The suggestion for mosquito control methods on mosquito habitat by land cover map classification (토지피복도 분류체계와 연계한 모기 서식지와 적용가능 방제법 검토)

  • Mosquitos serve as vectors for diseases, causing inconvenience as well as a threat to human life and health. Concern about mosquitos introducing and spreading new diseases has been intensifying. We observed a variety of mosquito habitats based on land cover classification from Korea's Ministry of Environment, and the mosquito species that could appear were classified according to the each habitat type. Finally, we suggested the best control methods for each type of habitat considering habitat characteristics and the ecological traits of mosquitos. Urban areas harbor various habitats for pests, contributing significantly to mosquito habitats. Control must be performed regarding larva and adults because various sources for habitats exist. Public mosquito control programs such as educational training, as well as information brochures can be effective in managing mosquito populations and public health. Agricultural areas show high densities of mosquito larva to lentic zones such as reservoirs, wetlands, paddy fields. So, biological control using natural predators may be effective in controlling mosquito populations. Forests are major habitats for Aedes albopctus, so physical controls should be deployed for residents living nearby, and excessive deforestation should be minimized. Other areas including aquatic ecosystems should be adopted regarding biological control using Bti (Bacillus thuringiensis var. israelensis) and chemical control for eradicating mosquitos. We classified habitats into four types of land cover patterns considering ecological traits and habitat preference, and suggest adequate control methods for each habitat type. Our suggestion can be used to positively contribute toward effective managing mosquito's density and reducing the damage to public health.

Control Effects against Mosquitoes Larva of Bacillus thuringiensis subsp. israelensis CAB199 isolate according to Different Formulations (Bacillus thuringiensis subsp. israelensis CAB199균주의 제형에 따른 모기유충방제 효과)

  • Among 18 Bacillus thuringiensis isolates with spherical parasporal inclusion from soils, B. thuringiensis subsp. israelensis CAB199 was selected. It was showing over 90% mortality against Aedes aegypti and Culex pipiens moletus. It was confirmed that this B. thuringiensis subsp. israelensis CAB199 isolate also had a insecticidal activity against Culex inatomii that was occurred in the marsh. Because most of mosquito larva were primarily situated or shifted from under- to surface water, we need to select long floating formulations on surface water for controlling mosquito larva. It was tested the pesticidal and control effects in the laboratory and wetland with two formulation types of B. thuringiensis subsp. israelensis, for example, wettable power (WP) and suspension concentrate type (SC). Laboratory test showed that SC formulation type was relatively faster and more effective against 3 tested mosquito species, C. pipiens, Aedes aegypti, and C. inatomii. Otherwise, the control efficacy of SC formulation type was more rapidly appeared against C. inatomii in the wetland.

Knowledge, Attitudes, and Practices Regarding Dengue Prevention Among Health Volunteers in an Urban Area - Malang, Indonesia

  • Objectives: The dengue prevention program known as "One House One Mosquito Larva Inspector" involves health volunteers who play a crucial role in the surveillance of mosquito larvae and reporting their findings to local public health officials. This study aimed to identify factors related to the knowledge, attitudes, and practices (KAP) of dengue prevention behavior among these health volunteers. Methods: A study was conducted in 5 sub-districts in Malang, an urban area in Indonesia. We employed a cross-sectional design and utilized a semi-structured questionnaire to assess the KAP of 400 health volunteers. Data were collected through face-to-face interviews. Results: Multiple logistic regression analysis revealed that individuals with a more positive attitude (odds ratio [OR], 1.69; p<0.05) and those with family sizes greater than five persons (OR, 1.90; p<0.05) were more likely to engage in effective dengue prevention practices. Additionally, possesing good knowledge was significantly assocated with more positive attitude (OR, 2.24; p<0.001). Furthermore, 40% reduction in positive attitude was observed in those over 45 years (OR, 0.60; p<0.05). The best practices most frequently reported by the majority of respondents included always reporting their surveillance activities (75.8%) and cleaning the water container in the bathroom at least once a week (65.2%). However, only 52.2% of respondents regularly checked for mosquito larvae in their neighborhood. Conclusions: Sustainable promotion and training for the "One House One Mosquito Larva Inspector" initiative are necessary, particularly among young health volunteers, to improve dengue prevention behaviors both within their own homes and in the surrounding environment.

Bioassay of Environment-friendly Insecticides for Management of Mosquito, Culex pipieos molestus (지하집모기 (Culex pipiens molestus)의 방제를 위한 환경친화적 살충제의 생물검정)

  • Bioassay of mosquito, Culex pipiens molestus, larva was investigated by several environment-friendly insecticides. These insecticides were Novaluron as chitin synthesis inhibitor, Metho-xyfenozide as ecdysone agonist, Pyriproxyfen as juvenile hormone mimic, and Spiromesifen as lipid biosynthesis inhibitor. The 50% lethal concentration $(LC_{50})$ of these insecticides were 0.00039, 0.07193, 0.65006 and 0.04839 ppm, respectively. Novaluron has lower concentration than any other insecticide. To determine the treatment time against larval stages, insecticides were applied to different larval stages of C. pipiens molestus. Mortality ratios of mosquito larva treated with Novaluron were 100.0, 84.5, 71.0 and 48.5% on 2, 4, 7 and 10 days after hatching from eggs, respectively. Otherwise, with the other insecticides, mortality ratios were under 80% with 2 days old larva. When exposure periods were tested to 3 or 4 days old larva against 4 insecticides, at least 3 hours were needed to 100% control effect against Novaluron, and over 12 hours with other insecticides.

  • The migut epitheluim of the last instar larva in the mosquito larvae, Anopheles sinensis was observed with electron microscopes. The midgut epitheluim of the mosquito larva is composed of a single-layered columnar absorptive cells, regenerative cells and secretory granular cells. The free surface of the columnar absorptive cells has a regular array of microvilli 'brush border', while cell membranes close to the basal lamina are extrmely infolded and a lot of mitochondria are concentrated in those processes. The columnar absorptive cells also contain cell organelles expected to be found in absorptive cell. Midgut regenerative cells which are positioned basally in the epithelium form the groups, which are called 'nidi', composed of 1 or $2{\sim}3$ cells, they show darker appearance than the columnar absoptive cells. The secretory granular cells contain numerous electron dense granules, $200{\sim}400$ nm in diameter. The cone shaped secretory granular cells are located in the basal portion of the midgut epitheluim. The epithelium is surrounded by the subepithelial space and muscle bundles. The subepithelial space, which is filled with fibrous connective tissue, is innervated by many axon cells and tracheoles.

The effect of four different temperatures on the growth of Aedes albopictus larva (네 가지 다른 온도가 흰줄숲모기(Aedes albopictus) 유충 생장에 미치는 영향)

  • We investigated to know the growth patterns of Aedes albopictus larva at the four different temperature conditions. Each of 120 individuals was placed into a $20m{\ell}$ vial and 12 sets (a set of 10 vials) were separated into 12 water tanks ($17{\times}24{\times}18cm^3$). Each water tank was composed of 3 the $1^{st}$ instar, 3 the $2^{nd}$ instar, 2 the $3^{rd}$ instar, and 2 the $4^{th}$ instar. Three sets of water tanks were placed under the four different incubator temperatures ($17^{\circ}C$, $21^{\circ}C$, $24^{\circ}C$, $28^{\circ}C$). We found that the eclosion rates were $20.00{\pm}5.77%$ at $21^{\circ}C$ and $3.33{\pm}3.33%$ at other temperatures. For the mosquito larva mortality rate, $1^{st}$ instar was $19.24{\pm}3.65%$, $2^{nd}$ instar was $16.48{\pm}3.25%$, $3^{rd}$ instar was $23.54{\pm}5.06%$, and $4^{th}$ instar was $40.74{\pm}7.08%$. The lowest mortality rate in growth stages according to temperature was $13.33{\pm}6.67%$ at $17^{\circ}C$ in $1^{st}$ instar larva, $7.41{\pm}7.41%$ at $21^{\circ}C$ at $2^{nd}$ instar larva, $10.74{\pm}6.43%$ at $24^{\circ}C$ in $3^{rd}$ instar larva, and $20.37{\pm}5.46%$ at $28^{\circ}C$ in $4^{th}$ instar larva. The survival period of mosquitoes in underwater were $26.33{\pm}0.67days$ at $17^{\circ}C$, $23.33{\pm}1.33days$ at $21^{\circ}C$, $20.00{\pm}2.52days$ at $24^{\circ}C$, and $11.67{\pm}1.20days$ at $28^{\circ}C$. From our results the most effective temperature to the normal growth of mosquito larva was $21^{\circ}C$, and the highest mortality rate was shown at the $4^{th}$ instar stage of larva growth. Our results would provide the basic data for the mosquito larva's growth pattern.

Mosquito Larvicidal Activity of Synechocystis PCC6803 Transformed with the cry11Aa gene to Culex tritaeniorhynchus and Anopheles sinensis (Cry11Aa 유전자로 형질전환된 Synechocystis PCC6803의 작은빨간집모기와 중국얼룩날개모기 유충에 대한 살충효과)

  • Bacillus thuringiensis produces crystal proteins toxic to medically and agriculturally important pests during sporulation. To improve the activity of insecticidal crystal protein in applying to mosquito larval control, an expression vector, pSyn4D harboring the mosquitocidal cry11Aa gene under control of psbA promoter of Amaranthus hybridus was constructed. This expression vector was transformed into Synechocystis PCC6803 and a transformant, Tr2C was selected with kanamycin. The mosquitocidal cry11Aa gene was stably integrated Into genomic DNA of Tr2C in PCR detection using cry11Aa-specific primers. The transformant expressed 72-kDa Cry11Aa protein and median lethal time (LT$\sub$50/) was approximately 2.1 days for Culex tritaeniorhynchus larvae and 0.7 day for Anopheles sinensis larvae, respectively. These results suggest this transformant can be used for mosquito larval control as a biological control agent.

Occurrence of Culex pipiens (Diptera, Culicidae) and Effect of Vent Net Sets for Mosquito Control at Septic Tanks in South-eastern Area of the Korean Peninsula (한반도 동남부 지역 정화조 내의 빨간집모기군(群)(Culex pipiens complex) 발생과 정화조 환기통의 방충망 설치에 의한 방제효과)

  • Culex pipiens complex larvae were found at the septic tanks in Busan and Ulsan cities located in South-eastern area of Korean peninsula. The larvae were collected in 25, 35 and 75 septic tanks out of 3,527, 622 and 1,111 septic tanks which the breeding rates were averages of 0.7%, 5.6% and 6.8% in Busan from January to May, 2003; Busan from February to March, 2004; and Ulsan from January to April, 2005, respectively. The average numbers of the mosquitoes in the tanks were 38.8, 14.9 and 13.7 larvae/dip with a 355-ml dipper. The larval densities were from 25.4 larvae/dip in January to 43.7 larvae/dip in April. 2003 which were not significantly different between them. It was the first report to find mosquito breeding at septic tink in Republic of Korea. Culex pipiens larvae were found in 6 out of 379 septic tanks at complex buildings (over $2,000m^2$ in size) in Ulsan from June to September, 2005, having an average of 1.6% for mosquito breeding rate. The mosquito larvae were collected in 8 and 530 septic tanks out of 210 and 3,729 septic tanks at large (over 300 homes) and small apartments (below 300 homes) with 3.8% and 14.2% of the average breeding rates, respectively At the septic tanks of private houses for below 50 persons, 708 out of 7,178 septic tan had Cx. pipiens larvae and pupae with 9.9% of the average breeding rate. Total average of the mosquito breeding rate was 10.9% of the investigated septic tanks of residential buildings in Ulsan. The number of mosquito larvae in the septic tanks were 15 to 75 larvae/dip. The bent net sets of septic tanks prevented from mosquito breeding to the rate of 93.1%.

Susceptibility of Medically Important Mosquito Larvae and Larvivorous Fishes to Abate and Abate-S in Korea (국내 주요 모기유충과 천적어류의 Abate 와 Abate-S 에 대한 감수성)

  • A study was performed on the susceptibility of Abate@ and Abate-S@ against 3rd instar larvae of 5 species of mosquitoes including Anopheles sinensis, Aedes dorsalis, Culex inatomii, Cx. pipiens pallens, Cx. tritaeniorhynchus and mosquito larvivorous fishes such as Chinese muddy loaches (Misgurnus muzolepis) and crucian carps (Carassius carassi~4s) in September, 1998. Cx. pipiens larvae showed that the LC5os of both Abatea and Abate-Sa were 0.006 ppm. The LC95 values of this mosquito species appeared 0.070 pprn of Abate@ and 0.035 pprn of Abate-S@ which were more susceptible than the dosage (1.0 ppm) recommended from WHO. An. sinensis larvae appeared 0.009 pprn of LC50 and 0.025 pprn of LCgs. The larvae of Cx. inatomii and Ae. dorsalis showed the most highly susceptible to Abate-S@ among the 5 mosquito species with LC50 values of 1.9 x pprn and 1.1 x pprn respectively, and the LC95 values were 5.2 x 10-hppm and 1.4 x ppm, respectively. On the other hand, Cx. tritaeniorhynchus appeared least affected by Abate-Sa. and was brought 0.048 pprn of LC50 and 0.808 pprn of LCg5. These two insecticides were much less toxic to the fishes than mosquito larvae showing the LC, value of Chinese muddy loaches at 24.145 pprn of Abate and 10.750ppm of Abate-S@. This fish species showed that the LCsos of Abate@ and Abate-S@ were 27.567 pprn and 14.775 ppm, respectively. In case of crucian carps, their LC, values were 7.914 pprn of Abate@ and 6.480 pprn of Abate-S@. Therefore, the maximum safe dosages of the insecticides to the fishes were 8 times as high as the values of LCg5 of the mosquito larvae. This study suggests that Abate@ and Abate-S@ demonstrated highly potential insecticides for further larger scale operational integrated control in some proper aquatic places if other aquatic invertebrates are also safe to the insecticides.

Characterization of Bacillus thuringiensis subsp. tohokuensis CAB167 Isolate against Mosquito Larva (모기유충에 활성 있는 Bacillus thuringiensis subsp. tohokuensis CAB167 균주의 특성)

  • Eight Bacillus thuringiensis strains activated against mosquito larva were compared their characterization. Spherical-shaped parasporal inclusion of B. thuringiensis subsp. tohokuensis CAB167 was observed by phase-contrast microscopy and scanning electron microscopy. $LC_{50}$ values of B. thuringiensis subsp. tohokuensis CAB167 against Culex pipiens molestus, Culex pipiens pallens, and Aedes aegyti were 173, 190 and 580 ng/ml, respectively. B. thuringiensis subsp. tohokuensis CAB167 had a parasporal inclusion containing 4 major protein components, for example, 135, 80, 49 and 28-kDa by SDS-PAGE. Otherwise, after trypsin digestion of parasporal inclusion, SDS-PAGE was showed new protease-resistant peptides at 72 and 63-kDa. Activated toxins of isolated CAB167 were different from other reference strains on a serological by immuno-diffusion test.
