다년생 초본류의 향기성분 분석 (Analysis of Essential Oil from Perennial Herbaceous Plants)

  • The chemical composition of essential oil from the perennial herbaceous plants (Houttuynia cordata, Filipendula glaberrima, Peucedanum japonicum, and Ainsliaea acerifolia) was determined by GC/MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were $\alpha$-phellandrene (18.97%), $\gamma$-terpinene (12.32%), decanal (8.72%), 1-decanol (10.92%), decanoic acid (12.12%), and 2-undecanone (12.32%) from H. cordata, farnesol (2.83%), l-$\alpha$-terpineol (2.72%), benzenmethanol (2.03%), (Z)-3-hexen-1-ol (4.32%), and T-muurolol (2.07%) from F. glaberrima, $\alpha$-phellandrene (14.25%), endobornyl acetate (3.84%), heptanal (47.52%), octanal (2.65%), (E,E)-2,4-decadienal (2.75%), and octanoic acid (4.52%) from P. japonicum, and geyrene (9.74%), $\beta$-cubebene (11.15%), berkheyaradulen (22.32%), $\beta$-elemene (6.21%), (-)-A-selinene (4.85%), benzaldehyde (4.52%), and benzenacetaldehyde (3.40%) from A. acerifolia.

화학적 수식에 의한 Bacillus stearothermophilus $\beta$-D-Xylosidase 의 연구 (Chemical Modification of the $\beta$-D-Xylosidase from Bacillus stearothermophilus)

  • Essential amino acids involving in the catalytic mechanism of the $\beta$-D-xylosidase of Bacillus stearothermophilus were determined by chemical modification studies. Among various che- mical modifiers tested N-bromosuccinimide (NBS), $\rho$-hydroxymercurybenzoate (PHMB), N-ethylma- leimide, 1-[3-(di-ethylamino)-propyl]$-3-ethylcarbodi-imide (EDC), and Woodward's Reagent K(WRK)inactivated the enzyme, resulting in the residual activity of less than 20%. WRK reduced the enzyme activity by modifying carboxylic amino acids, and the inactivation reacion proceeded in the form of pseudo-first-order kinetics. The double-lagarithmic plot of the observed pseudo-first- order rate constant against the modifier concentration yielded a reaction order of 2, indicating that two carboxylic amino acids were essential for the enzyme activity. The $\beta$-D-xylosidase was also inactivated by N-ethylmaleimide which specifically modified a cysteine residue with a reaction order of 1, implying that one cysteine residue was important for the enzyme activity. Xylobiose protected the enzyme against inactivation by WRK and N-ethylmaleimide, revealing that carboxylic amino acids and a cysteine residue were present at the substrate-binding site of the enzyme molecule.

HRT-mediated Turnip crinkle virus Resistance in Arabidopsis

  • Turnip crinkle vims (TCV) inoculation onto resistant Arabidopsis ecotype Dijon(Di-17) leads to a hypersensitive response (HR) on the inoculated leaves. A dominant gene, HRT, which confers an HR to TCV, has been cloned from Di-17 plants by map-based cloning. HRT is a LZ-NBS-LRR class resistance gene and it belongs to a small gene family that includes RPP8, which confers resistance to Peronospora parasitica Emco5. Outside of the LRR region, HRT and RPP8 proteins share 98% amino acid identity while their LRR regions are less conserved (87% identity). HRT-transformed Arabidopsis plants developed an HR but generally remained susceptible to TCV due to a dominant RRT allele, which is not compatible with resistance. However, several transgenic plants that over-expressed HRT much higher than Di-l7 showed micro-HR or no HR when inoculated with TCV and were resistant to infection. Both the HR and resistance are dependent on salicylic acid but independent of NPRI, ethylene, or jasmonic acid. Arabidopsis plants containing both TCV coat protein gene and HRT developed massive necrosis and death in seedlings, indicating that the TCV coat protein is an avirulence factor detected by the HRT.

Purification and Characterization of an Alkali-Thermostable Lipase from Thermophilic Anoxybacillus flavithermus HBB 134

  • An intracellular lipase from Anoxybacillus flavithermus HBB 134 was purified to 7.4-fold. The molecular mass of the enzyme was found to be about 64 kDa. The maximum activity of the enzyme was at pH 9.0 and 50℃. The enzyme was stable between pH 6.0 and 11.0 at 25℃, 40℃, and 50℃ for 24 h. The Km and Vmax of the enzyme for pNPL substrate were determined as 0.084 mM and 500 U/mg, respectively. Glycerol, sorbitol, and mannitol enhanced the enzyme thermostability. The enzyme was found to be highly stable against acetone, ethyl acetate, and diethyl ether. The presence of PMSF, NBS, DTT and β-mercaptoethanol inhibited the enzyme activity. Hg2+, Fe3+, Pb2+, Al3+, and Zn2+ strongly inhibited the enzyme whereas Li+, Na+, K+, and NH4+ slightly activated it. At least 60% of the enzyme activity and stability were retained against sodium deoxycholate, sodium taurocholate, n-octyl-β-D-glucopyranoside, and CHAPS. The presence of 1% Triton X-100 caused about 34% increase in the enzyme activity. The enzyme is thought to be a true lipase since it has preferred the long-chain triacylglycerols. The lipase of HBB 134 cleaved triolein at the 1- or 3-position.

배 검은별무늬병 감염과 저항성 방어반응 연관 전사체 프로파일 (Transcriptomic Profile in Pear Leave with Resistance Against Venturia nashicola Infection)

  • The molecular understanding of resistance and susceptibility of host plants to scab, a most threatful disease to pome fruit production worldwide, is very limited. Comparing resistant line '93-3-98' to susceptible one 'Sweet Skin' at seven time points of 0, 0.5, 1, 2, 3, 4, 8 days post inoculation, RNA-sequencing data derived from infected and mock-inoculated young leaves were analyzed to evaluate the tolerant response and to mine candidate genes of pear to the scab pathogen Venturia nashicola. Analysis of the mapped reads showed that the infection of V. nashicola led to significant differential expression of 17,827 transcripts with more than 3-fold change in the seven pairs of libraries, of which 9,672 (54%) are up- and 8,155(46%) are down-regulated. These included mainly receptor (NB-ARC domains-containing, CC-NBS-LRR, TIR-NBS-LRR, seven transmembrane MLO family protein) and transcription factor (ethylene responsive element binding, WRKY DNA-binding protein) related gene. An arsenal of defense response of highly resistant pear accessions derived from European pear was probably supposed no sooner had V. nashicola infected its host than host genes related to disease suppression like Polyketide cyclase/dehydrase and lipid transport protein, WRKY family transcription factor, lectin protein kinase, cystein-rich RLK, calcium-dependent phospholipid-binding copine protein were greatly boosted and eradicated cascade reaction induced by pathogen within 24 hours. To identify transcripts specifically expressed in response to V. nashicola, RT-PCRs were conducted and compare to the expression patterns of seven cultivars with a range of highly resistant to highly susceptible symptom. A DEG belonging to the PR protein family genes that were higher expressed in response to V. nashicola suggesting extraordinary role in the resistance response were led to the identification. This study provides the first transcriptional profile by RNA-seq of the host plant during scab disease and insights into the response of tolerant pear plants to V. nashicola.

광섬유를 이용한 레이저섬광의 균일분포 증진효과에 따른 열확산계수 측정에 관한 고찰 (A Study on Thermal Diffusivity Measurement by Improvement of Laser Flash Uniformity Using an Optical Fiber)

  • When thermal diffusivity is measured by laser flash method, the thermal diffusivity call be calculated front the assumption of the uniformly heated whole surface of the specimen. It has been known that the approximate 5% error is made by the non-uniform energy distribution on the specimen surface of laser pulse heat source. In this study, to obtain the highly-uniformed laser beam, which has both the low non-uniform heating error from non-uniform laser beam and the energy loss, research was carried out on no transmitting loss by optical fiber and high repetitions. In addition, heating error and thermal diffusivity were measured as the measuring positions were varied and compared with the results using the uniform and the non-uniform laser beams. In addition, dole to using the uniformalized laser beam, the whole surface of the specimen was heated uniformly and as a result, it was the thought that this was very effective to reduce the variations of the errors of the thermal diffusivity as the measuring positions were varied. It can be obtained that when the thermal diffusivity of POCO-AXM-5Q1 of SRM in NBS was measured with both the uniform and the non-uniform laser beams, the dispersion error of the former was from 2 to 2.5%, which was more improved than that of the latter.

  • Acid protease produced from Aspergillus tubingensis was pruified by ethanol fractionation, dialysis, and DEAE cellulose column chromatography. As a result of purification its specific activity increased to 5.4 times, and percent recovery was 39. The kinetic constants of the enzyme were studied. The Roles of Tryptophan and Histidine Residues in the Catalytic Activities $\beta$-Cyclodextrin Glucanotransferase from Bacillus firmus var. alkalophilus

  • In order to investigate the critical amino acid residues involved in the catalytic activities of $\beta$-cyclodextrin glucanotransferase ($\beta$-CGTase) excreted by Bacillus firmus var. alkalophilus, the amino acid residues in $\beta$-CGTase were modified by various site-specific amino acid modifying reagents. The cyclizing and amylolytic activities of $\beta$-CGTase were all seriously reduced after treatment with Woodward's reagent K (WRK) modifying aspartic/glutamic acid, N-bromosuccinimde (NBS) modifying tryptophan, and diethylpyrocarbonate (DEPC) modifying histidine residues. The roles of tryptophan and histidine residues in $\beta$-CGTase were further investigated by measuring the protection effect of various substrates during chemical modification, comparing protein mobility in native and affinity polyacrylamide gel electrophoresis containing soluble starch, and comparing the $K_m$ and $V_{max}$ values of native and modified enzymes. Tryptophan residues were identified as affecting substrate-binding ability rather than influencing catalytic activities. On the other hand, histidine residues influenced catalytic ability rather than substrate-binding ability, plus histidine modification had an effect on shifting the optimum pH and pH stability.

  • 츄잉껌 품질향상의 일환으로 저장기간 및 여러 가지 상대습도하에서 츄잉껌 향기성분의 변화를 GC/MS로 분석하였다. 츄잉껌 향기성분을 시료전처리로 포집하고 tetradecane을 내부표준용액으로 이용해서 각 성분과의 peak area ratio로써 향기성분함량의 감소량을 측정한 결과 저장초기에는 53% R.H 저장분 보다 33, 75% R.H에서 감소속도가 빨라으나 그 이후는 차이가 별로 없었으며 16주 후에는 $45{\sim}49%$의 잔존량을 나타내었다.

NPE법을 이용한 절연재료의 유효열확산계수의 과도측정에 관한 연구 (A Study on the Transient Measurement of the Effective Thermal Diffusivity of Insulation Materials by NPE Method)

  • The objective of this study is to present an efficient measurement method of the effective thermal diffusivity for the fibrous insulation material. The non-linear parameter estimation (NPE) method is adapted for this analysis because of its accuracy and its results are compared with those by other direct methods such as CTP, CHP and STD method. A experimental system is constructed with bell-jar vaccum chamber, diffusion pump, tube type furnace, control unit and data acquisition system included with A/D converter and IBM XT/AT personal computer. The typical results obtained from this study are as follows; 1) NPE method can be recommended as an useful and accurate method to measure the effective thermal diffusivity of insuation material because it is shown that the measurement error compared with those by other direct methods is reduced for standard material, NBS-1450b. 2) NPE method can minimize the effects of ill-measured temperature due to external disturbance, because the final value is found by point to point estimating. 3) NPE method dose not depend on the kinds of heat flux, since the surfac temperature are used to estimate the thermal diffusivity. 4) With NPE method, compared with the steady state method, a measuring time and a sample size could be reduced.
