The Chemistry of Secondary Products from Acanthopanax Species and their Pharmacological Activities

  • The chemistry of secondary products from Acanthopanax species and their pharmacological activities were reviewed. A nitrogenous compound, a furan compound, a quinoid, benzoids, coumarins, phenylpropanoids, lignans, flavonoids, terpenoids, phytosterols, polyacetylenes, a pyrimidine, cyclitols, monosaccharides and an aliphatic alcohol have been isolated from Acanthopanax species and have been shown to have various levels of activities such as anti-bacterial, anti-cancer, anti-gout, anti-hepatitis, anti-hyperglycemic, anti-inflammatory, anti-leishmanicidic, anti-oxidant, anti-pyretic, anti-xanthine oxidase, choleretic, hemostatic, hypocholesterolemic, immunostimulatory and radioprotectant effects, etc.

  • Four flavonoid glycosides, flavaprin (7), evodioside B (8), vitexin (11), and hesperidin (12), as well as the coumarins bergapten (1), xanthotoxin (2), and isopimpinellin (3), the lignan simplexoside (10), the steroids ${\beta}-sitosterol$ (4) and daucosterol (5), the limonoids isolimonexic acid (6) and limonin (9), and uracil (13) and myo-inositol (14) have been isolated from Euodia daniellii. The structures of these compounds were established from spectral data. Among the isolates, bergapten showed cyclooxygenase-2 inhibitory activity with an $IC_{50}$ value of $6.2{\;}{\mu\textrm{g}}/ml. Flavonoids isolated from this plant exhibited no cytotoxic activity against the human tumor cell lines, A549, SKOV-3, SKMEL-2, XF498, and HCT15.

  • Background: Breast cancer is the serious health concern in India causing the highest mortality rate in females, which occurs due to uncontrolled cell division and can be metastasize to other parts of the human body. Interactions with estrogen receptor (ER) alpha are mainly responsible for the malignant tumors with regulation of the transcription of various genes as a transcription factor. Most of the drugs currently used for the breast cancer treatment produce various side effects and hence we focused on natural compounds which do not exhibit any toxic effect against normal human cells. Materials and Methods: Structure of human ER was retrieved from the Protein Data Bank and the structures of flavonoid compounds have been collected from PubChem database. Molecular docking and drug likeness studies were performed for those natural compounds to evaluate and analyze the anti-breast cancer activity. Results: Finally two compounds satisfying the Lipinski's rule of five were reported. The two compounds also exhibited highest binding affinity with human ER greater than 10.5 Kcal/mol. Conclusions: The results of this study can be implemented in the drug designing pipeline.

  • Seven prenylated flavonoids and two pterocarpans together with a saponin were isolated from the roots of Echinosophora koreensis. They were identical with echinoisosophoranone, echinoisoflavanone, isosophoranone, sophoraisoflavanone A, kenusanone C, kenusanone A, sophoraflavanone D, 4-hydroxy-3-methoxy-(6aR,11aR)-8,9-methylenedioxy-pterocarpans, (-)-maackiain, and $3-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}2)-{\beta}-D-galactopyranosyl(1{\rightarrow}2)-{\beta}-D-glucuronopyranosyl$ kuzusapogenol A. Among them, a pterocarpan, 4-hydroxy-3-methoxy-(6aR,11aR)-8,9-methylenedioxypterocarpan, and a saponin, $3-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}2)-{\beta}- D-galactopyranosyl(l{\rightarrow}2)-{\beta}-D-glucuronopyranosyl$ kuzusapogenol A, were isolated for the first time from this plant.

Effects of Flavonoids on Pollen Tube Growth in Arabidopsis thaliana

  • Arabidopsis mutants deficient in flavonoid and sinapate ester (tt4 and fah1-7, respectively) were evaluated in vivo and in vitro to study the possible role of flavonoid compounds in pollen tube growth. In vivo, we investigated pollen tube growth in the pistils of the mutants and wild type(Ler). The growth of pollen tubes was significantly different among the three genotypes. In the fal1-7 pistils, the tubes grew to a greater length relative to those of the wild type or tt4. To examine in vitro pollen tube growth, a solid medium was devised for pollen germination and subsequent growth. In vitro, the identical result was obtained; fahl-7 pollens developed the longest tubes and elongated most rapidly. Therefore, the growth response of pollen tubes to phenolic compounds was examined by adding quercetin or sinapate ester in various concentrations to the media. Quercetin enhanced both germination rate and tube growth in the pollens of the mutants and the wild type, especially in tt4. In contrast, sinapate ester inhibits pollen germination and pollen tube growth in three genotypes. These results suggest that flavonoids and related phenolic compounds have physiological role in the plant reproductive system.

Natural Compounds with Antioxidant Activity: Recent Findings from Studies on Medicinal Plants

  • Reactive oxygen species potentially cause damage to cellular components including lipids, protein and DNA; this oxidative damage plays a key role in the pathogenesis of neurodegenerative disease, cardiovascular disease, metabolic disease and cancer. On the basis of the oxidative stress hypothesis, a number of studies have been performed to search for an efficient and safe antioxidant. Although in vitro studies have provided promising results, only a limited number of natural and synthetic antioxidants have been developed for clinical application due to their low efficacy and side-effects. Thus, the discovery of new antioxidants with marked efficacy and safety has attracted worldwide attention in recent decades. Since plants are recognized as important sources of natural antioxidants, our research has focused on the discovery of new naturally occurring antioxidants from medicinal plants. The purpose of this review is to open a new prospect in the field of search for natural antioxidants from medicinal plants by summarizing our recent findings. Using in vitro bioassay systems such as 2,2-diphenyl-1-picrylhydrazyl, superoxide radical scavenging tests and lipid peroxidation models, we have tested over than 350 species of medicinal plants for their antioxidant activity and selected several of them for further investigation. During the research on the discovery of effective natural antioxidants from the medicinal plants selected, we have isolated several new and known antioxidant compounds that include stilbene glycosides, phenolic glycosides, flavonoids, oligostilbenes, and coumarins. Our results suggest that the presence of antioxidant compounds in the medicinal plants might be associated with the traditional use to treat inflammation, cardiovascular disease and various chronic diseases.

Effects of dietary flavonoids on performance, blood constituents, carcass composition and small intestinal morphology of broilers: a meta-analysis

  • Objective: This study aims to evaluate the influence of dietary flavonoids on the growth performance, blood and intestinal profiles, and carcass characteristics of broilers by employing a meta-analysis method. Methods: A database was built from published studies which have reported on the addition of various levels of flavonoids from herbs into broiler diets and then monitored growth performance, blood constituents, carcass proportion and small intestinal morphology. A total of 42 articles were integrated into the database. Several forms of flavonoids in herbs were applied in the form of unextracted and crude extracts. The database compiled was statistically analyzed using mixed model methodology. Different studies were considered as random effects, and the doses of flavonoids were treated as fixed effects. The model statistics used were the p-values and the Akaike information criterion. The significance of an effect was stated when its p-value was <0.05. Results: Dietary flavonoids increased (quadratic pattern; p<0.05) the average daily gain of broilers in the finisher phase. There was a reduction (p<0.01) in the feed conversion ratio of the broilers both in the starter (linear pattern) and finisher phases (quadratic pattern). The mortality rate tended to decrease linearly (p<0.1) with the addition of flavonoids, while the carcass parameter was generally not influenced. A reduction (p<0.001) in cholesterol and malondialdehyde concentrations (both linearly) was observed, while super oxide dismutase activity increased linearly (p<0.001). Increasing the dose of flavonoids increased (p<0.01) the villus height (VH) and villus height and crypt depth (VH:CD) ratio (p<0.05) in the duodenum. Similarly, the VH:CD ratio was elevated (p<0.001) in the jejunum following flavonoid supplementation. Conclusion: Increasing levels of flavonoids in broilers diet leads to an improvement in growth performance, blood constituents, carcass composition and small intestinal morphology.

가금 생산에서 천연 항산화제로서 식물성 Flavonoids의적용 (Application of Plant Flavonoids as Natural Antioxidants in Poultry Production)

  • 최근 닭은 고도화된 육종개량과 함께고밀도사육, 열스트레스, 환경 및 사료 오염 등과 같은내외적스트레스요인에 의해 체 조직에서 과도한 활성산소(ROS) 발생으로 산화적 스트레스가 매우 높다. 산화적 스트레스는 지질과 산화작용으로 DNA 손상, 세포사 등을 유발하여 닭의 건강과 복지에 심각한 문제를 유발한다. 닭은 이러한 내외적 산화적 스트레스에 따라 항산화 효소와 비효소적 항산화 물질에 의해 활성산소를 제거하는 방어 작용을 한다. 최근 영양학적 방법으로서 산화적 스트레스를 억제하는 식물성 flavonoids 사료첨가제 개발에 주목하고 있다. Favonoid를 닭에게 급여 시 소화기관 및 근육에서 지질과산화도 감소, 항산화력 및 항산화 효소 활성도가 증가하는 것으로 항산화제로서 가치가 높다. Flavonoids는 구조화합물이 활성산소와 결합하여 안정화할 수 있는 구조로서 ROS를 소거 또는 항산화 효소의 발현을 통해 ROS를 제거한다. 또한 금속 이온과 결합하 는 킬레이트화 작용, 산화효소 및 NO 생성 억제 등 다양한 항산화 작용이 있다. 그러나 문제점으로 flavonoid 화합물은 용해도가 낮아 소장 흡수세포에서 흡수율이 저하됨으로 이를 극복할 수 있는 체내 운반 기술의 개발이 필요하다. 또한 사료 섭취 시 flavonoid 농도가 가장 높은 곳이 위장관으로 이곳에서 명확한 항산화 효과를 나타내는 용량을 구명하여 첨가 수준을 결정하여야 한다. 항산화 작용기전은 스트레스에 따라 다양한 전사인자들(transcrptional factors)을 통해 스트레스 반응 유전자(vitagenes)의 발현을 유도한다. Flavonoid의 항산화 작용기전으로 Nrf-2와 Nf-kB 등과 같은 전사인사의 발현 과정과 vitagenes을 통해 효율적 항산화 효과를 나타내는 방법에 관한 연구도 중요하다. 닭에서 육종 및 사육 환경 요인에 따라 발생하는 산화 스트레스는 경제성과 더불어 동물복지 측면에서도 심각한 문제를 유발한다. Flavonoid 함유 천연 항산화제 개발과 적용은 가금 산업에서 향후 중요한 분야가 될 것으로 전망된다.

  • Aster species has been used in traditional chinese medicine for treatment of a bruies and asthma. On reviewing the literatures of this species, monoterpene glycosides, diterpenoids, triterpene glycosides, cyclic pentapeptides, oligopeptides and flavonoids 1) were isolated and some pharmacological activites were investigated 2).   • From the 70% EtOH extract of the roots of Astragalus membranaceus (Leguminosae), eleven flavonoid derivatives and a lignan, were isolated and identified as liquiritigenin (1), daidzein (2), formononetin (3), sophorophenolone (4), calycosin (5), methylnissolin (6), isomucronulatol (7), isomucronulatol 7-O-glucoside (8), methylnissolin 3-O-glucoside (9), calycosin 7-O-glucoside (10), (+)-syringaresinol O-${\beta}$-D-glucoside (11), and isomucronulatol 7,2'-di-O-glucoside (12), by spectroscopic methods. This is the second report of the isoflavonoid derivatives sophorophenolone (4) and isomucronulatol 7,2'-di-O-glucoside (12) from a natural source, as well as the first report of compounds liquiritigenin (1), daidzein (2) and (+)-syringaresinol O-${\beta}$-D-glucoside (11) from the species A. membranaceus.
