MicroRNA-451 Inhibits Growth of Human Colorectal Carcinoma Cells via Downregulation of Pi3k/Akt Pathway

  • MicroRNAs (MiRNAs) play important roles in coordinating a variety of cellular processes and abnormal expression has been linked to the occurrence of several cancers. The miRNA miR-451 is downregulated in colorectal carcinoma (CRC) cells, suggested by several research groups including our own. In this study, synthetic miR-451 mimics were transfected into the SW620 human CRC cell line using Lipofectamine 2000 and expression of miR-451 was analyzed by real time PCR, while expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2 was analyzed by Western blot, and cell growth was detected by MTT assay. In comparison to the controls, a significant increase in the expression of miR-451 was associated with significantly decreased expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2. The capacity of cell proliferation was significantly decreased by miR-451 expression, which also inhibited cell growth. Our study confirmed that miR-451 has a repressive role in CRC cells by inhibiting cell growth through down-regulating the P13K/AKT pathway.

Factors Influencing Adult Media Engagement of Adolescents from Micro-systemic Perspective: Mediation Analysis of Cell-Phone Dependency (미시체계적관점에서의 청소년 성인매체 몰입에 미치는 영향요인: 휴대폰 의존의 매개효과 검정)

  • Purpose: The purpose of this study was to find the factors influencing adolescents' adult media engagement. The study is based on Agnew's General Strain Theory (GST) and a micro-systemic perspective. Methods: The study examined 479 middle school freshmen who participated in the fourth wave of the Korean Children & Youth Panel Survey (KCYPS) conducted by National Youth Policy Institute. Data were analyzed through descriptive statistics, factor analysis, and reliability analysis using SPSS 22.0 Ver. Also, hypothesis testing was performed via covariance-based structure equation modeling (CB-SEM) using Amos 21.0 Ver. Results: Strain factors had positive effects on adolescent depression, and depression had a positive effect on adult media engagement. Also, cell-phone dependency had a positive mediation effect between depression and adult media engagement. Conclusion: The study's result identified the factors affecting adult media engagement. Therefore, we suggest intervention programs considering adolescents' strains, depression and cell-phone dependency to improve their health.

Characterization of Sol-gel Coated Pb(ZrTi)O3 Thin film for Piezoelectric Vibration MEMS Energy Harvester (압전 MEMS 진동에너지 수집소자를 위한 졸겔 공법기반의 Pb(ZrTi)O3 박막의 특성 분석 및 평가)

  • In this paper, sol-gel-spin coated $Pb(ZrTi)O_3$ thin film with $ZrO_2$ buffer-layer and $PbTiO_3$ seed-layer was investigated for vibration MEMS energy harvester to scavenge power from ambient vibration via d33 piezoelectric mode. Piezoelectric thin film deposition techniques on insulating layer is the important key for $d_{33}$ mode of piezoelectric vibration energy harvester. $ZrO_2$ buff-layer was utilized as an insulating layer. $PbTIO_3$ seed-layer was applied as an inter-layer between PZT and $ZrO_2$ layer to improve the crystalline of PZT thin film. The fabricated PZT thin film had a remanent polarization of 5.3uC/$cm^2$ and the coercive field of 60kV/cm. The fabricated energy harvester using PZT thin film with PTO seed-layer generated 1.1uW of electrical power to $2.2M{\Omega}$ of load with $4.4V_{pvp}$ from vibration of 0.39g at 528Hz.

  • In-plane tension fatigue tests (R = 0.05) were carried out to investigate the initiation and propagation behaviors of micro-surface-fatigue cracks on smooth surfaces of a mild steel. Also, the investigations of saturated cyclic strain which can be obtained by the fatigue tests have been made via the cyclic strain intensity factor, .DELTA. $K_{\epsilon}$/, for the purpose of unifying two approaches of the study of fatigue; the one approach is based on the fracture mechanics concept and the other on lowcycle fatigue concept. Some of the results are as follows; The growth rate, d(2a)/dN, of small cracks cannot be represented by one straight line as a function of .DELTA.K for various of the nominal stress range, .DELTA..sigma., and is higher than that of a larger through crack. The rearrangement of the d(2a)/dN by .DELTA..epsilon..root..pi.s( = .DELTA. $K_{\epsilon}$/) with the stress range .DELTA..epsilon. in .DELTA.K replaced by .DELTA..epsilon., strain range, gives one straight line of the .DELTA. $K_{\epsilon}$-d(2a)/dN relation for various values of stress range .DELTA.$_{\epsilon}$../.X>../.

Post-transcriptional and translational regulation of mRNA-like long non-coding RNAs by microRNAs in early developmental stages of zebrafish embryos

  • At the post-transcriptional and translational levels, microRNA (miRNA) represses protein-coding genes via seed pairing to the 3' untranslated regions (UTRs) of mRNA. Although working models of miRNA-mediated gene silencing are successfully established using miRNA transfections and knockouts, the regulatory interaction between miRNA and long non-coding RNA (lncRNA) remain unknown. In particular, how the mRNA-resembling lncRNAs with 5' cap, 3' poly(A)-tail, or coding features, are regulated by miRNA is yet to be examined. We therefore investigated the functional interaction between miRNAs and lncRNAs with/without those features, in miRNA-transfected early zebrafish embryos. We observed that the greatest determinants of the miRNA-mediated silencing of lncRNAs were the 5' cap and 3' poly(A)-tails in lncRNAs, at both the post-transcriptional and translational levels. The lncRNAs confirmed to contain 5' cap, 3' poly(A)-tail, and the canonical miRNA target sites, were observed to be repressed in the level of both RNA and ribosome-protected fragment, while those with the miRNA target sites and without 5' cap and 3' poly(A)-tail, were not robustly repressed by miRNA introduction, thus suggesting a role as a miRNA-decoy.

Morphological Variation and Luminescence Properties of ZnO Micro/Nanocrystals Synthesized by Thermal Evaporation Method

  • ZnO micro/nanocrystals with different morphologies were synthesized by thermal evaporation of various zinc source materials in an air atmosphere. Zinc acetate, zinc carbonate and zinc iodide were used as the source materials. No catalysts or substrates were used in the synthesis of the ZnO crystals. The scanning electron microscope(SEM) image showed that the morphology of ZnO crystals was strongly dependent on the source materials, which suggests that source material is one of the key factors in controlling the morphology of the obtained ZnO crystals. Tetrapods, nanogranular shaped crystals, spherical particles and crayon-shaped crystals were obtained using different source materials. The X-ray diffraction(XRD) pattern revealed that the all the ZnO crystals had hexagonal wurtzite crystalline structures. An ultraviolet emission was observed in the cathodoluminescence spectrum of the ZnO crystals prepared via thermal evaporation of Zn powder. However, a strong green emission centered at around 500 nm was observed in the cathodoluminescence spectra of the ZnO crystals prepared using zinc salts as the source materials.

  • It is well established that there is a heritable element of susceptibility to chronic human ailments, yet there is compelling evidence that some components of such heritability are transmitted through non-genetic factors. Due to the complexity of reproductive processes, identifying the inheritance patterns of these factors is not easy. But little doubt exists that besides the genomic backbone, a range of epigenetic cues affect our genetic programme. The inter-generational transmission of epigenetic marks is believed to operate via four principal means that dramatically differ in their information content: DNA methylation, histone modifications, microRNAs and nucleosome positioning. These epigenetic signatures influence the cellular machinery through positive and negative feedback mechanisms either alone or interactively. Understanding how these mechanisms work to activate or deactivate parts of our genetic programme not only on a day-to-day basis but also over generations is an important area of reproductive health research.

  • In this paper, we implemented LED beam forming communication system controlled by stepping motor. ATMega1284 was used as a MCU of main control board which has two main external IO, one is RS232 for connection with PC, the other is PORT for connection with motor driving board. Stepping motor rotated 360 degree when provided 160 clock and its rotation radius was increased by Archimedian Spiral. So LED can provide its light anywhere in the space and its beam forming was controlled by PC connected with RS232 of main control board. The action of beam forming was verified via actual HW/SW implementation.

Repressive effects of red bean, Phaseolus angularis, extracts on obesity of mouse induced with high-fat diet via downregulation of adipocyte differentiation and modulating lipid metabolism

  • Obesity is generally caused by quantitative changes in adipocyte differentiation and fat metabolism. Only a few studies have been determined the effect of red beans extract on obesity and plasma cholesterol concentration. We have been studied the functional activities of red-bean extracts including anti-oxidative effect against DNA and cell damages. Histological study including micro CT analysis showed that the accumulation of fat in hepatocytes and intestines was significantly decreased in red bean extract treated group. In addition, plasma cholesterol and triglyceride levels were decreased in blood samples. In addition, it was confirmed that the red bean extract inhibited the expression of $PPAR{\gamma}$, Fabp4 and RETN genes, which regulate total adipocyte differentiation and lipid metabolism. Red bean extract inhibits the expressions of transcription factors associated with adipocyte differentiation in a dose-dependent manner, thereby inhibiting fat accumulation and decreasing blood lipid levels in obese mice induced by high fat diet.

Development of a micro-scale Y-Zr-O oxide-dispersion-strengthened steel fabricated via vacuum induction melting and electro-slag remelting

  • In this paper, the CLAM steel strengthened by micro-scale Y-Zr-O was prepared by vacuum induction melting followed by electroslag remelting (VIM-ESR). Yttrium (Y) and zirconium (Zr) were easy to aggregates into massive yttrium-zirconium-rich inclusions in the steel melted by vacuum induction melting (VIM), which would interrupt the continuity of the matrix and reduce the mechanical properties of steel. Micron-sized Y-Zr-O inclusions would be produced with the removal of original blocky Y-Zr-rich inclusions and the submicron-sized inclusions smaller than $0.2{\mu}m$ could be retained in the steel. The small grain size and the better refinement and distribution uniformity of Y-Zr-O inclusions after remelting would be responsible for the better yield strength and toughness. For VIM-ESR alloy, the ultimate tensile strength is 749 MPa and the yield strength is 642 MPa at room temperature, meanwhile they are 391 MPa and 367 MPa at $600^{\circ}C$, respectively. Meanwhile, the ductile-brittle transition temperature (DBTT) reduced from $-43^{\circ}C$ (VIM) to $-76^{\circ}C$ (VIM-ESR).
