Engineering and Application of Zinc Finger Proteins and TALEs for Biomedical Research

  • Engineered DNA-binding domains provide a powerful technology for numerous biomedical studies due to their ability to recognize specific DNA sequences. Zinc fingers (ZF) are one of the most common DNA-binding domains and have been extensively studied for a variety of applications, such as gene regulation, genome engineering and diagnostics. Another novel DNA-binding domain known as a transcriptional activator-like effector (TALE) has been more recently discovered, which has a previously undescribed DNA-binding mode. Due to their modular architecture and flexibility, TALEs have been rapidly developed into artificial gene targeting reagents. Here, we describe the methods used to design these DNA-binding proteins and their key applications in biomedical research.

Influencing Factors of Bone Mineral Density in Men (남성의 골밀도 영향요인)

  • Purpose: Osteoporosis is often considered as a women's disease, but men's osteoporosis is also increasingly recognized as an important public health problem. This study was carried out to identify the important modifiable risk factors for osteoporosis in men. Methods: Two hundred and fourteen men were selected among those who checked bone mineral density by dual energy x-ray absorptiometry in lumbar vertebrae from June to September, 2009 at 3 general hospitals in Busan. Self-administrated questionnaires and BMD results of lumbar vertebrae were used for data. Collected data were analyzed by using SPSS 14.0 program. Results: The sports activity level in 30's, body weight, and fracture history of mother were the most powerful influencing factors on the bone mineral density of lumbar vertebrae. Conclusion: These results highlight the need to participate vigorously in sports activity during 30s as a means of increasing peak bone mass in lumbar vertebrae in order to prevent osteoporosis and fracture in men.

Family Factors Influencing on Adolescents′Self-Efficacy (청소년의 자기효능감에 영향을 미치는 가족변인)

  • This research was proposed to investigate the relative influences of family variables affecting adolescents'self-efficacy. For accomplishing the aim of this study, the data has been collected from 392 junior middle school students. Statistical methods for data analysis were mean, frequency, t-test, Pearson correlation, and multiple regression. Major findings were as following: 1. The average of self-efficacy was 3.07 and there was not found any significant difference between boys and girls in 4 sub -dimensional areas of self-efficacy. 2. Parental support, socioeconomic levels, and family functioning variables were significantly proved to be influencing on adolescents'self-efficacy. Among the variables, parental support was the most powerful variable on self-efficacy.

  • The EVMS(Earned Value Management System) comes from C/SCSC which was first released by the United States Department of Defense in December 1967, and preyed very powerful and efficient project management tool from a lot of practices. Although it is an excellent tool, we can not be succeed appling foreign system due to the differences of construction culture and law between the Korean and US construction industries. EAC(Estimate at Completion) is one of the most important functions in the EVMS. The purpose of this study is to propose the improved EAC method according to the performance indices better than old that and to prove from examples. In advance, the improved EAC method is to estimate more exactly costs and to promote efficiency in construction projects.

Factors Influencing Drinking Problems in Female University Students (여대생의 음주문제 관련 요인)

  • Purpose: The purpose of this study was to identify the factors influencing drinking problems in female university students to provide basic data for a nursing intervention program to improve health and prevent drinking problems. Method: Data were collected from September 17 to 21, 2007 through a questionnaire survey of 325 female college students in G City. Collected data were analyzed using descriptive statistics, pearson correlation coefficients and stepwise multiple regression. Result: The most powerful predictor of drinking problems was refusal self efficacy. The combination of the influence of friends, perceived stress, personality of novelty seeking, and alcohol expectancy accounted for 23.7% of the variance in drinking problems. Conclusion: From the results, I recommend that refusal self efficacy, influence of friends, perceived stress, personality of novelty seeking, and alcohol expectancy should be contained in developing nursing intervention programs for preventing drinking problems in female university students.

Marital Instability of Wives Who Experienced Abuse from Their Husbands (학대 경험 아내들의 결혼불안정성에 관한 연구)

  • This study was to investigate the relationships of personal resources, individual psychological variables, and relational characteristics with marital instability of wives who experienced physical and/or psychological abuse from their husbands, and to analyze the relative significance of related variables on marital instability. Self-administered questionnaire method was used to collect data from 500 wives residing in Chonbuk-do province, and data from 306 wives who reported to be psychologically and/or physically abused during last three years were used in the final analyses. Results showed that there were significant differences in the levels of marital instability according to wives' employment status, number of children, and occupational status of husbands. Correlational analyses indicated that wives' economic dependence, all individual psychological variables, and three relational variables were significantly correlated with marital instability of abused wives. Finally, multiple regression analyses showed that marital comparison level and degree of abuse from husband were the most powerful predictors on marital instability, and all these variables accounted for 61% of the total variance of abused wives' marital instability.

Analysis of Gender-Specific Relationships among Children's Important Self-Domain, Self-Evaluation and Global Self-Esteem (아동의 중요자아영역과 자기평가 및 자아존중감 간의 관계: 성별 분석)

  • The purpose of this study was to investigate the relationships among children's important self-domain, self-evaluation and global self-esteem by gender. Information was collected on 376 5-6th graders from elementary schools in Busan. The major findings were that 1) The self-evaluation of peer domain was the most powerful determinant on self-esteem in both boys and girls 2) The percentage that valued domain of family self was higher than the other groups in both boys and girls(boys 36.3%, girls 55.4%). 3) In boys' domains of peer self and computer self, important self-domain moderated the effect of self evaluation on global self-esteem.

An Analysis of Operating Policies for Multi-Product Unit Load AS/RS (다품종 단위적재 자동창고 시스템의 운영정책 분석)

  • In the past few years, increasing numbers of automatic storage/retrieval system (AS/RS) using computer controlled storage/retrieval machine have been installed. This paper introduces two modeling approaches to determine the best operating policy for AS/RS : an M/G/1 queueing model and a computer simulation model. The operating policy consists of three elements. : the operation command cycle, the storage location method, and the operation dispatching rule. The analysis based on M/G/1 model is suitable for a quick and approximate evaluation, due to its inherent strict assumptions. The computer simulation can be used to perform a more realistic analysis. It is shown through the study that a significant improvement in the throughput and/or the space requirement can be expected by determining the best operating policy to a particular system. Most important, the computer simulation demonstrates its powerful capability in evaluating dynamic stochatic systems with imperfect information.

  • Convolutional coding with Viterbi decoding is known as a powerful method for forward error correction among many kinds of channel coding methods. This paper presents a soft decision Viterbi decoder which has systolic array trace-back architecture[1]. Soft decision is known as more effective method than hard decision and most of digital communication systems use soft decision. The advantage of using a systolic array decoder is that the trace-back operation can be accomplished continuously in an array of registers in a pipe-line fashion, instead of waiting for the entire trace-back procedure to be completed at each iteration. Therefore it may be suitable for faster communication system. We described operations of each module of the decoder and showed results of the logic synthesis and functional simulation.

  • The power flow calculations(PFc) are the most important and powerful tools in power systems engineering. The conventional power now problem is solved generally with numerical methods such as Newton-Raphson. The conventional numerical method generally have some convergency problem, which is sensitive to initial value, and numerical stability problem concerned with matrix inversion. This paper presents a new power flow calculation algorithm based on the genetic algorithm(GA) which can overcome the disadvantages mentioned above. Some case studies with IEEE 6 bus system also presented to show the performance of proposed algorithm.
