Effects of a Forced Air-Flow System for Recovery of Turfgrass after Intensive Traffic Injury (집중적 답압 피해에서의 잔디 회복을 위한 강제 흡.호기 순환식 설비의 효과)

  • Korea and Japan hosted the FIFA World Cup Soccer Game in 2002. Ten stadiums had been built and more than 30 soccer grounds for practice were renovated in Korea. Sport fields in both countries had problems on cool-season turfgrass growth and quality by summer decline during humid and warm climate especially followed by intensive uses. We measured the effects of air-flow system, which is designed to optimize rootzone soil gas and moisture levels to promote the growth and maintenance turfgrass. This experiment was carried out to verify the effects of the system on soil gas exchange, ground resilience, and turfgrass recovery in turfgrass rootzone. Within 1 or 2hr of operation of the system, rootzone soil gas ($CO_2$, $O_2$) levels returned to natural atmospheric levels completely Soil $CO_2$ levels began to decrease within the first 10 min of operation of the system. The levels were reduced from 1.3 to 0.06% after 30 min, and natural atmospheric levels within 1 hr. When the system was turned off, $CO_2$ levels increased to 0.36% and 0.7% after 5 and 20 hr, respectively. The application of the system did not affect the resilience of turf surface after traffic treatments. Higher traffic treatment resulted in higher surface resilience especially in zoysiagrass plots. Operation of the system had a significant beneficial impact on turf recovery by increased root dry wight and improved turf quality, as compared with the non-operated check plots.

Effects of exercise on myokine gene expression in horse skeletal muscles

  • Objective: To examine the regulatory effects of exercise on myokine expression in horse skeletal muscle cells, we compared the expression of several myokine genes (interleukin 6 [IL-6], IL-8, chemokine [C-X-C motif] ligand 2 [CXCL2], and chemokine [C-C motif] ligand 4 [CCL4]) after a single bout of exercise in horses. Furthermore, to establish in vitro systems for the validation of exercise effects, we cultured horse skeletal muscle cells and confirmed the expression of these genes after treatment with hydrogen peroxide. Methods: The mRNA expression of IL-6, IL-8, CXCL2, and CCL4 after exercise in skeletal muscle tissue was confirmed using quantitative-reverse transcriptase polymerase chain reactions (qRT-PCR). We then extracted horse muscle cells from the skeletal muscle tissue of a neonatal Thoroughbred. Myokine expression after hydrogen peroxide treatments was confirmed using qRT-PCR in horse skeletal muscle cells. Results: IL-6, IL-8, CXCL2, and CCL4 expression in Thoroughbred and Jeju horse skeletal muscles significantly increased after exercise. We stably maintained horse skeletal muscle cells in culture and confirmed the expression of the myogenic marker, myoblast determination protein (MyoD). Moreover, myokine expression was validated using hydrogen peroxide ($H_2O_2$)-treated horse skeletal muscle cells. The patterns of myokine expression in muscle cells were found to be similar to those observed in skeletal muscle tissue. Conclusion: We confirmed that several myokines involved in inflammation were induced by exercise in horse skeletal muscle tissue. In addition, we successfully cultured horse skeletal muscle cells and established an in vitro system to validate associated gene expression and function. This study will provide a valuable system for studying the function of exercise-related genes in the future.

Quality Evaluation of Dried Red Pepper After Copper Plate Contact (구리표면 접촉에 의한 건조 고춧가루의 특성 평가)

  • Copper possesses antimicrobial properties against certain bacteria and fungi. In this study, changes in microbial population, color and the antioxidant properties of dried red pepper powder (RPP) by contact with the surface of a copper plate were investigated. Lightness of RPP increased after treatment on the copper plate for 2 h at 55℃. The numbers of total aerobic mesophilic bacteria in RPP increased by 0.61 and 0.50 log on the surface of stainless steel and copper plates during 24 h at 37℃, respectively. The population of yeast and mold was reduced by 90% in the RPP treated on both copper and stainless steel plates under the same condition. Yeast and mold were significantly reduced on the copper plate during 2 h at 55℃. Scavenging activities of RPP treated on the copper plate for 24 h at 37℃ against DPPH radical were decreased; those for 2 h at 55℃ against ABTS radical were rather enhanced. The results suggest that copper might be applied in the processing and storage equipment in relation to RPP products for antimicrobial purposes.

Antioxidative effects of hesperidin and hesperetin under cellular system (Hesperidin과 hesperetin의 cellular system에서의 항산화 효과)

  • In this study, we investigated the antioxidant activity of hesperidin and hesperetin, which are the active compounds from Citrus junos, in the cellular system. Under cellular model of oxidative damage using LLC-$PK_1$ renal epithelial cell, the oxidative damage induced by 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH) led to the loss of cell viability, while treatment of hesperidin and hesperetin increased significantly the cell viability as dose-dependent manner. In addition, NO-induced cellular oxidative damage by sodium nitroprusside were significantly recovered by the treatment of hesperidin and hesperetin, showing the increase of cell viability. But hesperidin and hesperetin showed no significant protective effect on $O_2{^-}$-induced cellular oxidative damage. The present study indicates that hesperidin and hesperetin protect against free radical, especially AAPH-induced peroxyl radical. In particular, hesperetin has stronger protective effect against oxidative stress than hesperidin.

Sewage Treatment Using Natural Systems and Effluent Reuse for Crop Irrigation in Small Communities

  • A pilot study was performed from July 1998 to December 2002, including winter performance, to examine seasonal performance of a constructed wetland and subsequent pond system for treatment of sewage in small communities of Korea. Pond was operated as a intermittent-discharge pond during winter period, and continuous flow system during growing season; its effects was evaluated from December 2001 to April 2003. The subsurface flow (SSF) wetland was satisfactory for treating sewage with good removal efficiency even during the winter period. The wetland effluent concentrations of $BOD_5$ and TSS were often higher in winter than in the growing season, but this was explained by the higher loading rates, rather than lower removal efficiency. The relatively poor-quality wetland effluent was further polished during winter in the pond. The upper layer of the pond water column became remarkably clear immediately after ice melt. In the growing season, ponds could be operated as a continuous flow system to remove nutrients and pathogens, and the effluent of pond could be reused as a supplemental irrigation water without risk of infection by sewage-borne pathogens as well as causing adverse effect on growth and yield. Overall, the wetland system was found to be adequate for treating sewage with stable removal efficiency, and the intermittent-discharge pond was found to be effective for further polishing if necessary. Therefore, the combination of a wetland and subsequent pond system and reuse of effluent as crop irrigation water is recommended as a practical alternative to treat sewage in Korean small communities, and partial discharge of pond water in March is suggested.

Alpha-emitting Radioisotopes Production for Radioimmunotherapy (방사면역치료를 위한 알파 방출 방사성 동위원소 생산)

  • This review discusses the production of alpha-particle-emitting radionuclides in radioimmunotherapy. Radioimmunotherapy labeled with alpha-particle is expected to be very useful for the treatment of monocellular cancer (e.g. leukemia) and micrometastasis at an early stage, residual tumor remained in tissues after chemotherapy and tumor resection, due to the high linear energy transfer (LET) and the short path length in biological tissue of alpha particle. Despite of the expected effectiveness of alpha-particle in radioimmunotherapy, its clinical research has not been activated by the several reasons, shortage of a suitable a-particle development and a reliable radionuclide production and supply system, appropriate antibody and chelator development. Among them, the establishment of radionuclide development and supply system is a key factor to make an alpha-immunotherapy more popular in clinical trial. Alpha-emitter can be produced by several methods, natural radionuclides, reactor irradiation, cyclotron irradiation, generator system and elution. Due to the sharply increasing demand of $^{213}Bi$, which is a most promising radionuclide in radioimmunotherapy and now has been produced with reactor, the cyclotron production system should be developed urgently to meet the demand.

NO3-N Removal of A Reed Wetland Cell Constructed for Purifying Effluent from A Night Soil Treatment Plant During Its Initial Operating Stage (분뇨처리장 방류수정화 갈대습지셀의 초기운영단계 질산성질소 제거)

  • $NO^3$-N removal was examined from July 2002 to December 2002 of a surface-flow constructed treatment wetland cell, which was a part of a treatment wetland system composed of four wetland cells and one distribution pond. The system was established on rice paddy near the Kohung Estuarine Lake located at the southern part of the Korean Peninsula. The lake and the paddy were formed by a salt marsh reclamation project. Effluent from a secondary-level treatment plant was funneled into the system. The investigated cell was created in June 2002. Its dimensions were 87 m in length and 14 m in width. It had an open water zone at its center, which was equivalent to 10 percent of its total area. Reeds(Phragmites australis) were transplanted from natural wetlands into the cell and their stems were cut at about 40 cm height from their bottom ends. Average 25 $m^3$/day of effluent from the plant was funneled into the cell by gravity flow and average 24.2$m^3$/day of its treated effluent was discharged into the Sinyang Stream flowing into the lake. Its water depth was maintained about 0.2 m and its hydraulic detention time averaged 5.2 days. The average height of the reed stems was 45.2 cm in July 2002 and 80.5 cm in September 2002. The number of stems averaged 40.3 stems/$m^2$ in July 2002 and 74.5 stems/$m^2$ in September 2002. The reeds were established initially well. $NO_3$-N loading rate of influent and effluent averaged 173.7 and $93.5mg/m2{\cdot}day$, respectively. Removal of $NO_3$-N averaged $80.2mg/m2{\cdot}day$ and its removal rate by mass was about 50 %. Considering the initial operation of the cell and the inclusion of the cold months of November and December in the analysis period, the $NO_3$-N removal rate was good.

Removal of Phosphorus and NOM in Wastewater Effluent Using Ejector.BAF System (분사배출 고도 응집.생물여과 공정을 이용한 하수처리수 중의 TP 및 NOM 제거)

  • While the existing sewage treatment facilities are mainly being operated by biological processes, winter-time efficiency improvement and additional phosphorus treatment equipment using chemicals have been required to follow the effluent criteria of TP (0.2, 0.3 and 0.5 mg/L for the zone of I, II and III respectively) and $BOD_5$ (5.0 mg/L) which is intensified from 2012 in Republic of Korea. We made an investigation into actual condition of biological treatment process and calculated the optimal chemical input amount by jar test of supernatant of secondary sedimentation tank to evaluate the process improvement for the intensified criteria. Ejector BAF system for removing TP, $BOD_5$ of sewage effluent was suggested. The concentration of TP from biological process is 0.3-0.8 mg/L, and the input amount of optimal chemical coagulant was above Al/P ratio of 3(1.9 mg/L as Al) to meet the criteria of TP for secondary treatment effluent. From the results of this experiment, the best Al/P ratio for Ejector BAF system was about Al/P ratio of 1, and LV of BAF process for intensified criteria of $BOD_5$ and TP was below 1.97m/hr.

Anti-norovirus activity of natural compounds and its potential in food application (항노로바이러스 천연물을 이용한 식품개발)

  • Control of food pathogens is critical in food safety field. Norovirus is one of the major causes of gastroenteritis and food poisoning worldwide, however, currently, there is not a vaccine or a specific drug available for its treatment. There are several methods to inactivate norovirus during food processing by chemical and physical treatments, however, the use of natural substance has been suggested as an optional strategy due to their safety and consumer preference. In this study supported by Ministry of Food and Drug Safety in Korea, we identified novel plant-derived substances with significant anti-norovirus activities. The aim of this project was to determine the antiviral activity of a wide range of natural substances, including plant-derived extracts and essential oils, using a norovirus surrogate system, human norovirus replicon-bearing cells, and mouse in vivo experiments. During the activity screening test, we identified novel anti-norovirus substances or oils using plaque assay with MNV-1. Six selected substances were formulated into an optimum mixture and used as an ingredient for salad sauce of which anti-novovirus activity was confirmed(pending for patent and paper submission). The potential application of selected natural substances as a metal surface sanitizer was also tested. Interestingly, the mixture of selected natural compounds showed a significant inhibitory effect against norovirus. These results suggest that these substances may be used as food ingredient with anti-norovirus antivity or components for surface sanitizers to prevent norovirus contamination.

Antioxidant and cytotoxic activities of curcumin and its analogs: An exploration of structure-activity relationships (Curcumin과 관련 성분들의 산화방지활성과 세포독성 분석 및 구조와 활성 연관성 조사)

  • Bioactivities of curcumin, a major pigment of Curcuma longa L., have been widely investigated. In this study, the antioxidant and cytotoxic properties of curcumin and its analogs including ferulic acid, dibenzoylmethane (DBM), and tetrahydrocurcumin (THC), and their structure-activity relationships were assessed. Ferulic acid, THC, and curcumin showed strong scavenging activities against several radicals and exhibited considerable lipid peroxidation inhibitory activity. Curcumin showed the strongest cytotoxic activities against HeLa, HCT-116, IEC-6 and INT 407 cells, whereas ferulic acid did not show any cytotoxic effect up to 100 µM against these cell lines. Cytotoxicity of curcumin and THC was significantly enhanced by superoxide dismutase and diminished by N-acetylcysteine. The combination treatment of curcumin and ferulic acid enhanced the cytotoxicity, whereas the combination of curcumin and DBM offset their toxicity. These results suggest that methoxy phenolic and β-diketon moieties are crucial for the antioxidant- and cytototoxic activities of curcumin, respectively.
