Characteristics of Citrus By-Product Ferment Using Bacillus subtilis as Starter Extracts. (Bacillus subtilis를 이용하여 발효시킨 감귤 가공부산물 추출물의 특성)

  • In this study, we investigated the biological activity of antioxidant and antibacterial activity of citrus by-product ferment. Hot water extracts and 80% methanol extracts from citrus byproduct of ferment were screened for antibacterial activity pathogenic bacteria by paper disc method. Among the various hot water extracts and 80% methanol extracts the Bacillus subtilis showed relatively strong antibacterial activities in the order. The reducing activity on the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and ${\cdot}OH$ radical scavenging potential were sequentially screened, in search for antioxidant activities of citrus by-product ferment.

Neuroprotective Effect of Extracts from Root Bark of Morus alba on Glutamate-induced Cytotoxicity in Neuronal Cells. (Glutamate가 유도하는 세포독성으로부터 신경세포를 보호하는 상백피 추출물의 효과)

  • This study evaluated the neuroprotective effect of extracts from the root bark of Morus alba (MA) against glutamate-induced cytotoxicity in neuronal cells. Glutamate-induced cytotoxicity was shown by MTT reduction assay. The neuroprotective effects of methanol, ethanol, and acetone extracts from MA against glutamate-induced cytotoxicity were measured. Among the three extracts, the methanolic extracts showed the highest protective effect, as determined by the results of an morphological assay, a lactate dehydrogenase release assay. Furthermore, the methanol extracts were fractionated sequentially with hexane, diethyl ether, ethyl acetate, and water layer according to degree of polarity. The hexane fractions exhibited a neuroprotective effect against glutamate-stressed N18-RE-105 cells. Therefore, these results suggest that extracts of MA could be a new potential candidate as a protective substance against glutamate-induced cytotoxicity.

Antibacterial and Antioxidant Activities of Solvent Extracts from Different Parts of Hagocho (Prunella vulgaris) (하고초 부위별 용매추출물의 항균 및 항산화 활성)

  • This study was designed to investigate the effects of antioxidant and antibacterial activities of water, ethanol and methanol extracts from root, stem and flower stalk of Hagocho (Prunella vulgaris). The solvent extract yields from root were higher than those from stem and flower stalk, and water extract showed the highest extraction yield against ethanol and methanol extracts. The contents of total phenolic and flavonoid in ethanol extract were significantly higher in stem extract compared with those of root and flower stalk. In the case of water and methanol extracts, however, the contents were the highest in flower stalk. The electron donating ability and reducing power in all test groups were significantly increased in a dose-dependent manner, and antioxidant activities were the highest in methanol extract. In extracts from different parts of Hagocho, the antioxidant activity was the highest in flower stalk followed by stem and root. ABTS radical scavenging ability of water and methanol extracts was above 50% at $100\;{\mu}g/mL$ concentration. Antibacterial activities did not show significant differences depending on parts of Hagocho. However, antibacterial activity of ethanol extract was higher than those of other extracts.

  • The root extract of pulsatilla koreana has been used as antibacterial, antiparasitic and anti-inflam-matory analgesic agents in Traditional Medicine in Korea. Thus anti-inflammatory and analgesic actions of the methanol and water extracts of the root were investigated by administration in oral and intravenous route. From the results, it is concluded that the extracts exhibited the potent anti-inflammatory and analgesic actions in intravenous administration, but did not show the actions in oral administration in animals.

  • Chestnut (Casranea crenata S. et Z.) leaves and flowers were extracted with 80% methanol and then fractionated with ethylacetate, methanol and water. Their antimicrobial activities in each fraction were investigated. Methanol fraction of the chestnut leaves and flower showed strong antimicrbial activities against both of Gram positive and Gram negative bacteria. The ethylacetate and water fraction, however, showed only weak antimicrobial activities when the antimicrobial activities were occurred. Minimal inhibitory concentration (MIC) of the methanol extracts of the chestnut leaves and flowers against 5 strains of Gram positive and Gram negative bacteria were at $60\;{\mu}g/disc$. The extracts of the chestnut leaves and flowers inhibited the growth of Bacillus subtilis at 0.5% (w/w) concentration. In order to investigate the effect of extraction methods on the B. subtilis, scanning electron microscope was used. The B. subtilis was damaged when the methanol extracts of the chestnut leves and flowers were at 500 ppm.

Antioxidative Activities of Fruit Extracts of Paulownia tomentosa Stued (참오동나무(Paulownia tomentosa Stud.) 열매 추출물의 항산화 활성)

  • The concentrations of total polyohenolic compound from extract of Paulownia tomentosa Stued. were water-soluble extract 7.98%, methanol extract 12.30% and ethanol extract 11.63%. The free radical scavenger activities of three extracts at the levels of 0.5% were higher than that of BHT(butylated hydroxytoluene). In antioxidative activities measured the concentrations of TBARS(thiobabituric acid reactive substances) in tissues microsome induced with Fe$\^$++/ascorbate, the water-soluble extract was errdctively suppressed in liver and spleen. In antioxidative activities determined by thiocyanate method against lipid peroxidation using linoleic acid, the methanol extract showed the higher compared with other extracts, but BHT showed the highest antioxidative activity. These results suggest that fruit extracts of Paulownia tomentosa Stued. showed to have relatively high concentrations of polyphenolic compounds and antioxidative activities.

  • The inhibition of rat lens aldose reductase (AR) in vitro using methanol extracts from Korean folk plants was investigated. Among them, the extracts of Saussurea grandifolia and Rumex crispus showed highest inhibition of AR. $IC_{50}$ values of the extracts from S. grandifolia and R. crispus were demonstrated 0.07 and 0.05 mg/ml, respectively. Korean folk plants such as S. grandifolia and R. crispus has a possibility of new natural resources for the inhibition of AR.

  • To evaluate the protective effects of extracts of silkworm on carbon tetrachloride-induced hepatotoxicity, serum glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase and lactic dehydrogenase activities, malondialdehydes values and glutathione-S-transferase activity were measured in ICR mice. Extracts of silkworm was administered orally at 30min after the administration of CCl4 Mice were sacrificed at 24h after the administration of extracts of silkworm. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride treatment were markedly increased than normal control but those levels were decreased by the treatment of butanol soluble fraction of silkworm methanol extract. Glutathione S-transferase activity was decreased by carbontetrachloride than control, but also inhibited by the treatment of butanol soluble fraction of silkworm methanol extract.

  • Twenty seven plant extracts were selected on the basis of ancient literature search for rheurnatoid arthritis or similar syndrome. Methanol extract of each plant was prepared and administered orally to rats everyday at a dose of 200 mg/kg/day. Experimental arthritis was induced by subplantar injection of heat-killed Mycobacterium butyricum to right hind paw of rats. This treatment provoked swelling of the treated paw in two phases, acute primary swelling and secondary arthritic swelling. An inhibition of secondary swelling was considered to be antiarthritic activity. Several plant methanol extracts such as Akebia quinata (caulis), Ephedla sinica (herba) and 5ophorae subprostratd (radix) were found to show significant inhibitory activity against secondary swelling at the dose tested. Our results strongly suggested an antiarthritic potential of these plant extracts.

Effect of Artemisia Capillaris Extract on the Growth of Food-Borne Pathogens (인진쑥 추출물이 식중독 유발 세균의 성장에 미치는 영향)

  • In this study, Artemisia capillaries, which has been used as a folk remedy, was investigated for its antimicrobial activity. First, the Aremisia capillaris was extracted with methanol at room temperature, and fractionation of the methanol extracts from Artemisia capillaris was carried out using petroleum ether, chloroform, and ethyl acetate. Second, the antimicrobial activity of the Artemisia capillaris extracts was determined using a paper disc method and minimum inhibitory concentration of ethyl acetate extracts from Artemisia capillaris against food-borne pathogens and food spoilage bacteria was measured. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Artemisia capillaris against Staphylococcus aureus and Salmonella typhimurium. The ethyl acetate extract of Artemisia capillaris showed strong antimicrobial activity against S. typhimurium at a concentration of 1,000 ppm. The 3,000 ppm of ethyl acetate extract from Artemisia capillaris retarded the growth of S. aureus and S. typhimurium for up to 6 hours.
