Study of Viral Effects of the Mycovirus (LeV) and Virus-Free Commercial Line in the Edible Mushroom Lentinula edodes

  • dsRNA was found in malformed cultures of Lentinula edodes strain FMRI0339, one of the three most popular sawdust cultivated commercial strains of shiitake, and was also found in healthy-looking fruiting bodies and actively growing mycelia. Cloning of the partial genome of the dsRNA revealed the presence of the RdRp sequence of a novel L. edodes mycovirus (LeV), and sequence comparison of the cloned amplicon showed an identical sequence to known RdRp genes of LeV found in strain HKA. The meiotic stability of dsRNA was examined by measuring the ratio of the presence of dsRNA among sexual monokaryotic progeny. More than 40% of the monokaryotic progeny still contained the dsRNA, indicating the persistence of dsRNA during sexual reproduction. Comparing the mycelia growth of monokaryotic progeny suggested that, although variations in the growth rate existed among progeny and virus infection was observed in highly actively growing progeny, there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny. This study attempted to cure the edible mushroom L. edodes strain FMRI0339 of the L. edodes mycovirus (LeV) in order to obtain an isogenic virus-free fungal strain as well as a virus-infected strain for comparison. Mycelial fragmentation, followed by being spread on a plate with serial dilutions resulted in a virus-free colony. Viral absence was confirmed with gel electrophoresis after dsRNA-specific virus purification, Northern blot analysis, and PCR using reverse transcriptase (RT-PCR). Once cured, all of fungal cultures remained virus-free over the next two years. Interestingly, the viral titer of LeV varied depending on the culture condition. The titer from the plate culture showed at least a 20-fold higher concentration than that grown in the liquid culture. However, the reduced virus titer in the liquid culture was recovered by transferring the mycelia to a plate containing the same medium. In addition, oxygen-depleted culture conditions resulted in a significant decrease of viral concentration, but not to the extent seen in the submerged liquid culture. Although no $discernable phenotypic changes in colony morphology were observed, virus-cured strains showed significantly higher growth rates and mycelial mass than virus-infected strains. We were also explored effects of LeV on fruiting body formation and mushroom yield. The fruiting body formation yield of virus-free L. edodes was larger than virus-infected L. edodes. These results indicate that LeV infection has a deleterious effect on mycelial growth and fruiting body formation. In addition, we have been investigated host-parasite interaction between L. edodes and its mycovirus interaction to study viral mechanism by establishment of proteomics.

Mycelial growth and fruit body cultural characteristics of a new Grifola frondosa variety, 'Bakyeon' (잎새버섯 신품종 '백연' 균사배양 및 자실체 생육 특성)

  • This study was conducted to reduce the phenomenon of the biased cultivation of certain mushroom varieties and to develop a competitive variety of Grifola frondosa. We developed the first Korean white commercial mushroom strain, 'Bakyeon', by crossing monokaryons derived from brown strains. We have collected and tested the characteristics of mushrooms from domestic and international genetic resources since 2018. We bred the unique domestic variety, 'Bakyeon', which has the following characteristics. The optimal temperature for mycelial growth was 25~28℃ and the optimal temperature for fruit body growth was 16~18℃. The new variety was similar to the control variety (Daebak) in terms of the pileus, which formed a pine cone shape, and the number of days of cultivation. The yield was 94.1 g/bottle, which was 23% lower than the 108.5 g/bottle yield of the control variety. When incubating the parent and control varieties, the replacement line was clear. Moreover, polymerase chain reaction analysis of mycelial DNA resulted in different band patterns between the parent and control varieties, confirming the hybrid species.

Cultural Characteristics of Fomitella fraxinea (Fr.) Imaz. (장수버섯의 배양적(培養的) 특성(特性))

  • A good mycelial growth of F. fraxinea was observed on CDA medium. The optimum temperature and pH for the mycelial growth of F. fraxinea was at $30^{\circ}C$ and pH 6.0, respectively. Glucose was found to be the best carbon source and arginine was favored as nitrogen source. When the basal medium was supplemented with organic acids, the best growth was shown in succinic acid and the poor growth was shown in oxalic acid. Thiamine.HCl showed the best results on the growth of this fungus on basal medium. Mycelial growth of F. fraxinea was quite good when oak tree sawdust was used to cultural substrates. The best mycelial growth was observed when 20% of rice bran was added as a supplement on sawdust substrates. Higher yield of F. fraxinea was observed on the medium with oak tree and acacia sawdust.

Varietal characteristics of cross-bred Cordyceps militaris 'Dowonhongcho' (교배육종에 의한 밀리타리스 동충하초 '도원홍초'의 품종특성)

  • Cordyceps militaris is being studied and cultivated as a medicinal mushroom having many valuable biological and pharmaceutical activities. In the breeding of new C. militaris mushroom, single ascospores were isolated and examined their mycelial growth, mycelial density, and production of stroma and perithecia. Among them selected isolates were crossed and hybrids were produced showing high quality fruiting bodies on artificial media. Mycelial growth rate of new strain 'Dowonhongcho' was higher than that of better on 'Yedang 3' on SDAY at $10-25^{\circ}C$. The stromata of new strain were club-shaped and bright orange-red. Its height was 6.1 cm and the cordycepin content was 0.34% on average. The new strain showed 9% higher yield than 'Yedang 3' with producing firmer fruit bodies. The optimum temperature for mycelial growth was $22{\sim}25^{\circ}C$ and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. Fruiting bodies were began to produce 45 days later after inoculation. This new cultivar may serve as a valuable one for artificial cultivation and industrial-scale production of C. militaris.

Effects on yield of Pleurotus ostreatus(Suhanneutari 1ho) according to afterripening conditions with bottle cultivation (후숙 조건에 따른 느타리(수한느타리버섯 1호)의 생육특성)

  • This study of afterripening conditions during oyster mushroom (Suhanneutari 1ho) cultivation in bottles was investigated. Medium materials were used poplar-sawdust (68%), cotton seed peel (16%), Beet pulp (8%) and cotton seed cake (8%). Mix of materials was used as a percentage of the volume and to adjust moisture content (67%). Autoclaved mediums were placed in low temperature storage ($20^{\circ}C$) and then moved in inoculation room and conducted mechanical inoculation. Mycelial culture temperature was maintained at $20{\sim}21^{\circ}C$ and cultured during 18 days. The afterripening period were 6days, 9days, 12days and 15 days at $25{\sim}27^{\circ}C$. The yield of fruit body was higher for 9 days (163.1g/bottle) and 12 days (160.7g/bottle) than that of other afterripening period. Second, in the changes in moisture content of the medium according to the afterripening period, no significant changes were observed during mycelial grwoth. The longer afterripening period was showed slightly lower weight of media. The moisture content of media after harvest at afterripening for 9 days had the biggest reduction than any other treatments. In addition, weight of media and yield of afterripening for 9 days were the lowest and highest, respectively.

Mycelial and cultural characteristics of a new high yield oyster mushroom variety,Mantari, for bottle culture (다수성 병재배용 느타리 신품종 『만타리』의 균사배양 및 생육특성)

  • 'Mantari' is a new variety of oyster mushroom for bottle culture. It was bred by crossing monokaryons isolated from 'DM11732' and 'Chunchu-2ho'. The optimum temperature for the mycelial growth was $26{\sim}29^{\circ}C$ on PDA medium, and that for the primordia formation and the growth of fruiting body of 'Mantari' on sawdust media was $18^{\circ}C$ and $16^{\circ}C$. It took 32 days to finish spawn running, 4 days to finish primordia formation, and 3 days to finish fruiting body growth in the bottle culture. The fruitcharacteristics: the pileus was round and gray-black in color, and the stipe was long, thin, and light gray in color. The yield per bottle was 179 g/900 ml and was 5% higher than that of the control variety (Chunchu-2ho). The physical properties of the fruit body were as follows: springiness, cohesive, gumminess, and brittleness of the stipe tissue were 96%, 76%, 160 g, and 15 kg, respectively.

Mycelial and cultural characteristics of a new high-yield Pleurotus pulmonarius cultivar 'Santari' for bottle culture (다수성 병재배용 산느타리 신품종 '산타리'의 균사배양 및 생육특성)

  • Oyster mushrooms are one of the most popular edible mushrooms in Korea. The 'Santari' cultivar bred in this study, which belongs to the species Pleurotus pulmonarius, is a new oyster mushroom cultivar for bottle culture. It was bred by mating monokaryons isolated from 'GMPO20404' and 'Hosan'. The optimum temperature for 'Santari' mycelial growth was $26-29^{\circ}C$ on PDA medium, and the temperatures for primordium formation and for growth of the fruit body on sawdust medium were $22^{\circ}C$ and $20^{\circ}C$, respectively. It took 34 days to complete the spawn run, 3 days to form primordia, and 3 days to finish fruit body growth in the bottle culture. The fruit body pilei were round in shape and brownish, whereas the stipes were long, thick, and white. The yield per bottle of 'Santari' was 172 g/1,100 mL, which was 43% higher than that of the reference cultivar ('Hosan'). The springiness, cohesiveness, gumminess, and brittleness of the stipe tissue were 87%, 82%, 193 g, and 16 kg, respectively. These physical property values of 'Santari' were lower than those of the control cultivar, except for the cohesiveness.

Studies on the Yield, Ecological and Morphological Characteristics of Several Strains of Lentinus edodes (표고의 각(各) 계통별(系統別) 발생양(發生量)과 생태적(生態的) 및 형태적(形態的) 특징(特徵)에 관한 연구(硏究))

  • Thirteen strains or crosses of Lentinus edodes obtained from home and abroad were compared for their mycelial characteristics on artificial media. Eleven of them were selected and inoculated on logs in order to study for their productivity, and the morphological characteristics of the mushrooms were investigated. 1) Seven strains, i. e., 11001, 12001, 13005, 13012, 13015, 12004 and 20101 were grown well on the sawdust medium. 2) The best mycelial growth and establish­ment of infection on logs were obtained from strains 20101 and 11001. 3) The highest yield of the mushrooms on logs during the six year periods were obtained from strains 11001, 12001 and 20101. 4) The highest yield of the mushrooms usually was obtained on the fourth year after inoculation. 5) The fruit body productivity in terms of dry weight recovery was the highest in strains 13005 and 20101. 6) The most productive period of the mushrooms during the years depended on strains. Most prevalent types of the strains were summer and autumn growing types, and more mushrooms were produced during these periods. 7) Morphological characteristics of the mushroom were more or less dependent on environmental conditions. 8) Thicker fruit bodies and relatively uniform shapes of the mushroom were produced by strain 20101.

Breeding of a new cultivar 'Dadam' for Lentinula edodes sawdust cultivation (표고 톱밥재배용 신품종 '다담' 육성)

  • A new cultivar of oak mushroom 'Dadam' was bred from monokaryotic strains of 'GMLE36062-4' and 'GMLE36288-34'. The optimum temperature for mycelial growth of the new cultivar 'Dadam' on potato dextrose agar was 19-22℃. Total cultivation period of the new cultivar, from innoculation to its first harvest, was 135-139 days, similar to that of the control cultivar 'Hwadam'. The pileus color and stipe thickness of the new cultivar were darker and thinner than those of 'Hwadam'. Total yield of 'Dadam' was 621 g per 3 kg substrate, and is higher than that of 'Hwadam' (371 g). In farmhouse field test, it showed that the period of mycelial growth, browning, and fruiting body formation were the same as those of the control cultivar L808. The number of available fruiting bodies of 'Dadam' was 15, and is lower than that of the control (47), therefore, it was possible to save workforce in thinning. The total yield for 2 flush was 480 g for 'Dadam', similar to the 473 g of the control cultivar.

Growth characteristics of oyster mushroom following the addition of wheat straw pellet as a substitute for beet pulp (비트펄프 대체 밀짚펠렛 첨가에 따른 느타리 생육 특성)

  • The present study investigated the optimum additive ratio of wheat straw pellet as a substitute for beet pulp during oyster mushroom cultivation. The chemical properties across treatments were pH of 4.8-5.5, total carbon content of 45.9-46.5%, total nitrogen content of 1.5-1.7%, and carbon-to-nitrogen ratio of 27.8-31.0. Mycelial growth was the fastest in a medium containing 20% wheat straw pellet (at 88.2 mm in 'Heuktari' and 70.3-79.6 mm in 'Suhan-1Ho'); however, there were no significant differences in mycelial density among the treatments. The characteristics of fruiting bodies by variety were as follows. In 'Heuktari', the valid stipe number per bottle (1,100 cc) was 22.5 and yield was 177.1 g/1,100 cc in the mixed medium containing 10% wheat straw pellet, with the values being comparable to those of controls. In 'Suhan-1Ho', the valid stipe number per bottle (1,100 cc) was 14.0 and yield was 151.2 g/1,100 cc in the mixed medium containing 10% wheat straw pellet, with the values being comparable to those of controls.
