연안환경 모니터링을 위한 홍합(Mytilus coruscus)의 체내 수은 함량 분석 (Determination of Mercury in Korean Mussels (Mytilus coruscus) for Marine Environmental Monitoring)

  • Marine ecosystems are frequently exposed to a variety of chronic and acute pollutants derived from anthropogenic production and consumption activities. Mussels are sessile (can provide location-specific information), medium-sized (have enough tissue matrix for chemical analysis) filter-feeders (show accumulation of pollutant chemicals from seawater). These biological and ecological characteristics make mussels virtually ideal for pollution monitoring. In this study, Korean mussels (Mytilus coruscus) were collected from nine different sites situated along the coasts of the western sea to the eastern sea of Korea in 2017. Total mercury concentration was highest (mean ${\pm}$ standard error, $92.7{\pm}3.5ng/g\;dry$) on Baengnyeongdo Island in the western sea, and the sites in the southern sea showed the lowest mean concentrations (42.3 - 44.5 ng/g dry). These results were discussed in terms of possible pollution sources.

  • The heat shock proteins (Hsps), one of the most highly conserved groups of proteins, play crucial roles in protecting cells against environmental stressors, such as temperature, salinity, heavy metals and pathogenic bacteria. The glutathione S-transferases (GST) have important role in detoxification of oxidative damage, environmental chemicals and environmental stress. The purpose of this study is to investigate the gene expression of Hsp70 and GST on change of temperature and salinity in Mytilus coruscus. The M. coruscus was cultured in incubator of separate temperature and salinity (8, 20, $30^{\circ}C{\times}20$‰, 25‰, 30‰) for 28 days. Ten individuals in each group were selected after each 14 and 28 days exposure. Results that the expression of Hsp70 mRNA was no significant changed in M. coruscus exposed to temperature ($8^{\circ}C$, $20^{\circ}C$, $30^{\circ}C$) and salinity (20‰, 25‰, 30‰) for 14 days. Whereas the expression of Hsp70 mRNA was increased in exposure to temperature $30^{\circ}C$ and salinity (20‰, 25‰, 30‰) for 28 days. The expression of GST mRNA was increased in exposure to temperature $30^{\circ}C$, salinity (25‰, 30‰) for 14 days and temperature ($8^{\circ}C$, $20^{\circ}C$, $30^{\circ}C$), salinity (20‰, 25‰, 30‰) for 28 days. These results suggest that Hsp70 and GST were played roles in biomarker gene on the thermal and salinity stress.

배건담치의 정미성분에 관한 연구 (THE TASTE COMPOUNDS OF BROILED DRIED SEA MUSSELS)

  • This study was attempted to evaluate the taste of broiled-dried sea mussel which is a traditional dried Sea food in Korea. The contents of such compounds as free amino acids, nucleotides and their related compounds, TMAO, betaine, and nonvolatile organic acids were analysed. From the results of analysis of nucleotides, the contents of AMP and ADP in broiled-dried Mytilus coruscus and Mytilus edulis appeared higher than other nucleotides and tended to increase slightly after cooking, drying, and storage which might be due to both their stability and the addition of degradation of nucleic acids during cooking. In the free amino acid composition of fresh samples abundant amino were taurine, glycine, serine, glutamic acid, alanine, arginine, threonine, tyrosine, lysine and valine in order. Such amino acids as histidine, leucine, methionine and isoleucine were poor and both proline and phenylalanine were merely trace. The free amino acid composition in the extract of Mytilus corucus and Mytilus edulis after broil-drying was not changed. These amino acids, taurine, glycine, feline, glutamic acid, alanine and arginine were abundant in dried samples as well as in the fresh. The total free amino acid was greatly reduced after cooking and drying ranging from $76.3\%$ to $79.7\%$ loss to that of tile fresh. The content of betaine shelved the same tendency as in total free amino acid while TMA slightly increased relating to TMAO decrease during broil-frying. Such nonvolatile organic acids as succinic, lactic, malic and fumaric acid were abundant in both fresh and broiled-dried samples whereas oxalic and pyruvic acid were poor. It is found that the taste compounds of broiled-dried Mytilus coruscus and Mytilus edulis were composed of amino acids as glycine, serine, alanine, glutamic acid, arginine, and betaine, TMAO, ADP, AMP, and organic acids such as succinic acid, lactic acid, malic acid and fumaric acid. No significant differences in the taste compounds between male and female as well as between Mytilus coruscus and Mytilus edulis was not observed.

울산연안 해수 및 홍합 중의 중금속 함량 (Some Heavy, Metal Concentrations of Seawater and Mytilus coruscus in Ulsan Seaside)

  • 울산 연안의 공업단지를 비철금속 공업을 주로 하는 온산공단과 자동차 및 조선업을 주로 하는 미포공단, 석유화학 공업을 주로 하는 석유화학공단으로 나누어, 이러한 공업단지별로 해수 및 홍합중의 중금속 함량을 측정하고 이에 따른 홍합의 각 중금속 생물농축계수를 산정해 보았다. 시료는 1995년 4월 6일에 채취하였고, 분석방법은 원자흡광광도법에 따랐다. 해수중의 납 함량은 석유화학공단 해역 (1.8${\pm}$2.4ppb)이 가장 높았고, 홍합중의 납 함량도 석유화학공단 해역(0.800${\pm}$0.418ppm)이 가장 높았다. 해수중의 카드뮴 함량은 미포공단해역(1.3${\pm}$1.4ppb)이 가장 높았고, 홍합중의 카드뮴 함량은 석유화학공단 해역(0.282${\pm}$0.07ppm) 이 가장 높았다. 해수중의 수은은 전 해역에서 검출되지 않았고, 홍합중의 수은은 석유화학공단 해역(0.025${\pm}$0.0011ppm)이 가장 높았다. 해수중의 구리 함량은 미포공단 해역이 가장 높았고(0.5${\pm}$0.9ppb), 홍합도 미포공단 해역 (2.132${\pm}$0.526ppm)이 가장 높았다. 해수중의 망간 함량은 미포공단과 석유화학공단 해역이 비슷한 수준요도 나타났고(2.7${\pm}$2.1ppb, 2.4${\pm}$1.5ppb), 홍합에서는 석유화학공단 해역이 가장 높게 나타났다(3.212${\pm}$2.445ppm). 아연함량은 해수와 홍합 모두 미포공단 해역에서 가장 높게(55.6${\pm}$9.4ppb, 19.560${\pm}$9.979ppm) 나타났다. 울산연안 해역에 서식하는 홍합의 중금속 농축계수는 납, 카드뮴, 망간의 경우 평균 1,070배, 1,370배, 1,300배로 비슷하게 나타났고, 수은이 가장 낮은 80배, 구리는 6,940배로 가장 높은 농축률을 보였다. 울산연안 해수의 중금속 농도는 인근 공단별로 차이를 나타내고 있었으며, 카드뮴, 구리, 망간, 아연은 미포공단이 가장 높았고, 납은 석유 화학공단해역이 가장 높았다. 납, 구리, 아연의 경우 해수중의 농도가 높은 지역에서는 홍합중의 농도도 높게 나타났다. 그러나 카드뮴 경우 해수중의 농도는 미포공단에서 가장 높았으나 홍합중의 농도는 석유화학 공단에서 가장 높게 나타나 차이를 보였다. 이상의 결과로 볼 때, 연안 오염의 동태를 파악하기 위해서는 해수보다는 홍합과 간은 부착 패류를 지표종으로 설정하는 것이 타당한 것으로 보인다.

홍합의 사육조건과 성장 (Culture Condition and Growth of Larvae of the Mytilus Coruscus Gould)

  • The larvae of Mytilus coruscus were grown at the room temperature of approximately 15.1C under several different sulture conditions, i.e., salinity, population of the larvae, density and kind of food organisms, etc. (1) The egg of Mytilus coruscus obtained in the laboratory measured about 73.0${\mu}$ in diameter. The embryos gradually developed into larvae up to 179.0${\mu}$ shell length with the shell height of 135.9${\mu}$ even in the absence of the algal food. Beyond this size, however, the growth of larvae was considerably retarded, indicating that the better growth could be expected if the food began to be fed four days after spawning. (2) The larvae began settling upon reaching 281.4${\mu}$ to 310.9${\mu}$ in shell length or 264.3${\mu}$ to 301.9${\mu}$ in shell height. When the shell length reaches 322.6${\mu}$ to 337.1${\mu}$, the shell height also reaches about the same, i.e., 321.5${\mu}$ to 346.2${\mu}$. (3) Daily rate of food consumption was determined by the size of the larvae and the species of the algal food. Regardless of the species of food given, the rage of food consumption remained almost the same until the larva reached the straight-hinge stage, and marked variations were found as the larvae grew larger. Daily rate of food consumption was shown as follow; Chaetoceros calcitrans : Y=2.99167e$\^$0.000018243x$\^$2// Cyclotella nana : Y=3.00324e$\^$0.000015481x$\^$2// Monochrysis lutheri : Y=3.000056e$\^$0.000014485$\^$2// (4) Suitable amount of the food to be given was about five times of the consumed food by Mytilus coruscus. (5) When the numbers of the larvae was higher than ten per milliliter, the growth was significantly retarded. (6) Monochrysis lutheri and Cyclotella nana were much better than Chaetoceros calcitrans as the food of Mytilus coruscus, and even the same food organism showed some difference with the age of the organism. (7) Sea water of higher salinity showed the better result in the growth of the larvae and the water with the specific gravity of 1.020 or below was dangerous for the larvae. 홍합, Mytilus coruscus의 성 성숙과 생식주기 (Sexual Maturation and Gametogenic Cycle of the Hard Shelled Mussel, Mytilus coruscus (Bivalvia: Mytilidae))

  • 2000년 1월부터 12월까지 남해안 한산만 해역에서 채집한 홍합의 생식소 발달과 생식주기에 관하여 조직학적 방법을 이용하여 조사하였다. 홍합은 자웅이체이며, 생식소는 암 수 모두 다수의 배우자형성소낭으로 이루어져 있었다. 생식소지수(GI)는 2월(3.40)에 가장 높았으며, 8월(1.18)에 가장 낮았다. 비만도(CI)는 생식소지수(GI)와 유사하게 변화하였다. 홍합의 주 산란기는 2∼3월이며, 생식주기는 휴지기(8∼11월), 분열증식기(11∼12월), 성장기(12∼1월), 성숙기(1∼2월), 부분방출기(2∼4월), 완전방출 및 퇴화흡수기(4∼7월)로 구분할 수 있었다. 성비(암:수)는 1:0.96으로 조사되었다.

추자도산 홍합, Mytilus coruscus (Gould, 1861) 의 Condition Index 산정에 대한 연구 (Evaluation of Methods Used in the Calculation of Condition Index Using the Mussel, Mytilus coruscus (Gould, 1861) Collected from Chuja Island, Cheju, Korea)

  • Condition index(Cl)는 일반적으로 이매패류의 영양상태와 건강 등의 생리적인 상태를 알기 위한 중요한 지수이다. 이 연구는 제주도 추자 연안에 서식하는 홍합(M.coruscus)의 Cl를 다음과 같은 세 가지 방법으로 산출한 뒤, 이를 비교하였다: (1) Cl-vol=[tissue dry weight(g) 1000]shell cavity volume(ml). (2) Cl-wt=[tissue dry weight(g) 1000]internal shell cavith capacity(g). (3) Cl-size= tissue wet weight(g)/[shell length (mm)]$^{3}$. Cl-vol 산정을 위한 패각내 부피는 아르키메데스의 원리를 이용하였으며, 전체 부피와 육질을 제거한 패각의 부피 차를 이용하여 산출하였다. 월 별 세 가지 Ci들의 변화는 Pearson의 상관계수, 분산분석(ANOVA)과 Duncan의 다중검정법을 사용하여 세 지수의 유의성과 처리평균간의 근사 정도를 검정하였다. 실험에 이용된 홍합의 크기는 각장 50-180 mm 이었다. 세 지수의 범위는 67.48-140.61(CL-vol), 74.67-118.02(Cl-wt), 1.40$^{-5}$-1.60$^{-5}$(Cl-size)였다. Cl-vol은 Cl-wt, Cl-size와는 다르게 계절에 따른 유의차를 보였으며(p<0.05) 8월에 가장 높은 수치를 보였다. 그러므로 패각내 부피를 이용한 Cl는 홍합의 상태를 제시하는데 있어 가장 효율적인 것으로 사료된다.

홍합 (Mytilus coruscus) 족사 cuticle의 초미세구조 연구 (An ultrastructural study of the cuticle in the byssus of marine mussel (Mytilus coruscus))

  • Mussel byssus is a bundle of threads used to attach mussels to wet substrates. Recently, a thin cuticle layer on the byssus has attracted public attentions due to its remarkable toughness - stiff as epoxy resin and extensible as rubber. Here, we observed ultrastructure of the cuticle layer in a far eastern mussel (Mytilus coruscus) to understand underlying mechanisms for the mechanical properties. The cuticle layer observed by TEM was composed of submicron-sized granular inclusions in a continuous matrix phase. In addition, ultrastructural study in the presence of tertiary amine (Tetraethylammonium, TEA) showed an evidence that the cuticle is stabilized by cation-${\pi}$ interaction.

Ultrastrucure of Germ Cells during Spermatogenesis and Some Characteristics of Sperm Morphology in Male Mytilus coruscus (Bivalvia: Mytilidae) on the West Coast of Korea

  • The ultrastructure of germ cells during spermatogenesis and some characteristics of sperm morphology in male Mytilus coruscus, which was collected on the coastal waters of Gyeokpo in western Korea, were investigated by transmission electron microscope observations. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves in that it contains a short midpiece with five mitochondria surrounding the centrioles. The morphologies of the sperm nucleus type and the acrosome shape of this species have an oval and modified cone shape, respectively. In particular, the axial rod is observed between the nucleus and acrosome of the sperm. The spermatozoon is approximately $45-50{\mu}m$ in length including a sperm nucleus (about $1.46{\mu}m$ in length), an acrosome (about $3.94{\mu}m$ in length) and tail flagellum (approximately $40-45{\mu}m$). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9+2 structure. Some special charateristics of sperm morphology of this species in the genus Mytilus are (1) acrosomal morphology, (2) the number of mitochondria in the midpiece of the sperm, and (3) the existence of a satellite. The axial rod appears in the acrosome and sperm nucleus as one of the characteristics seen in several species of the subclass Pteriomorphia, unlikely the subclass Heterodonta containing axial filament instead of the axial rod. The number of mitochondria in the midpiece of the sperm of this species in the family Mytilidae are five, as one of common characteristics appeared in most species in the family Mytilidae. Most of Mytilus species contain a satellite body which is attached to the proximal centriole in the middle piece of the sperm, as one of common characteristics of sperm morphology in the family Mytilidae.

해양오염의 진단을 위한 생화학적 오염지표에 관한 연구 X. 남해산 홍합 (Mytilus coruscus)의 산소 라디칼 및 제거효소의 변화 (Study on Biochemical Pollutant Markers for Diagnosis of Marine Pollution X. Changes in Oxygen Radicals and Their Scavenger Enzymes of the Mussel (Mytilus coruscus) in the South Sea)

  • 해양오염의 진단을 위한 생화학적 오염지표 설정의 기초연구의 일환으로, 오염이 심각한 남해산 홍합(Mytilus coruscus)의 근육중의 활성산소종 및 그들의 제거효소의 활성을 $분석{\times}평가하였다.$ 이들 홍합의 근육중의 단백질의 향량은 남해안 양식산 홍합의 근육중의 단백질 함량이 동해안의 포항의 자연산 홍합(대조군)의 단백질 함량에 비해 4~14%나 유의적으로 감소하였다.(p<0.05-0.01).남해안의 양식산 홍합의 근육중의 수퍼옥시드 라디칼의 생성량은 대조군으로 사용한 동해안 포항의 자연산 홍합의 근육중의 수퍼옥시드 라디칼의 생성량 대비 82~138%정도나 높앗다. 또한 남해안의 양식산 홍합의 근육중의 히드록시 라디칼의 생성량은 대조군으로 사용한 동해안 포항의 자연산 홍합의 근육중의 히드록시 라디칼의 생서량 대비 9~25%정도나 높았다.남해안의 양식산 홍합의 근육중의 SOD의 활성은 동해안 포항의 자연산 홍합의 근육중의 SOD의 활성대비 16~28%정도나 낮았다. 이상의 영구결과에서 볼 때 남해안 홍합의오염 지표로서는 단백질 함량의 감소, 활성산소종의 증가 및 활성산소의 제거효소로서 SOD의 활성감소로 평가각 가능하다라는 사실을 확인할 수 있었다.
