Effect of Solvents of Extraction on the Biological Activities of Phyllostachys Nigra Munro (추출 용매에 따른 오죽(Phyllostachys nigra Munro) 추출물의 생리활성 효과)

  • In order to research the effect of solvents of extract on biological activities of Phyllostachys nigra Munro, antioxidative activity, superoxide dismutase (SOD)-like activity, nitrite scavenging activity, tyrosinase inhibitory activity, angiotensin-converting enzyme (ACE) inhibibitory activity in vitro were investigated. When ethyl acetate and hexane as solvents were used, the total phenols concentrations were 44.1 and 47.3 mg/kg, respectively, which was about 2.0 fold higher than that of water. The antioxidive activity was affected by solvents of extraction. The antioxidative activity was increased in order of hexane > ethyl acetate > n-butanol > methanol > water extraction. On the other hand, in the case of SOD-like activity, it was in the order of methanol > n-butanol > hexane > ethyl acetate extraction. The nitrite scavenging ability showed the most remarkable activity at pH 1.2 irrespective of solvents. Especially, when pH was increased from 1.2 to 6.0 using ethyl acetate extraction, it was decreased from 69.2 to 7.8%. The tyrosinase inhibitory activity was in the range of 15.2~21.3% and was increased in order of water > methanol > n-butanol>hexane > ethyl acetate extract. These results suggest that hexane and ethyl acetate extraction of Phyllostachys nigra Munro can be used in bioactive and functional materials.

The Effect of Flow Rate on the Process of Immiscible Displacement in Porous Media (다공성 매체 내 비혼성 대체 과정에서 주입 유량이 거동 양상에 미치는 영향)

  • A series of experiments using transparent micromodels with an artificial pore network etched on glass plates was performed to investigate the effects of flow rate on the migration and distribution of resident wetting porewater (deionized water) and injecting non-wetting fluid (n-hexane). Multicolored images transformed from real RGB images were used to distinguish n-hexane from porewater and pore structure. Hexane flooding followed by immiscible displacement with porewater, migration through capillary fingering, preferential flow and bypassing were observed during injection experiments. The areal displacement efficiency increases as the injection of n-hexane continues until the equilibrium reaches. Experimental results showed that the areal displacement efficiency at equilibrium increases as the flow rate increases. Close observation reveals that preferential flowpaths through larger pore bodies and throats and clusters of entrapped porewater were frequently created at lower flow rate. At higher flow rate, randomly oriented forward and lateral flowpaths of n-hexane displaces more porewater at an efficiency close to stable displacement. It may resulted from that the pore pressure of n-hexane, at higher flow rate, increases fast enough to overcome capillary pressure acting on smaller pore throats as well larger ones. Experimental results in this study may provide fundamental information on migration and distribution of immiscible fluids in subsurface porous media.

Anti-inflammatory and Anti-cancer Effects of Sterol-rich Fraction from Nannochloropsis oculata by using Saponification (해양미세조류(Nannochloropsis oculata)로부터 saponification을 통한 Sterols 분획물의 항염증 및 항암 효능 평가)

  • The green microalga of the genus Nannochloropsis (class Eustigmatophyceae) is a leading candidate for biofuel production due to its ability to accumulate high oil content (28.7% of cellular ash-free dry weight). We investigated the anti-inflammatory and anticancer activities of sterol-rich fraction from nannochloropsis oculata n-hexane (NOH) extract after saponification of the microalga. Among the fractions with n-hexane, chloroform and ethyl acetate, the n-hexane fraction showed the highest anti-inflammatory activity in LPS-stimulated RAW 264.7 macrophage as well as anticancer activity against human leukemia HL-60 cells without the cytotoxity. And the sterol-rich fraction was obtained from the n-hexane fraction by open silica column under the gradient solvent condition with 100% hexane (1L), hexane : ethyl acetate (20 : 1, 10 : 1, 5 : 1, 1 : 1, v/v). Among the four fractions (NOH-1~4), especially NOH-1 contained the highest content of sterols. NOH1 showed the highest HL-60 (about 85%) and NO inhibitory activities at the concentration of $100{\mu}g/mL$. These results demonstrated that the sterol-rich fraction from N. oculata might be a useful candidate as anti-inflammatory and anticancer agents for anti-inflammatory and anticancer activity.

A Study on Antimicrobial Activity of Lysimachia clethroides Duby Root Extracts against Methicillin-resistant Staphylococcus aureus

  • Methicillin-resistant Staphylococcus aureus (MRSA) is a bacterium responsible for a number of infections in humans that are difficult to treat, and as a result, is a substantial contributor to morbidity and mortality. In the present study, in search of natural products capable of inhibiting this multidrug-resistant bacterium, we investigated the antimicrobial activity of Lysimachia clethroides Duby root. The antibacterial activities of EtOH extract of Lysimachia clethroides Duby root and its n-hexane, EtOAc, n-BuOH and water fractions were evaluated against 15 strains of methicillin-resistant staphylococcus aureus (MRSA) and 1 standard methicillin-susceptible S. aureus (MSSA) strain by using the minimal inhibitory concentrations (MICs) assay, colorimetric assay using MTT test, checkerboard dilution test. Antimicrobial activity of n-hexane fraction of Lysimachia clethroides Duby root was remarkable. Against the 16 strains, the minimum inhibitory concentrations (MICs) were in the range of $31.25-62.5{\mu}g/ml$ and FICI values for n-hexane fraction of Lysimachia clethroides Duby root+AM and n-hexane fraction of Lysimachia clethroides Duby root+OX were checkerboard method performed using the MRSA, MSSA and one clinical isolate strains via MICI 0.12-1 and 0.25-0.75, showing the increase of synergistic effect. When combined together, these antibiotic effects were dramatically increased. These effective combinations could be new promising agents in the management of MRSA.

Antifungal Activity of Wood Extracts of Larix leptolepis and Pseudotsuga menziesii againstTrichoderma spp. (Trichoderma 속에 대한 낙엽송 및 미송 추출물의 항균활성)

  • This study was undertaken to determine inhibitory compounds from extracts of the softwood (larix leptolepis, Pseudotsuga menziesii) sawdust against Trichoderma spp. The sawdust of L. leptolepis and P. menziesii were hot water extracted, which were with fraction extracted organic solvents. The organic solvent extractions were carried out by n-hexane, methylene chloride, ethyl acetate. The antifungal activity of hot water extracts of L. leptolepis sawdust was determined to be 20.6% inhibition at a concentration of 1,000 ppm against Trichoderma spp. The antifungal activity of P. menziesii sawdust was outstanding about 60.3% against Trichoderma spp. The yields of the fractions of n-hexane soluble, methylene chloride soluble and ethyl acetate soluble from the hot water extract of L. leptolepis sawdust were 4.0%, 6.0% and 8.0%, repectively. However, the yields of the fractions of three solvents of P. menziesii sawdust were 8.0%, 13.0 and 14.0% correspondingly. The antifungal activity of n-hexane soluble fraction from hot water extracts of L. leptolepis sawdust was highest to about 68.5% to 79.9% against Trichoderma spp. compared to others. The antifungal activities of n-hexane soluble fraction from hot water extracts of P. menziesii sawdust showed 68.5%, 71.4%. 71.9%, 75.7% and 82.3% against T. aggressivum, T. atroviride, T. harzianum, T. koningii and T.viride, respectively. The n-hexane soluble fraction revealed much higher antifungal activity than the other fractions did. This study demonstrated that the n-hexane fraction of the hot water extracts of L. leptolepis and P. menziesii exhibited the greatest antifungal activity against Trichoderma spp.

Characterization of Hexane Biodegradation by Rhodococcus sp. EH741 (Rhodococcus sp. EH741에 의한 Hexane 생분해 특성)

  • As a strain EH741, having an excellent hexane degradability, was isolated from bacterial consortium using hexane as a sole carbon and energy source. EH741 was identified as a Rhodococcus sp. and the addition of a surfactant Pluronic F68(PF68), for increasing hexane solubility couldn't enhance the specific growth rate of the isolate EH741 n the mineral salt medium supplemented with hexane as a sole carbon source(hexane-BH medium). In the hexane-BH medium, the maximum specific growth rate(${\mu}_{max}$) of this strain was $0.04h^{-1}$, and the maximum hexane degradation rate($V_{max}$) and saturation constant($K_s$) were$161{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$ and 10.5 mM, respectively. Rhodococcus sp. EH741 was one of excellent microorgamisms for hexane biodegradation processes.

Antimicrobial Activity of Psoralea corylifolia, Schisandra chinensis, and Spatholobus suberectus Extracts (보골지, 오미자, 계혈등 추출물의 항균활성)

  • This study was performed to identify safe natural food preservatives from medicinal herbs and to evaluate the antimicrobial effects of medicinal plants against microorganisms. Medicinal herbs were extracted 3 times with methanol at $45^{\circ}C$ for 3 h and fractionated with n-hexane. The antimicrobial effects of the fractions were determined by measuring the diameter of the inhibition zone by using an agar-well diffusion assay. The MIC of fractions for the inhibition of microorganisms was determined using a microplate reader. The antimicrobial effects of fractions were greater against gram positive bacteria than against gram negative bacteria, but the difference was not significant. The antimicrobial effects of fractions were concentration dependent. While these results have implications, the underlying mechanisms of microbial inactivation need to be further elucidated. The results showed the possibility of developing safer food preservatives.

Application of Flory-Treszczanowicz-Benson model and Prigogine-Flory-Patterson theory to Excess Molar Volumes of Isomers of Propanol with Cyclohexane or n-Hexane

  • Excess molar volumes ($V_m^E$) of binary mixtures of 1-propanol or 2-propanol (1) + cyclohexane or n-hexane (2) were measured with V-shaped dilatometer at 303.15 K. The $V_m^E$ data for these mixtures varied as: 2-propanol > 1-propanol and were higher for cyclohexane than n-hexane for both propanol systems. The experimental data were correlated with Redlich-Kister polynomial. The $V_m^E$ data were interpreted qualitatively as well as quantitatively in terms of Flory-Treszczanowicz-Benson model and Prigogine-Flory-Patterson theory. Both models correctly described the sign and shape of $V_m^E$ vs $x_1$ curves. The values calculated by both the models agree well with the experimental data.

토양 중 브롬화다이옥신류의 분석법 고찰

  • The last few decades have seen drastic growth in the use of brominated flame retardants(BFRs). There is the problems of polybrominated dibenzo-p-dioxins(PBDDS) and polybrominated dibenzofurans(PBDFs) generation as by-products in combustion process of BFRs. PBDDs/PBDFs are highlighted the new pollutants of environmental contamination, recently. In this study, the 10 kinds of PBDDs/PBDFs standards were used to perform the experiments of recoveries. The analysis of PBDDs/PBDFs in soil environmental samples. was carried out. In silica gel column cleanup, PBDDs/PBDFs eluted until 80mL of n-hexane, and the recovery was obtained 73∼105%. In multi-layer silica gel column cleanup, the recovery was obtained 56∼125% with various elution solvents. In alumina clean process, the standard mixture(PBDES, PBDDs/PBDFs) eluted the first fraction of 2% dichloromethane with n-hexane in the range of 0∼100mL, PBDEs compounds eluted in the second fraction of 50% dichloromethane with n-hexane. In activated carbon column cleanup, the PBBEs eluted to the first fraction(n-hexane) and second fraction(toluene), but PBDDs/PBDFs only eluted to the second fraction. The analytical methods of PBDDs/PBDFs for soil was established based on the experimental results of the environmental samples.

  • Reverse osmosis(RO) and pervaporation(PV) membrane processes were compared with each other theoretically by using Paul and Ebra-Lima model. From this model the concentrations of liquid within the membrane when pressure was applied to the upper compartment(for PV case, the applied pressure is infinite) were calculated for rubber membrane-n-hexane and rubber membrane-benzene systems. The permeabilities of RO and PV were also calculated and compared for polyethylene film-n- hexane and polyethylene film- benzene systems Theoretically, the permeabilities of PV membrane were greater than those of RO membrane.
