아토피 피부염 동물모델에서 피부에 도포한 한약 복합추출물의 염증 억제 효과 (Topical Application of Herbal-mixed Extract on back skin suppresses inflammation in Ovalbumin-induced Atopic Dermatitis)

  • Objective : This study is conducted to evaluate the efficiency of ethanol extracts of 4 mixed herbs(CP001 or CP002) on mouse model of atopic dermatitis induced by ovalbumin. Methods : Female BALB/c mice were internally sensitized with ovalbumin($20{\mu}g$) plus aluminum hydroxide hydrate(4mg) once per week. After 3 weeks, they were dermally challenged with patches containing ovalbumin ($100{\mu}g$) plus aluminum hydroxide hydrate(20mg) every other day for 3 weeks. After induction of atopic dermatitis, mice back skin were gently rubbed with CP001 or CP002(200mg/$m{\ell}$, $100{\mu}{\ell}$) for two weeks(every 2 days). Results : In CP001 or CP002 treated group, there was a remarkable reduction in infiltration of eosinophils on the skin areas and diminution of mast cells and total T cells in blood samples as compared with control group. Cutaneous expressions of interleukin-13, 17 were also decreased by CP001 or CP002. Moreover, blood immunoglobulin E level was decreased by drug administration while there was no decrease in OVA sensitization group. Conclusion : In summary, our result shows that herbal extracts(CP001 and CP002) could be potential candidates for the treatment of chronic atopic dermatitis.

  • Oxidative stress is considered to play an important role in a variety of neurodegenerative disorders of central nervous system. The immortalized mouse hippocampal cell line, HT22, phenotypically resembles neuronal precursor cells but lacks functional ionotropic glutamate receptors, thus excluding excitotoxicity as a cause for glutamate triggered cell death. Therefore, HT22 cells are a useful model for studying oxidative glutamate toxicity. In this study, we examined whether the methanol extracts of some native plants at Mt. Baekdu could protect HT22-immortalized hippocampal cells against glutamate-induced oxidative stress. Seventy-eight plants sources were collected at Mt. Baekdu, and extracted with methanol. These extracts had been screened the protective effects against glutamate-induced oxidative damage in HT22 cells at the 100 and 300 ${\mu}g/ml$. Of these, thirteen methanolic extracts, Acer mono (leaf), Artemisia stolonifera (aerial part), Carduus crispus (aerial part), Carex mongolica (whole plant), Clematis hexapetala (whole plant), Galeopsis bifida (aerial part), Galium verum (whole plant), Ganoderma lucidum (whole plant), Ixeris chinensis (whole plant), Malva verticillata (aerial part), Polygonum senticosum (whole plant), Rebes mandshricum (branch), and Taraxacum mongolicum (aerial part), showed significant protective effects against glutamate-induced oxidative damage in HT22 cells.

  • Bitter melon (Momordica charantia) is used in traditional herbal medicine in many Asian countries for the treatment of several diseases such as diabetes, eczema, night blindness, psoriasis, and rheumatism. Especially, most reports concerning the biological activities of bitter melon have focused on its effects on diabetes and hyperglycemia. Also, bitter melon is regarded as a longevity food, suggesting that it has several beneficial effects on anti-aging and the maintenance of a healthy state. Thus, we investigated whether bitter melon could increase the capacity of exercise in this study. Interestingly, bitter melon fruit extract activated AMP-activated protein kinase (AMPK), which is important for regulating glucose homeostasis, mitochondrial content and exercise capacity. In addition, bitter melon extract increased the expression of enzymes involved in fatty acid oxidation such as mitochondrial uncoupling protein 3 (UCP3), carnitine palmitoyl transferase 1b (CPT1b), and pyruvate dehydrogenase lipoamide kinase isozyme 4 (PDK4). Moreover, exercise tolerance was much more enhanced in bitter melon treated animals compared to the non-treated control group. These results suggest that bitter melon is a promising candidate for the development of functional foods beneficial for physical strength and the enhancement of exercise capacity.

Increased Frequency of Foxp3+ Regulatory T Cells in Mice with Hepatocellular Carcinoma

  • The CD4+CD25+ regulatory T cell (Treg) is a special kind of T cell subset. Studies have showed that Treg cells are involved in a number of physiological processes and pathologic conditions such as autoimmune diseases, transplantation tolerance and cancer. Tregs with unique capacity for immune inhibition can impair anti-tumour immunity and help tumor cells to escape from immune surveillance. The aim of our study was to investigate whether Tregs are involved in hepatocellular carcinoma (HCC). A BABL/C mouse with HCC in situ model was established to evaluate the Treg existence in carcinoma tissues and the changes of Tregs in spleen using flow cytometry and immunohistochemistry methods. Granzyme B expression in carcinoma tissues was analyzed by immunohistochemistry to investigate the tumor local immune status.The proportion of CD4+CD25+/CD4+ spleen lymphocytes of tumor bearing mice ($18.8%{\pm}1.26%$) was found to be significantly higher than that in normal mice ($9.99%{\pm}1.90%$) (P<0.01 ). Immunohistochemistry of spleen tissue also confirmed that there was an increase in Treg in tumor-bearing mice, while in carcinomas it showed Treg cells to be present in tumor infiltrating lymphocyte areas while Granzyme B was rarely observed. Anti-tumour immunity was suppressed, and this might be associated with the increase of Tregs. Our observations suggest that the CD4+CD25+Treg/CD4+ proportion in spleen lymphocytes can be a sensitive index to evaluate the change of Tregs in hepatocellular carcinoma mice and the Treg may be a promising therapeutic target for cancer.

The Effect of Growth Hormone on mRNA Expression of the GABAB1 Receptor Subunit and GH/IGF Axis Genes in a Mouse Model of Prader-Willi Syndrome

  • Purpose: Growth hormone (GH) therapy substantially improves several cognitive functions in PWS. However, the molecular mechanisms underlying the beneficial effects of GH on cognition remain unclear in PWS. In this study, we investigated the effects of recombinant human GH on the gene expression of GABAB receptor subunits and GH/insulin-like growth factor (IGF) axis genes in the brain regions of PWS-mimicking mice (Snord116del). Methods: Snord116del mice were injected subcutaneously with 1.0 mg/kg GH or saline, once daily for 7 days. The collected brain tissues were analyzed for mRNA content using quantitative PCR (qPCR) in the cerebellum, hippocampus, and cerebral cortex. Results: GH increased the mRNA expression level of the $GABA_{B1}$ receptor subunit ($GABA_{BR1}$) and IGF-1R in the cerebellum. Furthermore, a significant positive correlation was found between the level of $GABA_{BR1}$ mRNA and the expression of the IGF-1R transcript. GH also induced an increase in the mRNA expression of IGF-2 and IGF-2R in the cerebellum. Conclusion: These data indicate that GH may provide beneficial effects on cognitive function through its influences on the expression of $GABA_{BR1}$ and GH/IGF-1 axis genes in PWS patients.

제스쳐 인식을 이용한 DID 인터페이스 구현 (Implementation of DID interface using gesture recognition)

  • 본 논문에서는 DID 시스템에서 사용할 수 있는 제스쳐 인식을 이용한 비접촉식 인터페이스를 구현하였다. 비접촉식 인터페이스는 별도의 부착물 없이 키넥트 카메라만을 사용함으로, 사용자의 편의와 공간적인 활용도를 높일 수 있다. 손 동작인식에는 사용자의 손 움직임의 기울기와 속력을 인식하는 방향성 기반의 인식 기법을 채용하였고 손 모양인식을 위해서 YCbCr 칼라모델을 이용한 손 영역 추출과 손 넓이의 원을 이용한 영상처리 기술로 손가락의 수를 인식하였다. 이러한 손 동작인식과 손 모양인식을 이용하여 다음 페이지, 이전 페이지, 화면 위로, 화면 아래로, 커서 움직임, 클릭 등의 이벤트를 발생시켜 DID 시스템 제어 명령으로 사용하였으며, 구현한 시스템을 갖고 동작 실험한 결과 93%의 명령 인식률을 보여 실용화의 가능성을 확인할 수 있었다.

인진, 울금, 구기자, 여정자 복합추출물(ACLL)의 항이상지질혈증 효능에 대한 실험적 연구 (Anti-dyslipidemic Effect of ACLL extract (Artemisia capillaris Thunb, Curcuma longa L, Lycium chinense, Ligustrum lucidum Aiton complex extract) on High Cholesterol Diet-induced Dyslipidemia in Mice)

  • Objectives : This study was aimed to investigate the effect of ACLL extract (Artemisia capillaris Thunb, Curcuma longa L, Lycium chinense, Ligustrum lucidum Aiton complex extract) in a dyslipidemia animal model induced by a high-fat diet. Methods : ACLL extracts was administered at 200 mg/kg, and 400 mg/kg to Apoe mice, where arthritis was induced by high cholesterol diet. After mice were treated with GO for 4 weeks, we measured AST, ALT, BUN and lipid levels (total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride), atherogenic index(AI) and cardiac risk factor(CRF). Results : Serum AST, ALT, creatinine, BUN levels were not changed by ACLL extract do not show any toxic effects. Also, ACLL extract groups were found to have atherogenic Index and cardiac risk factor as well as lipid metabolism improvement (total cholesterol, LDL cholesterol and triglyceride decrease). Conclusions : We suggest that ACLL extract may have the control effects of Dyslipidemia by improving lipid metabolism.

형개련교탕발효방(荊芥連翹湯醱酵方)이 NC/Nga mouse 동물병태에 미치는 영향 (Effect of Hyunggaeyunkyotangbalhyobang (HYBH) on Atopic Dermatitis in NC/Nga Mice Model)

  • Various related factors and tissue changes in vitro and in vivo were observed to investigate the efficacy of HYBH on atopic dermatitis. The results are described below. HYBH improved the atopic dermatitis symptoms by naked eye examination, and significantly decreased dermatitis clinical index at 14 weeks. HYBH significantly decreased CD4+/CD45+, CD4+, CD8+, CD3+/CD69+ immune cell ratios in PBMC by 28%, 16%, 30%, 26% and 22% respectively. HYBH significantly decreased CD11b+/Gr-1+, CD3 immune cell ratios in dorsal skin by 35.3% and 67.5% respectively. HYBH significantly decreased the expression of IL-4 and IFN-${\gamma}$ in spleen by 23% and 15% respectively. HYBH significantly decreased the production rate of IL-5, IL-13 and histamine in serum by 17%, 23%, and 8.8% respectively and increased IL-17 production by 17%. HYBH significantly decreased immunoglubulins IgG1 and IgE production in serum. The results above indicated that treatment of HYBH improved atopic dermatitis symptoms by anti-oxidant activity and immune modulation activity as a clinical evidence. Also, different fermentation conditions using various microbial strains should be accumulated as the clinical evidence for broad application in the future.

C57BL6 생쥐에서 NDM 헤어토닉의 모발성장 촉진 효과 (Effect of NDM Hair Tonic on Hair Growth Promotion in C57BL6 Mice)

  • NDM hair tonic is composed of several plant extracts which are known to be used in oriental medicine. This study was carried out to investigate the effect of NDM hair tonic on hair regrowth in an alopecia model of C57BL6 mice. Hair of six-weeks old mice were trimmed by electric clippers and electric shavers, so as not to damage the skin. The next day, mice without visible scraches were selected, randomized and separated in groups of 6 mice. There were four experimental groups including saline (negative control), 50% ethanol (vehicle control), 3% minoxidil (MXD), and NDM tonic. The test compounds were topically treated with 0.15 $m\ell$ per mouse per day for 2 weeks. The hair regrowth was determined photographically and histologically and the quantity of endocrine factors, IGF-1 and TGF-$\beta$, in the skin of mice was measured by PCR. No clinical signs were found in all animals. The topical treatment of NDM tonic for 2 weeks to dorsal skin accelerated hair regrowth faster than either the controls or MXD treatment. The NDM tonic treatment also promoted hair follicle development and elongation as compared with the controls or MXD treatment. Both NDM tonic and MXD treatment significantly increased the expression of IGF-1 and TGF-$\beta$ in the skin of C57BL6 mice as compared with the controls (p<0.05). These results suggest that NDM tonic has a hair growth activity and be useful for the treatment of baldness or alopecia.

Effects of Korean Red Ginseng Extract for the Treatment of Atopic Dermatitis-Like Skin Lesions in Mice

  • Atopic dermatitis (AD) is an allergic, inflammatory skin disease characterized by chronic eczema and mechanical injury to the skin, caused by scratching. Korean red ginseng (RG) has diverse biological activities, but the molecular effects of RG on allergic diseases, like AD, are unclear. The present study was designed to investigate whether RG inhibits 1-chloro-2,4-dinitrobenzene (DNCB)-induced AD in a mouse model. DNCB was applied topically on the dorsal surface of Balb/c mice to induce AD-like skin lesions. We observed the scratching behavior and examined the serum IgE level and interleukin (IL)-4 and IL-10 in splenocytes compared with dexamethasone. We also evaluated the DNCB-induced mitogen-activated protein kinases (MAPKs), NF-${\kappa}B$, and Ikaros activities after RG treatment using reverse transcriptase-polymerase chain reaction, Western blotting, and ELISA. Our data showed that the topical application of RG significantly improved the AD-like skin lesions and scratching behavior. RG decreased not only the mRNA expression of IL-4 and IL-10, but also the secretion of IL-4 protein and serum IgE in mice. Additionally, RG treatment decreased the DNCB-induced MAPKs activity and subsequent Ikaros translocation irrespective of NF-${\kappa}B$. We suggest that RG may be useful as a therapeutic nutrition for the treatment of AD.
