The Effects of Isocaloric High-Fat Diet and Endurance Exercise on Insulin Resistance and Mitochondria Biogenesis (Isocaloric high-fat diet와 지구성 운동이 인슐린 저항성과 미토콘드리아 생합성에 미치는 영향)

  • The purpose of this research was to find effects of high fat diet and endurance exercise on mitochondria biosynthesis and insulin resistance in male Wister rat. Th rats were randomly separated into 4 groups: A, Isocaloric high fat diet group (50% of calories from fat), B. Chow group, C. Isocaloric high fat diet with endurance exercise (treadmill running, slop 8%, 23 m/min, 120 min/day, 5 days per week), D. Chow group with endurance exercise. Both Isocaloric high fat diet group and chow group were given an equal caloric composition with 3.2kcal/g. equal amount of food were checked every day and given to both groups. 4 weeks of high fat diet did not show any change in body weight and amount of body fat. Further the level of glucose and insulin in blood, and insulin-stimuilated glucose transport rate in epitrochlearis muscle was not affected by Isocaloric high fat diet. However, the endurance exercise showed statistically significant change in the level of insulin in blood. Although either Isocaloric high fat diet or endurance exercise alone did not change on mitochondria biogenesis marker, Isocaloric high fat diet with endurance exercise could induce the increased level of marker (p<0.05). Also, plasma free fatty acids were increased in this group. From this investigation, Isocaloric high fat diet with moderate-intensity endurance exercise is effective way to induce mitochondrial biogenesis.

Possible Relevance of Blind-side Skin Hypermelanosis in Cultured Olive Flounders, Paralichthys olivaceus to Chronic Stress (양식산 넙치, Paralichthys olivaceus의 무안측 착색증 발현에 있어 만성적 스트레스 관련성)

  • In the present study, to clarify a possible relevance of blind-side hypermelanosis to chronic stress in cultured flounders, P. olivaceus, a serial experiment was tried with comparison of biochemical stress factors between wild type and hypermelanic type in cultured olive flounders. The mean size of experimental animal was total length 21.5±0.42 cm and body weight 87.5±6.1 g. The initial malpigmented area rate on the blind side skin was 0.63±0.12% and 16.7±4.7%, respectively, in the wild type and the hypermelanic type. The stress factors surveyed in the experiment were glucose (GLU), total protein (TP), cortisol, free type-thyroid hormones (FT3 and FT4) in plasma, and also moisture, crude protein, crude lipid, and crude ash in body muscle. As a result, GLU and TP were higher in hypermelanic type than in wild type. Plasma cortisol was also higher in hypermelanic type than in wild type. In FT3 and FT4, any difference between two groups was not observed. In body nutrient factors, the moisture and the crude ash in body composition were not different between two groups, but the crude protein was low and the crude lipid was high in the hypermelanic type. Therefore, it is concluded that the blind-side hypermelanosis of cultured flounders could be related with a chronic stress.

Role of Gait Variability and Physical Fitness as a Predictor for Frailty Status in Older Women (여성노인의 허약 상태 예측을 위한 보행변동성 및 체력의 역할 검증)

  • This study examined the association of gait variability and physical fitness with frailty status in older women. In a cross-sectional design, 168 elderly women, aged 65 years and older (75.07±5.40 years), measured body composition, gait parameters gait variability, physical fitness variables, MMSE-DS and CES-D. Subjects were classified as robust, pre-frail, and frailty based on the Fried et al.(2001) criteria for frailty weight loss, exhaustion, low hand-grip strength, low gait speed, and physical inactivity. Logistic regression analyses were used to determine the odds ratio (ORs) and 95% confidence interval (CI) of frailty status for having gait variability and physical fitness levels. Compared to the robust group (OR=1), the frailty group had significantly higher ORs of having terminal double limb stance (OR=1.48, 95% CI=0.10-2.21, p=.049), step cadence (OR=2.06, 95%CI=1.20-3.43, p=.009) variability, and significantly lower ORs of having upper-strength (OR=0.49, 95%CI=0.31-0.77, p=.002) even after adjusting for age, education, comorbidity, K-IADL, MMSE-KC and CES-D score. The finding of this study suggested that terminal double limb stance, step cadence and upper body muscular strength were independent predictors of frailty.

The Effect of Baekhogainsam-tang on Metabolism through Modulation of the Gut Microbiota and Gene Expression in High-Fat Diet Induced Metabolic Syndrome Animal Model (고지방식이로 유도된 대사증후군 모델 동물에서 백호가인삼탕(白虎加人參湯)의 장내미생물 및 유전자 발현 조절을 통한 대사 개선 효과)

  • Objectives We aimed to find out the improvement effect of Baekhogainsam-tang (Baihu Jia Renshen-tang, BIT) on metabolic syndrome and alteration of microbiota and gene expression. Methods We used male C57BI/6 mice and randomly assigned them into three groups. Normal control group was fed 10% kcal% fat diet, high-fat diet (HFD) group was fed 45% kcal% fat diet and 10% fructose water. BIT group was fed same diet as HFD group and treated by BIT for once daily, 6 days per week, total 8 weeks. We measured their body weight and food intake every week and performed oral glucose tolerance test 1 week before the end of the study. Then we collected the blood sample to measure triglyceride, total cholesterol, high-density lipoprotein cholesterol, insulin, and hemoglobin A1c. We harvested tissue of liver, muscle, fat, and large intestine for quantitative polymerase chain reaction (qPCR) and histopathological examination. Fresh fecal samples were collected from each animal to verify alterations of gut microbiota and we used RNA from liver tissue for microarray analysis. Results The body weight and fat weight of BIT group were reduced compared to HFD group. The qPCR markers usually up-regulated in metabolic syndrome were decreased in BIT group. Bacteroides were higher in BIT group than other groups. There were also differences in gene expressions between two groups such as Cyp3a11 and Scd1. Conclusions We could find out BIT can ameliorate metabolic syndrome and suggest its effect is related to gut microbiota composition and gene expression pattern.

Evaluation of coat color inheritance and production performance for crossbreed from Chinese indigenous Chenghua pig crossbred with Berkshire

  • Objective: This work was to determine coat inheritance and evaluate production performance for crossbred pigs from Berkshire×Chenghua (BC) compared with Chinese indigenous Chenghua (CH) pigs. Methods: The coat color phenotypes were recorded for more than 16,000 pigs, and the genotypes of melanocortin 1 receptor (MCIR) gene were identified by sequencing. The reproductive performance of 927 crossbred BC F4 gilts and 320 purebred CH gilts was recorded. Sixty pigs of each breed were randomly selected at approximately 60 days of age to determine growth performance during fattening period, which lasted for 150 days for BC pigs and 240 days for CH pigs. At the end of the fattening period, 30 pigs of each breed were slaughtered to determine carcass composition and meat quality. Results: The coat color of BC pigs exhibits a "dominant black" hereditary pattern, and all piglets derived from boars or sows genotyped ED1 ED1 homozygous for MC1R gene showed a uniform black coat phenotype. The BC F4 gilts displayed a good reproductive performance, showing a higher litter and tear size and were heavier at farrowing litter and at weaning litter than the CH gilts, but they reached puberty later than the CH gilts. BC F4 pigs exhibited improved growth and carcass characteristics with a higher average daily live weight gain, lower feed-to-gain ratio, and higher carcass lean meat rate than CH pigs. Like CH pigs, BC F4 pigs produced superior meat-quality characteristics, showing ideal pH and meat-color values, high intramuscular fat content and water-holding capacity, and acceptable muscle-fiber parameters. C18:1, C16:0, C18:0, and C18:2 were the main fatty acids in M. longissimus lumborum in the two breeds, and a remarkably high polyunsaturated/saturated fatty acid ratio of ~0.39 was observed in the BC F4 pigs. Conclusion: The BC F4 pigs exhibit a uniform black coat pattern and acceptable total production performance.

Effects of dietary supplementation of polysaccharide from Agaricus blazei Murr on productive performance, egg quality, blood metabolites, intestinal morphology and microbiota of Korean quail

  • Objective: This study aimed to investigate the effects of dietary supplementation with Agaricus blazei polysaccharide (ABP) at varying concentrations on the performance, egg quality, blood biochemistry, intestinal morphology, and microflora of quail. Methods: The study involved a total of 2,700 Korean quails, which were randomly divided into three groups. The measured variables encompassed productive performance, egg parameters, carcass parameters, serum metabolites, immune response parameters, antioxidative properties, and gut microbiome. Results: The addition of ABP did not have a significant effect on average daily feed intake. However, it was found to increase the average daily egg weight and egg production rate, reduce the feed-egg ratio. There were no significant impacts on egg quality measures such as egg shape index, egg yolk index and color, egg yolk and protein content. However, ABP supplementation significantly increased the Hough unit (p<0.01) and decreased the rate of unqualified eggs (p<0.01). Regarding serum parameters, the inclusion led to an increase in total protein concentration (p<0.05) and a reduction in low-density lipoprotein cholesterol (p<0.05). There were no significant effects observed on immune indicators such as immunoglobulin A (IgA) and IgM. ABP supplementation increased the levels of serum antioxidant indicators, including glutathione peroxidase, total superoxide dismutase (p<0.05), and total antioxidant capacity colorimeter (p<0.05). Furthermore, ABP supplementation significantly elevated the intramuscular fatty acid content in quail meat. Additionally, ABP supplementation demonstrated a significant improvement in the diversity of gut microbiota and induced alterations in the composition of the gut microbiota. Conclusion: The findings of this study indicate that dietary supplementation of ABP enhanced production performance and antioxidant capacity while increasing the levels of polyunsaturated fatty acids in quail muscle.

Effects of Diet Food Containing Jerusalem Artichoke's Inulin, Lotus Leaf, and Herb on Weight and Body Fat of Obesity University Students (돼지감자의 이놀린, 연잎, 허브의 다이어트제제가 비만인의 체중 및 체성분에 미치는 영향)

  • This study was conducted to assess the effects of diet food containing Jerusalem artichoke's inulin, lotus leaf, and herb on weight and body fat. Participants in this study were selected based on the following criteria: BMI over $25kg/m^2$, body fat percentage higher than 25%, abdominal obesity level of 0.85 measured by body composition measurement unit (ZEUS 9.9 PLUS, Korea) total 24 female and male university students over 20 of age were assigned to two different groups: control group and diet group and the study was carried out for 30 days. When we measured what the tester's body weight and height, we used an automatic measure machin which is called IMI-1000 from Immanuel company and when we measured what the hip and wist circumference, we used a tape measure. When we measured body mss index (BMI), body Int, body mass, lean mass, waist/hip ratio (WHB), obesity index, we used a ZEUS9.9 PLUS-Korea which is based on bioelectrical impedance analysis, The food intake was checked by means of diet record method to be input into CAN program in order to analyze nutrient intake. Our findings indicated that the diet group, as compared to the control group, lost weight of approximately 2.5 kg and showed statistically significant difference. In addition the level of body Int, muscle, abdominal obesity, obesity, waist and bottom measurement all showed significant decrease after study period. However, there was no big difference in body fat percentage because both body fat level and muscle level dropped together, Putting all these together, diet food in this study containing Jerusalem artichoke's inulin, lotus leaf powder, and herbs powder including nettle, eucalyptus was found to be effective in significant reduction of weight and body fat and obesity-related body indicators. Also, it is considered that this diet food has potential to prevent and improve effectively obesity from abnormal fat accumulation.

Age Variation in Extractive Nitrogenous Constituents of the Cultured Ascidian, Halocynthia roretzi Muscle (양식산 우렁쉥이, Halocynthia roretzi 함질소 엑스성분의 연령차)

  • The muscle extracts of the ascidian, Halocynthia roretzi cultured for two and three years old on the southern coast near Chungmu and the eastern coast near Pohang of Korea, were analyzed for extractive nitrogen (EN), free amino acids (FAA), combined amino acids (CAA), nucleotides and related compounds (NRC), quaternary ammonium bases and guanidino compounds using specimens collected in February 1989 and in April 1989, and compared for those contents with each other. As for the amount of EN, no remarkable difference was found between two- and three-year-old samples collected at St. 1 in the spring and winter seasons, while at St. 2 in the spring season the two-year-old sample was distinctly lower than the three-year-old one. Taurine, proline, glutamic acid, glycine and alanine were the major FAA in every sample. The amount of taurine, the most prominent FAA, was higher in three-year-old sample than in two-year-old one regardless of sampling station and season. Most of the other major FAA showed a similar tendency to EN at both sampling stations in both seasons. Adenosine 5'-triphosphate (ATP), adenosine 5'-diphosphate (ADP), adenosine 5'-monophosphate (AMP), inosine 5'-monophosphate (IMP), inosine (Ino) and hypoxanthine (Hyp) were detected in all the samples and ATP, ADP and AMP were the major ingredients. The amounts of total NRC were in parallel with those of EN and total FAA. As for the contents of betaines, two- and three-year-old samples collected in the winter season exhibited a great discrepancy each other, the former being clearly lower than the latter, but no remarkable difference was observed between two samples of two groups in the spring season. In proximate composition of the muscles, the two-year-old sample was considerably higher in moisture content and lower in protein and glycogen contents than the three-year-old one at St. 2 in the spring season. The large discrepancies observed between two- and three-year-old samples from St. 2 seems to be attributable to the difference in size of samples rather than to the difference in age.

The Effect of Change in Meat Quality Parameters on Pig Longissimus dorsi Muscle by the Addition of Fermented Persimmon Shell Diet

  • The objective of the present study was to investigate the effects of growth performance, chemical composition and meat quality parameters by the addition of fermented persimmon shell diets (FPSD). The experimental animals were ninety-six Berkshires. The pigs were allotted at 8 per pen in front-open building with three replicate pens per treatment. Until $61{\pm}1kg$ live weight at 140 days, the animals were fed growing diet, after which, experimental samples were fixed at 0, 3, 5 and 7% FPSD as C, T1, T2 and T3 in the finishing diets. Pigs of $103{\pm}1kg$ live weight were slaughtered by electrical stunning. In growth performance, ADG increased more (p<0.05) in T2 than C and T1. ADFI (kg/day) was higher (p<0.05) in T2 than in other groups. Feed conversion ratio was lower (p<0.05) in T2 than in other groups. On longissimus dorsi muscle (LM), the content of moisture was lower (p<0.05) in T3 than in other treatments. Crude fat increased (p<0.05) by addition of FPSD. pH at 24 h ($pH_{24}$) decreased more (p<0.05) in T2 and T3 than C and T1 by addition of FPSD. WHC decreased (p<0.05) by addition of FPSD. In meat, Hunter $L^*$ and $a^*$ were lower (p<0.05) in C than in other treatments. Hunter $b^*$ was higher (p<0.05) in T3 than in other treatments. In back-fat, Hunter $L^*$ was higher (p<0.05) in C than in other treatments. Hunter $a^*$ was not different in C and T1 but increased (p<0.05) in treatments by addition of FPSD. Hunter $b^*$ increased (p<0.05) by addition of FPSD. In sensory evaluation scores, for fresh meat, the value of meat color was higher (p<0.05) in T2 and T3 than in C and T1. The value of marbling was lower (p<0.05) in C than in other treatments. The value of drip loss was higher (p<0.05) in C than in other treatments and was lower (p<0.05) in T2 and T3 than in other treatments. The results of cooked meat, the value of tenderness was not different in C and T1 but increased (p<0.05) in other treatments. The value of flavor was significantly higher (p<0.05) in T2 and T3 than in C and T1. The value of overall acceptability was increased (p<0.05) by addition of FPSD. Hence we can conclude that addition of FPSD affected growth performance and meat quality parameters, in particular, it improved crude fat, hunter $L^*$ and $b^*$, and sensory evaluation on pork and was also seen to affect $pH_{24}$ and WHC. FPSD can be used for improvement of meat quality parameters.

Purification and Characterization of the Red Carotenoprotein from the Muscle of Blue Mussel, Mytilus edulis (진주담치 근육중의 적색 Carotenoprotein의 정제 및 특성)

  • Most of carotenoprotein complexes have been extracted by using buffered solutions. However, in this study carotenoprotein from the muscle of Blue mussel(Mytilus edulis) was extracted by a detergent such as Triton X-100. It was purified and characterized by $20\%$ (w/v) $(NH_4)_2SO_4$, DEAE-cellulose ion exchange and Sephacryl S-300 gel filtration. The carotenoprotein(${\lambda}_{max}=462nm$) had an approximate M. W. of 372KDa(gel filtration). SDS-PAGE analysis of the carotenoprotein indicated the presence of four polypeptides of 60KDa($23.70\%$), 46.9KDa($9.14\%$), 26KDa($49.14\%$) and 13KDa($18.02\%$). Carotenoprotein denaturated by treatment with SDS to a final concentration of $0.2\%$ (w/v) caused a hypsochromic shift of ${\lambda}_{max}$ from 462nm to 456nm. The carotenoprotein contained lipids as structure units. The amino acid composition of the carotenoprotein contained large essential amino acid amounts of $62.8\%$, and the content of threonine($35.9\%$) was higher than other amino acids, but histidine, methionine and proline were not present. In the carotenoprotein, the major fatty acids were $C_{16:4},\;C_{16:0},\;C_{20:5}\;and\;C_{22:6}$. The percentages of polyunsaturated fatty acids($62.4\%$) were higher compared to other fatty acids(saturated fatty acids $19.6\%$, monounsaturated fatty acids $18.0\%$). Carotenoid was extracted from the carotenoprotein by acetone and it was separated into five different components by preparative TLC(benzene:petroleum ether:acetone=69:17:14). The major components of carotenoid were mytiloxanthin($74.79\%$) and 3,4,3'- trihydroxy-7',8'-didehydro-${\beta}$-carotene($18.26\%$), and they were at least presented as prosthetic groups of carotenoprotein.
