  • Bone is so crucial anatomy for human body. Many researchers study deep into a subject about bone regeneration. There is no standard analysis for quantitative Neo-bone regeneration on calvarial defected model. Micro CT is so useful method to quantitative analysis of Neo-bone regeneration. This study was show that how to quantitative analysis of Neo-bone regeneration with ${\mu}-CT$ Micro CT was possible to quantitative analysis for Neo-bone regeneration on Calvarial defected model. futhermore Not only was Micro CT possible for qualitative analysis but quantitative analysis on the mouse calvarial model. This study will provide bone biology researchers with accurate quantitative analysis.

초고해상도 미세영상 기법을 이용한 Mouse 뇌의 자기공명영상 연구 (High-Resolution MRI Study on Mouse Brain Using Micro-Imaging)

  • 목적 : 본 연구는 핵자기공명 분광기를 개조한 미세영상 기법을 이용하여, 동물실험에 주류를 이루는 mouse를 대상으로, 0.1 mm 이내의 초고해상도 자기공명영상을 5분 정도 시간 안에 획득할 수 있는 방법을 개발하고자 하였다. 대상 및 방법 : 사용된 mouse는 C57BL/6로서 무게 50 그램 이내의 mouse를 사용하였다. 본 연구에 활용된 초전도 자석은 구경 89 mm, 4.7 T의 자기장 세기를 가진 수직형 자석이며, 사용된 샘플 코일의 직경은 30 mm 이고, 사용된 펄스시퀀스는 fast spin echo (FSE) 및 gradient echo (GE) 기법들이다. 결과 : 최적의 자기공명영상 파라미터를 확보하면서 2차원 영상으로서 수소밀도 및 T2 강조 영상을 획득하였다. 영상으로부터 mouse 뇌의 미세부분까지 상세히 해부학적 구조를 확인할 수 있었고, 또한 입체적인 정보를 획득하기 위하여 3D 영상도 부가적으로 획득하였다. 조영제를 이용한 dynamic contrast 연구에 3D 영상이 매우 유용하였다. 결론 : 본 연구를 통하여 mouse 뇌에 대한 고해상도 자기공명영상 획득을 위한 최적의 파라미터를 확보할 수 있었고, 또한 성공적인 자기공명영상도 획득하였다. 즉, 사람이나 다른 소동물뇌의 경우와 같이 mouse 뇌 조직의 다양한 부위의 미세부분을 확인할 수 있는 충분한 고해상도의 영상을 획득하였다. 최근 국내에서 mouse를 이용한 자기공명영상 연구가 시작되었으나 아직 초기단계라고 평가할 수 있고, mouse는 다른 동물에 비하여 취급/관리하기 쉬우므로 향후 mouse를 이용한 뇌 연구가 활성화 될 것으로 사료된다.

Administration of red ginseng regulates microRNA expression in a mouse model of endometriosis

  • Objective: Red ginseng (RG) exerts anti-inflammatory, anti-proliferative, and immunomodulatory effects on endometriosis through the regulation of microRNA (miRNA) expression. It may also ameliorate endometriosis by affecting the expression of multiple miRNAs simultaneously, rather than acting on a single miRNA at a given time. Since studies on the overall effects of RG on endometriosis via the regulation of miRNA expression are lacking, the current study aimed to explore the global effect of RG on miRNA expression in a mouse model of endometriosis. Methods: To establish the mouse model, the uterine horn of donor mice was implanted into the lateral side of the recipients' peritoneum, followed by vehicle or RG treatment for 8 weeks. Results: To confirm the effects of RG on the established mouse model, the size of the implanted uterus was measured; it was found to be lower in mice from the RG group than in mice from the control group. miRNA expression profiles in the implanted uterus of the mouse model of endometriosis after vehicle or RG administration were analyzed using microarray technology. Thereafter, seven candidate miRNAs and 125 candidate genes (miRNA targets) were identified through a bioinformatics analysis. Conclusion: The present findings suggest that RG regulates the expression of multiple miRNAs and mRNAs, thereby alleviating endometriosis in a mouse model of the disease.

A Sperm Factor Inducing Second Polar Body Formation in Mouse Secondayy Oocyte

  • A sperm factor(s) for oocyte activation during fertilization has not been clearly identified. In this study to elucidate an oocyte activation factor(s), mouse sperm were sonicated and ultra-filtered with a 30 kilo-daltons (KD) cutoff membrane and the ultra-filtrate was then sequentially fractionated over Suporose 12 column and Superdex column, The recovered fractions were micro-injected into Mⅱmouse oocytes and second polar body formation (PBF) was examined. Suporose fraction RV2.10 prepared from sperm extract significantly increased PBF. Of Superdex fractions re-separated from Suporose fraction RV2.10, fraction RV2.12 also had the strongest PBF activity. By analyzing with micro-reverse phase column (URPC), the Superdex fraction RV2.12 appeared to be glutamic acid. In microinjection test, glutamic acid significantly increased PBF. This study suggests that glutamic acid should be a type of sperm factor for second polar body formation related to oocyte activation.

The effects of circadian disturbances induced by night shifts on the mouse peripheral tissues

  • We have examined the effect of circadian disturbances induced by night shifts (NSs) on the phenotypes of the tibiae and abdominal adipose tissues (ADTs) in a mouse model by using in vivo micro-computed tomography (micro-CT). We found that the volumes of total and visceral ADTs in the night-shifted group of mice were significantly larger (69 and 92%, respectively) than those in the control. The mean polar moment of inertia, cross-sectional thickness, and bone mineral density of the cortical bone in the night-shifted group of mice were less (13, 5, and 3%, respectively) than those in the control. Moreover, the volume and the thickness of growth plates (GPs) of the tibiae in the night-shifted mice were significantly smaller (22 and 20%, respectively) than those in the control. Taken together, our results indicate that disturbances in the mouse circadian rhythms induced by NSs affect the morphological characteristics of cortical bone, the volume and the thickness of GPs, and the volume of ADTs.

This paper designed, fabricated and tested a digital hearing aid directivity testing device in which a micro-mouse-like the stepping motor with a speaker rotates around an examinant. Both ears of the examinant were fixed with ITE hearing aids in order to response to receiving sound. The diameter of the directivity testing device was 2 [m] and the micro-mouse was precisely controlled by PICBASIC micro processor.

생쥐 및 소 수정란의 분할방법에 관한 연구 (A Study on the Splitting Methods in Mouse and Bovine Embryos)

  • This study was carried out to obtain the basic information on splitting and culture of mouse and bovine embryos. Two-, four-, eight-, cell and morula mouse embryos were digested with pronase, splitted in vitro by micro-glass needel with hand, and bovine embryos were splitted by micromanipulator. The splitted embryos were cultured under 5% of CO2 gas in air at 37$^{\circ}C$ for 48-72 hours. The results obtained in this study were summarized as follows: 1. The mouse and cattle were superovulated by 5IU of PMS and HCG, and 2500IU of PMS and 25mg of PGF2$\alpha$, respectively. The average number of embryos after superovulation were 32.5$\pm$8.2 and 7.5$\pm$3:1, respectively. 2. Out of total 122 embryos splitted, the successful splitting rate was 75.0%, 66.7%, 68.4% and 71.4% in 2-, 4-, 8- and morula embryos in mouse, respectively. There was no different splitting rate between mouse(71.4%) and bovine embryos(66.7%) in morula. 3. The successful culture rate of splitted embryos was 68.0% and 67.9% in mouse and bovine embryos, respectively.

  • 실험상의 어려움으로 인하여 생체역학 분야에서 유한요소법은 최선의 대안으로 여겨지고 있다. 또한, CT를 비롯한 여러 디지털 장비로부터 얻은 영상데이터를 이용하여 유한요소모델을 생성하는 방법으로 모델 생성 속도의 향상을 가져와 더욱 효율을 높이고 있다. 그러나, 일반 CT의 해상도를 가지고는 골(bone) 변화의 주를 이루고 있는 해면골(trabecular bone)의 변화를 파악하기에는 큰 어려움이 따른다. 이에 본 논문에서는 10~20$\mu\textrm{m}$ 정도까지의 해상도를 가지는 Micro-CT의 영상데이터를 이용하여 유한요소 모델을 생성하고 해석하여 보았다.(중략)
