receptor, a GPCR protein, binds with the hormone kiss peptin. They are important in the neuroendocrine regulation of reproduction and in the secretion of gonadotrophin-releasing hormone. Thus, analysing the structural features of the receptor becomes important. However, the three dimensional structure of the protein is unavailable. Hence in this study, we have performed the homology modelling of KiSS1-derived peptide receptor with 5 different templates. 30 models were constructed using two platforms - Easymodeller and ITasser. The optimal models were chosen based on the model validation. Two models were selected after validation. The developed models could provide useful for analysing the structural features of KiSS1-derived peptide receptor and their pathophysiological role in various disorders related to them.

분변내 성호르몬의 분석을 통한 멸종위기 사육 오랑우탄에서의 번식형태 (The Reproductive Patterns of Endangered Captive Orangutans by Analysing the Sex Hormones in Feces)

  • To monitor the reproductive patterns of endangered orangutan, Time-resolved fluorescence immuno assay (TR-FIA) were used to analyze metabolites of sex hormones in feces. Orangutan had long-term interbirth intervals (amenorrhea) during lactation period in which the secretion of sex hormones was repressed. The concentration of progesterone in the serum of pregnant orangutan was 30fold higher than that in non-pregnant orangutan. However, the concentration of hCG during pregnant period was different from the result of other primates. The present study suggested that age is not the important factor in determining the reproduction capability, because it is rather greatly influenced by the combination of various factors. Tracing metabolites of sex hormones in orangutan feces seems to be provide the effective solution for the infertility in orangutan. This study result shows the basic data in operating conservation project for endangered orangutan.

  • Allocation patterns of energy and mineral elements were investigated with phaseolus multiflorus grown in the environmental gradients. The result showed different energy allocation patterns according to relative light intensities and nutrients. The optimal switching time of energy allocation from vegetative to resproductive growth was delated as decreasing relative light intensity. The switch of the shift to reproduction was timed earlier in phosphorus treatment and delayed in nitrogen treatment. Analyzing the mineral elements to various organs, patterns of energy allocation were different from those of mineral allocation. There was no significant difference for allocation patterns in relative light intensity gradients. it was shown that n and p were distributed over the reproductive organs, k mainly in stems, ca in leaves and na in roots. mg was evenly distributed in each organs.

PAF Regulate Blastocyst Development to Hatching Stage through PKC Activity in the Mouse

  • The developmental regulation of the preimplantation mammalian embryos is a fundamental step for preparing the implantation and it may be regulated by several autocrine and paracrine factors including platelet-activating factor. PAF improved the embryonic survival and implantation but its role during blastocyst development is still largely unknown. In this study, the effects and the possible pathway of PAF on developmental regulation of blastocyst to hatching stage were investigated. Developmental pattern in hatching embryo was a concentration-response curve showing maximal activity at 1 nM PAF, with decreasing activity at higher concentrations. $50{\mu}M$ 1-(5-isoquinolimnesulfonyl)-2-methylpiperazinme dihydrochloride (H-7), a PKC inhibitor, inhibited the progression of blastocyst to hatching embryo. In addition H-7 blocked the PAF effects on the blastocyst development. Besides tetradecanoylphorbol acetate (TPA), a PKC activator stimulated development of blastocyst to the hatching stage. These finding revealed that PAF support the blastocyst development to the hatching embryo. Also it is suggested that PAF action pathways in hatching supporting include the PKC signaling pathway.

  • PDP벽걸이형 대형 평판 디스플레이로서 최근 시각적 극대화에 도움이 되는 대형화의 가능성과 자연스러운 색감 표현력 등의 장점 때문에 주목받고 있다. 하지만 저계조 표현, 의사 윤곽 문제등의 화질 저하 문제로 우위에 있지 못하고 다른 디스플레이 장치들과 치열한 경쟁을 하고 있어 더욱 많은 화질 연구가 필요하다. 본 논문은 42" PDP의 차질 개선 연구를 위한 Verilog HDL을 이용한 소프트 웨어 테스트-베드 구현과 FPGA를 이용한 자체 제작 보드에서 대해 설명한다.

First Record of Ulva pertusa Kjellman (Ulvales, Chlorophyta) in the Pacific Coast of Mexico

  • Based on samplings during 2006-2007 on the intertidal zone of Playa Tampico, Eréndira and Punta Baja, Baja California, Ulva pertusa Kjellman (Ulvales, Chlorophyta) was recorded for the first time for the Pacific coast of Mexico. An alga native to Asia, U. pertusa has just recently been recorded in the southern coast of California (USA). The identification of U. pertusa was based on the observation of the morphology and reproductive characters of the collected specimens. Furthermore, the species identity was confirmed by molecular comparison between nuclearencoded ITS2 sequences of the Mexican samples and those of other Ulva species in GenBnak data. Habitat and distribution of U. pertusa along the Pacific coast of North America are also described. Considering our new report of its occurrence in Mexico, we conclude that U. pertusa is expanding its geographical distribution by trans-ocean introductions.

Evolutionary Viewpoint on GnRH (gonadotropin-releasing hormone) in Chordata - Amino Acid and Nucleic Acid Sequences

  • GnRH (gonadotropin-releasing hormone) is a supreme hormone regulating reproductive activity in most animals. The sequences of amino acid and nucleic acid of GnRH reported up to now are examined from the evolutionary framework of Chordata. All identified GnRH are classified into GnRH1, GnRH2, or GnRH3. In all three forms of GnRH both N-terminal and C-terminal are conserved, which allows for effective binding to their receptors. The three amino acids in the middle of GnRH1 sequence have altered diversely from the primitive Chordata, which is indicative of the adaptation process to the ambient environment. GnRH2 and GnRH3 sequences are well conserved. There are more diverse modifications in the nucleic acids than in amino acid sequence of GnRH1. These variations can result from meiosis, mutation, or epigenetics and indicate that GnRH is the product of natural selection.

  • The experimental manipulation of protooncogenes and their gene products is a valuable research tool for the study of human neoplasia. In this study, the recently identified human cervical cancer protooncogene (HccR-2) was expressed in transgenic mice under the control of the tetracycline regulatory system. The phenotype observed was similar in many respects to human chronic neutrophilic leukemia (CNL). Thus, the HccR-2 transgenic mouse model is important not only for investigating the biological properties of the HccR-2 protooncogene in vivo, but also for analyzing the mechanisms involved in the progression of CNL.

  • In a conventional sense, dried-spermatozoa are all dead and motionless due to the lost of their natural ability to penetrate oocytes both in vivo and in vitro. However, their nuclei are completely able to contribute to normal embryonic development even after long-term preservation in a dried state when the dried-spermatozoa are microinjected into the oocytes. In this sense, dried spermatozoa must still be alive. Thus, defining spermatozoa as alive or dead seems rather arbitrary. Several drying method of sperm including freeze-drying, evaporative/convective-drying and heat-drying were represented in this review. Although the drying protocol reported here will need further improvement, the results suggest that it may be possible to store the male genetic resources.

  • Sex ratio of a population is a key factor to determine reproductive rate of the population. If the population has skewed sex ratio to the male, the population size may decline gradually at an area which it inhabits, and, furthermore, if a species plays an ecologically important role in its community (e.g., a keystone species in Paine, 1966), its potential reproductive rate will be to form a characterized community because community structure is strongly influenced by the abundance of the ecologically important species (Hughes, 1986). A number of workers (e.g., Bryan et al., 1986 in Nucella lapillus; Spence et al., 1990 in Thais haemastoma) suggested that high frequency and/or late stage of imposex might affect the female mortality and normal reproduction adversely. Consequently, the imposex could affect sex ratio of natural population of some neogastropod including thaisid species. (omitted)
