Vertical Distribution of Vascular Plants in Namdeogyusan, Mt. Deogyu National Park by Temperature Gradient (덕유산국립공원내 남덕유산 관속식물의 고도별 온도구배에 따른 수직분포)

  • In order to investigate vertical plant distribution on Namdeogyu of Mt. Deogyu National Park by temperature change, vascular plants of nine areas with 100-meter-high difference were surveyed from the Deogsangyo (650m alt.) to the Namdeogyusan (1,507m alt.). A total of 455 taxa belonging to 99 families, 280 genera, 402 species, 5 subspecies, 43 varieties, 4 forms and 1hybrid were vegetated on survey areas. Around 700 m high did species diversity of vascular plants decrease rapidly. The Detrended correspondence analysis (DCA) divided distribution of vascular plants into five groups; areas below 700m alt., 700~1,100m alt., 1,100~1,300m alt., and areas above 1,300m alt.. These results showed that vegetation of vascular plants on investigated areas has high correlation with climate elements of temperature. Vascular plants should be crowded within their own optimal ranges of vegetation. Climate change would result in shift of these distribution ranges, and thus vegetation shift will be happened accordingly.

Morphometric Characteristic between Diploid and Spontaneous Triploid Carp in Korea

  • We used flowcytometry to ploidy verification after that investigate difference between diploid and spontaneous triploid through the truss dimension and classical dimension at crucian carp, Carassius auratus, crucian carp C. cuvieri and common carp, Cyprinus carpio collected from Hangang river, Hantangang river, Imjingang river, Geumgang river, Yeongsangang river and Nakdonggang river, Korea. There were significant differences among the three species for the truss dimensions anterior origin of dorsal fin $(2){\times}$ anterior origin of anal fin (5), $2{\times}$ anterior origin of pelvic fin (6), $2{\times}$ origin of pectoral fin (7), posterior origin of dorsal fin $(3){\times}5$, $3{\times}6$, and $3{\times}7$ (P<0.05). There were no significant differences among the three species in the truss dimensions dorsal fin length ($2{\times}9$) and eye diameter (ED)(P>0.05). On the other hand, there were no significant differences in the several classical dimensions of each species (P>0.05). Three classical dimensions, most anterior extension of the head $(1){\times}2$, $1{\times}6$ and $2{\times}$ most posterior scale in lateral line (4) did not differ between the C. auratus diploid and spontaneous triploid (P>0.05). Two classical dimensions, $1{\times}6$ and longest length between most anterior extension of the head and gill cover ($1{\times}8$) did not differ between the C. cuvieri diploid and spontaneous triploid (P>0.05). One classical dimensions, $1{\times}2$ did not differ between the common carp diploid and spontaneous triploid (P>0.05). There were significant differences in the each diploid and triploid species (P<0.05). These results suggest that the classification of each species and classification between diploid and spontaneous triploid morphometrical parameters used in this study are useful indices of morphometrical status in the each species from major river of Korea.

  • Blooms of the small centric diatom Stephanodiscus is quite occasional in winter season in temperate freshwater ecosystems. Often, it leads to degradation of water quality and affects quality of supplied drinking water. In previous studies, naphthoquinone (NQ) compounds have been shown to be effective and selective for controlling winter bloom species Stephanodiscus hantzschii. We conducted a 5 ton scale mesocosm experiment to investigate the effects of NQ on native Stephanodiscus sp. collected from Nakdonggang River in water. After treatment with NQ 4-6 compound (0.2 μM), the cell density of Stephanodiscus sp. was rapidly reduced from 5 × 103 cells mL-1 to 0.2 × 103 cells mL-1 for 10 days. Additionally, NQ 4-6 compound did not affect physicochemical factors (water temperature, dissolved oxygen, pH, conductivity, nutrients) and biological factors (bacteria, heterotrophic nanoflagellates, zooplankton). Therefore, these findings suggest that the NQ 4-6 compound has potential as an alternative algicidal substances to effectively mitigate natural Stephanodiscus sp. blooms, and the application of NQ 4-6 compound will restore the healthy aquatic ecosystems.

The study for grading of the mudflat by birds (조류에 의한 갯벌의 등급화 연구)

  • This study was carried out to grade and evaluate the intertidal mudflat by birds. The mudflat was graded by two methods such as the scoring and ranking. First, for the scoring of the mudflat, five factors were estimated; population size, the number of protected species, the individuals of protected birds, the number of species(or subspecies) over 1% level of the survival population, and 1% level of the domestic population. After scoring of these five factors, the grade was scored by total. Second, for the ranking, six factors were estimated; population size, the density of birds, the index of species diversity, species richness, species evenness, and population stability. After ranking of these six factors, the grade was relatively ranked. Five grade by scoring was the important mudflat for the value of international preservation, and the most mudflats of the estuary gained V grade. The higher grades by ranking were the mudflats of Nakdonggang and Mankyunggang river estuaries. And the inner mudflats were more valuable than the outer mudflats in Mankyunggang & Dongjingang river estuaries. The grading of the mudflat by birds can be used for estimating and appointing of the important mudflats, and suggested the objective of criteria for the effective preservation and management.

Estimating Spatio-Temporal Distribution of Climate Factors in Andong Dam Basin (안동댐 유역 기상인자의 시공간분포 추정)

  • This study investigates characteristics of time series spatial distribution on climate factors in Andong Dam basin by estimating precise spatio-temporal distribution of hydro-meteorological information. A spatio-temporal distribution by estimating Semi-Variogram based on spatial autocorrelation was examined using the data from ASOS and 7 hydro-meteorological observatories in Andong Dam basin of upper Nakdonggang River, which were installed and observed by NIMR(National Institute of Meterological Research). Also, temperature and humidity as climate variables were analyzed and it was recognized that there is a variability in watershed area by time and months. Regardless of season, an equal spatial distribution of temperature at 14 o'clock and humidity at 10 o'clock was identified, and nonequal distribution was noticed for both variables at 18 o'clock. From monthly spatial analysis, the most unequal distribution of temperature was seen in January, and the most equal distribution was detected in September. The most unequal distribution of humidity was identified in May, and the most equal distribution was seen in January. Unlike in forest, seasonal spatial distribution characteristics were less apparent;but temperature and humidity had respective characteristics in hydro-meteorology.

Production of Lactulose by Biological Methods and Its Application (생물학적 방법을 통한 기능성 이당 lactulose의 생산과 응용 연구)

  • Lactulose (4-O-${\beta}$-D-galactopyranosyl-D-fructose) is a non-digestible synthetic ketose disaccharide which can used in food and pharmaceutical fields due to its useful functions for encephalopathy, chronic constipation, hyperammonemia, etc. Therefore, the lactulose is regarded as one of the most important disaccharides and have been concentrated much interesting as an attractive functional material in the current industry. From this reason, the research related on the production of lactulose has been carried out various academic and industrial research groups. To produce lactulose, two main methods, chemical production and enzymatic production have been used. Commercially lactulose produced by alkaline isomerization of lactose as chemical production method but it has many disadvantages such as rapid lactulose degradation, purification, and waste management. From these reasons, lactulose produced by enzymatic method which solves these problems has been suggested as a proper method for lactulose production. Two different enzymatic methods have been reported as methods for lactulose production. Lactulose can be obtained through hydrolysis and transfer reaction catalyzed by a ${\beta}$-galactosidase which requires fructose as co-substrate and exhibits a low conversion. Alternatively, lactulose can be produced by direct isomerization of lactose to lactulose catalyzed by cellobiose 2-epimerase which requires lactose as a single substrate and achieves a high lactulose yield. This review summarizes the current state of lactulose production by chemical and biological methods.

A new combination for Saxifraga octopetala (Saxifragaceae) and its phylogenetic relationship (구실바위취의 신조합명 및 계통 유연관계)

  • This study was conducted to address the taxonomic status of the Korean endemic species Saxifraga octopetala, which is sometimes considered conspecific to Micranthes manchuriensis. Extensive molecular phylogenetic analysis using nrITS sequences as well as morphological examination of type specimens of the two species were undertaken to ascertain the phylogenetic position and species delimitation of S. octopetala. In the resulting nrITS trees, a total of 65 accessions representing S. octopetala grouped together and nested within the Micranthes clade, exhibiting a close relationship with M. nelsoniana and M. manchuriensis. Multiple accessions of M. manchuriensis collected from China and Russia also formed a clade, showing a sister group relationship with M. nelsoniana var. pacifica and M. fusca. The ambiguous species entity of S. octopetala is thought to have originated from Nakai's misinterpretation of Wilford's collection (type specimens of M. manchuriensis), which is a complex collection including an inflorescence of M. nelsoniana. In spite of apparent morphological similarity between S. octopetala and M. manchuriensis, they differ in the presence and absence of underground stolons. The distinct position of S. octopetala within the Micranthes clade on the nrITS tree suggests that it should retain species status in Micranthes. Thus a new combination (Micranthes octopetala) is proposed.

Spatial and temporal variation on fruit set in Epipactis thunbergii (Orchidaceae) from southern Korea (한국남부 자생 닭의난초 (난초과)의 시 공간에 따른 결실률 변이)

  • Spatio-temporal variation in fruit set in orchids would affect long-term population viability and will influence genetic diversity over many generations. The aim of this study was to examine the breeding system of the nectariferous terrestrial orchid Epipactis thunbergii, to specifically determine levels of fruit set in terms of time and space under natural conditions. We examined pollination under natural conditions and conducted hand pollination experiments during a 2-year survey in four populations located along 1.5 km of coastal line in Jinguiri (rual village) [Jeollanam-do (province), southern Korea]. We found that, over a 2-year period, levels of percentage of fruit set were similar within patches of the four populations. By contrast, we detected significant differences in the percentage of fruit set among patches. We also found that plants with larger inflorescence size produced significantly more fruits than plants with fewer flowers. Over a 2-year period, the percentage of fruit set for E. thunbergii was similar but low (14.1%) compared to that averaged for eighty-four rewarding species (37.1%). However, an increase in fruit set was achieved by hand-pollinations: artificial self-pollination (90.5-95.2%), artificial geitonogamy (94.7-95.0%), and cross-pollination (artificial xenogamy, 91.3-91.4%). No emasculated flowers produced fruits and no automatic pollination was found in E. thunbergii. Our findings suggest that E. thunbergii is a self-compatible terrestrial orchid that depends on pollinators (insects) to achieve fruit set in natural habitats, and that local environmental conditions were similar over a period of 2 years in the study area. Our results also highlight the cryptic variation of fruit production in time, but more pronounced variability in space.

Population Genetic Structure of the Korean Endemic Species, Iksookimia pacifica (Pisces: Cobitidae) Distributed in Northeast Korea (한국고유종 북방종개(어류강, 미꾸리과)의 집단유전학적 구조)

  • Population genetic studies of 10 groups of Iksookimia pacifica were conducted to investigate the genetic diversity and population genetic structure across its known range in South Korea. Population DNA sequences of one mitochondrial gene (mtCOI) and three nuclear genes (IRBP, EGR2B, RAG1) were examined in samples collected from ten streams that flow into the East Sea. Both mitochondrial and nuclear sequences exhibited significant differentiation among populations except a few cases. The Bayesian analysis of the multi-locus genotypes inferred from the DNA sequences of nuclear genes clustered the individual fish largely into two geographical groups: a northern group (from Baebong stream to Cheonjin stream) and a southern group (Yangyangnamdae stream to Gangneungnamdae stream). Given that the streams flowing into the East Sea are geographically isolated water systems, such separation of genotypes can be interpreted by the geographical separation of common ancestors into north and south that had colonized South Korea. Since the initial geographical separation of the ancestral population by north and south, the ancestral groups seem to have experienced further differentiation into the current genetic clusters through the physical isolation of streams by the East Sea in each region. It is notable that many individuals in the Jasan stream formed a genetic cluster with those of Yangyangnamdae and Gangneungnamdae streams which are distant from each other. In addition, mitochondrial gene showed low genetic differentiation between some neighboring populations and very low level of genetic diversity in several populations. The present population genetic study will provide valuable information for the conservation and management of the Korean endemic fish species, I. paicifica.

Gibberellin A7 production by Aspergillus tubingensis YH103 and cultural characteristics of endophytic fungi isolated from Tetragonia tetragonoides in Dokdo islands (독도 번행초에서 분리된 내생균류의 배양적 특성과 Aspergillus tubingensis YH103의 gibberellin A7의 생산)

  • Coastal plant species Tetragonia tetragonoides (Pall.) Kuntze native to the Dokdo islands was sampled and then 17 endophytic fungi were purely isolated based on morphological differences. The fungal isolates were characterized by their growth properties under NaCl concentration or pH gradient. Culture filtrates of the 17 fungal isolates were treated to Waito-c rice (WR) seedlings for verifying plant growth-promoting activity. As the results, YH103 strain showed the highest plant growth-promoting activity among them. Phylogenetic analysis of the isolates was done by the maximum likelihood method based on partial internal transcribed spacer region (ITS region: contaning ITS1, 5.8S, and ITS2), beta-tubulin (BenA), and calmodulin (CaM) gene sequences. Chromatographic analysis of the strain YH103 culture filtrate showed the existence of gibberellins ($GA_4$, $GA_7$, $GA_8$, and $GA_{19}$). Finally, the strain YH103 was identified as Aspergillus tubingensis by microscopic observation and molecular analysis and, to our knowledge, this is the first report of GAs producing A. tubingensis.
