Breeding of early maturing mulberry cultivar 'Suhyang' (Morus alba L.) for mulberry fruit production (조숙계 오디 생산용 뽕나무 품종 '수향' 육성)

  • We bred a mulberry cultivar named Suhyang through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test was carried out at four places (Suwon, Kongju, Wanju and Jinju) for six years from 2007 to 2012. This is belonging to (Morus alba L.) crossed between Josaenghongpisang (Morus alba L.) and Kaeryang (Morus alba L.) among mulberry genetic resources preserved in Suwon. Suhyang was high yielding cultivar in fruit productivity by 12% compared to control cultivar 'Chungil (Morus alba L.)' for four years. Although Suhyang was higher in sugar content of mulberry fruits, it was bigger than that of 'Chungil'. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.

This is tetraploidy variety belonging to (Morus alba L.) selected among mulberry genetic resources preserved in Suwon. Shimheungppong was high yielding cultivar in fruit productivity by 5% compared to control cultivar 'Chungilppong(Morus alba L.)' for four years. Although Shimheungppong was a little lower in sugar content of mulberry fruits, it was bigger than that of Chungilppong. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.

Evaluation of Biological Activity and Analysis of Functional Constituents from Different Parts of Mulberry (Morus alba L.) Tree (뽕나무(Morus alba L.) 부위별 생리활성 측정 및 기능성 물질 분석)

  • Evaluation of biological activity and analysis of functional constituents from water and ethanol extracts of four different parts of mulberry (Morus alba L.) tree were carried out to develop functional ingredients and foods using extracts of mulberry tree. The water and ethanol extracts of four different parts of mulberry tree were prepared and their biological activities and functional constituents determined by in vitro assays and HPLC, respectively. In general, ethanol extracts showed stronger biological activities and higher functional constituents than water extracts. Ethanol extracts of mulberry fruit, root bark, and twig showed stronger antioxidant ($IC_{50}=128.4{\mu}g/mL$), ${\alpha}$-glucosidase ($IC_{50}=12.0{\mu}g/mL$), and lipoxygenase ($IC_{50}=36.3{\mu}g/mL$) and tyrosinase ($IC_{50}=410.3{\mu}g/mL$) inhibitory activities, respectively, than those of other parts. Mulberry fruit and leaf showed the highest contents of anthocyanin (cyanidin 3-glucoside: 213.20 mg/100 g) and chlorogenic acid (514.97 mg/100 g), and especially ethanol extract of mulberry leaf contained higher quercetin 3-O-(6-O-malonyl)glucoside (143.25 mg/100 g) and kaempferol 3-O-(6-O-malonyl)glucoside (30.25 mg/100 g) contents without water extract of mulberry leaf. Meanwhile, mulberry twig contained both oxyresveratrol glycoside (48.90 mg/100 g) and its aglycone (21.88 mg/100 g), whereas mulberry root bark contained mostly oxyresveratrol glycoside (724.05 mg/100 g). Additionally, mulberry root bark and leaf contained much higher ${\gamma}$-aminobutyric acid (223.90 mg/100 g) and 1-deoxynojirimycin (86.07 mg/100 g) contents, respectively, than other parts of mulberry tree. These results suggest that high quality processed foods and functional foods using mixtures of mulberry fruits, leaves, twigs, and root barks should be developed for prevention and inhibition of several pathological disorders.

Comparison of Functional Constituents and Biological Activity of the Seed Extracts from Two Mulberry Fruits

  • The seeds from two mulberry fruits [Morus alba (MA) and Cudrania tricuspidata (CT)] were examined for their oil content, and fatty acid, phytosterol and tocopherol compositions and contents. Moreover, polyphenolic compounds and biological activity of the two defatted seed residue extracts were also evaluated. Oil contents of MA and CT seeds were 29.36% and 16.69%, respectively, while MeOH extracts of the defatted MA and CT seed residues were 5.10% and 6.22%, respectively. The two seed oils were composed of 81.4 and 74.37% linoleic, 5.75 and 11.39% oleic, 8.40 and 10.18% palmitic acid, and 3.52 and 3.0% stearic acids, and two other minor fatty acids, such as linolenic and arachidic acids. MA seed had higher contents of phytosterols (507.59 mg/100 g of oil), tocopherols (99.64 mg/100 g of oil), and total flavonoid (106.50 mg/100 g of seed) than CT seed, whereas CT seed had higher levels of total polyphenol than MA seed. The MeOH extract of MA seed residue showed higher antioxidant, anti-diabetic, and anti-melanogenic activity than that of CT seed residue. trans-Resveratrol (9.62 mg/100 g), quercetin (54.83 mg/100 g), and 4-prenylmoracin (48.70 mg/100 g), were found to be the main polyphenolic components in the MeOH extract of MA seed residue. These results indicate that MA seeds are good sources of essential dietary phytochemicals with antioxidant, anti-diabetic and anti-melanogenic activity.

Inhibitory Effects of Various Mulberry Fruits (Morus alba L.) on Related Enzymes to Adult Disease (품종이 다른 오디(Morus alba L.)의 성인병 관련 효소 억제효과)

  • The objective of this research was to develop the functional material of water and 60% ethanol extracts from nine kinds of mulberry fruits (Morus alba L.) that influence the inhibitory activity on angiotensin-converting enzyme, xanthine oxidase, ${\alpha}$-amylase, and ${\alpha}$-glucosidase. The total phenolic contents in the water extracts were over 2 mg/g in two species (Cheongilppong and Kangwon III) and five species (Daeyoupchosaeng, Cheongilppong, Kangwon III, Hihak, and Cataneo) of 60% ethanol extracts. The inhibitory activity against the angiotensin-converting enzyme was determined with them. Baekwoon III was $90.9{\pm}4.5%$ in the water extracts, and Hihak was $81.8{\pm}4.5%$ in the 60% ethanol extracts. The inhibitory activity of Kuksang 20 against xanthin oxidase was about 10% in the water extracts, and Cataneo was $21.4{\pm}2.3%$ in the 60% ethanol extracts. Six of the species (Daeyoupchosaeng, Suwonppong, Cheongilppong, Kangwon III, Hihak, and Kuksang 20) in the water extracts showed inhibitory activities against ${\alpha}$-amylase, as 100%, respectively. The inhibitory activity of ${\alpha}$-glucosidase was determined for these nine species. Four species (Baekwoon III, Daeyoupchosaeng, Cheongilppong, Kangwon III, Hihak, and Kuksang 20) in the water extracts and three species (Daechoukmyeun, Kangwon III, and Kuksang 20) in the 60% ethanol extracts showed inhibition of over 20%. The results revealed strong biological activity in spite of little total phenolic contents. These water and 60% ethanol extracts with high-quality biological activity from various mulberry fruits (Morus alba L.) are expected to represent good candidates for the development of antihypertentive and antidiabetes sources.

Occurrences of Insect Pests on Fruit-producing Mulberry Plants in Jeonbuk Province (전북지역 오디 생산용 뽕나무 해충의 발생양상)

  • Thirty-three species of insect pests including of two-spotted spider mite, Tetranychus urticae Koch, and air-breathing land snail, Acusta despecta Sowerby, were observed on mulberry for producing mulberry fruits in Jeonbuk province from 2015 to 2016. Of these, 8 species of insect pests, including of mulberry thrips, Pseudodendrothrips mori Niwa, are regarded as major insect pests. Of the 25 species observed before harvesting, 6 species-P. mori, Anomoneura mori, Pseudaulacaspis pentagona, Baris deplanata, Menophra atrilineata and T. urticae-are regarded as major insect pests. In contrast, of the 20 species observed after harvesting, 5 species-P. mori, P. pentagona, Glyphodes pyloalis Walker, Hyphantria cunea Drury and T. urticae-are regarded as major insect pests. Eight of the major insect pest species and nine other insect pest species-including Hexcacentrus unicolor Serville, Malcus japonicus Ishihara et Hasegawa, Halyomorpha halys stal, Xylosandrus germanus Blandford, four species of Repidoptera (Bombyx mandarina Moore, Euproctis subflava Bremer, Acronicta major Bremer, Agrotis trifurca Butler), and Acusta despecta Sowerby-were observed at all sites studied, although occurrence of the insect pests observed on mulberry varied from site to site.

Characteristics of Newly Bred Mulberry Cultivar "Daesungppong" (Morus Lhou(Ser.) Koidz.) for Mulberry Fruit Production (새로 육성한 오디 생산용 뽕품종 "대성뽕"의 특성)

Local adaptability test had been carried out at three places(Suwon, Konju and Cheongju) for three years since 2002. This is tetraploidy variety belonging to (Morus Lhou(Ser.) Koidz.) made by colchicine treatment on growing point of winter buds. Daeseongppong was high yielding in productivity by 48% compared to control cultivar "Chungilppong" for two years. Although Daesungppong was a little lower in sugar content of mulberry fruits, it contains more bioactive materials like C3G, rutin and amino acids than Chungilppong. It is adaptable to every where except the places where cold damage and sclerotial disease happen frequently.

Microbial Population of Foodborne Pathogens during Fermentation of Mulberry Wort (오디 발효액의 발효기간 동안 식중독 세균수의 변화)

  • Mulberry is considered a healthy food for antioxidants and many other beneficial nutrients. However, food safety concerns exist as this commodity scarcely passes through a sanitizing step due to the fragile nature of mulberry fruits. Fermented mulberry wort is one traditional way to utilize and preserve mulberries by mixing with sugars although hygienic practices are not often implemented. The purpose of this study was to investigate the fate of foodborne pathogens in fermented mulberry wort. Microbial population of inoculated E. coli in mulberry wort showed a decreasing pattern as the fermentation progressed. A quicker decrease was observed in the mulberry wort mixed with sugar at 1 to 0.8 ratio (w/w, mulberry: sugar) compared to 1 to 1 ratio, which could be due to the amount of acids generated during the fermentation process. When fully-fermented mulberry wort was contaminated with foodborne pathogens, they all decreased in population although their deceasing patterns varied depending on the species of tested bacteria. Steep deceases in population of inoculated foodborne pathogens were observed when the fermented wort was stored at $30^{\circ}C$ in comparisons to the other storage temperature, 5 and $20^{\circ}C$, regardless of bacterial species. This study necessitates the optimization of a sanitizing process during fermentation and storage of mulberry wort.

Mori Fructus Induces Cell Death through ROS-dependent Mitochondrial Apoptotic Pathway in Human Glioma Cells

  • Mulberry has been reported to contain wide range of polyphenols and have chemopreventive activity. However, little has been known regarding the effect of mulberry fruits on cell viability in human glioma cells. The present study was undertaken to examine the effect of mulberry fruit (Mar; Fructus) on cell viability and to determine its underlying mechanism in human glioma cells. Cell viability and cell death were estimated by MTT assay and trypanblue exclusion assay, respectively. Reactive oxygen species (ROS) generation was measured using the fluorescence probe DCFH-DA. The mitochondrial transmembrane potential was measured with $DiOC_6$(3). Bax expression and cytochrome c release were measured by Western blot analysis. Caspase activity was estimated using colorimetric kit. Mori Fructus resulted in apoptotic cell death in a dose- and time-dependent manner. Mori Fructus increased ROS generation and the Mori Fructus-induced cell death was also prevented by antioxidants, suggesting that ROS generation plays a critical role in Mari Fructus-induced cell death. Western blot analysis showed that Mori Fructus treatment caused an increase in Bax expression, which was inhibited by the antioxidant N-acetylcysteine (NAC). Mori Fructus induced depolarization of mitochondrial membrane potential and its effect was inhibited by the antioxidants NAC and catalase. Mori Fructus induced cytochrome c release, which was inhibited by NAC. Caspase activity was stimulated by Mori Fructus and caspase inhibitors prevented the Mori Fructus-induced cell death. These findings suggest that Mori Fructus results in human glioma cell death through ROS-dependent mitochondrial pathway in human glioma cells.
