식물은 다양한 병원성 미생물에 대하여 효과적인 방어 기제를 발전시켜 왔다. 최근 유전체와 다중 오믹스 기술의 발전은 우리에게 미생물 인자에 의한 식물 면역을 폭넓게 이해할 수 있는 단초를 제공해 주었다. 하지만 아직까지는 이러한 기술을 병 방제 전략에 이용한 적은 많지 않다. 그래서 본 리뷰에서 식물 면역의 기본 개념을 소개하고 최근 얻어진 결과들을 소개하였다. 덧붙여 이미 논문에서 발표된 진균, 세균, 바이러스 유래 결정인자에 의한 생물적 방제 가능한 방법에 대해 기술하였다. 특히 미생물 결정인자인 chitin, glucan, LPS/EPS, 미생물분자패턴, 항생제, 식물유사호르몬, AHLs, harpin, 비타민, 휘발성물질에 대한 결과를 자세하게 기술하였다. 이 리뷰를 통하여 많은 과학자들과 농민들이 미생물 결정인자 기반의 생물적 방제에 대한 지식이 폭넓어지고, 다양한 미생물 결정 인자가 앞으로 농업현장의 종합적인 병방제 전략의 하나로 자리매김하기를 바란다.
Allyl isothiocyanate (AITC), as a fumigant, plays an important role in soil control of nematodes, soil-borne pathogens, and weeds, but its effects on soil microorganisms are unclear. In this study, the effects of AITC on microbial diversity and community composition of Capsicum annuum L. soil were investigated through Illumina high-throughput sequencing. The results showed that microbial diversity and community structure were significantly influenced by AITC. AITC reduced the diversity of soil bacteria, stimulated the diversity of the soil fungal community, and significantly changed the structure of fungal community. AITC decreased the relative abundance of dominant bacteria Planctomycetes, Acinetobacter, Pseudodeganella, and RB41, but increased that of Lysobacter, Sphingomonas, Pseudomonas, Luteimonas, Pseudoxanthomonas, and Bacillus at the genera level, while for fungi, Trichoderma, Neurospora, and Lasiodiplodia decreased significantly and Aspergillus, Cladosporium, Fusarium, Penicillium, and Saccharomyces were higher than the control. The correlation analysis suggested cellulase had a significant correlation with fungal operational taxonomic units and there was a significant correlation between cellulase and fungal diversity, while catalase, cellulose, sucrase, and urease were the major contributors in the shift of the community structure. Our results will provide useful information for the use of AITC in the assessment of environmental and ecological security.
Membrane bioreactor (MBR) provides the benefits on high effluent quality and construction cost without the secondary clarification. Despite of these advantages, fouling, which clogs the pore in membrane modules, affects the membrane life span and effluent quality. Studies on the laboratory scale MBR were focused on the control of particulate fouling, organic fouling and inorganic fouling. However, less studies were focused on the control of biofouling and microbial aspect of membrane. In the full scale operation, most MBR produces high effluent quality to meet the national permit of discharge regulation. In this study, the performance and microbial community analysis were investigated in two MBRs. As the results, the performance of organic removal, nitrogen removal, and phosphorus removal was similar both MBRs. Microbial community analysis, however, showed that Azonexus sp. and Propionivibrio sp. contributed to indirect fouling to cause the chemical cleaning in the DX MBR.
본 논문에서는 복합 미생물 배양기의 제어시스템을 제안하였다. 제안하는 제어시스템은 복합 미생물 배양기의 제어부, 통신부, 전원부, 제어시스템 등으로 구성된다. 복합 미생물 배양기의 제어부는 아날로그 신호와 디지털 신호의 변환, LCD 패널을 이용한 디스플레이, 수위센서, 온도센서, pH 농도센서 등과 같은 센서들의 신호 제어를 하도록 설계 및 제작한다. 사용하는 수위센서는 기존 수위센서가 거품과 같은 이물질 등으로 인해 측정이 어려운 문제점을 해결하고자 직진성이 우수한 IR 레이저 방식을 사용하여 정확한 수위 측정이 가능하도록 설계 및 제작한다. 온도센서는 열 저항 원리를 사용하여 측정함으로써, 높은 정확도와 누적 저항 오차가 없도록 설계하여 사용한다. 통신부는 2개의 LAN 포트와 1개의 RS-232 포트로 구성하여 복합 미생물 배양기에서 사용되는 LCD 패널, PCT 패널, 로드셀 컨트롤러 등의 신호를 제어부에 전달할 수 있도록 설계 및 제작한다. 전원부는 제어부와 통신부가 원활하게 동작할 수 있도록 24V, 12V 5V 등 3개의 전압 공급 단자로 구성하여 전원을 공급하도록 설계 및 제작한다. 복합 미생물 배양기의 제어시스템은 PLC를 사용하여 pH 농도센서, 온도센서, 수위센서 등의 센서값과 배양에 사용되는 써큘레이션 펌프, 써큘레이션 밸브, 로터리 펌프와 인버터 로드셀 등의 동작을 제어한다. 제안된 복합 미생물 배양기의 제어시스템의 성능을 평가하기 위하여 공인인증기관에서 실험한 결과는 수위 측정감도의 범위가 -0.41mm~1.59mm로, 물 온도의 변화 폭이 ±0.41℃로 현재 상용으로 판매되는 제품들 성능보다 우수한 성능으로 동작됨이 확인되었다. 따라서, 본 논문에서 제안한 복합 미생물 배양기의 제어시스템의 효용성이 입증되었다.
고춧가루의 LED(적색, 황색, 청색, 녹색광) 처리시 저장기간 중 미생물 저감화, 수용성 색소, capsanthin 함량, 표면 색도(Hunter L, a, b, ${\Delta}E$) 및 관능평가를 실시하였다. 저장기간 중 총 호기성 세균은 유의적인 차이를 나타내지 않았으며, 효모 및 곰팡이의 경우 황색 LED 처리군에서 1.76 log 감소되었다. 표면 색도는 저장기간이 경과함에 따라 모든 시료군에서 명도(L), 적색도(a) 및 황색도(b)가 유의적으로 감소하는 경향을 나타내었다. 전반적인 색차는 황색 LED 시료에서 ${\Delta}E$ 값이 3.0 이하로 나타나 다른 시료군에 비해 색도의 변화가 적은 것으로 나타났다. 수용성 색소는 저장기간 중 유의적인 차이는 나타나지 않았으며, capsanthin 함량은 저장기간이 경과함에 따라 감소하였으나 적색 LED 시료와 황색 LED 시료는 저장 10일 후 초기와 유의적인 capsanthin 함량 차이를 나타내지 않았다. 관능적 품질은 대조군의 경우 외관, 색 및 상품성이 저장기간 중 유의적으로 감소하는 경향을 나타내었으나 황색 LED 시료와 녹색 LED 시료는 유의적인 차이가 나타나지 않았다.
Bioethanol production from lignocellulose is considered as a sustainable biofuel supply. However, the low cellulose hydrolysis efficiency limits the cellulosic ethanol production. The cellulase is strongly inhibited by the major end product cellobiose, which can be relieved by the addition of ${\beta}$-glucosidase. In this study, three ${\beta}$-glucosidases from different organisms were respectively expressed in Saccharomyces cerevisiae and the ${\beta}$-glucosidase from Saccharomycopsis fibuligera showed the best activity (5.2 U/ml). The recombinant strain with S. fibuligera ${\beta}$-glucosidase could metabolize cellobiose with a specific growth rate similar to the control strain in glucose. This recombinant strain showed higher hydrolysis efficiency in the cellulose simultaneous saccharification and fermentation, when using the Trichoderma reesei cellulase, which is short of the ${\beta}$-glucosidase activity. The final ethanol concentration was 110% (using Avicel) and 89% (using acid-pretreated corncob) higher than the control strain. These results demonstrated the effect of ${\beta}$-glucosidase secretion in the recombinant S. cerevisiae for enhancing cellulosic ethanol conversion.

NdgR, a global regulator in soil-dwelling and antibiotic-producing Streptomyces, is known to regulate branched-chain amino acid metabolism by binding to the upstream region of synthetic genes. However, its numerous and complex roles are not yet fully understood. To more fully reveal the function of NdgR, phospholipid fatty acid (PLFA) analysis with gas chromatography-mass spectrometry (GC-MS) was used to assess the effects of an ndgR deletion mutant of Streptomyces coelicolor. The deletion of ndgR was found to decrease the levels of isoleucine- and leucine-related fatty acids but increase those of valine-related fatty acids. Furthermore, the defects in leucine and isoleucine metabolism caused by the deletion impaired the growth of Streptomyces at low temperatures. Supplementation of leucine and isoleucine, however, could complement this defect under cold shock condition. NdgR was thus shown to be involved in the control of branched-chain amino acids and consequently affected the membrane fatty acid composition in Streptomyces. While isoleucine and valine could be synthesized by the same enzymes (IlvB/N, IlvC, IlvD, and IlvE), ndgR deletion did not affect them in the same way. This suggests that NdgR is involved in the upper isoleucine and valine pathways, or that its control over them differs in some respect.
A microbial fuel cell (MFC) and bioelectrochemical systems are novel bioprocesses which employ exoelectrogenic biofilm on electrode as a biocatalyst for electricity generation and various useful chemical production. Previous reports show that electrogenic biofilms of MFCs are time varying systems and dynamically interactive with the electrically conductive media (carbon paper as terminal electron acceptor). It has been reported that maximum power point tracking (MPPT) method can automatically control load by algorithm so that increase power generation and columbic efficiency. In this study, we developed logic based control strategy for external load resistance by using $LabVIEW^{TM}$ which increases the power production with using flat-plate MFCs and MPPT circuit board. The flat-plate MFCs inoculated with anaerobic digester sludge were stabilized with fixed external resistance from $1000{\Omega}$ to $100{\Omega}$. Automatic load control with MPPT started load from $52{\Omega}$ during 120 hours of operation. MPPT control strategy increased approximately 2.7 times of power production and power density (1.95 mW and $13.02mW/m^3$) compared to the initial values before application of MPPT (0.72 mW and $4.79mW/m^3$).
Objective: This study was to determine the bacterial diversity and monitor the community dynamic changes during storage of vacuum-packaged sliced raw beef as affected by Lactobacillus sakei and Lactobacillus curvatus. Methods: L. sakei and L. curvatus were separately incubated in vacuumed-packaged raw beef as bio-protective cultures to inhibit the naturally contaminating microbial load. Dynamic changes of the microbial diversity of inoculated or non-inoculated (control) samples were monitored at $4^{\circ}C$ for 0 to 38 days, using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). Results: The DGGE profiles of DNA directly extracted from non-inoculated control samples highlighted the order of appearance of spoilage bacteria during storage, showing that Enterbacteriaceae and Pseudomonas fragi emerged early, then Brochothrix thermosphacta shared the dominant position, and finally, Pseudomonas putida showed up became predominant. Compared with control, the inoculation of either L. sakei or L. curvatus significantly lowered the complexity of microbial diversity and inhibited the growth of spoilage bacteria (p<0.05). Interestingly, we also found that the dominant position of L. curvatus was replaced by indigenous L. sakei after 13 d for L. curvatus-inoculated samples. Plate counts on selective agars further showed that inoculation with L. sakei or L. curvatus obviously reduced the viable counts of Enterbacteraceae, Pseudomonas spp. and B. thermosphacta during later storage (p<0.05), with L. sakei exerting greater inhibitory effect. Inoculation with both bio-protective cultures also significantly decreased the total volatile basic nitrogen values of stored samples (p<0.05). Conclusion: Taken together, the results proved the benefits of inoculation with lactic acid bacteria especially L. sakei as a potential way to inhibit growth of spoilage-related bacteria and improve the shelf life of vacuum-packaged raw beef.
This study researched the microbial change of quality according to various phases of product flow of Dongtae-Jeon (a pan-fried dish) and rolled egg in packaged meals. In order to carry out the study, the time required, temperature, water activity and microbial quality were measured at various phases of production flow of Dongtae-Jeon and rolled egg in packaged meals, and the effects of these factors on microbial multiplication was analyzed. According to the phases in product flow of Dongtae-Jeon, it was shown that the time required is 12.5hrs and water activity is distributed 0.932-0.980. These conditions were suitable for microbial multiplication. According to the phases in product flow of rolled egg, it was shown that the time required is 3.3hrs. In addition, qualitative analysis of pathogenic microorganisms (Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus) detected no such microorganisms in any of the samples.
