Comparison of scopolamine-induced cognitive impairment responses in three different ICR stocks

  • Cognitive impairment responses are important research topics in the study of degenerative brain diseases as well as in understanding of human mental activities. To compare response to scopolamine (SPL)-induced cognitive impairment, we measured altered parameters for learning and memory ability, inflammatory response, oxidative stress, cholinergic dysfunction and neuronal cell damages, in Korl:ICR stock and two commercial breeder stocks (A:ICR and B:ICR) after relevant SPL exposure. In the water maze test, Korl:ICR showed no significant difference in SPL-induced learning and memory impairment compared to the two different ICRs, although escape latency was increased after SPL exposure. Although behavioral assessment using the manual avoidance test revealed reduced latency in all ICR mice after SPL treatment as compared to Vehicle, no differences were observed between the three ICR stocks. To determine cholinergic dysfunction induction by SPL exposure, activity of acetylcholinesterase (AChE) assessed in the three ICR stocks revealed no difference of acetylcholinesterase activity. Furthermore, low levels of superoxide dismutase (SOD) activity and high levels of inflammatory cytokines in SPL-treated group were maintained in all three ICR stocks, although some variations were observed between the SPL-treated groups. Neuronal cell damages induced by SPL showed similar response in all three ICR stocks, as assessed by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay, Nissl staining analysis and expression analyses of apoptosis-related proteins. Thus, the results of this study provide strong evidence that Korl:ICR is similar to the other two ICR. Stocks in response to learning and memory capacity.

Optimization of Processing Conditions of Chinese Smoke-cured Bacon (Larou) with a New Natural Coating Solution during Storage Period

  • The study aims to determine the optimum sterilization rate and water activity of Chinese traditional smoke-cured bacon product (Larou) in the preservation with natural coating solution. With the response surface methodology (RSM), we analyzed 3 factors of processing conditions (the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan) and 2 response factors (sterilization rate and water activity). Sterilization rate and water activity of Larou were largely affected by the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan. The final optimum concentrations of lysozyme, sodium alginate, and chitosan were 0.09, 1.40, and 1.60% and realized the high sterilization rate. Water activity of sliced Larou was significantly correlated with the sterilization rate. Low-field nuclear magnetic resonance analysis verified the optimum processing conditions. The coating resulted in 99.69% rate of reducing bacteria after 30-day storage. The data of the total number of colony, peroxidation value, moisture content, pH, and sensory evaluation provided the theoretical basis for extending the shelf life of Chinese traditional smoke-cured bacon product (Larou) with natural coating solution.

Screening of Natural Herb Resources for Anti-oxidative Effects in Korea (국내 자생 자원식물들의 항산화 활성 검색)

  • In the present study, the anti-oxidative properties of 80% ethanol extracts from 140 kinds of natural plants were investigated. Persicaria perfoliata had the highest anti-oxidative relative potency(VitaminE, $ORAC_{PE}$=1.0), followed by Sorbaria sorbifolia var. stellipila, Rosa multiflora, Lysimachia vulgaris var. davurica, Quercus aliena, Quercus mongolica and Lespedeza bicolor ($ORAC_{PE}$ ${\geq}$ 1.50). Among them, the anti-oxidative activity values of the n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH, and water fractions of the 80% EtOH extracts of P. perfoliata and S. sorbifolia in the ORAC assay system were $1.13{\pm}0.002$, $1.10{\pm}0.004$, $3.67{\pm}0.232$, $1.62{\pm}0.049$ and $1.05{\pm}0.003$; $1.02{\pm}0.007$, $1.00{\pm}0.016$, $2.94{\pm}0.130$, $1.80{\pm}0.019$ and $1.04{\pm}0.012$, respectively, with marked anti-oxidative activity in the ethyl acetate fraction from selected the 2 plant extracts.

Numerical Investigation of Pressure Fluctuation Reducing in Draft Tube of Francis Turbines

  • For a prototype turbine operating under part load conditions, the turbine output is fluctuating strongly, leading to the power station incapable of connecting to the grid. The field test of the prototype turbine shows that the main reason is the resonance between the draft tube vortex frequency and the generator natural vibration frequency. In order to reduce the fluctuation of power output, different measures including the air admission, water admission and adding flow deflectors in the draft tube are put forward. CFD method is adopted to simulate the three-dimensional unsteady flow in the Francis turbine, to calculate pressure fluctuations in draft tube under three schemes and to compare with the field test result of the prototype turbine. Calculation results show that all the three measures can reduce the pressure pulsation amplitude in the draft tube. The method of water supply and adding flow deflector both can effectively change the frequency and avoid resonance, thus solving the output fluctuation problem. However, the method of air admission could not change the pressure fluctuation frequency.

Variation in Carotenoid Composition in Carrots during Storage and Cooking

  • High-performance liquid chromatography (HPLC) was applied to determine the carotenoid composition of carrots during storage and cooking. Analyses were conducted immediately after harvest and 1, 2, 4, and 8 weeks after harvest. During the course of the storage, the carotenoid levels generally decreased, and this decrease was found to be greater during the first week for $\beta$-carotene (all-trans-$\beta$-carotene) and lutein, and during the second week for $\alpha$-carotene. Additionally, the amount of the $\alpha-$ and $\beta$-carotenes in carrot leaves changed slightly within the first 2 weeks of harvest when stored at $4^{\circ}C$. Specifically, the level of lutein, the main component of carrot leaves, increased from 233.8$\pm$11.7 to $346.2\pm26.7{\mu}g$/g DW during the first 2 weeks. In addition, the change in carotenoid contents was observed during the home-processing of one Korean cultivar. Carrots fried in oil showed the highest amount of $\beta$-carotene ($164.3\pm6.6{\mu}g$/g DW) and $\alpha$-carotene ($50.1\pm0.4{\mu}g$/g DW), while carrots that were prepared by sauteing, pressure-cooking in water and microwaving had the second highest levels. The greatest loss of in carotenoids occurred in response to boiling in water containing 1% NaCl, braising and baking. The content of lutein increased slightly after boiling in water containing 1% NaCl ($9.3\pm0.4{\mu}g$/g DW), while a loss in lutein occurred after preparation using other home-processing methods. A cis-isomer of all-trans-$\beta$-carotene, 13-cis-$\beta$-carotene, was present in detectable amounts in all processed samples, but not in raw roots. Another isomer, 9-cis-$\beta$-carotene, was detected in carrots that were prepared by boiling, frying and pressure-cooking.

Antimicrobial activities in the Korean Traditional Leaf Mustard, Brassica juncea Coss.

  • As part of developing natural food preservatives, the antimicrobial effect of ethanol and water extracts from the Cruciferous vegetable was examined Korean traditional Brassica juncea Coss. was used widely as an ingredient of Kimchi, a natural flavoring and spice for a long time. Antimicrobial activities were examined against 15 microorganisms which were food-born pathogens and/or food poisioning microorganisms and food-related bacteria and yeasts. Ethanol extract from leaf mustard showed an antimicrobial effect in most of the strains used in the present study. The lowest minimum inhibitory concentration (MIC) were seen in Bacillus subtilis and Bacillus natto at 10 mg/ml. MIC of water extract was 40-60 mg/ml for bacteria and yeast. Antimicrobial activity of the ethanol extract was not disappeared by the heating at $121^{\circ}C$ for 15 min and not affected by pH.

  • Since the middle of the 70s, the countries with the marine resources declared the exclusive water zone of two hundred miles. As a result, our country lacked many foreign fishing grounds. This awakened our perception of fishing grounds. The fishing ground is an important factor in the economical effectiveness of fisheries. The fishing ground must have the following basic conditions. 1. It must be the place the fishery resources inhabit and wander. 2. Their catching or aquaculturing must be possible by the technical methods. 3. It must have economic worth in the management. The fishing ground that achieves the above basic conditions is affected by the following factors. This is the factor of the fishing ground location. A. Natural factor 1). Suitable climate 2).Abundant resources 3). Good quality of the sea-bottom 4). Not to be pollutted area B, Economic factor 1). Be adjacent to fishing port and fisheries market 2). Good working conditions 3). Be abundant of economical resources C. Social factor 1). Be possible to work legally 2). Be permitted in the political relations 3). Be acquainted with the customs of foreign countries The fishing ground location is mainly affected by the natural factor of the above factors. The abundance of resources depends on the natural resources. The resources in the fishing ground have the cycle of developing period, maturing period, and decaying period according to the fishing efforts. The point that we sustainably yield the maximum of fishery resources is the changing point of maturing period and decaying period. We call this point MSY, so the resources of fishing ground should be managed by the MSY. Therefore, when we select the fishing ground location. We should judge the natural factor, the economic factor and the social factor synthetically according to the situation and environment, so we can achieve the efficient management.

  • Anhui Province, with a total north-south length of 570km and an east-west width of 450km and a total area of 139.6 thousand km2, accounts for 1.45% of China's total area. The landform and land feature of Anhui Province is diverse, and generally it can be divided into 5 natural regions: (1) Huaibei Plain; (2) Jianghuai Hillocks; (3) Dabie Mountains in the West of Anhui Province; (4) Yanjiang Plain, (5) Mountain Area of southern Anhui Province. Anhui Province is located in the transitional zone of warm and humid zone and subtropical zone, and its mean annual precipitation is 800-1800mm. The province, which has diverse climate, multiple land forms and many rivers and lakes, passes three basins (Huaihe River, Yangtze River and Xin'an River) and has large differences in the time distribution and regional distribution of water resource. Therefore, the development and usage conditions of the water resource in different regions are different.

Flow duration change in downstream of reservoir by selective deficit supply method (선택적 부족분 공급방식에 따른 댐 하류하천의 유황 변화 분석)

  • Currently, South Korea implements water resources management policies focusing on integrated water quantity, quality and hydro-ecology management. In particular, rehabilitation of natural rivers has become a major issue. As for reservoir operation during non-flood season, efforts have been made continuously to apply the Deficit Supply Method that can maximize water supply to address droughts and increase in water demand. When Deficit Supply Method is applied, the water supply capacity of reservoir can be maximized. However, downstream water flow would remain constant. In consideration that a natural stream, a long-time-created hydro-ecology, can be significantly influenced by flow variability, the Deficit Supply Method-based reservoir operation can generate effective water supply. Still, it may trigger adverse effects from the aspects of natural rehabilitation and hydro-ecology recovery. The main objective of this study is to analyze impacts on downstream flow duration through reservoir simulation by comparing the Firm Supply Method, the Deficit Supply Method and the Selective Deficit Supply Method, and examining each method's effects on reservoir operation. This study found that the Firm Supply Method could maintain water flow variability, but could not maximize water supply capacity. When the Deficit Supply Method was applied, water supply capacity could be increased while remaining vulnerable regarding water flow variability, as a difference between average flow and low flow was negligible at downstream. In comparison, the Selective Deficit Supply Method was found to sustain time-based reliability at 95% or higher, whereas downstream flow duration could be maintained at a level similar to the level generated by the Firm Supply Method.

  • 본 연구에서는 GIS 기법을 통하여 구축된 지형 자료와 수문지형학적 기법을 사용하여 미계측 소유역에서의 강우-유출 특성을 분석하는 기법을 제시하였으며, 실제로 경상남도 낙동강 제1 지류인 양산천과 지리산의 대원사 유역에 적용하여 보았다.
