Different parts of the mulberry plant are described to be potential sources of polyphenolics exhibiting strong antioxidant activity. In this study, we prepared various aqueous extracts of mulberry root bark by subjecting to steam at different temperatures and time intervals (45℃, 15 h; 70℃, 15 h; 95℃, 6 h and 95℃, 15 h) followed by extracting at 80℃ for 1 h. The total polyphenolic content ranged from 66.82-101.20 mg gallic acid equivalent (GE)/g of extract whereas the flavonoids were in the range of 13.03-25.23 mg catechin equivalent (CE)/g of extract. The extracts also exhibited strong antioxidant activities (0.99-1.66 mg trolox equivalent (TE)/g of extract in DDPH assay and 10.65-16.26 mg TE/g of extract in ABTS assay). This study clearly showed an improvement in the antioxidant activity of the water extract of mulberry root bark by the steam treatment, which can be used as a tea or health-promoting materials.

Morus alba root extract (MARE) has been used to treat hyperglycaemic conditions in oriental medicine. Here, we studied whether MARE possesses a cytotoxic effect on neuroblastoma. To check the cytotoxicity generated by MARE was whether relatively higher against the cancer cells rather than normal cells, we chose a neuroblastoma cell line (B103) and a normal cell line (Rat-2). A CCK assay revealed that MARE ($10{\mu}g/ml$) reduced cell viability to approximately 60% compared to an untreated control in B103 cells. But in Rat-2 cells, MARE induced relatively lower cytotoxicity. To investigate the mechanisms underlying the cytotoxic effect of MARE, we used flow cytometry combined with immunoblot analyses. We found that MARE-treatment could accumulate ROS and depolarize mitochondria membrane potential of B103 cells. Further treatment with MARE in B103 cells also could damage DNA and induce apoptosis. An expression study of p-Akt also suggested that there was a reduction in cellular proliferation and transcription along with the process of apoptosis, which was further evidenced by an increase in Bax and cleaved-caspase 3 activity. Together, our findings suggest that MARE produces more cytotoxicity in cancer cells while having a relatively attenuated effect on normal cells. As such, MARE may be a safer option in cancer therapeutics, and it also shows potential for the patients with symptoms of hyperglycemia and cancer.

The effects of four extracts of medicinal herbs, Rubus coreanum, Acorus calamus, Morus alba and Rehmannia glutinosa on hair growth activity and acnes control were investigated. In the course of screening natural extracts for hair growth, we found that the extract of dried root of Rubus coreanum has the hair growth promoting effect. After topical application of these extracts to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by treatment of these extracts. Furthermore these extracts do not possesspotent inhibitory effect on $5{\alpha}-reductase$ I and II activity and anti-bacterial effect on Escherichia coli , Propionibacterium acnes, Pityrosporum ovale, Staphylococcus aureus, Staphylococcus epidemidis, and Candida albicans. RT-PCR analysis showed that these extracts did notinduce mRNA levels of growth factors such as insulin-like growth factor-I, keratinocyte growth factor, hepatocyte growth factor and vascular endothelial growth factor in dermal papilla cells. These results suggest that Rubus coreanum has hair growth promoting effect. However, the effects of these materials on the hair growth promotion are not mediated through inhibition of $5{\alpha}-reductase$ I and II activity, stimulation of hair follicle cells and expression of growth factors in the dermal papilla cells.
Objectives : The root bark of Morus alba L. (Mori Cortex Radidus; MCR) has been traditionally used to reduce heat from the lungs, soothe asthma, and edema and to promote urination. In this study, we investigated the effect of MCR ethanol extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : Mice were sensitized at day 0, 7 and 14 with 0.2% OVA and then airway challenged at day 21, 23, 25, 27 and 29 to induce allergic asthma. MCR extracts at doses of 50 and 100 mg/kg body weight (bw) were orally administered during OVA challenge once per a day. The levels of allergic mediators such as histamine, OVA-specific IgE, IFN-${\gamma}$ and IL-4 were measured in the sera of mice by ELISA. The histopathological change of lung tissues was observed with hematoxylin and eosin (H&E) staining. Results : MCR extract significantly decreased not only the serum levels of histamine, OVA-specific IgE, and IL-4 compared with those of OVA control group, but significantly increased the serum level of IFN-${\gamma}$. In H&E staining, MCR extract inhibited the infiltration of inflammatory cells and bronchiolar damage with epithelial thickening in lung tissues of OVA-induced asthma mice. Conclusions : These results indicate that MCR extract inhibits lung damage by asthma through regulating the allergic immune response, suggesting that MCR may be used as a useful agent for the treatment of allergic asthma.

This study was performed to examine the shelf life and qualities of wet noodle with Morus alba L. root and Curcuma aromatica extracts (MCE) during storage at $4^{\circ}C$. Lightness and redness of wet noodle were decreased with increasing amounts of MCE added in noodle while yellowness was increased. The viable cell and molds count of wet noodle with MCE was reduced about 1~2 log cycle as compared with control during storage time. Also the TBARS (thiobarbituric acid reactive substance) value of wet noodle with MCE was lower than that of control. Hence the wet noodle with MCE has shown remarkable antioxidation effect. In sensory evaluation, the wet noodle containing the ratio of 2.5:0.02 of M. alba : C. aromatica was preferred than the control. From these results, the addition of 2.5% of M. alba and 0.02% of C. aromatica extracts in wet noodle had a good effect on improvement of preservation and development of quality.
Root system and distribution of soil components (pH, available P, K, Ca and Mg) were studied in a 20-year old, low-cutting mulberry field. The variety, kairyonezumigaeshi (Morus alba L.), was planted in a spacing of 1.8m${\times}$0.6m(740 tiess/10a). Roots and soil samples were taken from each of 196 blocks in a demension of 26${\times}$17${\times}$10cm(as shown in Fig. 1) from the soil between rows. 1. Whereas root system concentrated towards the stock, chemical components, pH, available P, Ca and Mg, were increasing toward the center of the row spacing. 2. Root system and distribution of chemical components between trees were not significantly different among blocks. 3. Roots were distributed most densely in a depth of 10∼20, intermediate 20∼30 and 30∼40, and least 0∼10cm. However, the concentrbtion of chemical components was the highest in a depth of 0∼10cm and decreased with soil depth. The soil. a depth of 0∼10cm on the center of row spacing, was the lowest in the root system and the richest in a chemical components. The results indicate that special fertilizinng management is required for the efficient absorption of nutrients.
One hundred and twenty six Vietnamese traditional herbals belonging to 59 families were screened for their free radical (DPPH) scavenging activity and inhibitory effect on lipid peroxidation. Of these, MeOH extracts of seven plants, including Euphorbia thymifolia (leaf), Gnetum montanum (stem), Heterosmilax erythrantha (root), Morus alba (leaf), Syzygium formosum (leaf), Jussiaea repens (aerial parts), and Camellia sinensis (leaf), exhibited significant antioxidant activities. All of these herbs showed remarkable free radical scavenging activities with $IC_{50}$ values of 11.0, 14.5, 17.0, 13.6, 10.8, 7.7, and $8.5\;{\mu}g/ml$, respectively, and significant inhibitory effects on lipid peroxidation with 79.7, 83.8, 78.9, 82.5, 88.8, 88.0, and 96.2% inhibitions, respectively, at the concentration of $50\;{\mu}g/ml$.
Anti-obese effects of mulberry (Morus alba L.) root bark was investigated in vitro by measuring its inhibitory effect against 3T3-L1 preadipocyte differentiation and digestive enzymes such as ${\alpha}$-amylase, ${\alpha}$-glucosidase and pancreatic lipase. Ethanol extract of mulberry root bark (MRE) showed the potent inhibitory activities on ${\alpha}$-amylase, ${\alpha}$-glucosidase and pancreatic lipase with $IC_{50}$ values of $7.86{\pm}0.36$, $0.12{\pm}0.03$ and $7.93{\pm}0.11mg/mL$, respectively. Furthermore, MRE significantly suppressed cellular lipid accumulation in 3T3-L1 cells in a dose-dependent manner. To elucidate the mechanism of MRE, we performed qRT-PCR and Western blotting for the expression of genes related with adipogenesis and lipogenesis. Treatment of MRE markedly suppressed the protein expression of $PPAR{\gamma}$, $C/EBP{\alpha}$ and SREBP-1c, as well as FAS and ACC, which are the key transcription factors and metabolic enzymes in adipogenesis and lipogenesis. In addition, qRT-PCR analysis indicated that the anti-adipogenesis effect of MRE might be due to its inhibition at transcription levels. These results demonstrate that MRE can effectively suppress adipocyte differentiation and inhibit key enzymes related to obesity. Our findings suggest that mulberry root bark may have a potential benefit in preventing obesity.
To make clear the changes of the amount of reserve substances in the bark of stem and root of mulberry graftages after planting, studies were carried with mulberry graftages classified into three groups of 7.0-7.9mm, 9.0-9.9mm and 11.0-11.9mm in diameter of stems at 3cm above their base. The results obtained were as follow : 1. The dry weight of stem and root bark gradually decreased with growth of new shoots till about five weeks after planting, and from then it increased. 2. The duration of new shoots growth depending on the reserve substance was limited for 6-7 weeks after planting. In no fertilized group, the length of new shoots increased up to 6-7 weeks, form then decrease. 3. Dry leaf yield three weeks after planting was found to be no difference in the same diameter of saplings between with and without fertilization. 4. The content of chlorophyll in the leaves was higher in thicker group and in fertilized group than thinner and no fertilized one, respectively. 5. There was no difference in the amount of carbohydrates of the bark of stem and root among the stem diameters. There was a transient increase in the amount of total sugar in the bark of stem and root in the first three weeks after planting, followed by gradual decline up to five weeks. The amount of reducing sugar in the bark of stem and root increased both in fertilized and in no fertilized group up to three weeks after planting, and then it decreased. There was a gradual decline in the content of starch in the bark of stem and root both in fertilized and in no fertilized group up to five weeks after planting, followed by gradual increase.
In the present study, ten herbal extracts, Citrus unshiu Markovich, root and stem of Berberis koreana, Morus alba, Dendrobium moniliforme, Aster gramineus, A. scabar, Alisma canaliculatum, Fallopia japonica and Origanum (O.) vulgare were determined to examine anti-mycoplasmal activity. Among them, O. vulgare extract (OVE) showed strong anti-mycoplasmal activity and was analyzed by gaschromatography/ mass spectrometry (GC/MS). As the results, OVE was consisted of carvacrol (68.78%), o-cymene (9.80%), terpinene (7.61%) and thymol (4.03%) as main ingredients. To investigate inflammatory activity by intact pathogenic Mycoplasma hyopneumoniae (M. hyo) at 30 $\mu$g/ml, we examined induced transcription of proinflammatory cytokines such as cyclooxygenase-2, tumor necrosis factor-a, interleukin (IL)-1, IL-6 and inducible nitric oxide synthase in RAW 264.7 cells. With the above results, we further investigated whether OVE could reduce inflammation induced by M. hyo at minimal inhibitory concentration. The result showed that 32 $\mu$g/ml of OVE inhibited nitric oxide production by 60%. This study also evaluated the combination of OVE with antibacterials against M. hyo for application. Based on these results, it could be concluded that M. hyo induces inflammation in RAW 264.7 cells and OVE protects this inflammation, indicating that OVE may be useful for industrial animals.
