Current Status of Registered Korean Patents Related to the Health Functional Food for Skin Care (피부미용 관련 건강기능식품 국내 특허 등록현황)

  • Proper skin care promotes good metabolism and the biological activity of skin, helps maintain the skin in a healthy and beautiful state when combined with nutritional nourishment. Health functional foods are being used for the purpose of enhancing efficient skin care. Health functional foods related to skin care maintain the epidermis, dermal fibroblast layer and subcutaneous tissue that form the skin. Efficient functional foods alleviate the signs of endogenous aging that come with getting older and exogenous aging caused by sunlight. Even though the field of skin care related to health functional foods has received less attention and been the subject of less research compared to functional cosmetics which are developing widely, this area of skin care that maintains and improves the layer of dermal fibroblast through the intake of food, is expected to progress with the commercialization of products in many fields when the related technical research development is galvanized and the related patents are applied for. Research into health functional foods related to skin care in Korea started rather late in comparison to other advanced countries where patents for relevant techniques have been applied for since 1990's together with research conducted on how they might be used for practical purposes. This study used the key words 'skin care, health functional food' to search for Korean patents that have been applied for at the Korean Intellectual Property Office from 1983 to 2011 using the KIPRIS database, in order to help researchers in the related fields by organizing the patented formulas for health functional foods for skin care which have gained attention recently. According to the number of registrations in each industry field, the total number of patents was 1,120 we screened the major patents among them, the field with the largest number was food foodstuff non-alcoholic beverage and preparation or treatment(A23L) with 135, then natural plant medicine & medical supplies and cosmetics(A61K) with 112, microorganisms or enzymes(C12N) with 63, heterocyclic compounds(C07D) with 44, horticulture or cultivation of sea weeds, forestry(A01G) 16 times, listed from the highest to the lowest number of applications. It has been revealed that food, foodstuff, non-alcoholic beverage and preparation or treatment(A23L) was the industry field where the most patents for health functional foods related to skin care were registered.

Anti-inflammatory effect of ethanol extract of Syzygium aromaticum (정향(Syzygium aromaticum) 에탄올 추출물의 항염 효과)

  • The purpose of this study is for checking anti-inflammatory effects of Syzygium aromaticum ethanol extract. For this, we investigated biological active evaluation about anti-inflammatory effects by Syzygium aromaticum ethanol extract. Syzygium aromaticum was extracted with 70% ethanol. This extract was tested for the cell viability on RAW 264.7 cell line by MTT assay, nitric oxide inhibitory activity and expression of inflammatory mediators. The extract showed low cytotoxicity as more than 90% cell viability in under 25 ㎍/ml concentration. On LPS-induced RAW 264.7 cell line, nitric oxide inhibition activity result showed that the extract reduced NO productions in a concentration-dependent manner. Expression of inflammatory cytokines as PGE2, TNF-α and IL-1β decreased in a concentration-dependent manner and iNOS and COX-2 proteins expression were decreased significantly in western blot analysis. We confirmed that the Syzygium aromaticum ethanol extract has excellent anti-inflammatory effect. This results suggested that extract of Syzygium aromaticum may have value as the cosmetic materials.

The Coffee Sliver Skin Extracts from Coffee Beans Exhibited Cosmetic Properties with Antioxiant Activity and Inhibitory Effects for Elastase, Collagenase and Tyrosinase (커피 은피 추출물의 항산화 효과와 엘라스타제, 콜라게나제 및 티로시나제 저해효과)

  • The coffee silver skin is a part of coffee beans. We report that the coffee sliver skin extracts exhibited cosmetic properties of antioxidant, anti-winkle and whitening effects. The ethanol extracts of silver skin showed free radical scavenging activity up to 92.26% in $50{\mu}g/mL$, especially against DPPH radical and ABTS radical cation. The silver skin extracts showed inhibitory effects for tyrosinase activity and DOPA oxidation in a dose-dependent manner, suggesting the extracts retain for the whitening property in cosmetics. The coffee silver skin extracts effectively inhibited the elastase and collagenase. Cytotoxicity of the coffee silver skin extracts was measured by the colorimetric MTS assay. The viability of the human keratinocytes (HaCaT) treated with the coffee silver skin extracts was same as that of untreated cells, indicating the extracts are safe to human cells. Here, we suggest that the silver skin extracts of coffee bean could be a potential natural substance for anti-winkle, whitening, antioxidant properties for cosmetics.

Mechanism of Melanogenesis Inhibition by Melanoston Isolated from Yeast (효모에서 분리한 멜라닌 생성 억제 물질의 작용 기전)

  • Melanocytes synthesize melanin within discrete organelle termed melanosomes which are transferred to the surrounding keratinocytes and can be produced in varying sizes, numbers and densities. Skin whitening products have become increasingly popular in the past few years. The most successful natural skin whitening agents are: Arbutin, Vitamin C, Kojic acid, Mulberry, which are all tyrosinase inhibitors. In this work, melanoston, a melanogenesis inhibitor isolated from yeast was studied to understand its mechanism of melanogenesis inhibition. It was found that melanoston was not a tyrosinase inhibitor, while when melanoston was applied to the B16 melanoma cell culture media, the intracellular tyrosinase activity was decreased by more than 30%, When B16 melanoma was stimulated with ${\alpha}$-MSH, cell morphololgy was dramatically changed to have lots of dendrites on the cell membrane surface. On the other hand, B16 was treated with ${\alpha}$-MSH and melanoston, simultaneously, the change of cell morphology was not so great. This inhibition effect of melanoston was found to be related to the inhibition of intracellular activation and transportation of tyrosinase, which was observed by immunostaining of B16 melanoma using anti-tyrosinase antibody. From these results, melanoston was regarded as an inhibitor to the differentiation of melanoma cells.

Study of Antioxidation Action of Lenonuri herba Extract (익모초 추출물의 항산화 작용에 관한 연구)

  • Motherwort (Leonurus sibiricus L), a hemp nettle (Labiatae) plant, grows about 1.5${\~}$2 m high spontaneously to wildness in Korea, China, Japan, and other Asian place. Its medical applications includes women's uterine disease, urination, bloody stool, bloody urine, and hemorrhoids. It's also effective for high blood pressure, heart stimulation, and anti-cancer activity. We first prepared chlroform and methanol extracts of motherwort and then they were fractionated using water, $30\%$ methanol, $60\%$ methanol, and $100\%$ methanol, respectively. Each fractionates is assayed for free radical scavenging activities against DPPH and anti-oxidant activity by TBARS assay measuring lipid peroxidation using LDL. The $30\%$ and $60\%$ methanol fractionates of methanol extracts showed strong anti-oxidant activity compared to vitamin C. They also had more Potent SOD activity using pyrogallol at 250 ppm than that of vitamin C. These results suggest that anti-oxidant activities of motherwort may be applicable to development of natural anti-oxidant cosmetics. Possibility of nature anti-oxidation ability cosmetics is seen possibility low official.

A Study on the Surface-Modification of Barium Sulfate/TiO$_2$/Dimethicone Composite Powder and its Application in Color Cosmetics (바륨설페이트/티타늄디옥사이드/디메치콘의 복합화 및 메이크업에서의 응용)

  • Sensor and optical properties have become critical features in powder foundation. The flaky barium sulfate powder shows good smooth texture, adhesion and natural looking characteristics. However, it has limitations abilities in UV shielding, hiding and blooming effect. Thus we adopt TiO$_2$ that has excellent hiding power and blooming effect as well as UV shielding ability, but TiO$_2$ has still intrinsic problems in dispersion and texture. To overcome this disadvantages, the barium sulfate/TiO$_2$/dimethicone composite powder was prepared. The flaky barium sulfate powder was coated with TiO$_2$ in nanoscale and followed by coated with dimethicone. When this surface-modified powder was applied for make-up cosmetics, especially in powder foundation, the powder gave powder foundation more good characteristics than the original flaky barium sulfate, Ti02 powder, dimethicone in abovementioned optical and sensory properties. To characterize the distinctive features of this surface-modified powder, we measured its characteristics with UV in vitro tester, hiding powder test method, goniophotometer, consumer panel test and so on.

Enhancement of the Cosmeceutical Activity by Nano-encapsulation of Thiamine Di-lauryl Sulfate (TDS) with antimicrobial efficacy (항균 효능이 있는 비타민 B1 유도체(Thiamine Dilauryl Sulfate:TDS)의 나노입자화를 통한 기능성 향장 활성 증진)

  • This study was to improve cosmetical activity of thiamine di-lauryl sulfate (TDS) by encapsulation of nanoparticle with lecithin. Results showed that most of the nanoparticles containing the TDS were well formed in round shape with below 150 ~ 200 nm diameter as well as they were fairly stable in various pH ranges by measuring zeta potentials. The nanoparticles of TDS resulted in 85% cell viability of human normal fibroblast cells (CCD-986sk) when added at the highest concentration (1.0 mg/mL). The nanoparticles of Acer mono sap showed highest free radical scavengering effect as 88.1% in adding sample (1.0 mg/mL), compared to TDS solution of non-encapsulation (81.6%). The nanoparticles of TDS reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 41.4%. The TDS solution and nanoparticles showed significant anti-microbial activities agaionst the salmonella typhimurium and listeria monocytogenes at 5 and 6 days as compared with control. Anti-microbial activities of TDS nanoparticles were similar to positive control. These results indicated that TDS nanoparticles may be a source for functional cosmetic agents capable of improving cosmetical activity such as antioxidant, whitening, and anti-wrinkling effects and can be further developed as natural preservative in cosmetics.

Antioxidative and Cellular Protective Effects of Dolwoe (Gynostemma pentaphyllum Makino) Extracts against Oxidative Stress (돌외(Gynostemma pentaphyllum Makino) 추출물의 산화적 스트레스에 대한 항산화 및 세포보호효과)

  • In this study, we investigated the total phenolic and flavonoid contents, component analysis, antioxidative activity and cellular protective effects against oxidative stress on human skin cells in 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Gynostemma pentaphyllum (G. pentaphyllum) Makino. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activites ($FSC_{50}$) of the 50% ethanol extracts, ethyl acetate fraction and aglycone fraction were 246.8, 147.2, $128.9{\mu}g/mL$, respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the 50% ethanol extract, ethyl acetate fraction and aglycone fraction on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were 37.15, 10.74, $7.19{\mu}g/mL$, respectively. We investigated the cellular protective activity and the results showed that treatment of aglycone fraction ($0.05-0.39{\mu}g/mL$) protect human skin cells in a concentration-dependent manner when the skin cell damages were induced by treating them with $H_2O_2$. These results suggest that extract/fractions of G. pentaphyllum Makino may be applicable as natural antioxidants in cosmetics.

Evaluation of Biological Activities on the Extractives of Pinaceae (소나무과 일부 수종의 에탄을 추출물에 대한 생물학적 평가)

  • Most of the cells, specially in the skin, free radicals and reactive oxygen species induced aging, accompanying with diseases. The pinaceae family is very familiar plant with Korean spirit. However, those plants were not estimated for natural products in manufactural process. We employed 4 different plants, Pinus koraienis S et Z. Pinus banksiana LAMBERT. Pinus rigida MILL. Pinus densiflora S. et Z. in pinceae family. The ethanol extracts were prepared for three different parts of plant, wood, bark and leaf, respectively. In this study, we carried out free radical and superoxide radical scavenging assay to investigate the anti-oxidative activity The bark and leaf part of plants showed similar anti-oxidant effect. L-ascorbic acid and $\alpha$-tocopherol at $10{\mu}g/mL$. Also we tried to investigate inhibitory effects of elastase activity in in vitro experiments on pinaceae plant extract as a matters for functional cosmetics. Among those plants, the bark of Pinus rigida MILL. and Pinus densiflora S. et Z. showed inhibitory effect. The cell viability was evaluated with MTT assay. The potential relationship was shown between the cell viability and anti-oxidant effect because the anti-oxidant effects were positively correlated wvith the cell growth in MTT assay. As the results in our experiments, we expect the potential activities of pinaceae as a material of functional cosmetics.

Activating the Proliferation of Keratinocyte Stem Cells by Paeonol, a Compound from Natural Herb (Paeonol에 의한 표피줄기세포 활성화)

  • Epidermis is continuously regenerated by keratinocyte stem cells (KSCs) residing in basement membrane, which is critical to the survival of an organism. KSCs are believed to persist during the whole lifetime and generate an enormous number of keratinocytes, required for the maintenance of epidermis, through transit amplifying cells dividing definite times until they become differentiated. In this report, we have developed a phenolic compound, paeonol, purified from Moutan Cortex, as a KSC proliferation activator, by screening about 350 herbal compounds. The cell proliferation activation by paeonol is specific for KSC not for keratinocyte, and no significant difference in the expression of p63 protein, a KSC marker, in KSCs treated with paeonol was observed in FACS analysis with anti-p63 antibody. In the colony forming assay, paeonol-treated KSC showed improved colony forming activity more than 1.3 fold. In addition, the result of PCR array shows that the activity of paeonol is through several signal pathways involving stem cell functions. These results suggest that paeonol could enhance KSC proliferation activity without reduction in stemness and could be applied to cosmetics as a KSC activating ingredient.
