The white button mushroom (Agaricus bisporus) is one of the most widely cultivated species of edible mushroom. Despite its economic importance, relatively little is known about the genetic diversity of this species. Illumina paired-end sequencing produced 43,871,558 clean reads and 69,174 contigs were generated from five offspring. These contigs were subsequently assembled into 57,594 unigenes. The unigenes were annotated with reference genome in which 6,559 unigenes were associated with clusters, indicating orthologous genes. Gene ontology classification assigned many unigenes. Based on genome data of the five offspring, 44 polymorphic simple sequence repeat (SSR) markers were developed. The major allele frequency ranged from 0.42 to 0.92. The number of genotypes and the number of alleles ranged from 1 to 4, and from 2 to 4, respectively. The observed heterozygosity and the expected heterozygosity ranged from 0.00 to 1.00, and from 0.15 to 0.64, respectively. The polymorphic information content value ranged from 0.14 to 0.57. The genetic distances and UPGMA clustering discriminated offspring strains. The SSR markers developed in this study can be applied in polymorphism analyses of button mushroom and for cultivar discrimination.
세계의 버섯 생산량은 매년 10-20% 증가해 왔으며 다품목화 되어가고 있다. 최근에는 큰느타리, 백령느타리 등이 새로운 품목으로 재배 면적이 증가하고 있다. 우리나라에서는 고려시대 저술한 김부식의 삼국사기(1145년)에 처음으로 금지(영지)와 서지가 기록되었고, 조선시대에는 16종류 이상의 농서 또는 의학서에서 버섯의 이용이 기록되었다. 상업적으로 이용되는 버섯으로는 지금까지 25종의 160품종이 보급되었다. 하지만 품종보호등록은 8품종만이 이루어져 아주 적은 편이다. 버섯산업이 1960년대 수출산업으로 육성되면서 자실체 생산량은 계속 증가해왔다. 2003년에는 181,828톤으로 생산가액 약 8,000억원을 능가할 것으로 추정된다. 버섯의 유효성분이 많이 알려지고 있는데 항균, 항염증, 항종양, 항바이러스, 항세균과 항기생물, 혈압조절, 심장혈관 장애 방지, 콜레스테롤 과소혈증과 지방과다 혈증 방지, 항당뇨, 면역조절, 강신장, 간장 독성 보호, 신경섬유 활성화, 생식력 증진의 효과가 있다. 따라서 버섯 의약품, 건강음료, 가공식품, Biotransformation에 의한 신기능성 제품 개발 등이 이루어지고 있다. 또한 버섯은 환경정화 기능이 뛰어나 환경오염 물질의 정화가 가능하며, 버섯재배후 배지를 이용하여 퇴비생산, 가축사료 생산에 이용된다. 한국의 버섯산업은 이미 가장 빨리 성장하는 농업 투자분야로 되었다. 그러나 버섯산업의 국제 경쟁력 강화를 위하여 국내고유의 품종개발에 의한 품종보호등록이 시급하다. 또한 저렴한 생산기술 개발과 유통구조 개선에 의한 생산량 조절이 이루어져야 버섯산업이 더욱 발전할 수 있을 것이다.
Two kinds of mushrooms, Gumji (金芝; Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; B.C 57~A.D 668; written by Bu Sik Kim in 1145) in Korea-dynasty. Many kinds of mushrooms were also described in more than 17 kinds of old books during Chosun-dynasty (1392~1910) in Korea. Nowadays, mushroom cultivation has been increased through out the world last decade years. Production of mushrooms has also been increased 10-20% and many varieties have been cultivated. Similar trends were also observed in Korea. Approximately two hundred commercial strains of 37 species in mushrooms were developed and distributed to cultivators. Somatic hybrid variety of oyster mushroom 'Wonhyeong-neutari' were developed by protoplast fusion, and distributed to grower in 1989. The fruiting body yield index of somatic hybrids of Pleurotus ranged between 27 and 155 compared to parental values of 100 and 138. In addition, more diverse mushroom varieties such as Phellinus baumi, Auricularia spp., Pleurotus ferulae, Hericium erinaceus, Hypsizigus marmoreus, Grifola frondosa, Agrocybe aegerita and Pleurotus cornucopiae have been attempted to cultivate in small scale cultivation. Production of mushrooms as food was 190,111 metric tons valued at 800 billion Korean Won (one trillion won if include mushroom factory products; 1dollar = 1,040 Won) in 2011. Major cultivated species are Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Lentinula edodes, Agaricus bisporus, and Ganoderma lucidum, which cover 90% of total production. Since mushroom export was initiated from 1960 to 1980, the export and import of mushrooms have been increased in Korea. Technology developed for liquid spawn production and automatic cultivation systems lead to the reduction of the production cost resulting in the increasement of mushroom export. However some species were imported because of high production cost for these mushrooms requiring the effective cultivation methods. Developing of effective post-harvest system will be also directly related to mushroom export. In academic area, RDA scientists have been conducting mushroom genome projects. One of the main results is the whole genome sequencing of Flammulina velutipes for molecular breeding. An electrophoretic karyotype of of F. velutipes was obtained using CHEF with 7 chromosomes, with a total genome size of approximately 26.7 Mb. The mususcript of the genome of F. velutipes was published in PLOS ONE this year. For medicinal mushrooms, we have been conducting the genome research on Cordyceps and its related species for developing functional foods using this mushroom. In 2013, Korea Food and Drug Administraion (KFDA) approved Cordyceps mushroom for its value as an immune booster.

Bacillus thuringiensis 656-3 which was isolated from a soil sample of mushroom house and showed high toxicity to mushroom flies, Lycoliella mali and Coboldia fuscipes, was surveyed for insecticidal effect in the oyster mushroom house.3. thutingiensis 656-3 was mass-cultured in the fermenter containing soybean cake(2%) and wheat bran (2%) as media source. Semi-for-mutation of B. thuringiensis 656-3 was performed with metamorphic starch only. When the formulation suspension containing $5{\times}{10^7}$ cfu was sprayed on the mushroom in mushroom house, the insecticidal effect of B. thuringiensis 656-3 to mushroom flies,1. mali and C. fuscipes, was maintained over 90% by the fifth day after starting spraying. The yield of oyster mushroom house with B. thuringiensis 656-3 was significantly increased compared to control. B. thuringiensis 656-3 represents a powerful biological insecticide for the control of mushroom flies.

This study was carried out to determine the basic mycelial culture conditions for Poria cocos growth. According to colony diameter and mycelial density, suitable media for mycelial growth were Malt yeast extract, Potato dextrose agar, Yeast extract agar, and Yeast malt agar. The optimum temperature for mycelial growth was between 25 and $35^{\circ}C$, and the optimum pH value was between 4 and 7. Carbon and nitrogen sources were fructose and yeast extract. The optimum C/N ratio was about 10 to 1 with 2% glucose. Other minor components for optimal growth were thiamine-HCl and nicotinamide as vitamins, acetic and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $FeSO_4{\cdot}7H_2O$ as mineral salts.
본 연구에서는 유럽 양송이 자원들을 SSR marker를 통해 유전적 다양성과 집단 구조, 유전적 분화에 대하여 분석하였다. 본 연구에서 유럽의 양송이 자원들은 유전적 거리기반의 4개의 그룹으로 나뉘었고 집단구조 분석을 통하여 2개의 subpopulation으로 이루어져 있었다. 본 연구에서 사용한 SSR 마커로 유럽의 양송이 자원들은 지리적 그리고 갓색으로 구분되지 않았다. 유전적 다양성은 유전적 거리기반의 그룹에서는 Group 4, 집단구조 분석을 통한 subpopulation에서는 Pop. 2의 다양성이 높았다. 그리고 양송이 자원들은 유전적 분화가 매우 낮았다. 본 연구의 결과는 차후 양송이의 육종 등에 이용 할 수 있을 것이다.

Lignosus rhinocerus is a macrofungus that belongs to Polyporaceae and is native to tropical regions. This highly priced mushroom has been used as folk medicine to treat diseases by indigenous people. As a preliminary study to develop a culture method for edible mushrooms, the cultural characteristics of L. rhinocerus were investigated in a range of culture media under different environmental conditions. Mycelial growth of this mushroom was compared on culture media composed of various carbon and nitrogen sources in addition to C/N ratios. The optimal conditions for mycelial growth were $30^{\circ}C$ at pH 6 and 7. Rapid mycelial growth of L. rhinocerus was observed on glucose-peptone and yeast extract peptone dextrose media. Carbon and nitrogen sources promoting mycelial growth of L. rhinocerus were glucose and potassium nitrate, respectively. The optimum C/N ratio was approximately 10 : 1 using 2% glucose supplemented as a carbon source in the basal media.
단백질 및 기능성 식품인 Lentinus tuber-regium의 분말을 이용하여 버섯분말의 입도, 이화학적 특성과 제빵시 반죽의 이화학적 특성에 미치는 영향 및 제빵 적성 등에 미치는 영향에 대하여 조사하였다. 버섯분말 입자크기는 $38\;{\mu}m$에서 $450\;{\mu}m$까지 비교적 큰 입자로 골고루 분포되어 있었으며, 특히 밀가루 입자 크기의 범위에 해당되는 $150\;{\mu}m$이하는 약 64% 정도를 나타냈다. 버섯분말의 일반성분 분석결과 수분 11.32%, 조단백질 11.22%, 회분 2.10%, 조지방 1.60%의 함량을 보였다. 버섯분말 2-10% 첨가에 따른 복합분 반죽의 mixogram 특성 측정 결과 수분 흡수율은 버섯분말 첨가에 따라 67.0-79%로 증가하였으며, peak time, peak height, band width, seven minute height는 대조구과 비슷한 경향을 나타냈다. Sedimentation value와 PK value는 버섯의 첨가량이 증가할수록 그 값은 감소하는 경향을 나타냈다. 호화특성 중 호화개시온도는 10% 첨가구외는 대조구와 버섯분말 첨가시 큰 차이가 없었으며, 최고점도는 버섯분말 첨가량이 증가할수록 약간 증가하는 경향을 보였다. 버섯분말 첨가에 의한 제빵특성 중 부피변화는 버섯분말 2, 4% 첨가구가 703.50cc로 가장 큰 값을 나타냈으며, 6% 이상의 첨가구에서는 대조구보다 크게 감소하는 경향을 보였다. 무게 변화는 버섯분말 첨가에 의해 증가하는 경향을 보였다. 비체적은 버섯분말 2, 4% 첨가구가 각각 3.98, 3.95cc/g으로 대조구 3.94cc/g보다 높은 값을 보였으나, 버섯분말 6% 이상의 첨가구에서는 첨가량이 증가할수록 비체적이 감소하는 경향을 나타내었다. 버섯분말을 0-10% 수준으로 첨가하여 제조한 빵의 표피와 내부의 색도 변화는 crust 색의 경우 버섯분말 첨가량이 증가할수록 L값, b값이 증가하는 경향을 보였으며 a값은 버섯분말 첨가량이 증가할수록 값이 감소하는 경향을 보였다. Crumb 색중 L값의 변화는 crust에서와는 대조적인 결과로 버섯분말 첨가량이 증가할수록 감소하는 경향을 보였다. 버섯가루를 첨가시켜 제조한 빵의 조직감 측정 결과 springiness와 adhesiveness는 대조구와 버섯분말 첨가구 사이에 유의적 차이가 없었으며 chewiness와 gumminess는 버섯분말 첨가량이 증가할수록 그 값이 증가하는 경향을 보였다. Hardness의 경우 버섯분말 첨가량이 증가할수록 hardness도 증가하는 경향을 보였으며, 버섯분말 2% 첨가구는 대조구와 유의적 차이를 보이지 않았다. 버섯분말 첨가 빵의 $4^{\circ}C$ 및 $25^{\circ}C$ 저장 결과 버섯분말 첨가량은 노화의 진행속도를 빠르게 한다는 것을 알 수 있었으며, 버섯분말 첨가 빵의 관능검사 결과 버섯분말 첨가량이 증가할수록 모든 측정치의 값이 감소하는 결과를 나타냈다. 버섯분말을 제빵에 이용시 4%까지 이용할 수 있을 것으로 사료된다.
The antioxidant properties of mushroom and sea tangle extracts and mustard leaf added to Kimchi were investigated by total phenolic content, DPPH radical scavenging, ABTS anion scavenging, FRAP reducing power, and ORAC value. Total phenolic content of functional Kimchi (FK) was significantly higher than that of control Kimchi (CK). DPPH radical and ABTS anion scavenging activities of functional Kimchi were significantly higher than those of control Kimchi by 2.7 and 1.7 fold, respectively (p<0.05). Also, the FRAP reducing power and ORAC value of functional Kimchi increased compared to those of the control Kimchi by 1.6 and 1.1 fold, respectively (p<0.05). Our results suggest that functional Kimchi made by mushroom and sea tangle extracts and mustard leaf may be a potent antioxidant source and could be developed as a antioxidant functional food that may be for the effective treatment of oxidant conditions. Through continuous research and development of functional Kimchi by the use of mushroom and sea tangle extracts, it will be necessary to try to identify other functions that could be useful for preventing various diseases.
This study was conducted in order to perform efficient extraction of lignocellulolytic enzymes amylase (EC 3.2.1.1), cellulase (EC 3.2.1.4), laccase (EC 1.10.3.2), and xylanase (EC 3.2.1.8) from spent mushroom compost (SMC) of Pleurotus ostreatus, P. eryngii, and P. cornucopiae. Optimal enzyme recovery was achieved when SMCs were extracted with 50 mM sodium citrate (pH 4.5) buffer at $4^{\circ}C$ for 2 hr. Amylase, cellulase, and xylanase activities showed high values in extracts from P. ostreatus SMC, with 2.97 U/g, 1.67 U/g, and 91.56 U/g, respectively, whereas laccase activity and filter paper degradation ability were highest in extracts from P. eryngii SMC, with values of 9.01 U/g and 0.21 U/g, respectively. Enzymatic activities varied according to the SMCs released from different mushroom farms. The synthetic dyes remazol brilliant blue R and Congo red were decolorized completely by the SMC extract of P. eryngii within 120 min, and the decolorization ability of the extract was comparable to that of 0.3 U of commercial laccase. In addition, laccase activity of the SMC extract from P. eryngii was compared to that of commercial enzymes or its industrial application in decolorization.
