Effect of SPZZC, a Composition of Herb Extracts, on Atopic Dermatitis in BALB/c and NC/Nga Mouse (BALB/c 및 NC/Nga 마우스의 아토피성 피부질환모델에서 생약조성물 SPZZC의 치료효능)

  • The aim of this study was to evaluate if a composition of herb extracts, PLX-PLS was effective to treat atopic dermatitis (AD) in mice. SPZZC is a composition of herb extracts containing the roots of Scopolia parviflora and Paeonia lactiflora, the herb of Zizania caudiflora, the fruit of Ziziphus jujuba and the leaf of Chinese arborvitae. AD in BALB/c mouse was induced by patching ovoalbumin on the backside, while it in NC/Nga mouse was induced by repeated application of 1-chloro 2,4-dinitrobenzene (DNCB). Mice were topically treated with SPZZC or Domohorn ointment on the backside for 2 weeks (BALB/c) or 1 week (NC/Nga). Scratching behavior, clinical skin severity and the levels of WBC, neutrophil, eosinophil and total serum IgE were measured. After AD induction, scores of scratching behavior and clinical skin severity and the levels of WBC, neutrophil, eosinophil and total serum IgE were increased. Treatment with SPZZC significantly decreased scores of scratching behavior and clinical skin severity in a dose dependent manner in NC/Nga and BALB/c mice. Treatment with SPZZC 2% significantly decreased also the levels of WBC, neutrophil, eosinophil and total serum IgE. Especially, treatment of SPZZC 2% reduced more rapidly score of clinical skin severity than clobetasol cream. These results suggest that the SPZZC may be an alternative substance for the management of AD.

The Effect of Essential Oil on Atopic Dermatitis Model of NC/Nga Mice (라벤더, 로즈마리, 레몬밤을 이용한 생쥐의 아토피피부염모델에 대한 효과)

  • Purpose: This study was aimed to evaluate the effect of blending oils on atopic dermatitis (AD) model of NC/Nga mice. Methods: Adult NC/Nga mice were assigned to 1 of 3 groups: control group (C, n=8), experimental group [AD-induced group, blending oil-treated group (E=8, 8)]. Mice in the E group were given a treatment of blending oils such as Lavender, Rosemary and Lemon Balm (LRL) for 2 sessions (every day) for 6 days. Results: The AD-induced mice with LRL blending oils treatment showed a significant decrease in epidermal thickness, number of mast cells and degranulation, expression of TNF-${\alpha}$ and scoring of sensual assessment. Conclusion: LRL blending oils may be a putative resource for the cure of or treatment of AD by the diminution of AD-pathological factors such as the epidermal thickness, the number of mast cells and degranulation as well as the expression of TNF-${\alpha}$.

Cheonggi-san Inhibits Atopy Dermatitis in NC/Nga Mouse through Regulation of iNOS mRNA Expresssion & NO production (청기산(淸肌散)의 iNOS 발현과 NO 생성 억제가 NC/Nga 생쥐의 아토피 피부염에 미치는 영향)

  • Inducible nitric oxide synthase (iNOS) are important inflammation enzyme and severe up-nitric oxide (NO) production by this enzyme has been intricate with pathogenesis of atopy dermatitis. The present study was designed in order to determine whether Cheonggi-san could inhibit atopy dermatitis through modulation of iNOS mRNA expression and NO production. We found that iNOS mRNA expression and NO production in RAW 264.7 macrophages stimulated with lipopolysaccharide dose-dependantly decreased by Cheonggi-san extract treatment (0.5 - 2.0 mg/ml). The distribution of iNOS positive reacted cell in NC/Nga mice with atopy dermatitis were decreased by Cheonggi-san extract treatment (2.5 ml/kg/day) and apoptosis were increased. These data likely indicate that Cheonggi-san may act as inflammatory regulator for atopy dermatitis and may be possible to develop useful agent for chemoprevention of NO intricate inflammatory diseases.

Lonicera Japonica Inhibits Atopy Dermatitis in NC/Nga Mouse through Regulation of iNOS by NF-${\kappa}B$ Suppression (금은화의 NF-${\kappa}B$ 활성 억제를 통한 iNOS 조절이 NC/Nga 생쥐의 아토피 피부염에 미치는 영향)

We found that IKK mRNA and iNOS mRNA expression in RAW 264.7 macrophages stimulated with lipopolysaccharide dose-dependantly decreased by Lonicera japonica (0.4 - 1.0 mg/$m{\ell}$) and NO production decreased. The distribution of NF-${\kappa}B$ p65 and iNOS positive reacted cell in NC/Nga mice with atopy dermatitis were decreased by Lonicera japonica (45 mg/kg/day) and apoptosis were increased. These data likely indicate that Lonicera japonica may act as inflammatory regulator for atopy dermatitis through iNOS modulation by NF-${\kappa}B$B suppression and may be possible to develop useful agent for chemoprevention of NO intricate inflammatory diseases.

The Effects of Samulsopungeum and Prednisolone on NC/Nga Atopic Mice (사물소풍음(四物消風飮)과 Prednisolone이 NC/Nga mice의 아토피 피부염에 미치는 영향)

  • Objectives : Atopic dermatitis(AD) is a chronic recurrent skin disease which usually developed in infancy or childhood. AD often repeat improvement and relapse. The cause of AD is so indefinite that many methods of therapies(moisturizer, steroid ointment, antihistamine, immunomodulator, immunosuppressant, herbal medicine, alternative medicine, etc.) are tried. Recently, a lot of studies were made. But there is no report about the effect of Samulsopungeum(SM) and Prednisolone(PN) on AD. So, author aimed to investigate the effects of SM and PN on AD of NC/Nga mice. Methods : Thirty two mice(8 Balb/c mice and 24 NC.Nga mice) were divided into four groups; Balb/c mice was normal group. NC/Nga mice were divide into three group : control, PN, SM group. AD was induced in the control, PN, SM group by spreading DNCB. Then normal saline, PN and SM were orally administered three times in a week for 8 weeks to the control, PN, SM group, respectively. We observed changes of clinical skin severity score, serum IgE, IgG1, IFN-$\gamma$, IL-2, IL-4, IL-5, IL-10, IL-13, and so on. We used one-way ANOVA test statistically(p<0.01). Results : The clinical skin severity scores of PN group and SM group in 8th week were decreased compared to the control group. Serum IgE, IgGl levels of PN group and SM group were significantly decreased compared to the control group. Serum IFN-$\gamma$ in SM group was significantly increased compared to the control group. But, Serum IFN-$\gamma$ in PN group was significantly decreased compared to the control group. Serum IL-10 levels of PN group and SM group were significantly decreased compared to the control group. Serum IL-2, IL-4, IL-5, IL-13 levels of PN group and SM group were significantly decreased compared to the control group. mRNA expression levels of IL-2, IL-4, IL-5, IL-13 in the dorsal skin tissues of PN group and SM group were significantly decreased compared to the control group. According to biopsy reports of the ear and skin tissues showed that the tissue damage of PN group and SM group were highly reduced compared to the control group. Creatinine, BUN, ALT, AST levels of PN group and SM group were normal. Conclusion : According to the above results, it is considered that SM is effective treatment for the AD.

Effects of PGA-LM on CD4+CD25+foxp3+ Treg Cell Activation in Isolated CD4+ T Cells in NC/Nga Mice (NC/Nga 생쥐에서 분리한 T 세포에서 foxp3+ 세포 활성화에 대한 PGA-LM의 효과)

  • Poly-$\gamma$-glutamic acid ($\gamma$-PGA) was mixed natural flora of Bacillus subtilis, contaminated from cooked soybeans. Also, it was performed to find out the antiallergic activity by using NC/Nga mice, in vitro. The $\gamma$-PGA (PGA-HM : PGA-high molecular weight), Molecular weight 300 kDa, was decomposed and made PGA-LM (PGA-low molecular weight) which has molecular weight below 30 kDa by sonication. Therefore, it was same result between PGA-HM and PGA-LM, and reported PGA-LM as basic result. We found that PGA-LM contains antiallergic efficacy that inhibit B cells and Th2 cells activation from isolated CD4+T cells in NC/Nga atopic dermatitis model mice, and not show a cytotoxicity in the hFCs. To investigate the effects of these PGA-LM in vitro, isolation of splenic B cell and CD4+ T cells in atopic dermatitis mice were used. To elucidate the role of PGA-LM in anti-CD40+ interleukin-4 (IL-4)-mediated B-cell activation, showed that the capacity of B cells to expression IL-$1\beta$, IL-6, and TNF-$\alpha$ mRNA down-regulated, and IL-10 mRNA up-regulation by PGA-LM treatment, but it had no effect on TGF-$\beta$ expression. In addition to CD4+IFN-$\gamma$+ and CD4+CD25+foxp3+, the functions of PGA-LM in the development of the CD4+CD25+foxp3+ and CD4+IFN-$\gamma$+cells, the phenotype and functions of PGA-LM induced CD4+CD25+foxp3+, and CD4+IFN-$\gamma$+cells in CD4+T cells. These results suggested that PGA-LM could change cytokine production and generate CD4+CD25+foxp3+ Tregs in NC/Nga mice, and may be effective for immunotherapy in patients with AD.

Effects of SPDJTK(SoPungDoJeokTangKami) and Concurrent Administration of AJ (Atopy cream, Jawoongo) Plus SPDJTK on Atopic Dermatitis-like Skin Lesions in NC/Nga Mouse Induced by BMAC (아토피양(樣) 피부염 NC/Nga 생쥐에서 소풍도적탕가미(消風導赤湯加味)와 아토피크림, 자운고(紫雲膏) 및 소풍도적탕가미(消風導赤湯加味)의 병용투여가 피부염에 미치는 영향)

  • Objectives The purpose of this study is to investigate the effect of SPDJTK(SoPungDoJeokTangKami) and concurrent administration of AJ(Atopy cream, Jawoongo)+SPDJTK on atopic dermatitis-like skin lesions by using in NC/Nga atopic dermatitis mouse induced by BMAC-induced mice. Methods Clinical skin score, hematology and Serum total IgE and IgG1 of NC/Nga atopic dermatitis mice were evaluated. Moreover, the cytokine level, total cell number, Immunohistochemical staining and Histological features of axillary lymph node(ALN), draining lymph node(DLN), peripheral blood mononuclear cells(PBMCs) and dorsal skin tissue were used in NC/Nga mice. Results Orally administrated SPDJTK with concurrent administration of SPDJTK and AJ decreased the clinical skin score, total cell number of WBC, eosinophils in blood, serum total IgE & IgG1, IL-5, IL-13, IFN-$\gamma$. Also, total cell number of ALN and dorsal skin tissue, absolute cell number of CD4+, CD8+, CD3+CD69+, CD3+CCR3+, CCR3+, CD4+CXCR5+ in ALN, absolute cell number of CD3+CCR3+, CCR3+ in DLN, granulocytes in PBMCs, activation cell number of CD3+CD69+, CCR3+, total cell number of CD3+ T cell in dorsal skin tissue were significantly decreased. Furthermore, thickness of epidermis, infiltrated inflammatory immune cell and mast cell in dermis, amount of Eotaxin2 mRNA, CCR3 mRNA in dorsal skin tissue, gene expression of IL-5, IL-13 mRNA in ALN, CD4+ Th cell in dorsal skin tissue and CCR3+ eosinophils in ALN were all significantly decreased. However, total number of DLN, absolute number of CD3e+ T cell and CD19+ B cell, absolute number of CD4+, number of Th cell in DLN and gene expression of foxp3 mRNA were significantly increased significantly. Conclusions Concurrent administration of SPDJTK and AJ on atopic dermatitis in NC/Nga atopic dermatitis mouse was very effective treatment for atopic dermatitis.

Effects of Houttuyniae Herba Complex Prescription on Atopic Dermatitis in NC/Nga Mice (어성초복합방(魚腥草複合方)이 NC/Nga mouse 아토피 병태 모델의 관련 면역 세포 및 IgE 생성량에 미치는 영향)

  • To examine the effects of HHCP on atopic dermatitis and its various immunopathologic parameters was induced by DNCB in NC/Nga mice and the animals were orally administrated with HHCP. We summarized the results obtained from serum levels of IgE and the numbers of various immune cells as follow. HHCP has no cytotoxic effects at the range of concentration (1-400 ${\mu}g$/ml) on fibroblast isolated from lung of BALB/c mice. HHCP significantly lowered the serum levels of IgE compared with control at 16 and 20 week. HHCP significantly reduced the number of CD19$^+$ cell in spleen and DLN, as well as the number of B220$^+$ /IgE$^+$ cell in DLN compared with control. HHCP significantly reduced the number of ${\alpha}$${\beta}$ TCR$^+$ in spleen and DLN, the number of CD8$^+$ in spleen compared with control, and also significantly reduced the number of CD3$^+$, CCR3, CD3$^+$/CD69$^+$, CD3/ CCR3, CD4$^+$, CD3$^+$/ CD4$^+$/CD45$^+$ cell in DLN. HHCP increased the number of NK$^+$ cells in spleen compared with control, in contrast significantly decreased the number of CD11c$^+$/ Classll$^+$ cell and CD11b$^+$/Gr-1$^+$ cell in DLN. Taken together, these results suggested that HHCP has suppressive effects on atopic dermatitis through the inhibition of IgE production and modulation of immune cell population in NC/Nga mice.

  • Objective : The aim of this study is to investigate the effect of Chamaecyparis Obtusa Oil to NC/Nga mice induced in Atopic dermatitis-like skin lesions by DNCB Mthods.Methods : NC/Nga mice which have been induced to Atopic dermatitis-like skin lesions by DNCB are divided into 3 groups, the one is the mice which have been spread with Chamaecyparis Obtusa Oil 2% cream, the other is which have been spread with dexamethasone (Dexa.) 0.5% on their Atopic lesion, another is the control group. then we take the blood sample of each group to measure state of the dorsal skin, the number of immunocytes. and resect the skin lesion to analyze the state of cells.Result & Conclusion : There are meaningful results of measuring the number of IgE, IL-4, IFN-γ and the state of the skin lesion and cells in the group with hamaecyparis Obtusa Oil 2% cream in comparison with the other groups.

The Effects of Ionized Maifanite on Skin Damages of Nc/Nga Mice after Atopic Dermatitis Elicitation (이온화된 맥반석이 아토피 피부염 유발 Nc/Nga mice의 피부손상 및 염증 완화에 미치는 영향)

  • Objectives : Atopic dermatitis accompanies with severe pruritus and collapse of skin barrier, inflammation. Maifanite could be used as an ointment for skin disease. However, there have been few studies about maifanite uses for atopic dermatitis. We report the anti-inflammatory and promoting skin recovery effects of ionized maifanite on damaged skin barrier with experimentally elicited atopic dermatitis. Methods : Nc/Nga mice were divided into 3 groups: control group(CON), atopic dermatitis elicited group(AE group), ionized maifanite treated group after atopic dermatitis elicitation(MT group). After 5% SDS was applied D. pteronyssinus crude extract also applied for 3 weeks to elicit atopic dermatitis-like skin disease. MT group was treated for 3 weeks with ionized maifanite. Ionized maifanite was applied once a day and voluntarily administrated. AE group and control group were treated with normal saline in the same way. Results : In MT group, skin lesions like eczema were more improved than AE group. p-ERK1/2 positive reaction was reduced in MT group. MMP-9 and substance P positive reaction at dermal papillae was also reduced in MT group. With skin angiogram, capillary vessel decreased in MT group. Also, IL-4 positive reaction cell and STAT-6 positive reaction cell reduced more in MT group than in AE group. $NF-{\kappa}B$ p65 positive reaction cell and iNOS positive reaction cell also declined more in MT group than in AE group. Conclusions : It is supposed that ionized maifanite has anti-inflammatory effects on NC/Nga mice's atopic dermatitis with suppressing IL-4 production and Th2 cell differentiation, and controlling $NF-{\kappa}B$ activation.
