Effect of Kami-chungsimyeunjatang on atopic dermatitis-like skin lesions induced in NC/Nga mice by mite antigen stimulation (가미청심연자탕(加味淸心蓮子場)이 NC/Nga mice의 아토피양(樣) 피부염에 미치는 영향)

  • Objectives : The purpose of this study is to examine of the effect of Kami-chungsimyeunjatang(KCSYJT) medicine on the atopy eruption control. Methods : The expression of IgE, IL-4, IL-6, $TNF-{\alpha}$, IgG2b, IgM, IgG2a and IgG1 level in serum, and $IFN-{\gamma}$ production by KCSYJT were analyzed. CD3e+/CD69+, CD4+/CD25+, B220+/IgE+ and B220+/CD23+ positive cells by flow cytometry in splenocytes were assayed and the revelation of CD3e+/CD69+, CD4+/CD8+ and CD4+/CD25+ marker in PBMC, spleen and DLN were observed. The outturn of IL-4, eotaxin 2, CCR3, TARC mRNA in splenocytes werw observed. We also analyzed NC/Nga mice's ear, DLN and neck-back skin after biopy and dye by H&E, and toluidine staining (mast cells marker) method, measured about epidermis and dermis part in comparison with control group. Results : NC/Nga mice suffered from dermatitis very similar to human AD with IgE hyperproduction. Specially, result that measure IgE content in serum on 8 weeks, 12 weeks, 16 weeks decreased remarkably than control group. After experiment end, result that observe revelation CD3e+/CD69+, CD4+/CD8+ and CD4+/CD25+ marker in PBMC, spleen and DLN establishment observed recover as normal with political background. And decreased than result control group which measure IL-4, IL-6, $TNF-{\alpha}$, IgG2b, IgM, IgG2a, IgG1 level in serum, and $IFN-{\gamma}$ production secreted in Th1 cell displayed increase by KCSYJT medicines. Ear thickness decrease than control group in result that observe effect that get in ear of a NC/Nga mouse. Course inflammation immunocyte etc.. permeated of result that effect that KCSYJT medicines get to NC/Nga mouse's skin establishment analyzes ear, DLN and neck-back skin after biopy, and dye by H&E, and toluidine staining (mast cells marker) method decreased about epidermis. and inflammation of dermis part remarkably than control group. Immunohistochemical examination of the skin lesion showed decrease by KCSYJT medicines on numbers of mast cells (CCR3) and CD4+ T cells containing IL-4 necessary for IgE. Conclusions : Th1 cell and Th2 cell was observed to be shift by secretion amount of IL-4 and $IFN-{\gamma}$ by KCSYJT medicines. Therefore, the KCSYJT medicine turned out to be useful in allergy autoimmune disease.

Antiinflammatory and Antiallergic Activity of Fermented Turmeric by Lactobacillus johnsonii IDCC 9203 (Lactobacillus johnsonii IDCC 9203으로 발효한 울금의 소염 및 항알레르기 효과)

  • Although turmeric has numerous pharmacological effects, the poor water-solubility of curcuminoids, active components of turmeric, restricts their systemic availability in orally administered formulations and limits their therapeutic potential. In this study we attempted turmeric fermentation using several probiotic bacteria to improve its solubility, and also investigated the effects of turmeric and fermented turmeric on anti-inflammatory activity. Fermented turmeric, by L. johnsonii IDCC 9203, more strongly inhibited LPS-induced expression of the pro-inflammatory cytokines than non-fermented turmeric and fermented turmeric by other probiotic strains. We used an NC/Nga mouse model for mite antigen-induced atopic dermatitis to examine the efficacy of the fermented turmeric. Fermented turmeric-fed mice exhibited a significantly reduced serum IgE level and mitigated acute inflammation. When the fermented turmeric was pre-treated by oral administration, it had more preventive activity against acute anaphylactic reaction than the non-fermented group. In addition, we observed that fermentation of turmeric leads to increased water-solubility of curcumin and a change in the active components ratios for bisdemethoxycurcumin, demethoxycrucumin and curcumin. Taken together, these results strongly suggest that fermented turmeric by L. johnsonii IDCC 9203 could be used as a functional food ingredient for improving treatments for atopic dermatitis.

Effects of Humidity and Citrus Leaf Age on the Multiplication of Aculops pelekassi (Acari: Eriophyoidea) and Seasonal Population Abundances in Citrus Orchards (습도와 감귤 잎의 연령이 귤녹응애 증식에 미치는 영향 및 감귤원에서 발생소장)

  • The pink citrus rust mite, Aculops pelekassi (Keifer) (Acari: Eriophyoidea), is an important pest in the citrus orchards of Jeju, Korea. This study was performed to investigate the seasonal population abundance of A. pelekassi, and the effects of humidity and citrus leaf age on the multiplication of this pest. Relative humidity (RH) significantly affected the longevity and fecundity of A. pelekassi. Longevity was 7.5, 14.5 and 14.6 d and fecundity was 5.4, 21.5, and 27.1 eggs at 33, 75 and 84% RH, respectively. The leaf age of citrus significantly affected the multiplication of A. pelekassi. The population abundance on 40 day-old leaves was much higher more than 3 times that on 10-day old leaves at 4 weeks after introduction. Overwintered A. pelekassi adults between the bud scales of the citrus trees became active in late April; they were found on newly emerged leaves, followed by their settlement on young fruits in mid-June. The population levels of A. pelekassi peaked on the leaves in late June to July, and on the fruits in early August. The results obtained in this study should be useful for the control of A. pelekassi in citrus orchards in Jeju.

A Postharvest Control Technique of the Oriental Fruit Moth, Grapholita molesta, Infesting Apples Using CATTS (CATTS를 이용한 복숭아순나방 사과 수확 후 방제 기술)

  • Postharvest insect pest control is necessary for agricultural industry including domestic consumer markets and exporting products to meet quarantine issue. Especially, the organic or environmentally friendly agricultural products do not fit to the traditional chemical postharvest treatments mostly using methyl bromide. As an alternative, a physical treatment called CATTS (controlled atmosphere and temperature treatment) has been developed to control various insect and mite pests on apple and several stone fruits. This study was designed to determine the CATTS conditions to control the oriental fruit moth, Grapholita molesta, which is restricted in exporting the infested apples. To apply CATTS on this insect pest, the most heat-tolerant stage was determined. Among the immature stages locating on the fruits, the fifth instar larvae were the most tolerant to $44^{\circ}C$ for 20 min. The ramping rate (the time to increase chamber temperature from $25^{\circ}C$ to $46^{\circ}C$) was positively correlated with the CATTS efficiency under 15% $CO_2$ and 1% $O_2$. After the ramping step, the duration of CATTS was positively correlated with CATTS efficiency. In addition, the CATTS efficiency was highly dependent on the fruit internal temperature at $44^{\circ}C$. From all these parameters, we developed a standard protocol yielding 100% control efficiency of CATTS against apples infested by 4,378 larvae including 2,104 fifth instar individuals.

Incidence of Polyphagotarsonemus latus (Banks) (Acari: Tarsonemidae) on Green Perilla(Perilla frutescens var. japonica Hara) (잎들깨에서 차먼지응애의 발생특성)

  • The populations of Polyphagostarsonemus latus (Acari: Tarsonemidae) peaked two times on green perilla grown in greenhouses of Geumsan-Gun, Chungchungnamdo. The first peak of P. latus was in the middle of June, after it was first detected in late May. The population of P. latus peaked for the second time in mid July as its density stared rapidly increasing in early July. The application of chemical pesticides and eco-friendly agricultural materials, the two management methods used to control P. latus on green perilla, did not alter the occurrence patterns of P. latus; population size of the mite was much larger in greenhouses using chemical pesticides than in the ones using eco-friendly agricultural materials. This difference might be cuased by continuity of the management methods. Chemical control of P. latus should be limited owing to pesticide residue. The highest density of P. latus on green perilla plants was observed after 25 days after inoculation. The density was the highest in mid-aged leaves (e.g., the largest leaves) and the lowest in newly developed leaves (e.g., smallest leaves). However, there was no significant (P > 0.05) correlation between leaf size and density of P. latus. These results indicate that leaf size (e.g., leaf age) did not affect the occurrence of P. latus. Thus, any leaf of a green perilla plant is available as a sample unit for P. latus.

Changes of Causative Allergen in Patients with Allergic Diseases in Chonbuk Province (전북지역 알레르기 환자에서 원인 알레르겐의 변화)

  • Background: The prevalence of allergic disease has increased substantially over the past decades. The reasons for these trends are complex but include increased environmental allergen exposure. Environmental control measures are of particular importance in the prevention and management of allergic disease. Thus, we purposed to evaluate the changes of causative allergens in allergic patients in Chonbuk provincial area that began industralized recently. Method: To evaluate the allergens epidemiologically, we performed skin prick tests and RAST in 201 allergic patients from February, 1984 to February, 1985 and 339 patients from February, 1997 to February, 1998. Results: The Following results were obtained. 1) Total sensitization rates did not change significantly(74.2% vs 75.8%, p=0.664). 2) Positive reaction rates to pollens were similar(35.8% vs 30.1%, p=0.17). 3) Positive rates to HDM(house dust mite) were decreased(66.1% vs 56.3%, p=0.02). 4) Positive rates to fungi and animal danders were decreased significantly(p<0.001). Conclusion : Environmental allergens associated with hygine in Chonbuk provincial areas such as HDM and fungi are decreased significantly during 13 years.

Biological Control of Insect Pests with Arthropod Natural Enemies on Greenhouse Sweet Pepper in Winter Cropping System (파프리카의 겨울작형 시설재배에서 천적을 이용한 해충 밀도억제 효과)

  • This study reports the results of natural enemy application against four insect pests on greenhouse sweet pepper in winter cropping system. Orius laevigatus ($3.3/m^2$) was released at two different times (Nov. 2005 & Feb. 2006) to control Frankliniella occidentalis, western flower thrips (WFT). Throughout the cropping season, the lowest level recorded of WFT was less than 2.0 individuals per yellow sticky trap. Amblyseius swirskii ($232.3/m^2$) was released four times (Nov. 2007 ~ Aug. 2008) to control Bemisia tabaci, tobacco whitefly (TWF). Until July 2008, TWF population had been suppressed lower than 6.6/trap. Phytoseiulus persimilis ($44.5/m^2$) was released seven times (Apr. 2008 ~ Aug. 2008) to control Tetranychus kanzawai, tea red spider mite (TRSM). As a result, TRSM population was suppressed lower than 1.7/1eaf. To control aphids, Aphidius colemani ($9.5/m^2$) was released seven times (Oct. 2004 ~ Jan. 2005), with the transplantation of banker plants ($5.5pot/660m^2$). As a whole, aphid's population has been successfully reduced to less than 2.0/leaf.

Evaluation of Susceptibility of Red Poultry Mite, Dermanyssus gallinae (Acari: Dermanyssidae) in Five Regions to 11 Acaricides (11종의 살비제를 이용한 5개지역 닭진드기의 감수성 평가)

  • The susceptibility of Dermanyssus gallinae adults, collected from poultry farms in 5 regions, to 11 acaricides was investigated. When bifenthrin and formic acid with pyridaben were diluted 100 times, bifenthrin showed 100% acaricidal activity; however, formic acid with pyridaben showed less than 20% acaricidal activity for poultry farms in 3 regions (Gyeongju, Chilgok, and Geumsan) except Yeoncheon and Anseong. The carbamate compound, carbaryl showed 100% acaricidal activity in most of the regions, except Gyeongju. A phosphorus compound, dichlorvos showed 100% activity in most of the regions, yet 66.7% acaricidal activity in Yeoncheon. The susceptibility of D. gallinae from poultry farms in Yeoncheon and Anseong to most acaricides was high; however, in the other farms, the susceptibility of D. gallinae varied depending on the acaricide. Clothianidin, thiamethoxam, fenitrothion, and formic acid with pyridaben showed differences in acaricidal activity among regions. Therefore, farmers should concentrate during the selection of these acaricides. However, carbaryl, cartap hydrochloride, dichlorvos, and bifenthrin showed high activity against D. gallinae collected from poultry farms in five regions. Therefore, these acaricides could be used in most of these regions. To control D. gallinae effectively, the alternation of acaricides is necessary, and indiscriminate pesticide use should be avoided. Therefore, this study can serve as a basis for controlling D. gallinae.

Studies on Pear Phylloxera (Aphanostigma iakusuiense Kishida) (1) On the Control Methods (콩가루벌레 (Aphanostigma iakusuiense)에 관한 연구 (I) 방제시험을 중심으로)

  • Life history and control methods of Aphanostigma iakusuiense Kishida were studied during April to November, 1974, in the Naju pear orchard area, Cholla Namdo. The results are summarized as follows: 1. More than $60\%$ of wintering eggs were found at the height of 1.5 to 2.0m from the soil surface, with approximately $10\%$ at 0.5 to 1.5m and 1.5 to 2.5m respectively. 2. There was little difference in the size of egg masses and hatching ratio $(12.7-13.7\%)$ between the varieties of host trees, Mansamkil and Kooksoo. 3. Hatching was not observed until April 6, but hatches of $12.8\%$ on April 13 and $90.6\%$ on May 4 were found. 4. During the hatching season in early spring, 8 chemicals were applied to test effectiveness of control. C8514, Metasystox, Acar 338, and Parathion were more effective than either Sevin or Malix for control of this insect. 5. Highest populations of pear phylloxera ocurred from August 20 to September 20 at which time levels of eggs and larvae were simillar in abundance. 6. In tests for control of this insect after in paper bags, DDVP, Loxon, C8514, Sevin, and Binapacryl were more effective than either EPN or Otran. 7. Predacious mite, Protolaelape pygmaeus Miller, and several species of lady beetles (Coccinellidae) preying on this insect as natural enemies were collected. The predation ratio of mites varied according to region: that is 0.77 to $18.6\%$ in Bia Orchard; $7.88-48.7\%$ in Noan Orchard and almost none in Youngsanoo Orchard. 8. The varietal difference in fruit damage by this insect was in the order of Choseng Chuck, Kooksoo, followed by Sin Ko, Mansamkil, Keumchon Choo, and Changsimrang.

Occurrence Monitoring and Population Growth of Tomato Russet Mite, Aculops lycopersici Massee (Acari: Eriophyidae) Using Green Label Sticker (시설토마토에서 녹색 라벨 스티커를 이용한 토마토녹응애의 발생예찰과 개체군 생장)

  • This study is conducted to develop a new method of early occurrence monitoring for Aculops lycopersici Massee (Acari: Eriophyidae), a major pest in tomato. A. lycopersici causes damage to plants such as curling of leaflet edges, followed by leaflet bronzing. As feeding continues by the mites, the plant takes on a russeted, wilted appearance, eventually culminating in leaflet desiccation and finally plant death. Fruit damages in tomato were shown when the density of mites was very high. Also, A. lycopersici caused more damage to unripe fruits than ripe fruits. The time taken to detect A. lycopersici on green label sticker was 7.0 seconds, the shortest. Blue, orange and white label stickers took 17.1, 19.8 and 12.3 seconds respectively. The permeable microscope with side illumination was useful in observing A. lycopersici on label stickers. A. lycopersici began to occur in late-April and the density of the mites increased rapidly after mid-May. The density peaked in mid to late-June, and decreased after late-June. A. lycopersici was observed on lower stems 20 days after the release of mites and observed on higher stems 60 days after. The peaked densities were observed on stems 40 and 60 days after and on leaf 80 days after. The label sticker is enough to monitor early occurrence of A. lycopersici in tomato cultivation facilities.
