The synergy effect of trisodium phosphate (TSP) and ethanol against murine norovirus 1 (MNV-1), as a surrogate for human noroviruses, on fresh produces was evaluated. More than 2% (w/v) of TSP effectively inactivated MNV-1. The single treatment of 1% TSP or 30% ethanol for 30 min was not effective on MNV-1; however, cotreatment showed inactivation of MNV-1 on stainless steel and the produces of lettuce and bell pepper under 15 min. The results suggest that cotreatment of TSP and ethanol at a low concentration and a short time of exposure might be useful for the reduction of norovirus in some produce.

Flavonoids are phenolic compounds widely distributed in wide variety of edible plants including leafy vegetables, fruits, beverages. Quercetin is one of bioflavonoid compounds and has anti-tumor effect by suppressing tumor growth in vitro and in vivo, including multiple biological effects by antioxidant and effective anti-inflammatory agent. The present study investigated whether quercetin can enhance antioxidant enzyme activities (glutathione peroxidase: GPX, superoxide dismutase : SOD, catalase: CAT) and intracellular reactive oxygen intermediate (ROI) levels in the presence of taurine on B16F10 murine melanoma cells. From this result, the antioxidant enzyme activities of quercetin in the presence of taurine was enhanced. In addition, the same treatments decreased intracellular reactive oxygen intermediate levels on B16F10 murine melanoma cells. Taken together, these results demonstrate that the antioxidant effect of quercetin can enhance in the presence of taurine and it might play an important role in anti-tumor effect.
NAD(P)H:quinone reductase (QR), known as DT-diaphorase, is a kind of detoxifying phase II metabolic enzyme catalyzing hydroquinone formation by two electron reduction pathway from quinone type compounds, and thus facilitating excretion of quinoids from human body. With the usefulness of QR induction activity assay system for the modulation of toxicants, in the course of searching for cancer chemopreventive agents from natural products, the methanolic extracts of approximately two hundreds of oriental medicines were primarily evaluated using the induction potential of quinone reductase (QR) activity in cultured murine Hepa1c1c7 cells. As a result, several extracts including Hordeum vulgare, Momordica cochinchinensis, Strychnos ignatii, Houttuynia cordata, and Polygala japonica were found to significantly induce QR activity. In addition, the methylene chloride fraction of H. vulgare, one major dietary food source, showed potent induction of QR activity $(CD=6.4{\mu}g/ml)$. Further study for isolation of active principles from these lead extracts is warranted for the discovery of novel cancer chemopreventive agents.
To determine the effect of picolinic acid on NOㆍ production, murine macrophages were incubated with either medium, various concentrations of picolinic acid, or IFN-${\gamma}$ plus picolinic acid for 48 hr. Picolinic acid does not induce NOㆍ production by itself, it acted synergistically with INF-${\gamma}$ for the induction of reactive nitrogen intermediate production in murine macrophages. Thymidine incorporation appeared to be reciprocally related to nitrite levels, suggesting that IFN-${\gamma}$ plus picolinic acid induced NOㆍ synthesis exerted antiproliferative effects.
The retroviral Gag polyprotein directs the assembly of virion particles and plays an important role in some events after entry into a host cell. The Gag polyprotein of a virus mixture is responsible for inducing murine acquired immunodeficiency syndrome (MAIDS) when injected into susceptible strains of mice. In order to identify the host cellular proteins which interact with the MAIDS virus Gag proteins and possibly mediate the function of the Gag proteins, mouse T-cell leukemic cDNA expression library was screened using the yeast GAL4 two hybrid system. Of 11 individual positive clones, the clone Y1 was selected for the study of protein-protein interaction. Its DNA sequence revealed that it was an exact match to the murine SH3 domain-containing protein SH3P8. It is expressed as 2.4 kbp transcripts in testis at higher levels and in various tissues tested at lower levels. Glutathione S-transferase-Y1 fusion protein binds tightly to $Pr60^{def-gag}$ as well as $Pr65^{eco-gag}$.
The purpose of this research was to investigate effects of mixed extract of Ginseng Radix and Angelicae gigantis Radix on activity of murine splenocytes and macrophages. GAE (300 mg/kg) was administered p.o. for 7 days. GAE decreased the viability of murine splenocytes in vivo. Also, GAE enhanced the population of $Thy1^+$ cells in splenocytes and the population of splenic $CD4^+$ cells. Furthermore, GAE increased the production of ${\Upsilon}$-interferon from splenocytes. GAE enhanced the production of nitric oxide and the phagocytic activity of peritoneal macrophages. These results suggest that GAE regulates the immune response via activation of splenic Th1 cells and peritoneal macrophages.
The phagocytic activity of murine peritoneal macrophage, was determined by lucigenin chemiluminescence and engulfment of fluorescein-conjugated E. coli particle. The acti vity-guided fractionation upon the methylenechloride fraction of Aurantii immaturi pericarpium led to the isolation of a flavonoid, isosinensetin, as a suppressive component of phagocytosis. Isosinensetin suppressed the lucigenin chemiluminescence and the engulfment of fluorescein-conjugated E. coli particles and enhanced the production of nitric oxide in murine peritoneal macrophage.
Plant cell culture is emerging to express bioactive foreign proteins because it has several advantages in that it is safe, economical, genetically stable and eukaryotic expression system comparing with other expression systems. However several limitations such as slow growth rate, low expression level and lack of well established down stream process need to be answered. As a preliminary approach to produce the immunologically interested molecules through the plant cell culture, we tested if granulocyte-macrophage colony stimulating factors (GM-CSFs) from both murine (mGM-CSF) and human (hGM-CSF) are produced as a biologically active form through plant cell culture. The murine and human GM-CSF genes were cloned into the plant expression vector, pBI121, and Ti-plasmid mediated transformation of tobacco leaves was conducted using Agrobacterium tumefaciens harboring both recombinant GM-CSF (rGM-CSF) genes. Cell suspension culture was established from the leaf-derived calli of transgenic tobacco plant. Northern blot analysis indicated the expression of the introduced mGM-CSF gene in both transgenic plant and cell suspension cultures. In addition, the biological activities of both murine and human GM-CSF from plant cell culture were confirmed by measuring the proliferation of the GM-CSF dependent FDC-PI and TF-1 cells, respectively.
Ulcerative colitis (UC) is an inflammatory bowel disease of unknown etiology limited to the large intestine. The disease is prevalent in industrial societies and is associated with specific ethnic populations. A number of murine models, each focused on distinct aspects of the disease process, were developed over the past 20 years to further our understanding of the pathogenesis of UC. These models have been and remain our best resource for the study of the disorder as a result of their homology to human UC and the ease in which they can be manipulated and examined. This review examines and distills what has been learned from these models and how this information is related back to human UC.
The musk has been reported to have significant anti-inflammatory activities in clinical use and several animal models and we examined the effects of water soluble fraction(WSF) of the musk on murine peritoneal macrophages. WSF decreased the production of nitric oxide from the lipopolysaccharide(LPS)-treated murine peritoneal macrophages and also reduced the phagocytic activity of macrophages on the opsonized sheep red blood cells(SRBC). Transcriptional expression level of the inducible nitric oxide synthase(iNOS) was also decreased and the viability of the treated macrophages was not affected by WSF, suggesting that the effects could be partly explained by transcriptional regulation. Contrary to down-regulating iNOS expression, WSF slightly increased the release of tumor necrosis factor-alpha$(TNF-{\alpha})$, which implied its selective action on cellular pathways activated by LPS. Our results showed that anti-inflammatory activities of the musk could be partly explained by the inhibitory effects of the water soluble fraction on the macrophageal activation.
