Study on the immobilization of plant glutathione S-transferase for development of herbicide detection kit (제초제 검출 키트 개발을 위한 식물 해독효소 고정화 연구)

  • Glutathione S-transferase is known to play a crucial role in detoxification in many cases. To develop a herbicide detection biosensor, we in this study attempted to immobilize glutathione S-transferase enzyme on solid supports, polystyrene and agarose, and Na-alginate. These matrixes were attractive materials for the construction of biosensors and might also have utility for the production of immobilized enzyme bioreactors. We also compared the activities of glutathione-S-transferase immobilized OsGSTF3 and free OsGSTF3. The specific activity of the free enzyme in solution was 3.3 higher than the immobilized enzyme. These results suggest that 50% of the enzyme was bound with the catalytic site in polystyrene-alkylamine bead and immobilized enzymes showed 80% remaining activity until 3 times reuse.

The Structural and Microwave Dielectric Properties of the BMT Ceramics with Sintering Temperature and BCN Composition Ratio (소결온도와 BCN 초성에 따른 BMT 세라믹스의 구조 및 마이크로파 유전특성)

  • The microwave dielectric properties of Ba(Mg$_{1}$3/Ta$_{2}$3/O$_3$-Ba(Co$_{1}$3/Nb$_{2}$3/O$_3$[BMT-BCN] ceramics were investigated. The specimens were prepared by the conventional mixed oxide method with the sintering temperature of 15$25^{\circ}C$~1575$^{\circ}C$. It was found that Ba(Mg$_{1}$3/Ta$_{2}$3/O$_3$[BMT] and BCN formed a solid solution with complex perovskite structure. As increasing the mole fraction of BCN, dielectric constant increased while the temperature coefficient of resonant frequency was changed from positive to negative value. The highest value of quality factor, Q$\times$f$_{0}$=138,205GHz, obtained in the 0.9BMT-0.1BCN ceramics sintered at 1575$^{\circ}C$. In the range of x$\geq$0.4, the dielectric constant was about 30. The 0.55BMT-0.45BCN ceramics sintered at 15$25^{\circ}C$ for 5 hours showed the microwave dielectric properties of $\varepsilon$$_{r}$=30.21, Q$\times$f$_{0}$=85,789GHz and $\tau$$_{f}$=2.9015ppm/$^{\circ}C$.EX>.

  • The lower-order lithium organocyanocuprate compound, (THF)₃Li(NC)Cu($C_6$H₃-2,6-Mes₂) (1), and the bulky terphenyl Grignard reagent, Br(THF)₂Mg($C_6$H₃-2,6-Trip₂) (2), have been synthesized and structurally characterized both in the solid state by single crystal x-ray crystallography and in solution by multi-nuclear NMR and IR spectroscopy. The compound (1) was isolated as a monomeric contact ion-pair in which the C (organic ipso)-Cu-CN-Li atoms are coordinated linearly. The lithium has a tetrahedral geometry as a result of solvation by three THF molecules. The compound (1) is the first example of fully characterized monomeric lower order lithium organocyanocuprate. The bulky Grignard reagent (2) was also isolated as a monomer in which the magnesium, solvated by two THF molecules, has a distorted tetrahedral geometry. The crystals of (1) possess triclinic symmetry with the space group $P{\={1}}$, Z = 2, with a = 12.456(3) Å, b = 12.508(3) Å, c = 13.904(3) Å, α = 99.81°, β = 103.72(3)°, and γ = 119.44(3)°. The crystals (2) have a monoclinic symmetry of space group $P2_{1/C}$, Z = 4, with a = 13.071(3) Å, b = 14.967(3) Å, c = 22.070(4) Å, and β = 98.95(3)°.

The effects of cytokinin and plating density on protoplast culture of sunflower

  • Sunflower (Helianthus annuus L.) protoplasts were isolated from seven-day-old etiolated hypocotyls of 10 A line and four-week-old fully expanded young leaves of PI 441983 line in vitro seedlings using an enzymatic method. Purified protoplasts were collected by filtration and floatation in sucrose solution. Semi-solid protoplast culture was performed using the L4 regeneration protocol with various culture media and plating densities to achieve the highest efficiencies for protoplast culture of hypocotyl and mesophyll protoplasts of 10 A and PI 441983 lines, respectively. The concentrations in liquid L'4M medium and different plating densities were evaluated in two types of cytokinins, the adenine-type 6-benzyladenine (BA) and the phenylurea-type thidiazuron (TDZ). The highest colony formation was achieved in both sunflower lines when 0.5 mgL-1 BA and 0.5 mgL-1 TDZ were applied with a high plating density (3 × 105 protoplasts mL-1). These conditions led to 38.45% and 39.40% colony formation for hypocotyl protoplasts of the 10 A line and mesophyll protoplasts of the PI 441983 line, respectively. Moreover, many hypocotyl protoplast-derived colonies developed into micro-calli. In addition, superior development of both sunflower protoplasts was observed with all plating densities when BA was used in combination with TDZ. This finding will be applicable to future sunflower hybrid production via somatic hybridization.

  • To study the Cd adsorption in the presence of competing ions in soil-solution interphase, three soil samples from the Bt horizon were taken and analyzed for their physical and chemical properties. Adsorption of ethylene glycol monoethyl ether(EGME) and N, were determined to establish the specific surface area of the soils. We attempted to establish a qeneralizing competitive sorption isotherms for soils of entirely different composition of the solid phase, resulting in the routine use as a guidelines for the fate of reactive solute in soil profiles. Many physicochemical factors including competitive adsorption bettween solutes will affect the general adsorption phenomena as shown in a single not only on the soil:solution ratio used, but also on the surface areas of its respective soil samples. This phenomenon was attributed to competition Cd for sorption sites with Mg by different soil constituents. These adsorption isotherms are able to use as examples to demonstrate that this phenomenon can complicate the development of a standardized batch adsorption procedure as well as interpreting fate and adsorption of toxic inorganic compounds.

Efficiency and Mechanism of Pb(II) Removal from Aqueous Solutions Using Cornus controversa and Quercus mongolica Biomass Waste (층층나무와 신갈나무 폐바이오매스를 활용한 수용액 중 납 제거 효율 및 기작)

  • BACKGROUND: Enormous amounts of the wood biomass wastes have been produced through various wood processing. This study characterizes the surface characteristics of biomass powders of Cornus controversa (CC) and Quercus mongolica (QM) and investigates their removal efficiency and mechanism for Pb (II) in aqueous solution on which to base potential recycling alternative of the wood biomass. METHODS AND RESULTS: Batch experiments were conducted under different conditions of Pb concentrations, temperatures, time and solid/solution ratios. Adsorption isotherm of Pb by CC and QM biomass was explained significantly by the Langmuir model, indicating Pb was likely adsorbed on the monolayer of the surfaces. The adsorption kinetics were fitted significantly to the double first-order model consisting of rapid and slow steps. The respective rate constants (k1) of CC and QM for the rapid adsorption kinetic steps were 0.051 and 0.177 min-1, and most of the sorption reactions proceeded rapidly within 6-20 minutes. The maximum adsorption quantities (qmax) of Pb were 17.25 and 23.47 mg/g for CC and QM, respectively. Thermodynamic parameters revealed that adsorption of Pb on the biomass of CC and QM was a spontaneous endothermic reaction. CONCLUSION(S): Results demonstrate that biomass wastes of CC and QM can be used as Pb adsorbents judging from adsorption isotherm, kinetics, and thermodynamic parameters.

Plant Regeneration via Multiple Shoot Formation from Sucker Explant of Hybrid Blackberry (Rubus fruticosus L. ${\times}$ R. parvifolius L.) (교잡종 블랙베리(Rubus fruticosus L. ${\times}$ R. parvifolius L.)의 근맹아 절편체로부터 다경유도를 통한 식물체 재분화)

  • This study was carried out to induce plant regeneration via multiple shoot formation from sucker explants of Rubus fruticosus L. ${\times}$ R. parvifolius L. To induce adventitious shoots, the sucker explants were sterilized in 1.2% sodium hypochlorite (NaOCl) solution, and then were cultured on the full and 1/2 MS solid medium supplemented with BA (0.1, 0.5, 1.0, $2.0mg{\cdot}L^{-1}$). After 4 weeks of culture, the highest frequency (83.3%) of shoot formation from sucker explants was obtained from the full MS medium with $1.0mg{\cdot}L^{-1}$ BA. The highest shoot number (3.7) per explant was obtained from the full MS medium with $0.5mg{\cdot}L^{-1}$ BA. After 12 weeks of culture, the number of shoots (15.4) per explant was increased. The highest frequency (95%) of root formation was obtained from the 1/2 MS medium, when the explant with shoot were cultured on the full, 1/2 and 1/4 MS medium. The survival rate of the plantlets after transfer to plastic pots containing sand, soil, and vermiculite (1.1:1, vol.) was 95%. The results indicate that multiple shoot procedure can be applied for an efficient mass propagation of Rubus fruticosus L. ${\times}$ R. parvifolius L.

  • B$_2$O$_3$ added Ba(Mg$_{1}$3/Nb$_{2}$3/)O$_3$ (BBMN) ceramics were not sintered below 900 $^{\circ}C$. However, when CuO was added to the BBMN ceramic, it was sintered even at 850 $^{\circ}C$. The amount of the $Ba_2$B$_2$O$_{5}$ second phase decreased with the addition of CuO. Therefore, the CuO additive is considered to react with the B$_2$O$_3$ inhibiting the reaction between B$_2$O$_3$ and BaO. Moreover, it is suggested that the solid solution of CuO and B$_2$O$_3$ might be responsible for the decrease of the sintering temperature of the specimens. A dense microstructure without pores was developed with the addition of a small amount of CuO. However, a porous microstructure with large pores was formed when a large amount of CuO was added. The bulk density, the dielectric constant ($\varepsilon$$_{r}$) and the Q-value increased with the addition of CuO but they decreased when a large amount of CuO was added. The variations of those properties are closely related to the variation of the microstructure. The excellent microwave dielectric properties of Qxf = 21500 GHz, $\varepsilon$$_{r}$ = 31 and temperature coefficient of resonance frequency($\tau$$_{f}$) = 21.3 ppm/$^{\circ}C$ were obtained for the Ba(Mg$_{1}$3/Nb$_{2}$3/)O$_3$+2.0 mol%B$_2$O$_3$+10.0 mol%CuO ceramic sintered at 875 $^{\circ}C$ for 2 h.h.2 h.h.

Analysis of Sludges from Wastewater of Hanji Production (한지 생산공정에서 발생하는 슬러지의 분석)

  • Sludges from the wastewater of hanji production were analysed to see that sludges from the bleaching process, the bleach washing process, and the paper preparation process were fibrous, while sludges from the process cooking with NaOH and the process of final chemical and microbiological treatment were non-fibrous. Sludge solution from the bleaching process had the most amount of solid matters. Sludges from the bleaching process, the bleach washing process, and the final chemical and microbiological treatment process contained about 80-90 %(w/w) moisture. Owing to hypochlorite ion, pH of sludge from the bleaching process was higher than any other sludge samples. The inorganic components of sludges were determined by ICP-AES. The major inorganic component of sludge from the process cooking with NaOH was Na, while the sludge from the bleaching process had Na and Ca, major components in the final chemical and microbiological treatment process were Ca and Al. Trace amount of Mg, Fe, K, P, Mn and Ti were observed in sludges. Sludge from the bleaching process had ashes more than any other sludges. Shape of sludges observed by SEM was appeared to fibrous or crystalline, and the breadth of fibrous sludge was $5{\sim}50{\mu}m$. The sludge from the bleaching process had the most amount of matters soluble by cold and hot water and by alcohol-benzene.
