Isolation and Phosphate-Solubilizing Characteristics of PSM, Aeromonas hydrophila DA33

  • bacterium having high abilities to solubilize in-organic phosphate was isolated from cultivated soils. The strain was identified as Aeromonas hydrophila DA33, based on the physiological and biochemical properties. The optimum temperature and initial pH to solubilize insoluble phosphate in sucrose minimal medium were 3$0^{\circ}C$ and pH 5.0, respectively. In these conditions, phosphate-solubilizing activities of the strain against two types of insoluble phosphate were quantitatively determined. When glucose was used for carborn source, the strain had a marked mineral phospahte solubilizing activity. Inorganic phospahte solubilization was directly related to the pH drop by the strain. Analysis of the culture medium confirmed the production of gluconic acid as the main organic acid released by Aeromonas hydrophila DA33.

산초 정유성분의 식중독균에 대한 항균 활성 (Antibacterial Activities of Essential Oil from Zanthoxylum schinifolium Against Food-Borne Pathogens)

  • In this study, the antibacterial activities of essential oil from Zanthoxylum schinifolium against four Gram-positive bacteria and six Gram-negative bacteria were investigated. The antibacterial activity of the oils was determined using the agar-well diffusion assay, MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration). In particular, essential oil from Z. schinifolium showed higher antibacterial activity against Gram-positive bacteria than against Gram-negative bacteria. Essential oil from Z. schinifolium displayed large inhibition zones especially against Bacillus cereus (31 mm). At concentrations between 0 and $20\;{\mu}g/mL$ the oils showed an antibacterial effect against both Gram-negative and Gram-positive bacteria. The minimum inhibitory concentration (MIC) values against nine bacteria ranged from 1.25 to $5\;{\mu}g/mL$. The minimum bactericidal concentration (MBC) values against eight bacterial ranged from 2.5 to $20\;{\mu}g/mL$, except Shigella sonnei. Furthermore, our finding on the antibacterial activities of essential oils from Zanthoxylum schinifolium validated the use of this plant for medical purposes.

Improved Production of Curdlan with Concentrated Cells of Agrobacterium sp.

  • The addition of a limited concentration of yeast extract to a minimal salt medium (MSM) enhanced cell growth and increased the production of curdlan whereas nitrogen-limitation was found to be essential for the higher production of curdlan by Agrobacterium sp. ATCC 31749. As the amount of the inoculum increased, the cell growth as well as the production of curdlan also increased in the MSM without a nitrogen source. The cell growth and production of curdlan increased as the initial pH of the medium decreased as low as 5.0. The conversion rate and concentration of curdlan from 2% (w/v) glucose in the MSM with concentrated cells under nitrogen deletion was 67% and 13.4 g/L, respectively. The highest conversion rate of curdlan under the conditions optimized in this study was 71% when the glucose concentrations was 1% (w/v).

Evaluation of Benthic Macroinvertebrate Diversity in a Stream of Abandoned Mine Land Based on Environmental DNA (eDNA) Approach

  • Recently, environmental DNA (eDNA)-based metabarcoding approaches have been proposed to evaluate the status of freshwater ecosystems owing to various advantages, including fast and easy sampling and minimal habitat disruption from sampling. Therefore, as a case study, we applied eDNA metabarcoding techniques to evaluate the effects of an abandoned mine land located near a headwater stream of Nakdonggang River, South Korea, by examining benthic macroinvertebrate diversity and compared the results with those obtained using the traditional Surber-net sampling method. The number of genera was higher in Surber-net sampling (29) than in the eDNA analysis (20). The genus richness tended to decrease from headwater to downstream in eDNA analysis, whereas richness tended to decrease at sites with acid-sulfated sediment areas using Surber-net sampling. Through cluster analysis and non-metric multidimensional scaling, the sampling sites were differentiated into two parts: acid-sulfated and other sites using Surber-net sampling, whereas they were grouped into the two lowest downstream and other sites using eDNA sampling. To evaluate freshwater ecosystems using eDNA analysis in practical applications, it is necessary to constantly upgrade the methodologies and compare the data with field survey methods.

Establishment of an Incentive System for Prohibition of Lending Firefighting Qualifications and Rental Agencies

  • This study suggests a solution of an incentive system from the viewpoint of the external effect on the renting of firefighting qualifications and on rental agencies. Currently, the Human Resources Development Service of Korea does not have a penalty for neglecting to perform a search into qualification lending and rental agencies. In the short term, the penal clause should be strengthened so that companies and qualification holders cannot borrow or lend their qualifications. In addition, the Human Resources Development Service of Korea will increase the number of personnel dedicated to investigating the loaning of qualifications. It is necessary for the government to establish a penalty for neglecting to investigate the lending of qualifications for the Human Resources Development Service of Korea. In the long term, the state needs to make only minimal regulations. Also, the state needs to make companies and qualification holders feel like they benefit more when they are not borrowing or lending qualifications. The anticipated effect of this study is that the government will intervene appropriately by applying firefighting qualifications according to market principles. It can also be used as a basis for the revision of related laws on the renting of national technical qualification certificates and the investigation of rental agencies.

A Study on Antimicrobial Activity of Lysimachia clethroides Duby Root Extracts against Methicillin-resistant Staphylococcus aureus

  • Methicillin-resistant Staphylococcus aureus (MRSA) is a bacterium responsible for a number of infections in humans that are difficult to treat, and as a result, is a substantial contributor to morbidity and mortality. In the present study, in search of natural products capable of inhibiting this multidrug-resistant bacterium, we investigated the antimicrobial activity of Lysimachia clethroides Duby root. The antibacterial activities of EtOH extract of Lysimachia clethroides Duby root and its n-hexane, EtOAc, n-BuOH and water fractions were evaluated against 15 strains of methicillin-resistant staphylococcus aureus (MRSA) and 1 standard methicillin-susceptible S. aureus (MSSA) strain by using the minimal inhibitory concentrations (MICs) assay, colorimetric assay using MTT test, checkerboard dilution test. Antimicrobial activity of n-hexane fraction of Lysimachia clethroides Duby root was remarkable. Against the 16 strains, the minimum inhibitory concentrations (MICs) were in the range of $31.25-62.5{\mu}g/ml$ and FICI values for n-hexane fraction of Lysimachia clethroides Duby root+AM and n-hexane fraction of Lysimachia clethroides Duby root+OX were checkerboard method performed using the MRSA, MSSA and one clinical isolate strains via MICI 0.12-1 and 0.25-0.75, showing the increase of synergistic effect. When combined together, these antibiotic effects were dramatically increased. These effective combinations could be new promising agents in the management of MRSA.

  • 파장분할다중화방식 전광통신망에서는 신호가 먼 거리를 진행할 경우, ASE(Amplified Spontaneous Emission) 잡음이나 누화(Crosstalk)와 같은 물리적 제약으로 인해 신호가 손상된다. 소낫ㅇ된 신호를 회복시키기 위해서는 광신호를 중간 노드에서 재생(signal regeneration)해야 하지만, 신호 재생은 추가의 네트워크 자원을 필요로 하기 때문에 재생 노드(Signal Regeneration Node, SRN)는 다른 광경로의 블록킹을 최소화하도록 신중히 선택해야 한다. 이 논문에서는 물리적인 제약 하에서 다중홉 방식으로 광경로를 설정하는 문제에 대해 최소비용 알고리즘(Minimal-Cost Placement Algorithm, MCPA)과 휴리스틱 알고리즘들을 제안하다. 최소비용 위치선정 알고리즘은 다이내믹 프로그래밍을 이용하여 송수신기나 파장의 부족으로 인한 다른 광경로들의 블록킹을 최소화하도록 공식화하였다.

최적의 설계 자동화를 위한 최소자원 할당 알고리듬 (A Minimum Resources Allocation Algorithm for Optimal Design Automation)

  • 본 논문에서는 최적의 설계 자동화를 위한 최소자원 할당 알고리듬을 제안한다. 제안된 할당 알고리듬은 연산자 간을 연결하는 신호선이 반복적으로 이용되어 연결 신호선 수가 최소가 될 수 있도록 기능 연산자를 할당한다. 레지스터 할당 시 연결구조에 따라 가중치를 갖는 구간 그래프를 구성한다. 최소의 클러스터 파티션 알고리듬을 이용하여 생성된 최대 크기의 클러스터들에 연결구조가 고려된 레지스터들을 할당한다. 또한 연결구조에 대한 멀티플렉서의 중복 입력을 제거하고 연산자에 연결된 멀티플렉서 간의 입력을 교환하는 입력 정렬 과정으로 연결 구조를 최소화한다. 또한, 벤치마크 실험을 통하여 제안된 알고리즘의 효용성을 보인다.

한우 Lactoferrin 가수분해물의 항균 활성 (Antibacterial Activity of Enzymatic Hydrolyzates of Lactoferrin derived from Korean Native Cattle)

  • In this study, we carried out to isolate lactoferrin from Korean native cattle and Holstein cow by batch extraction, ion exchange chromatography, gel filtration, and affinity chromatography. The purity of the isolated lactoferrin was higher than that of lactoferrin purchased from Sigma, when determined by SDS-PAGE and HPLC analysis. Antibacterial activity of E. coli O111 by Korean native cattle lactoferrin was lower than that of Holstein lactoferrin. A minimal inhibitory concentration(MIC) of Korean native cattle lactoferrin and Holstein lactoferrin was 2.75 mg/ ml and 1.5 mg/ml respectively. The lactoferrin hydrolyzate of Korean native cattle exhibited antimicrobial activity at 0.25 mg/ml, whereas that of Holstein cow exhibited antimicrobial activity at 0.12 mg/ml. The antibacterial potency of the hydrolyzate was at least tenfold greater than that of undigeated lactoferrin with strains tested. The effect of hydrolyzate was bactericidal as indicated by rapid loss of viability of E. coli O111.

A Honey-Hive based Efficient Data Aggregation in Wireless Sensor Networks

  • The advent of Wireless Sensor Networks (WSN) has led to their use in numerous applications. Sensors are autonomous in nature and are constrained by limited resources. Designing an autonomous topology with criteria for economic and energy conservation is considered a major goal in WSN. The proposed honey-hive clustering consumes minimum energy and resources with minimal transmission delay compared to the existing approaches. The honey-hive approach consists of two phases. The first phase is an Intra-Cluster Min-Max Discrepancy (ICMMD) analysis, which is based on the local honey-hive data gathering technique and the second phase is Inter-Cluster Frequency Matching (ICFM), which is based on the global optimal data aggregation. The proposed data aggregation mechanism increases the optimal connectivity range of the sensor node to a considerable degree for inter-cluster and intra-cluster coverage with an improved optimal energy conservation.
