  • We examined the inhibitory activities against monoamine oxidase (MAO) of Prunella vulgaris in vitro and in vivo methods. Methanolic extract of P. vulgaris showed significantly Inhibitorγ activities on MAO-A and MAO-B that were prepared from rat brain and liver in vitro. The inhibitory activities were measured by serotonin and benzylamine as substrates, respectively. MAO-A and MAO-B activities were potently inhibited by ethylacetate extracts of P. vulgaris in vitro tests. It was observed that those activities in vivo tests have different tendency each other. MAO-A activity was increased by the oral administration of methanolic extract of P. vulgaris while MAO-B activity was decreased. Consequently, we suggest that P. vulgaris may have the effects on the inhibitory activities against MAO both in vitro and in vivo.

  • In the present study, we have tested the effects of 21 salt marsh plants on cell proliferation of mouse immune cells (spleen and thymus) using MTT assay in culture. The methanolic extracts of six salt marsh plants (Rosa rugosa, Ixeris tamagawaensis, Artemisia capillaris, Tetragonia tetragonoides, Erigeron annus, and Glehnia littoralis) showed very powerful suppressive effects of mouse immune cell death and significant activities of cell proliferation in vitro. Especially, the methanolic extract of Rosa rugosa was found to have fifteen times compared to the control treatment, demonstrating that Rosa rugosa may have a potent stimulation effect on immune cell proliferation. These results suggest that several salt marsh plants including Rosa rugosa could be useful for further study as an immunomodulating agent.

  • Background: Roots of Withania somnifera (WS) are a celebrated medicinal ingredient in Ayurvedic and many other indigenous systems of medicine. The present study investigates the effect of the phytochemical composition of the extracts on their antioxidant and reducing activities. Methods: WS roots were extracted with water, acetone, aqueous methanol (1:1), and methanol:-chloroform:water (1:1:1) to obtain aqueous, acetone, hydro-methanolic, and methanol-chloroform-water extracts. Thereafter, phytochemical constitution and antioxidant and reducing activities of the extracts were compared using different qualitative and quantitative tests. Results: Maximum extraction recovery was obtained with 50% aqueous methanol whereas extraction with acetone yielded the poorest recovery. Methanol-chloroform-water extract had the highest content of phytochemical constituents, except tannins, and also exhibited the highest antioxidant and reducing activities. Conclusion: Phytochemical composition and antioxidant and reducing activities of the extracts were positively associated with the use of organic solvents during the extraction process. Alkaloids and flavonoids were the most important contributors in the antioxidant and reducing activities of the extracts.

  • Euonymus alatus (EAE) and Euonymus alatus for. ciliatodentatus (Franch. & Sav.) Hiyama (EACHE) belong to the Celastraceae family and are one of the medicinal herbs used in traditional medicine as a therapeutic agent for blood circulation diseases. In this study, we prepared leaves methanolic extract of EAE (L-EAE), twigs methanolic extract of EAE (T-EAE), leaves methanolic extract of EACHE (L-EACHE) and twigs methanolic extract of EACHE (T-EACHE), and compared their anti-inflammatory activities. To analyze the anti-inflammatory activities, Raw 264.7 cells were used, since they are immature-macrophages and easily matured by lipopolysaccharide (LPS) stimulation. All extracts showed anti-inflammatory activities in the activated Raw 264.7 cells. That is, we showed that L-EACHE and T-EACHE are potent inhibitors of the production of nitric oxide and pro-inflammatory cytokines. These results are expected to serve as a guide for future studies on the ability of Celastraceae family to inhibit acute and chronic inflammatory diseases.

  • We investigated a total of 335 samples of Korean native mushroom extracts as part of our lipoxygenase (LOX) inhibitor screening program. Among the mushroom-methanolic extracts we investigated, 35 exhibited an inhibitory activity greater than 30% against LOX at a concentration of 100 ${\mu}g/mL$. Especially, Collybia maculata, Tylopilus neofelleus, Strobilomyces confusus, Phellinus gilvus, P. linteus, P. baumii, and Inonotus mikadoi exhibited relatively potent LOX inhibitory activities of 73.3%, 51.6%, 52.4%, 66.7%, 59.5%, 100.0%, and 85.2%, respectively. Bioassay-guided fractionation led to the isolation of inoscavin A from the methanolic extract of P. baumii, which showed the most potent activity and was identified by spectroscopic methods. Specifically, inoscavin A exhibited potent LOX inhibitory activity with an $IC_{50}$ value of $6.8{\mu}M$.

  • The total phenolic contents and the antioxidant activities of methanolic extracts of different parts (fruit, calyx, leaf, stem, and root) of Physalis alkekengi var. francheti were investigated using established in vitro systems including DPPH radical-scavenging, nitrite-scavenging, superoxide anion radical-scavenging activity, measurement of reducing power, and assessment of the metal-chelating effect. The highest extraction yield was from fruit (52.55%), whereas the lowest levels were obtained from root (10.49%) and stem (12.88%).The leaf extract had the highest total phenolic (58.47 mg/g) and total flavonoid (4.83 mg/g) contents, plus the greatest antioxidant activity, as shown by the DPPH radical-scavenging assay, and the highest levels of reducing power at concentrations of 1 and 5 mg/mL. In addition, the calyx also showed good antioxidant activity. These findings indicate that methanolic extracts of leaf and calyx may be useful in the food manufacturing and nutraceutical industries.

  • Despite considerable efforts, cancer remains an aggressive killer worldwide. Chemotherapeutic drugs that are currently in use lead to destructive side effects and have not succeeded in fulfilling expectations. For centuries, medicinal plants are used for treating various diseases and are also known to have anticancer activity. The main aim of this research was to evaluate antiproliferative activity of saffron, clove, fenugreek, and green tea on Vero and MDA-MB-231 cell lines and to subsequently analyze the effect of these extracts on IRAK-4, TAK1, IKK-alpha, IKK-beta, NF-Kappa B, IRF3, IRF7 genes in Toll Like Receptors (TLRs) pathway. Antiproliferative assay was done by Neutral Red Dye uptake assay. Methanolic extract of green tea was found to be most effective against both cell lines as IC50 was achieved at least concentration of the extract. For molecular studies, MDAMB-231 cells were sensitized with methanolic extract of green tea at same IC50, and RT-PCR was performed to determine the relative expression of genes. Expression of IRAK-4, TAK1, IKK-beta, NF-Kappa B, IRF3 genes was down regulated and IRF7 and IKKalpha was upregulated. Green tea has a potential cytotoxic effect on both cell lines which was demonstrated by its effect on the expression of (TLRs) pathway genes.

  • Since reactive oxygen radicals play an important role in carcinogenesis and other human diseases including neurodegenerative states, antioxidants present in natural products have received considerable attention for alleviation of these disease states. Therefore, in order io identify antioxidants in plant extracts, fifty-seven methanolic extracts derided from indigenous Korean plants were primarily assessed for potential to scavenge stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals. As a result, nine plant extracts were found to exhibit the DPPH free radical scavenging activity in the criteria of $IC_{50}<40\;{\mu}g/ml$. In particular, the extracts of Melioma oldhami $(IC_{50}=0.1\;{\mu}g/ml)$, Myrica rubra $(IC_{50}=16.2\;{\mu}g/ml)$, Sympolocos paniculata $(IC_{50}=23.0\;{\mu}g/ml)$, Carpinus laxiflora $(IC_{50}=25.1\;{\mu}g/ml)$, and Cleyera japonica $(IC_{50}=26.2\;{\mu}g/ml)$ showed a potent radical scavenging activity. Further study for the identification of active compounds from these lead extracts might be warranted.

  • In continuous efforts for discovery of novel potent antitumor agents from natural products, fifty-seven methanolic extracts derived from indigenous Korean plants were primarily evaluated for in vitro cytotoxic activity in cultured human lung (A549) and colon (Col2) cancer cells. As a result, 16 plant extracts were found to be active against A549 cells and 15 extracts were active against Col2 cells in the criteria of $IC_{50}$<$50\;{\mu}g/ml$. In particular, the extracts of Calystegia soldanella $(IC_{50}$<$8.0\;{\mu}g/ml\;in\;A549;IC_{50}=27.4\;{\mu}g/ml\;in\;Col2)$, Heloniopsis orientalis $(IC_{50}=4.6\;{\mu}g/ml\;in\;A549; IC_{50}=4.5\;{\mu}g/ml\;in\;Col2)$, and Thuja koraiensis $(IC_{50}=1.2\;{\mu}g/ml\;in\;A549;IC_{50}=0.6\;{\mu}g/ml\;in\;Col2)$ showed a potent cytotoxic activity. Further study for the identification of active compounds from these lead extracts might be warranted.
