상백피 물 추출물의 항산화 활성 및 항천식 효과 (Antioxidative and Anti-asthma Effect of Morus Bark Water Extracts)

  • 상백피의 기능성식품 소재로서의 이용 가능성을 조사하기 위하여 상백피 물 추출물의 항산화 활성 및 in vitro에서의 항천식 효과를 확인하였다. 항산화 측정 결과는 총 페놀함량은 35.2 mg/g로 나타났으며, DPPH radical 소거능은 35.3~59.8%, hydroxyl radical 소거능은 62.1~78.8% 범위로 높은 항산화 효능을 나타내었다. DNP-IgE와 HSA로 활성화된 RBL-2H3 세포에서의 항천식 효과는 농도가 증가할수록 유의적으로 $\beta$-hexosaminidase를 감소시켰으며, 염증성 cytokine인 IL-4, TNF-$\alpha$ 사이토카인 분비량 또한 유의적(p<0.05)으로 감소시켰다. 결과적으로 상백피는 높은 항산화력을 가지며, 항천식과 관련된 지표들을 유의적으로 감소시킴으로써 천식 예방 및 개선을 위한 기능성 소재로서 이용가치가 높을 것으로 사료된다.

상백피 추출물의 향균력 및 최적추출조건 검토 (Investigation of Optimum Extracting Condition and Antimicrobial Activity of the Extract from the Root Bark of Morus alba)

  • In order to develop a natural food preservative, the root bark of Morus alba was extracted with several solvents, and then antimicrobial activity was investigated. The optimum extracting condition for the antimicrobial substance from the sample, minimum inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. The antimicrobial activity of the ethanol extract form the sample was stronger than those of the extracts by the other solvents such as water, methanol. ethyl acetate and acetone. The optimum extracting condition for antimicrobial substance from the sample was shaking extraction twice for 5 hours at room temperature in case of 7 times of absolute ethanol added to the crushed root bark of Morus alba. The ethanol extract from the root bark of Morus alba had strong B. cereus, L. monocytogenes and S. aureus. Especially, Bacillus species was the most susceptible to the extracted substance. The ethanol extract showed antimicrobial activity against Gram negative bacteria(MIC, 160~1600 ug/ml) and yeasts(MIC, 1600 ug/ml) such as C. albicans and S. acidifaeciens. The extract also showed growth inhibition against molds such as A. niger, A. parasiticus, A versicolar and T. viride.

상백피 추출물이 미생물의 균체성분 및 형태 변화에 미치는 영향 (Effects of Treatment with the Extract from the Root Bark of Morus alba on the Cell Composition and the Shape Change of Microorganisms)

  • The ethanol extract from the root bark of Morus alba showed the strongest antimicrobial activity on the growth of almost all the tested microorganisms which were food-borne pathogens and food-related microorganisms. Therefore, fatty acid composition, amino acid composition and shape change of microorganisms treated with the ethanol extract from the root bark of Morus alba were examined. In effects of treatment with the ethanol extract on the fatty acid compositions of B. subtilis, S. aureus and E. coli, fatty aicd compositions such as hexadecanoic acid (16:0) and octadecanoic acid (18:2) of the tested strains were increased but pentadecanoic acid (15:0) heptadecanoic acid (17:0) and acid (16:1) and octadecenoic acid (18:1) of E. coli were decreased. The ethanol extract did not significantly affect the aminn acid composition of the tested strains. Transmission electron micrographs of microorgani는 treated with the ethanol extract exhibited morphological changes that irregularly contracted cell surface in S. aureus and destructed cell walls in B. subtilis and E. coli.

상백피 추출물로부터 향균성 물질의 분리정제 (Purification of Antimicrobial Substance for the Extract from the Root Bark of Morus alba)

  • The ethanol extract from the root bark of Morus alba showed the strongest antimicrobial activity on the growth of almost all the tested microorganisms which were food-borne pathogens and food-related microorganisms. 1) In order to isolate and purify of antimicrobial substance extracted from the root bark of Morus alba, the antimicrobial substance from the ethanol extract which exhibited a strong antimicrobial activity was purified by solvent fractionation, silica gel column chromatography, TLC and HPLC. Among the fractions fractionated by 4 kind of solvents from the ethanol extract, the antimicrobial activity of ethyl acetate fraction had the strongest antimicrobial activity against B. subtilis. Unknown compound were isolated from the ethyl acetate fraction by silica gel column chromatography, TLC and HPLC and the compounds showed strong absorbance at 207, 217 and 285 nm, therefore, it was supposed to be a kinds of aromatic compound.

  • The root-bark of the mulberry tree has been used as a medicine with antitussive, antiphlogistic, diuretic etc. in Korea. Among many kinds of mulberry trees, Morus Lhou has been widely cultivated in our country. In this experiment, three substances were obtained from the root-bark of Morus Lhou removed its cork layer by column chromatography of methanol extract. One of the substances was supposed as ${\alpha}$-amyrin acetate, another was alyceride, the other was confirmed as ${\beta}$-sistosterol by chemical and physical methods.

  • Background: Inflammation plays an important role in various diseases, including ulcerative colitis, Behcet's disease, and rheumatoid arthritis. In this study, we investigated the anti-inflammatory effects of Morus alba L. extracts obtained using different extraction methods (water extraction or high temperature extraction) on RAW264.7 cells. Methods and Results: Extracts from the central part (including the heartwood, sapwood, cambiun, and phloem) and bark (including the periderm and cortex) of Morus alba L. were obtained using either water or high temperature extraction. The following extract were obtained: MA1, water extract from the central part of Morus alba L., MA2, high temperature extract from the central part of Morus alba L., MA3, water extract from the bark of Morus alba L., and MA4, high temperature extract from the bark of Morus alba L. None of these extracts was observed to be cytotoxic to RAW264.7 cells. The MA2 extract reduced the production of LPS-induced NO (nitric oxide), $PGE_2$ (prostaglandin $E_2$), $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$ production in LPS-stimulated RAW264.7 cells. Conclusions: These results indicated that the inflammatory response was moderated by MA2. Treatment with MA2 could be used as a natural medicine for treating diseases involving inflammation. However, further experiments are required to determine how the high temperature extraction method alters the active ingredients in the extract and influences the anti-inflammatory effects of Morus alba L..

상백피(Morus alba root bark)로부터 triterpenoid의 분리 및 동정 (Isolation and Identification of Triterpenoids from the Mulberry (Morus alba) Root Bark)

  • 뽕나무(Morus alba L.) 뿌리껍질을 실온에서 80% MeOH 수용액으로 추출하고 이 추출물을 EtOAC 분획, n-BuOH 분획, $H_2O$ 분획으로 나누었다. EtOAc 분획에 대하여 silica gel과 ODS column chromatography를 반복 실시하여 5종의 triterpenoid를 분리, 정제하였다. NMR, IR, 및 EI/MS 등을 해석하여, ${\alpha}$-amyrin (1), ${\alpha}$-acetyl amyrin (2), lupeol (3), betulinic acid (4) 그리고 glutinol (5)로 구조동정 하였다. 화합물 중 a-amyrin (1), lupeol (3) 그리고 glutinol (5)은 상백피로부터는 이번 실험에서 처음으로 분리되었다.

조선시대 소고기 연화제의 연화효과 비교분석 (Comparative Analysis on the Effect of Beef Tenderizers in Joseon Dynasty)

  • One of the main processes of tenderizing beef in Joseon Dynasty was chemical methods involving Apricot seeds, manchu cherry twig and leaves, bamboo skins, mulberry tree bark, mangsa (硭砂), salmiacum (磠砂), alcohol, fermented malt, and original honey. This study analyzed and compared the effect of broussonetia papyrifera, fermented malt, cherry trees, and mulberry tree bark from old cookbooks. Tenderizing beef with cherry trees was most effective in the experiment on shearing force, TPA, and electrophoresis of beef. According to sensory evaluation and electrophoresis test results, tenderized beef with mulberry tree bark was slightly more preferred over the method using cherry trees. However, in accordance with the above mentioned experiment, quantitative descriptive analysis showed that the most common tenderizing material was derived from morus alba powder.

뽕나무(Morus alba)와 꾸지뽕나무(Cudrania tricuspidata)의 수용성 추출물에 의한 항산화 활성 (Antioxidative Activities by Water-Soluble Extracts of Morus alba and Cudrania tricuspidata)

  • 뽕나무(Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata)의 잎과 줄기 껍질의 수용성 추출물에 대하여 in vitro 실험계에서 항산화 활성을 비교 검토하였다. 성장기 마우스의 간장 microsome을 이용한 생체막 지질 과산화 억제정도는 꾸지뽕나무 줄기 껍질(53%)>뽕나무 줄기 껍질(43%)>꾸지뽕나무 잎(38%)>뽕나무 및(34%) 순으로 나타났다. Linoleic acid산화 실험계에서는 꾸지뽕나무 줄기 껍질과 뽕나무 잎의 추출물이 강한 항산화 활성을 나타내었으며, 꾸지뽕나무 줄기 껍질의 추출물은 반응 8일째까지 강한 활성을 보였다. TBA method에서는 뽕나무 줄기 껍질이 다른 것에 비해 약간 강한 활성을 보였으며, 또한 DPPH에 의한 수소공여능은 각 시료첨가 0.05%에서 뽕나무 잎>꾸지뽕나무 잎>통나무 줄기 껍질>꾸지뽕나무 줄기 껍질 순으로 나타나 및 추출물에서 비교적 수소공여 작용이 강하였다. 이들 추출물의 총 폴리페놀 화합물의 함량을 측정한 결과 뽕나무 잎(1.32%), 뽕나무 줄기 껍질(1.28%), 꾸지뽕나무 잎(1.34%), 꾸지뽕나무 줄기 껍질(1.30%)로 큰 차이가 없었다.

상백피 품종에 따른 ${\gamma}$-Aminnobutyric acid 함량 비교 (Varietal Comparison of ${\gamma}$-Aminnobutyric acid Content in Mulberry Root Bark)

  • From mulberry root bark of 13 various varieties, ${\gamma}$-Aminnobutyric acid(GABA) contents were examined on HPLC analyzer. GABA contents in mulberry root bark were affected by genotypes. Average GABA content of the 13 mulberry varieties was 2.19 mg/gDW. GABA content of the varieties ranged from 1.70 to 2.62 mg/gDW. Among the three genotypes, GABA content of the Morus Lhou was higher than the other genotype. In GABA content Hongolppong was the highest with 2.62 mg/gDW on the 13 varieties of mulberry root bark.
