Nanofood and Its Materials as Nutrient Delivery System (NDS)

  • Incorporation of bioactive compounds such as vitamins, probiotics, bioactive peptides, and antioxidants into Nutrient Delivery System (NDS) for 'nanofood' provides simple way to develop novel functional foods that may have physiological benefits or reduce risks of diseases. As vital nutrient in nanofood, proteins possess unique functional properties including ability to form gels and emulsions, which allow them to be ideal nanofood materials for encapsulation of bioactive compounds. Based on protein physico-chemical properties, this review describes potential role of nanofood materials for development of NDS in hydrogel form, micro-or nano-particles. Applications of these nanofood materials to protect delivery-sensitive nutraceutical compounds are illustrated, and impacts of particle size on release properties are emphasized.

An Experimental Study on Estimate of the Optimal Grout Injection Ratio for Stabilization of Mudstone fill (이암 성토지반의 안정화를 위한 최적주입률 산정에 관한 실험적 연구)

  • Water glass chemical grouts are primarily used in ground injection projects. Natural and Durable Stabilizer (NDS), Space Grout Rocket (SGR), and other similar materials composing of inorganic accelerating agents with ultra fine cement have been gaining popularity as ground improvement material in South Korea. However, there are questions as to grouting results and environmental issues caused by NDS grout. This study uses the injection method in mudstone embarkment to evaluate the differences in strength, permeability, and optimum injection volume through the use of uniaxial and triaxial compression tests and fish poison tests for NDS and SGR materials. After 28 days, results showed SGR and NDS to have a 50% increase in strength and 50% decrease in permeability with the proper injection rate at 50%.

A Sense Amplifier Scheme with Offset Cancellation for Giga-bit DRAM

  • To improve low sense margin at low voltage, we propose a negatively driven sensing (NDS) scheme and to solve the problem of WL-to-BL short leakage fail, a variable bitline reference scheme with free-level precharged bitline (FLPB) scheme is adopted. The influence of the threshold voltage offset of NMOS and PMOS transistors in a latch type sense amplifier is very important factor these days. From evaluating the sense amplifier offset voltage distribution of NMOS and PMOS, it is well known that PMOS has larger distribution in threshold voltage variation than that of NMOS. The negatively-driven sensing (NDS) scheme enhances the NMOS amplifying ability. The offset voltage distribution is overcome by NMOS activation with NDS scheme first and PMOS activation followed by time delay. The sense amplifier takes a negative voltage during the sensing and amplifying period. The negative voltage of NDS scheme is about -0.3V to -0.6V. The performance of the NDS scheme for DRAM at the gigabit level has been verified through its realization on 1-Gb DDR2 DRAM chip.

A Study on Durability and Impermeability of Environmentally Friendly Inorganic Ground Injection Material (환경 친화적인 무기질계 지반주입재의 내구성 및 차수효과에 관한 연구)

  • Recently, the ground injection method using water glasses as one of the main resources and the products of these constructions have basic problems in terms of the method of constructions for the permanent foundation reinforcement and stopping leakage of water because they have some serious problems such as durability, compressive strength, injectant eluviation and so forth even though they are still used to stop leakage of water in the temporary structures. The purpose of this study is to demonstrate the strength characteristic and environment friendliness of NDS method by unconfined compressive strenth test, permeability test, length change test, leaching test, and assessment of environmental impact in comparison water glass type material. The test results show that NDS method has significant improvement of strength, permeability, volume change, and leaching. An assessment of environmental impact also demonstrates that the NDS material is environmentally friendly.

Strength Development Mechanism of Inorganic Injection Material (무기질계 주입재의 강도발현 메커니즘)

  • Recently, NDS(Natural and Durable Stabilizer)method and other similar methods are composed of inorganic accelerating agent and the ultra-super fine cement have been studied as the ground improvement material in Korea. However, in the existing research, the chemical changing process of NDS in the strength development mechanism with the elapsed curing time and the principles of strength development did not give an explanation. For the popularization of the inorganic grout material, it determined that the mechanism verifying of the curing process had to be clearly preceded. Therefore, unconfined compression test, SEM and XRD analysis were performed by the elapsed curing time and were analyzed. In addition, the same trial for SGR method, that is the representative example of the water glass grout material, was selected as comparative target in order to distinguish properties of NDS more clearly. The result of experiment, the strength development mechanism of NDS could be investigated through the close correlation of the unconfined compression strength - SEM - XRD analysis, and excellence of a performance was confirmed.

Development and Characterization of a Hydrolyzed Goat Milk Protein/Chitosan Oligosaccharide Nano-Delivery System (산양유 단백질 분해물/키토올리고당 나노 전달체 제조 및 물리화학적 특성연구)

  • The aims of this study were to manufacture a hydrolyzed goat milk protein (HGMP)/chitosan ologisaccharide (CSO) nano-delivery system (NDS) and to investigate the effects of production variables, such as sodium tripolyphosphate (TPP), HGMP, and CSO concentration levels, on the formation and physicochemical properties of the NDS. An HGMP/CSO NDS was produced using the ionic gelation method at pH 5.5. Transmission electron microscopy and a particle size analyzer were used to determine the morphological and physicochemical properties of NDSs, respectively. The size of the HGMP/CSO NDS decreased from 225 to 138 nm as HGMP and CSO concentration levels decreased. The NDS had a positive surface charge, with a zeta-potential value of +23 mV. The encapsulation efficiency (EE) of docosahexaenoic acid was enhanced as the HGMP concentration level increased. Additionally, increasing the concentration level of CSO resulted in an increase in the EE of resveratrol. The HGMP/CSO NDS exhibited good physical stability during freeze-drying. Thus, our findings showed that the HGMP/CSO NDS was successfully manufactured and that HGMP and CSO concentration levels were key factors affecting the physicochemical properties of the NDS.

Comparison of the Biological Activities of Electrodialysis-desalted Bioactive Compounds from the Halophyte Suaeda japonica (전기투석시스템을 활용한 염생식물 칠면초(Suaeda japonica) 추출물의 염분 표준화에 따른 주요 성분 및 생리활성 비교 연구)

  • To effectively remove salts from Suaeda japonica, extracts, an electrodialysis system was developed. The biological activities of non-desalted (NDS) and desalted S. japonica (DS) extracts were compared. The DS extract exhibited superior polyphenolic (6.26%) and carbohydrate (28.56%) contents. The IC50 values of the DS extract against DPPH radicals and hydrogen peroxide were 0.22 and 0.68 mg/mL, respectively, which was higher than that of the NDS extract. Neither the DS nor the NDS extract was cytotoxic in RAW 264.7 macrophages. Additionally, the DS extract had a higher NO inhibitory effect compared to the NDS extract in lipopolysaccharide (LPS)-activated RAW 264.7 macrophages. These data indicate that DS extracts have greater biological activity than do ND extracts, and application of the electrodialysis process may be useful in marine bioresource applications.

Polymorphic Diversity of UBX Domain D from cDNA Isolated from Pectoral Muscle of Korean Native Chicken

  • The objectives of this study are to identify specific functional genes which are related with growth and protein structure of the pectoral muscle of Korean native chicken. Pectoral muscle was isolated from three Korean native chickens (KNC, red brown, 12 months old, 2.41 ${\pm}$ 0.24 kg) and three Cornish chickens (16 month old, 2.76 ${\pm}$ 3.0 kg). The subtraction cDNA library was prepared in PCR4 Blunt-TOPO vector. The DNA sequence homology was compared with other breeds and species in GenBank. A clone NDS-81 was found to be unique for the DNA sequence homology with UBX family. Their partial sequence has high homology (98%) with chicken UBX domain D. Chicken UBX domain has chicken (93%), cattle (68%), dog (67%), mouse (64%) and, human (63%) nucleotide sequence homology. Several regions were mutated from T in chicken to C or G in the NDS-81 clone. The first site is LAD in chicken, but it was expressed as (L)RM in clone NDS-81. In this site, amino acids were changed from Ala to Arg, and from Asp to Met. The second site was changed from ER (Arg) in chicken to ED (Asp) in clone NDS-81. They are both containing functional side chains and play an important role in binding other proteins. Therefore, the clone NDS-81 could be a different candidate gene for the UBX family gene and could related with pectoral muscle structure of Korean native chicken.

Precise assembly and regulation of 26S proteasome and correlation between proteasome dysfunction and neurodegenerative diseases

  • Neurodegenerative diseases (NDs) often involve the formation of abnormal and toxic protein aggregates, which are thought to be the primary factor in ND occurrence and progression. Aged neurons exhibit marked increases in aggregated protein levels, which can lead to increased cell death in specific brain regions. As no specific drugs/therapies for treating the symptoms or/and progression of NDs are available, obtaining a complete understanding of the mechanism underlying the formation of protein aggregates is needed for designing a novel and efficient removal strategy. Intracellular proteolysis generally involves either the lysosomal or ubiquitin-proteasome system. In this review, we focus on the structure and assembly of the proteasome, proteasome-mediated protein degradation, and the multiple dynamic regulatory mechanisms governing proteasome activity. We also discuss the plausibility of the correlation between changes in proteasome activity and the occurrence of NDs.
