Detection of CTX-M and Clonal Analyses using MLST in Cefotaxime Resistant $Escherichia$ $coli$ Isolated from the Han-River, Korea

  • Bacteria resistant to various antibiotics have recently become an issue of the utmost importance. Resistant strains are not uncommon, even in municipal drinking water sources. The health threat posed by resistant, pathogenic bacteria has serious ramifications for both public health and agriculture. In this study, we isolated antibiotic resistant bacteria from water samples from the Han River, Korea, which is contaminated by the wastewater from many industrial complexes, hospitals, agricultural and animal husbandry estates, and from wastewater treatment facilities. We determined the degrees of resistance to various antibiotics exhibited by the isolated strains. The similarities between the isolated $E.$ $coli$ strains were examined, using the pulsed field gel electrophoresis and multilocus sequence typing, in order to trace their origins and to explore the syntechnic adaptations and pathogenicity of the various strains and relate these to their genetic sequence. A total of 25 $E.$ $coli$ strains were isolated from six stations along the Han River. All the 25 strains exhibited resistance to ampicillin. We also investigated resistance to amoxicillin, clavulanic acid, cefazolin, cofoxitin, cefotaxime, cefpodoxime, ceftriaxone, cefepime, nalidixic acid, aztreonam, ciprofloxacin, streptomycin, gentamicin, chloramphenicol and imipenem. Based on the ESBL detection, 14 strains belonged to the ESBL producing strains. The number of the clonal complex producing strains was 5 among the 14 isolated strains. The 5 strains were included in the 168, 23, 38, 469, 156 clonal complex, respectively. The rest 9 strains were not included in the clonal complex, but showed independent STs.

Analysis on the Dynamics of Distribution and Eradication of Nutria on Nakdong River Area (낙동강 생태계에서의 외래종 뉴트리아 확산과 근절의 동태성 분석)

  • Nutria(Myocastor coypus) is one of the most notorious invasive species in many countries. The Nutria's original range was South America like Brazil, Chile and Argentina. But it has been introduced to North America, Europe, Africa, and Asia including Korea, by fur ranchers. Like most invasive and disturbing species, nutria not only destroys habitats but also impacts many native species throughout most of its range with destructive feeding behavior. The purpose of this study is to investigate the dynamics of survival mechanism, population increase and spatial distribution, and eradication effect of Nutria on Nakdong River Area with the systems thinking. The result of this study revealed that Nutria can survive during winter with the behavior that the colder the winter was, the more deeply it burrowed. Further, the study uncovered the dynamics that nutria has survived more and more strongly during the extreme cold winter in almost every region and their population has increased dramatically as a result of the uncontrolled reproduction. Finally, the result showed that the action delay in eradicating Nutria, which was originated from the information delay of perception of nutria's survival and reproduction structure, reinforced the spread of Nutria across the nation.

Monitoring of Hydrological and Water Quality in Dongjin-River Hengjeong Bridge Watershed for Agricultural Watershed Non-Point Pollutant Sources Management (농업유역 비점오염 관리를 위한 동진강 행정교 유역의 수문·수질 모니터링)

  • This study was performed to investigate the stream water quality characteristics in Hengjeong Bridge Basin of Dongjin River during twelve months from October, 2010 to September, 2011. Also, pollutant loads were calculated on the basis of the water quality and runoff results. The results showed that ranges of water temperature, pH and EC were $6.2{\sim}23.90^{\circ}C$, 6.32~7.78, $84.4{\sim}126.5{\mu}S/cm$ respectively. The Concentration of DO, BOD, COD, SS, Tot-N and Tot-P were observed as 6.80~9.20 mg/L, 0.40~1.60 mg/L, 1.96~4.41 mg/L, 59.60~142.20 mg/L, 1.28~3.52 mg/L, 0.001~0.07 mg/L respectively. Tot-N showed correlativity with BOD, and Tot-P showed correlativity with SS. The runoff pollutant loading of Tot-N and Tot-P were 117.94 kg/ha and 2.06 kg/ha respectively, in Hengjeong bridge of Dongjin river watershed. In the case of the correlativity between runoff pollutant loads and concentrations, Tot-N and Tot-P show low significant relationships.

  • Rainfall-Runoff Analysis in Whangryong River Basin was made using HEC-HMS and HEC-GeoHMS. The Basin was divided into three sub-basins using HEC-CeoHMS and GIS. Then, GIS input data were derived from each sub-basins. SCS CN runoff-volume model, Snyder's UH direct-runoff model, exponential recession baseflow model and Muskingum routing model in HEC-HMS were used to simulate the runoff volume using selected rainfall event and the parameters were optimized. Peak flowrate calculated using optimized parameters was compared to the observed flowrate in the basin. The result proved to be good agreement with each other. Optimized parameters in this local basin can be used to calculate the peak flowrate in the future.

The Evaluation of Water Quality in Coastal Sea of Kunsan Using Statistic Analysis (통계분석기법을 이용한 군산연안해역의 수질평가)

  • This study was conducted to evaluate water quality in coastal sea of Kunsan using multivariate analysis. The analysis data in Coastal Sea of Kunsan use of surveyed data by the NFRDI from April 2000 to November 2002. Twelve water Quality parameter were determined on each sample. The results was summarized as follow ; Water quality in coastal sea of Kunsan could be explained up to 62.782% by four factors which were included in loading of nitrogen-nutrients by Keum river(24.688%), suspended solids variation (12.180%), seasonal climate variation (18.367%) and variation of DIP (10.546%). To analyze spatially and monthly variation by factor score, it was divided by inner area and outer area spatially, and spring and summer monthly. The result of time series analysis by factor score, inner area of Kunsan coastal sea(St.1 and St. 2) was the most affected by nitrogen-nutrient and suspended solids due to runoff by Keum river. It could be suggested from these results that it is important to reduce tile pollution loads from Kuem river for the control of the water quality in coastal sea of Kunsan.

  • The objective of this study is to develope a predictive model for flood forecasting in the tidal reaches of the Nakdong river and to analyze the tidal effects of major flood forecasting station of the Nakdong river by using the hydraulic flood routing. In the calibration process the optimum roughness coefficients as functions of channel reach and discharge were determined and the calibration results suggest that the unsteady hydraulic flood routing model simulated with the optimum roughness coefficients showed close agreement between the calculated and observed stage.

Dynamics of Fish Larvae in the Han River Estuary and Kyunggi Bay, Korea

  • Species composition and abundance of fish larvae were studied from May 1988 through August 1989 in the Han River Estuary and adjacent Kyunggi Bay, Korea. Of 23 taxa identified, Coilia nasus, llisha elongata, and Cyprinidae spp. were dominated. Maximum density (3,5771/1,000m)$^3$ occurred in August 1988 and minimum (3/1,m$^3$) in February 1989. O1igo- ormesohaline species dominated during ebb tides while polyhaline species during flood tides. Correspondence analysis revealed that three distinctive species groups; oligohaline species, Coilia nasus, Ilisha elongata and Cyprinidae spp., which dominated at both channels of Kwanghwa Island (stations 1, 2), mesohaline species, Sardinella zunasi and Gobiidae spp., at the mouth of Yeomha Channel (station 3), and polyhaline species, Engraulis japonicus and Syngnathus schlegeli, in the middle of Kyunggi Bay (station 4). Coilia nasus was the most abundant species and reported first time in the study area. Given the species composition and density of fish larvae, the Han River Estuary is considered to be a major spawning and nursery ground for brackish water species such as Coilia nasus, Ilisha elongata and some Cyprinidae spp.

Monitoring algal bloom in river using unmanned aerial vehicle(UAV) imagery technique (UAV(Unmanned aerial vehicle)를 활용한 하천 녹조 모니터링 평가)

  • The purpose of this study is to evaluate the fixed wing type domestic UAV for monitoring of algae bloom in aquatic environment. The UAV used in this study is operated automatically in-flight using an automatic navigation device, and flies along a path targeting preconfigured GPS coordinates of desired measurement sites input by a flight path controller. The sensors used in this study were Sequoia multi-spectral cameras. The photographed images were processed using orthomosaics, georeferenced digital surface models, and 3D mapping software such as Pix4D. In this study, NDVI(Normalized distribution vegetation index) was used for estimating the concentration of chlorophyll-a in river. Based on the NDVI analysis, the distribution areas of chlorophyll-a could be analyzed. The UAV image was compared with a airborne image at a similar time and place. UAV images were found to be effective for monitoring of chlorophyll-a in river.

Evaluation of instream flow in Han river according to the Imnam dam operation in North Korea (북한 임남댐 운영에 따른 북한강 하천유지유량 평가)

  • The objective of this study is to evaluate the instream flow in the North Han River basin according to the operation of Imnam Dam in North Korea. The water budget and instream flow satisfaction were analyzed using hourly, daily and monthly data of Water Management Information System (WAMIS) from Jan. 1991 to Dec. 2018. As a analysis result of water budget using hourly data in the North Han River basin, although inflows compared with dam release in the upstream basin of Peace Dam-Hwacheon Dam and Chuncheon Dam-Soyanggang Dam-Uiam Dam were calculated as negative values, the reasonable results using daily and monthly average data were estimated. It showed that the results of water budget analysis of dam inflow and total release may be different by time units of data. The monthly average inflow of Hwacheon Dam decreased significantly after the construction in 2003 of Imnam Dam, which confirmed that the operation of Imnam Dam had a significant effect on the dams in the North Han River basin. The operation of Imnam Dam is one of the main reasons for the lack of instream flow and total shortage amounts and shortage period increased up to +330% due to the decrease in inflow and total release of dams in the North Han River water after the operation of Imnam Dam. It is necessary to study various plans to secure instream flow including transboundary river management

A Correlation Analysis of the River Naturalness and Water Quality for Biological Habitat Evaluation (하천 생물 서식처 평가를 위한 하천 자연도와 수질의 상관성 분석)

  • In this study, the analysis of river naturalness and water quality were executed in the major rivers of the Nakdong River Basin. As a result, the assessment index of the General Evaluation was 1.428 to 4.107 as $1^{st}$ to $4^{th}$ grades. The River Shape was 1.929 to 4.429 as $2^{nd}$ to $4^{th}$ grades and the River Environment was 1.774 to 3.643 as $1^{st}$ to $4^{th}$ grades. As well, evaluation of water quality showed that concentration of pH was 7.102 to 8.497 mg/l, BOD was 0.748 to 5.271 mg/l, DO was 5.077 to 12.335 mg/l and 55 was 3.658 to 19.960 mg/l. The correlation between river naturalness and water quality was analyzed to investigate similarity and independence of river naturalness evaluation index. It was shown that coefficient of correlation was low with value of -0.1503 to -0.5886, therefore, was evaluated as independent.
