Antioxidant activities and physiological properties of Euphorbia humifusa extracts prepared using different solvents (추출용매에 따른 비단풀 추출물의 항산화 및 생리활성)

  • This study was conducted to investigate the antioxidant activities and physiological properties of Euphorbia humifusa extracts prepared using three different solvents (water, ethanol, and methanol). The highest total polyphenol content (293.25 mg/100 g) and total flavonoid content (21.05 mg/100 g) were observed in the methanol extract. The content of substances related to proanthocyanidin were highest in the water extract (8.42 mg/100 g), followed by methanol (5.70 mg/100 g) and ethanol (5.39 mg/100 g) extracts. The DPPH and ABTS radical scavenging activities of the methanol extract were 91.72% and 85.83%, respectively, at 50 mg% concentrations, which were higher than those of the other extracts. The extract reducing power decreased in the following order: ethanol > methanol > water. The methanol extract had relatively high antioxidant activity. The ${\alpha}-glucosidase$ and xanthine oxidase inhibitory activities of the methanol extract at a concentration of 10 mg% were somewhat higher than the other extracts. HMG-CoA reductase inhibitory activity in the water extract was slightly higher than in the methanol and water extracts. These results indicated that Euphorbia humifusa extracts were a high-value food ingredient due to their antioxidant activities and nutritional value.

Hepatoprotective Effect of the Methanol Fraction of Chinese Cabbage on Liver Injury in Rats Treated by bromobenzene (Chinese Cabbage 잎과 뿌리가 메탄올층의 Bromobenzene 간손상에 대한 보호효과)

  • Chinese cabbage is a vegetable of Cruciferous family. It was usually consumed as Kimchi. It was known to have amount of vitamin c. Recently the trend for the development of functional food combined with oriental herbs. For this aim the study was performed to evaluate the hepatoprotective effect via antioxidant activity of leaf and root of Sanchon Chinese cabbage(Brassica campetris L.) comparatively. The methanol extracts of Chinese cabbage were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzyme in vitro in bromobenzene-treated rats. The methanol extracts of chinese cabbage reduced bromobenzene-induced hepatic lipid peroxidation and inhibited the activity of xanthine oxidase. The methanol extracts of chinese cabbage did not activate amionopyrine N-demethylase, aniline hydroxylase and glutathione S-transferase. Epoxide hydrolase activity was decreased by bromobenzene, which was restored by pretreatment of the methanol extracts of chinese cabbage. The results suggest that the methanol extracts of Chinese cabbage is reduced by enhancing the activity of epoxide hydrolase.

Comparison of the Antioxidative Abilities of Commercial Gal Geun Tang (시판 갈근탕의 항산화능의 비교)

  • Each 3 g of five different commercial Gal Geun Tang extractive granules was mixed with 97 $m\ell$ of methanol or water for 1.5 hour at room temperature and filtered through a Whatman No. 1 filter paper, and the filtrate was used to determine antioxidant activity. Antioxidative abilities of each extracts were compared by measuring total phenol content (TPC), electron donating ability (EDA), inhibition of lipid peroxidation, and reducing power Average TPC of methanol extracts from five samples was 0.14 mM within the range of 0.07~0.23 mM, while that of water extracts was 0.23 mM, higher than that of methanol extracts. Average EDA of the methanol extracts was 85.54%, higher than that of the 0.1% BHT (72.26%). Average EDA values of the water extracts were 69.7%. Average value of inhibition against lipid peroxidation of the methanol extracts was 26.95%, and the range of five samples was 17.02~37.36%. Inhibition against lipid peroxidation of the water extracts showed relatively low value, 18.62%. Reducing power of the methanol extracts was 0.77 unit, which was 73.1% of 0.1% BHT, and that of water extracts was 0.88 unit. These results indicate that commercial Gal Geun Tang shows high antioxidant ability, though there are some differences depending on extract solvent and manufacturing company.

Antioxidant Activity of Soybean Extracts by Mixed Solvents Ethanol-Water and Methanol-Water (에탄올-물과 메탄올-물 혼합 용매를 이용한 대두 추출물의 항산화 효과)

  • The effective extraction of antioxidative substances from soybeans was studied by the mixed solvents of methanol or ethanol with water at 30$^{\circ}C$ and 85$^{\circ}C$. At accelerated peroxidation condition, POV (peroxide value) of soybean oil and TBARS (thiobarbituric acid reactive substances) value of egg yolk liposome with extracts added, were applied for the evaluation of antioxidative activity of the extracts. In the peroxidation of soybean oil, antioxidative substance of soybeans was extracted more effectively by methanol and at higher temperature 85$^{\circ}C$, and the deffated and roasted soybean extract (DRS) showed higher antioxidative activity. But in the Peroxidation of egg yolk liposome, methanol extract from the roasted soybeans (ROS) showed higher activity.

Screening on Cytotoxicity of Medicinal Plants against L1210 Cell (L1210 세포에 대한 약용 식물의 세포독성 검색)

  • For the research of cytotoxic natural products, 50 medicinal plants were extracted with benzene and methanol, separately, and screened against L1210 cells. From the results(Table I), 6 samples showed cytotoxicity both in benzene and methanol extracts of 17 samples in benzene extracts and 3 samples in methanol extracts, respectively. Generally, the cytotoxicity exhibited high frequency (34%) in benzene extract but low frequency in methanol extract (6%), it meant that active cytotoxic components had less polarity. $ED_{50}$ values less than $10\;{\mu}g/ml$ were observed in 17 medicinal plants.

  • We have examined radical scavenging effects, tyrosinase inhibition activities and anti-allergic activities on the methanol extracts of Opuntia ficus-indica cultivated in Cheju island. By the comparison of the four samples, methanol and 50% methanol extracts of respective fruits and stems of Opuntia, the most desirable activities were obtained in the sample of the stem's methanol extracts. Based on this analysis, further study to identify the active components in this sample is warranted.

Ammonia as Extractant and Reactant for Ginsenosides

  • In different approaches ginsenosides were extracted from Korean ginseng roots by ammonia and for comparison with methanol-water and water. The extracts have been analyzed qualitatively and quantitatively to evaluate yield and selectivity of extractions of ginsenosides. Water supplied the lowest yield. The yields of extracts with liquid ammonia were higher than those with methanol-water. However, this is partly due to the conversion of malonyl ginsenoside to normal ginsenosides by ammonia. It was proved by HPLC that malonyl-ginsenosides $m-Rb_1,\;m-Rb_2,$ m-Rc and m-Rd were converted to the corresponding neutral ginsenosides. Furthermore, ginsenosides from ginseng roots were extracted by alkaline methanol-water $(60\%)$ solutions. Alternatively, the extracts of the methanol-water $(60\%)$ extraction were treated with sodium hydroxide solution. Both methods also convert the malonyl-ginsenosides to neutral ginsenosides.

Antioxidative Activity of Feral Haw (Crataegus pinnatifida BUNGE) Seed Extracts Using Various Solvents (자생 산사(Crataegus pinnatifida BUNGE)씨의 추출 용매에 따른 항산화 활성)

  • The purpose of this study was to examine the antioxidative activity of feral haw (Crataegus pinnatifida BUNGE) seed extracts using 70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v/v), n-butanol and ethyl acetate (EA). The total phenol content of the five extracts ranged from 37.29 mg/g to 55.53 mg/g. Moreover, the content was high in the 70% methanol and, 70% ethanol extracts, but low in the n-butanol extract. On the contrary, the total flavonoid content decreased in the order of n-butanol (2.93 mg/g), EA (2.67 mg/g), 70% methanol (1.00 mg/g), 70% ethanol (0.88 mg/g) and CM (0.67 mg/g) extracts. The $NO_2$ radical scavenging activity, antioxidant activity by ${\beta}$-carotene bleaching assay and, superoxide dismutase (SOD) like ability decreased in the order of 70% methanol, 70% ethanol, CM, EA and, n-butanol extracts; further, a similar tendency was also observed in the total phenol contents. Overall, these results indicated that the antioxidative activity of feral haw seeds was closely related to the total phenol and flavonoid contents. Therefore, haw seeds might be usefully applied to natural antioxidants as well as functional foods.

Determination and Characterization of Inhibitory Activity of Mycelial Extracts from Several Korean Mushrooms against Helicobacter pylori (한국산 버섯추출물의 Helicobacter pylori에 대한 항균력 검색과 특성)

  • The effect of hot water and methanol extracts of 9 species from Korean mushrooms (Fomitopsis pinicola, Fomitella traxinea, Codyceps militaris, Phellinus linteus, Coriolus versicolor, Sparassis crispa, Ganoderma lucidum, Fomes fomentarius, Agaricus blazei) on the growth of Helicobacter pylori were examined using a Mueller-Hinton agar diffusion method. Hot water ($121^{\circ}C$) extracts from fruit bodies of F. traxinea, C. militaris, P. linteus, C. versicolor and F. pinicola showed $10{\sim}15\;mm$ inhibition zone against H. pylori. Methanol extracts of F. pinicola showed 44 mm inhibition zone, but another extracts showed no inhibition. Early fractions of DEAE-Sephadex A-25 column chromatography of methanol extracts from F. pinicola showed high inhibition activity against H. pylori.

Inhibitive Effects of Meju Extracts Made with a Single Inoculum of the Fungi Isolated from the Traditional Meju on the Human Leukemia Cell Line (전통 메주에서 분리된 단독균으로 제조한 메주추출물의 혈액암세포에 대한 저해효과)

  • In order to study the antitumoral effect of meju extracts, which was made with a single inoculum of the microorganism, the cytotoxicity effects on several human leukemia cells such as promyelocytic leukemia cell (HL60), histiocytic lymphoma cell (U937) and acute T-cell leukemia Jurkat cell, and lymphocyte were analyzed by MTT assay. Twenty one microbes, mainly fungal genera, were isolated from Korean traditional mejus of different regions. From those collected isolates, meju was manufactured and extracted with 80% methanol, respectively. Meju methanol extracts exhibited low activites in cytotoxicity tests on HL60 cell, but high antitumoral effects of meju methanol extracts were shown on U937 and Jurkat cells. Meju methanol extracts made with a genera of Mucor, Absidia and Aspergillus showed prominant cytotoxic activities, especially. However all these extracts had no inhibitory effects on the cell growth of lymphocyte under the same conditions.
