Full-length chloride intracellular channel protein 1 (CLIC1) is a member of the family of proteins related to bovine chloride intracellular channel p64. Mutations in the SEDL gene cause spondyloepiphyseal dysplasia tarda (SEDT), a rare X-linked chondrodysplasia. The link between the intracellular chloride channels and SEDL is an important step toward understanding their functional interplay. In the present study, CLIC1 protein was subcloned into the pGEX-KG vector and overexpressed in XL-1 blue cells. We developed a large-scale expression system composed of glutathione S-transferase (GST) fused with a 240-amino-acid CLIC1 protein in Escherichia coli. The soluble CLIC1 protein was successfully purified to homogeneity, and its purity, identity, activity and conformation were determined using SDS-PAGE, MALDI-MS, biophotometer and circular dichroism spectroscopic studies. The binding of both CLIC1 and SEDL proteins in vitro was detected by BIAcore biosensor and fluorescence measurements.
From the viewpoint of the structural design, the principal problem of the heat exchanger is the potentiality of structural instabilities due to the fluid loading effect during operations. Excessive fluid loading may give rise to permanent deformation of tube and would enentually result in collapse of heat exchanger, which would cause an obstruction of the fluid flow in the narrow channels. In this study, a fluid-structural interaction model was developed to investigate analtically the vibration characteristics of thin rectangular tube used in the heat exchanger. The model consists of two flat plates separated by fluid. The effects of the fluid in the tube was stuided. For analyses, the natural frequency coefficients of the model were investigated for the plate aspect ratios, channel heights, and boundary conditions. As conclusions, the natural frequency coefficients of the tube is found to be affected largely by the fluid loading and the channel heights.

This study estimates the region of freshwater influence (ROFI) by Han River discharge in the Yeomha channel, Gyeonggi Bay. A 3-D numerical model, which is validated for reproducibility of variation in current velocity and salinity, is applied in Gyeonggi Bay. Distance of freshwater influence (DOFI) is defined as the distance from the entrance of Yeomha channel to the point where surface salinity is 28 psu. Model scenarios were constructed by dividing the Han River discharge into 10 categories (200~10,000 m3/s). The relation equation between freshwater discharge and DOFI was calculated based on performing a non-linear regression analysis. ROFI in Yeomha channel expands from the southern sea area of Ganghwa-do to the northern sea area of Yeongheung-do as the intensity of Han River discharge increases. The discharge and DOFI are a proportional relationship, and the increase rate of DOFI gradually decreases as discharge increases. Based on the relation equation calculated in this study, DOFI in the Yeomha channel can be estimated through the monthly mean Han River discharge. Accordingly, it will be possible to respond and predict problems related to damage to water quality and ecology due to rapid freshwater runoff.

Bed material in natural streams is affected by geological characteristics of upstream and acting focre by flow of stream. Acting force in streams are also affected by bed slope and discharge of streams. Yamamoto(1994) suggested that channel reaches with similar slope share similar characteristics values in representative bed material size, tractive force, channel width and depth. He also suggested that main factors dominating channel characteristics are mean annual discharge, epresentative bed material size and bed slope. Bed material shows physical and ecological characteristics of channels. In this research, characteristics of bed material in natural or close to natural streams were studied and compared with Janpanese stream data. In the result, characteristics of gravel and sand material can be presented in the term of non-dimensional tractive force, bed slope and bed material size.

When a molten corium is relocated in a lower head of a reactor vessel, the ERVC (External Reactor Vessel Cooling) system is actuated as coolant is supplied into a reactor cavity to remove a decay heat from the molten corium during a severe accident. To achieve this severe accident mitigation strategy, the two-phase natural circulation flow in the annular gap between the external reactor vessel and the insulation should be formed sufficiently by designing the coolant inlet/outlet area and gap size adequately on the insulation device. For this reason, one-dimensional natural circulation flow tests were conducted to estimate the natural circulation flow under the ERVC condition of APR1400. The experimental facility is one-dimensional and scaled-down as the half height and 1/238 rectangular channel area of the APR1400 reactor vessel. As the water inlet area increased, the natural circulation mass flow rate asymptotically increased, that is, it converged at a specific value. And the circulation mass flow rate also increased as the outlet area, injected air flow rate, and outlet height increased. But the circulation mass flow rate was not changed along with the external water level variation if the water level was higher than the outlet height.
A multi-channel gas leakage monitoring system was designed and fabricated by using CPLD(complex Programmable Logic .Device) for monitoring and controlling the leakage of natural gas from supplying-pipes under the ground. Fabricated SnO$_2$thick film gas sensor elements were attached on safeguard steel plate of natural gas supplying pipes, and the local monitoring system in this study received the signal from the gas sensors. The monitoring system was implemented by using CPLD chip to reduce the development time and implement simple one chip system. The time division multi-channel system received the input signal from individual gas sensor at one of divided times by multiplexor and signal processed sequentially. The system reduced the size of peripheral circuit resulted in implementation of creditable simple system.

It is essential to obtain accurate and highly reliable streamflow data for water resources planning, evaluation and management as well as design of hydraulic structures. A new discharge estimation method, which is named 'non-dimensional velocity distribution and index-velocity method,' was proposed in this research. This method showed very close channel discharges which were calculated with the exiting velocity-area method. When velocity-area method is used to estimate channel discharge, it is required to observe point velocities at every desired point and vertical using a current meter like Price-AA. However 'non-dimensional velocity distribution and index-velocity method' is used, it become optional to observe point velocities at every desired point and vertical. But this method can not be applied for the cases of very complex and strongly asymmetric channel cross-sections because non-dimensional velocity distribution by entropy concept may be quite biased from that of natural rivers.
Theoretical and numerical study on heat transfer and evaporation in the vertical channel has been carried out and basic correlations have been derived for the heat transfer evaluation of PCCS. Analysis program was developed with low-Reynolds-number k-$\varepsilon$ model and surface transfer rates were calculated for the turbulent natural convection in the vertical channel. In relation to dry cooling by buoyancy-driven air, first, the system parameters which govern overall heat transfer rate are determined through the adequate nondimensionalization procedure. After comparison with existing experimental data, numerical results are used to derive heat transfer correlation by sensitivity calculations. In relation to wet cooling by falling water film, numerical analysis are carried out for evaporation process with real film surface conditions and evaporation correlation is derived through analogy concept and correction factors.

A two-dimensional analytical model for deriving the threshold voltage of a short channel fully depleted (FD) cylindrical/surrounding gate MOSFET (CGT/SGT) is suggested. By taking into account the lateral variation of the surface potential, introducing the natural length expression, and using the Bessel functions of the first and the second kinds of order zero, we can derive potentials in the gate oxide layer and the silicon core fully two-dimensionally. Making use of these potentials, the minimum surface potential can be obtained to derive the threshold voltage as a closed-form expression in terms of various device parameters and applied voltages. Obtained results can be used to explain the drain-induced threshold voltage roll-off of a CGT/SGT in a unified manner.
We investigated the effect of organic materials on membrane fouling in advanced drinking water treatment by a hybrid module packed with granular activated carbon (GAC) outside multi-channel ceramic microfiltration membrane. Synthetic water was prepared with humic acid and kaolin to simulate natural water resouces consisting of natural organic matter and inorganic particles. Kaolin concentration was fixed at 30 mg/L and humic acid was changed as 2~10 mg/L to inspect the effect of organic matters. Periodic back-flushing using permeate water was performed for 10 sec per filtration of 10 min. As a result, both resistance of membrane fouling (Rf) and permeate flux (J) were influenced highly by concentration of humic acid. It proved that NOM like humic acid could be an important factor on membrane fouling in drinking water treatment. Turbidity and UV254 absorbance were removed up to above 97.4% and 59.2% respectively.
