Antimutagenic and Antitumor Effects of Adenophora triphylla Extracts (잔대 추출물들의 항돌연변이 및 항종양 효과)

  • This study was carried out to investigate the mutagenic, antimutagenic, cytotoxicity and antitumor effects of Adenophora triphylla (AT). AT was extracted with 70% ethanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water. Antimutagenic, cytotoxicity and antitumor effects of AT extracts were measured by using Ames test, SRB method, and the tumor growth inhibition test. AT extracts did not show any mutagenicity in the Ames test; however, 70% ethanol extracts and its fractions had strong antimutagenic effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and 4-nitroquinoline-1-oxide (4NQO). The ethyl acetate fraction of AT (200 ${\mu}g$/plate) showed approximately 66.5% inhibitory effect on the mutagenesis induced by 4NQO against TA98 strain, whereas 83.3% and 75.1% inhibitions were observed on the mutagenesis induced by MNNG and 4NQO against TA100 strain. In anticancer effects, the cytotoxicity of AT extract and its fractions against cancer cell lines including human cervical adenocarcinoma (HeLa), human hepatocellular carcinoma (Hep3B), human breast adenocarcinoma (MCF-7), human gastric carcinoma (AGS), human lung carcinoma (A549) and transformed primary human embryo kidney (293) were investigated. The treatment of 1 mg/mL AT ethyl acetate faction had the highest cytotoxicity of 79.9%, 74.9%, 66.0%, 71.0% and 74.3% against HeLa, Hep3B, MCF-7, AGS and A549 cells, respectively. In contrast, the extract and its fractions showed only $3{\sim}36%$ cytotoxicity for a normal human kidney cell line (293). In vivo anti-cancer effect of Adenophora triphylla extract was tested using Balb/c mice transplanted sarcoma-180 cells. Adenophora triphylla ethyl acetate fraction showed the highest inhibition rate of 37.2% at the 50 mg/kg concentration.

Antimutagenic and Cytotoxic Effects of Acer ginnala Max. Bark Extracts (신나무 껍질 추출물의 항돌연변이원성 및 세포독성 효과)

  • In the present study, we investigated the antimutagenic and cytotoxic effects of Acer ginnala Max. bark extract on S. typhimurium TA98, TA100 and cancer cell lines with Ames test and SRB assay, respectively. They were extracted with methanol and then fractionated using hexane, chloroform, ethyl acetate, butanol, and water to obtain the fractions. The inhibition rate of methanol ($200\;{\mu}g/plate$) of Acer ginnala Max. bark extract in the Salmonella typhimurium TA100 strain showed $83.3\%$ against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). In addition, the suppression of methanol extract with same concentration of in the Salmonella typhimurium TA98 and TA100 strains showed $80.3\%\;and\;92.7\%$ inhibition against 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-1), respectively. The cytotoxicity effects of Acer ginnala Max. bark extract against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS), human hepatocellular carcinoma (Hep3B) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 mg/mL Acer ginnala Max. bark methanol extract of methanol showed strong cytotoxicities of $77.3\%,\;90.4\%,\;88.9\%,\;and\;83.7\%$ against A549, AGS, Hep3B and MCF-7, respectively.

Antioxidant and α-glucosidase inhibitory effects of ethanolic extract of Ainsliaea acerifolia and organic solvent-soluble fractions (단풍취 추출물 및 분획물의 항산화 및 α-glucosidase 저해 활성 평가)

  • Among the naturally occurring antioxidants, polyphenols are widely distributed in various fruits, vegetables, wines, juices, and plant-based dietary sources and divided into several subclasses that included phenolic acid, flavonoids, stilbenes, and lignans. As part of our continuing search for bioactive food ingredients, the antioxidant and ${\alpha}$-glucosidase inhibitory activities of the aqueous ethanolic extract from the aerial parts of Ainsliaea acerifolia were investigated in vitro. The antioxidant properties were evaluated via radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. In addition, the anti-diabetic effect of A. acerifolia extracts was tested via ${\alpha}$-glucosidase inhibitory assay. Furthermore, the total phenolic contents were determined using a spectrophotometric method. All the tested samples showed dose-dependent radical scavenging and ${\alpha}$-glucosidase inhibitory activities. In particularly, the ${\alpha}$-glucosidase inhibitory and radical scavenging properties of the ethyl acetate (EtOAc)-soluble portion from the aerial parts of the A. acerifolia were higher than those of the other solvent-soluble portions. These results suggest that A. acerifolia could be considered a new potential source of natural antioxidants and antidiabetic ingredients. More systematic investigation of the aerial parts of A. acerifolia will be performed for the further development of anti-oxidative and antidiabetic drugs.

Effects of Ethylacetate Fraction of Plantago asiatica L. on Hypercholesterolemia Induced by High Cholesterol Diet in Rats (질경이(Plantago asiatica L.) 에틸아세테이트분획이 흰쥐의 고콜레스테롤혈증에 미치는 효과)

  • Plantain was extracted with ethanol and fractionated systemically with n-hexane, chloroform, ethylacetate, n-butanol and water to study inhibitory effect on cholesterol synthesis in vitro. To screen the effect, inhibitory activities on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase obtained from Saccharomyces cerevisiae were examined using the five fractions of Plantain. The HMG-CoA reductase activity was inhibited most by ehylacetate fraction among the fractions, although the all five fractions had the effect To see the hypocholesterolemic effect of the ethylacetate fraction of Plantanin (PAE) in vivo, male Sprague-Dawley rats were received 5 types of diets for 6 weeks: normal diet group (NOR), high cholesterol diet group($1\%$ cholesterol and $0.25\%$ sodium cholate, CON), normal diet and PAE 70 mg/kg administered group(S1), high cholesterol diet and PAE 140 mg/kg administrated group(S2), and high cholesterol diet and PAE 140 mg/kg administered group(S3). Body weight gains of the CON were significantly increased compared to those of S1, S2 and S3. Activities of serum AST and ALT were tended to be increased in CON compared with NOR and reduced by the PAE administration. Concentrations of serum total cholesterol, free cholesterol, LDL-cholesterol, triglyceride and the atherogenic index were tended to be decreased in the PAE administered groups compared with the CON. HDL-cholesterol and phospholipid concentrations were significantly decreased in the CON and markedly increased by the PAE administered groups. Taten together, it is suggested that the ethylacetate fraction of Plantanin exerts antiatherosclerotic effect by reducing serum cholesterol concentrations in rats fed high cholesterol diets.

  • Defatted rice bran is mixed with diluted acid solution, the mixture is agitated some hrs. at constant temparature. After the mixture is filtered, thus filtrate is obtained. This filtrate is phytin extract solution. (Test-1) The alkali is added to this filtrate and filtered out, then the precipitation of phytin is obtained. (Test-2) At the test-1, the effect of kind of acid, conc. of acid, amount of extract sol'n., time of extraction, temp. of extraction, to the extract amount of phytin is tested. Consequently, the following facts are known. 1. Amount of phytin extract is greater HCI extraction than $H_{2}SO_4$ extraction. 2. At 0.3% HCI, the amount of phytin extract is greatest of all HCl extraction. 3. The sufficient amount of acid solution is 8-10 times of amount of defatted rice bran. 4. The time of extraction at room temperature is sufficient 8-12 hrs. 5. When extract temperature is $20-30^{\circ}C$, the amount of phytin extraction is greater of all temp. 6, When defatted rice bran 20 g is shanken with 160 ml of 0.3% HCl for 10 hr. at room temp., in this case the amount of phytin extract is 11.34% of defatted rice bran, it is 93% of theoretical yield. At the test-2 the effect of kind of precipitation agent, degree of nutralization to the amount of phytin prcipitation is tested. 1. Degree of nut. is best at pH 6.8-7.0. 2. When use of $Ca(OH)_2$ the amount of phytin precipitation is more than use of KOH, NaOH, or $NH_{4}OH$. 3. At pH 6.0-7.2, the solubility of phytin is followed. K-phytate > $NH_{4}-phytate$ > Na-phytate > Ca-phytate 4. When phytin extract solution is nutralized with $Ca(OH)_2$ to pH 7.0, the amount of phytin precipitation is 94.78% of theoretical yield.

Synthesis of Vitamin $D_3$ and It′s Analogues in Dorsal Skin of 8 Week-Old Broiler Chicks Irradiated by UV Light (자외선 조사에 의한 8주령 브로일러 병아리 등 피부에서의 비타민 $D_3$ 및 그의 유사물질 합성)

  • This study was conducted to investigate the synthesis of previtamin D$_3$(PreD$_3$), lumisterol$_3$($L_3$) and vitamin $D_3$ ($D_3$) in dorsal skin of 8 week-old broiler chicks irradiated by UV light. The Hubbard line day-old broiler chicks were fed with vitamin D-depleted ration (or 3 weeks and with medium broiler diet for 5 weeks. The 3$\times$3cm area of dorsal skin was defeathered one day before the experiment. The 310nm UV light was utilized to irradiate the dorsal skin, which was separated into epidermis and dermis after irradiation. The irradiated epidermis was extracted with 60% ethyl acetate in n-hexane and PreD$_3$, $L_3$, D$_3$ and 7-dehydrocholesterol (7-DHC) were analyzed by HPLC. When UV light was treated for 0, 90 or 150min. the L$_3$ content in the separated epidermis was 219, 298 or 248ng/$\textrm{cm}^2$, respectively. The irradiation for 90min produced 79ng/$\textrm{cm}^2$ of L$_3$, and the synthesizing rate was 0.9ng/$\textrm{cm}^2$/min. The PreD$_3$ content was 23, 33 or 12ng/$\textrm{cm}^2$ when exposed for 0, 60 or 150min The 10ng/$\textrm{cm}^2$ of PreD$_3$ was produced for 60min. The D$_3$ was generated by 27ng/$\textrm{cm}^2$ when exposed for 90min. The 7-DHC contents were decreased gradually from initial 123 to 35ng/$\textrm{cm}^2$ when exposed for 150 min, showing the rate of disappearance as 0.6ng/$\textrm{cm}^2$/min.

Inhibitory Substance on the Snake Venoms Produced by Penicillium sp. (사독의 조해물질에 관한 연구)

  • One strain of Penicillium sp. (175-66-B), isolated from soil, was able to produce a substance that has a strong inibition activity against the Agkistrodon and Trimeresurus venoms. In this experiment, the chemical and biological properties of the sample were investigated. As an inhibitory substance, it was effective to the proteinase, hemorrhagic and lethal factors of Agkistrodon and Trimeresurus venoms, and also effective to several fractions of the proteinases and hemorrhagic factors of Agkistrodon halys blomhoffi venom. Moreover, in the addition of prednisotone, it was more effective for the cure of the mouse envenomated with the venom amount of two fold of MLD$_{100}$. This substance was very stable to the acid, alkali and heat. Its melting point was high enough to sublime at 222$^{\circ}C$ without any decomposition. This sample was easily dissolved only in hot water, but not in several organic solvents except for a little dissolution in elate. It did not have the chelating activity. It had very strong specificity to the snake venoms. but its activity was depressed by the addition of zinc or cupric salts. This sample had no acute toxicity to the mouse. Its chemical formula was $C_{16}$ $H_{12}$$N_2$ $O_{10}$ with the molecular weight of about 392. It has two epoxy groups and four carboxyl radicals, but amino, nitrite and nitrate radicals, unsaturated bonds and aromatic ring were not detected. DISTRIBUTION OF SOME CHEMICAL POLLUTANTS IN SUYEONG BAY (수영만 인근해수의 오탁분포에 대하여)

  • In order to study the water pollution in Suyeong Bay, Busan, some chemical constituents were determined at 25 stations in the neap tides on 9 Aug. 1977 and spring tides on 30 Aug. 1977. Range and mean values of the constituents in the spring tides are as follows: $pH\;6.54\~8.06,\;7.54;$ electrical conductivity $0.413\~0.481\times10^5\;\mu\mho/cm,\;0.467\times10^5\;\mu\mho/cm;\;transparency\;0.2\~5.5m,\;2.2m;$ turbidity $1\~60ppm$, 14ppm, chlorosity $15.20\~18.11g/\ell,\;17.67g/\ell;$ fluoride ion $0.94\~1.03ppm$, 0.99ppm; dissolved oxygen $0.17\~7.60ppm$, 4.77ppm; sulfide $0\~0.46ppm$, 0.07ppm; chemical oxygen demand $1.20\~40.74ppm$, 6.11ppm; ammonia-nitrogen $0.060\~0.520ppm$, 0.180ppm; nitrite-nitrogen $0.001\~0.026ppm$, 0.009ppm; nitrate-nitrogen $0\~0.037ppm$, 0.014ppm; phosphate-phosphorus $0.002\~0.261ppm$, 0.050ppm; n-Hexane soluble $0.5\~5.4ppm$, 2.1ppm ; iron $1.0\~104.11\;ppb$, 24.15ppb ; copper $0\~27.45ppb$, 4.19ppb; lead $0\~2.50ppb$, 0.92ppb; zinc $0\~5.15ppb$, 1.47ppb ; cadmium $0\~0.26ppb$, 0.04ppb; and mercury $0.05\~0.37ppb$, 0.11ppb respectively. The variations of the contents of the chemical constituents in the spring tides were larger than in the neap tides. The contents of COD, sulfide, nutrient salts and heavy metals were the highest in the estuary of Suyeong River, and decreased in order of off Kwangan-Ri region, outer Bay and off Haeun-Dae region. The water quality in Suyeong Bay was particularly shown that the concentrations of COO, iron, copper and mercury were higher than those of other coastal aseas and deficiency in dissolved oxygen was observed in some parte of Suyeong Bay. In consideration of the relationship between the chlorosity and the concentrations of nutrient salts, COD and total heavy metals, water pollution of this area is considered due to the inflow of Suyeong River which was extremely polluted by sewage and industrial wastewaters.

Quality Characteristics and Antioxidant Activity of Prickly Pear Cactus Cladodes (손바닥 선인장 엽상경의 품질 특성과 항산화 효과)

  • Prickly pear cactus cladodes were extracted with hot water and 70% ethanol, followed by fractionation with n-hexane (HF), ethyl acetate (EF), n-butanol (BF), and distilled water. Total phenolics and total flavonoid contents as well as antioxidative and anti-inflammatory activities were then measured. Total phenolic contents were 784, 452, and 220 mg gallic acid equivalents (GAE)/g, whereas total flavonoid contents were 214, 76, and 113 mg quercetin equivalents (QE)/g in EF, BF, and HF, respectively. DPPH and ABTS radical scavenging activities ($IC_{50}$) were 103 and $105{\mu}g/mL$ in EF, 359 and $379{\mu}g/mL$ in BF, and 469 and $605{\mu}g/mL$ in HF, respectively. Oxygen radical absorbance capacity was highest at $391{\mu}M$ TE in EF (in decreasing order of $117{\mu}M$ TE in BF and $64{\mu}M$ TE in HF), whereas superoxide anion radical scavenging activity ($IC_{50}$) was highest at $40{\mu}g/mL$ in EF (in decreasing order of $69{\mu}g/mL$ in BF and $98{\mu}g/mL$ in 70% ethanol extract). Inhibitory activity ($IC_{50}$) of nitric oxide (NO) production induced by LPS-activated RAW264.7 cells was highest at $62{\mu}g/mL$ in HF (in decreasing order of $104{\mu}g/mL$ in EF and $465{\mu}g/mL$ in BF). The selectivity index (ratio of inhibitory activity of NO production to cell cytotoxicity) was highest at 4.63 in EF (in decreasing order of 3.37 in HF and 2.14 in BF). In conclusion, EF showed potent antioxidant and anti-inflammatory effects with high phenolic and flavonoid contents.

Analysis and risk assessment of polycyclic aromatic hydrocarbons (PAHs) in seafood from oil contaminated bay (유류 오염지역의 수산물 중 다환방향족탄화수소류 (PAHs) 분석 및 위해평가)

  • To assess health risk for the intake among residents after the Hebei Spirit oil spill, 16 Polycyclic Aromatic Hydrocarbons (PAHs) in seafood samples from oil contaminated bay were determined by Gas Chromatography-Mass Spectrometry (GC-MSD) and samples were personally collected and purchased by residents. Samples were hydrolyzed with KOH and extracted with methylene chloride. The extracted solution were cleaned up using silica/florisil column and 16 PAHs were eluted by methylene chloride : n-hexane (1:9) mixture and determined by GC-MSD in Selected Ion Monitoring (SIM) mode. The mean recoveries for 16 PAHs ranged from 79% to 85%. The 16 PAHs levels in 126 samples ranged from 0.17 to $6.04\;{\mu}g$/kg and the TEQBaP (Toxic EQuivalents) levels in 126 samples were calculated using benzo(a)pyrene toxic equivalency factor for individual 16 PAHs and ranged from 0 to $0.91\;{\mu}gTEQ$/kg. The average Benzo(a)pyrene dietary exposure of residents was $5.5{\times}10^{-8}\;mg/kg$ bw/day and the average PAHs chronic dietary exposure was $1.3{\times}10^{-5}\;mg$ TEQ/kg bw/day. The margin of exposure (MOE) and the excess cancer risk and were $1.8{\times}10^6$ and $9.8{\times}10^{-8}$, respectively. Therefore, the assessment result was considered as low concern for health risk.
