Bacillus sphaericus was mutagenized with UV light irradiation and dimethyl sulfate. Thirty-five conditional lethal temperature-sensitive(ts) mutants were isolated at the nonpermissive temperature of $42^{\circ}C$ and classified into three groups by their growth characteristics on the nutrient broth, peptone glucose yeast extract agar and mineral salts agar. First was the lethal ts group, 24 mutants, which did not grow at the nonpermissive temperature, the second, 9 mutants, was the less growth is group whose growth was restricted to one-half, and the third, 2 mutants, was the cold lethal ts group whose growth was restricted at the permissive temperature($25^{\circ}C$and $30^{\circ}C$)

Marine bacterial strains were isolated trom coastal regions of Goa and screened for the strains that produce the highest amount of mucous expolysaccharide (EPS). Our screening resulted in the identification of the strain Vibrio furnissii VB0S3 (hereafter called VB0S3), as it produced the highest EPS in batch cultures during the late logarithmic growth phase. The isolate was identified as VB0S3 based on morphological and biochemical properties. Growth and EPS production were studied in mineral salts medium supplemented with NaCl (1.5%) and glucose (0.2%). The exopolymer was recovered from the culture supernatant by using three volumes of cold ethanol precipitation and dialysis procedure. Chemical analyses of EPS revealed that it is primarily composed of neutral sugars, uronic acids, and proteins. Fourier-transform infrared (FT-IR) spectroscopy revealed the presence of carboxyl, hydroxyl, and amide groups, which correspond to a typical heteropolymeric polysaccharide, and the EPS also possessed good emulsification activity. The gas chromatographic analysis of an alditol-acetate derivatized sample of EPS revealed that it was mainly composed of galactose and glucose. Minor components found were mannose, rhamnose, fucose, ribose, arabinose, and xylose. EPS was readily isolated from culture supernatants, which suggests that the EPS was a slime-like exopolysaccharide. This is the first report of exopolysaccharide characterization that describes the isolation and characterization of an EPS expressed by Vibrio surnissii strain VB0S3. The results of the study contribute significantly and go a long way towards an understanding of the correlation between growth and EPS production, chemical composition, and industrial applications of the exopolysaccharide in environmental biotechnology and bioremediation.
This experiment was carried out to evaluate the carcass characteristics, chemical composition and fatty acid profile of the Longissimus muscle (LM) of three cattle genetic groups (Purunã, PUR, 11; 1/2 Purunã vs. 1/2 British, PUB, 6 and 1/2 Charolais vs. 1/2 Caracu, CHC, 10) finished in pasture systems. The field work took place at the Lapa Research Farm of the Agronomic Institute of Paraná, in the city of Lapa, south Brazil. The animals were fed during the winter with corn silage, cottonseed meal, cracked corn, urea, limestone and mineral salts as sources of protein, as well as an energy supplement, in pasture systems of Brachiaria decumbens Stapf. The animal groups were slaughtered at 20 months of age, at 50122.6 kg live weight. CHC bulls had higher (p<0.05) final weight than PUR and PUB bulls. Hot carcass weight was similar (p>0.10) between PUR and PUB. Hot carcass dressing percentage was higher (p<0.05) for PUB bulls than for PUR and CHC bulls. On the other hand, hot carcass dressing percentage was similar (p>0.05) between PUR and CHC bulls. Fat thickness was similar (p>0.10) among all genetic groups. However, the Longissimus area of CHC bulls was greater (p<0.05) than in PUR and PUB genetic groups. The genetic groups did not affect (p>0.10) the marbling of Longissimus. There was no observed difference (p>0.10) in moisture, ash, crude protein and total cholesterol contents among the three genetic groups. On the other hand, the total lipid percentage was higher (p<0.05) for the PUB genetic group in comparison with PUR and CHC. CLA percentage was highest for PUR animals. However, total CLA amounts were not altered by the different genetic groups.
Cordyceps cardinalis was reported in Japan and the USA in 2004, and its fruiting bodies have recently been cultured in Korea. Herbarium specimens preserved at the Cordyceps Research Institute, Mushtech, Korea were revised and identified as C. cardinalis, based on morphological characters and conidial structures. Most of the C. cardinalis specimens were collected from Mt. Halla in Jeju-do. The effects of various nutritional sources and environmental conditions such as temperature and pH on mycelial growth of C. cardinalis were studied. Oatmeal agar, Martin's peptone dextrose agar, and Schizophyllum (mushroom) genetics complete medium plus yeast extract resulted in the best mycelial growth. Among carbon sources, cereals, and nitrogen sources, maltose, oatmeal, and peptone resulted in the best mycelial growth respectively. Mineral salts helped to increase growth rate but only resulted in thin mycelial density, similar to water agar. A temperature of $25^{\circ}C$ and a pH of 7 resulted in the highest mycelial growth. Based on these results, a Cordyceps cardinalis composite medium (CCM) was formulated with 1% maltose, 2% oatmeal, 1% peptone, and 2% agar. Use of the CCM resulted in slightly better mycelial growth than that of other commonly used agar media. Only organic nitrogen sources imparted a reddish pigmentation to the agar media, but this character diminished after several subcultures. A 7 day culture duration resulted in the best mycelial growth.
Development of unconventional natural resources such as shale gas, shale oil and coal bed methane, has been activated and improved the productivity due to the recent technology advance in horizontal drilling and hydraulic fracturing. However, the flowback water mixed with chemical additives, and the brine water containing oil, gas, high levels of salts and radioactive metals is produced during the gas production. Potential negative environmental impact due to large volumes of the produced wastewater is increasingly seen as the major obstacles to the unconventional natural resource development. In this study an integrated framework for the flowback and brine water treatment is proposed, and we reviewed the upcoming state of the art technology in water treatment. Basic separation processes which include not only membrane, evaporation, crystallization and desalination processes, but the potential water reuse and recycling techniques can be applied for the unconventional natural resource industry.
In the paper insight is given in the legislation policy to restrain environmental pollution by pig husbandry, focused on The Netherlands (Mineral Accounting System). Besides, nutritional measures are presented to reduce environmental pollution by lowering excretion of N and P, emphasizing (multi) phase feeding, the use of low protein, synthetic amino acids supplemented diets, phytase and its effect on phosphorus and calcium digestibility, its interaction with phytic acid and proteins, and the environmental impact of the use of phytase in pig diets. Also, nutritional means are indicated to reduce ammonia volatilization from pig operations. It is concluded that nutrition management can substantially contribute to reduction of N and P excretion by pigs, mainly by lowering dietary protein levels, (multi) phase feeding and the use of microbial phytase, and that the use of phytase on a large scale in The Netherlands has a tremendous environmental impact. In 20 years the excretion of P in growing-finishing pigs has more than halved. Ammonia emission from manure of pigs can be reduced substantially by lowering dietary protein content, but also by including additional non-starch polysaccharides in the diet. A very promising method to reduce ammonia emission is to manipulate dietary cation-anion difference, e.g. by adding acidifying salts to the diet, which will lower pH of urine substantially. Further research is desirable. This also applies to determining dietary factors influencing the odour release from manure. Finally, some speculation on the future of pig farming from an environmental viewpoint is presented.
Recently, the production of taste and odor (T&O) compounds is a common problem in water industry. Geosmin is one of the T&O components in drinking water. However, geosmin is hardly eliminated through the conventional water treatment systems. Among various advanced processes capable of removing geosmin, adsorption process using granular activated carbon (GAC) is the most commonly used process. As time passes, however GAC process changes into biological activated carbon (BAC) process. There is little information on the BAC process in the literature. In this study, we isolated and identified microorganisms existing within various BAC processes. The microbial concentrations of BAC processes examined were $3.5{\times}10^5$ colony forming units (CFU/g), $2.2{\times}10^6CFU/g$ and $7.0{\times}10^5CFU/g$ in the Seongnam plant, Goyang plant and Goryeong pilot plant, respectively. The dominant bacterial species were found to be Bradyrhizobium japonicum, Novosphingobium rosa and Afipia broomeae in each plants. Removal efficiencies of $3{\mu}g/L$ geosmin by the dominant species were 36.1%, 36.5% and 34.3% in mineral salts medium(MSM) where geosmin was a sole carbon source.
Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.
Adventitious shoots were induced from cambial tissue cultures of 3-year-old seedlings using BLG mineral salts medium supplemented with 10 mM glutamine and 30 mM sucrose. The optimum growth regulator level for bud induction was 4,5 $\mu$M BA which produced average 25.5 shoots per cambium segment. Induced buds were elongated on GD medium supplemented with 30 mM sucrose followed by LMG medium supplemented with 30 mM sucrose for further shoot elongation. Elongated shoots were rooted on half-strength GD medium containing $0.54 ;\mu\textrm{M}$ NAA with the frequency of 20%. This system proved the high morphogenic potential of cambial tissue in larch.

Because the stone cultural properties located outdoors, they have been altered and deteriorated in external appearance due to environmental factors such as acid rain, extreme change in temperature, and salts. Damage to stone cultural properties is accelerated particularly due to recent industrial development and environmental pollution. An experimental study was conducted to evaluate the effect of environmental contaminants(sea water and air pollutants such as, $SO_2$, NO, $CO_2$) on the weathering of granite. And as part of the developing of conservation method, $TiO_2$ catalyst was prepared and tested. After expose a granite to sea water and air pollutants, the mineral compositions of the tested granite surface were lower then that of the fresh granite. And absorption capacity of the weathered granite was steadily increased. From these results, it was expected that granite could be weathered by sea water and air pollutants. For the $TiO_2$ coated granite, absorption capacity was slightly enhanced compared to the $TiO_2$ non-coated granite. Therefore, the $TiO_2$ coating method tested in this study considered to be a viable method to assist in the conservation of granite from environmental contaminants.
