Karyological Study of Japanese Quail (Coturnix coturnix japonica) (일본산 메추리(Coturnix coturnix japonica)의 핵형연구)

  • Chromosome complements of Japanese quail (Coturnix coturnix japonica) were studied using several tissues which involving testis, leukocytes and embryos. The diploid count was estimated to be 2n=78. Analyzing the metaphase of secondary meiosis in spermatocytes, the haploid count estimated to be n=39. Morphometric analysis were studied by the centromeric index and relative length of 8 macro-chromosomes and Z, W chromosomes The differences of morphological feature were not significant among tissues. Exceptionally the chromosome 4 shelved a considerable variety in the presence of it's short arm.

  • The chromosome of Cipangopaludina chinensis malleata Chunchon area in 1988 was analysed by using aceto-orcein squash techniques of spermatogonial tissues to obtain mitotic and meiotic chromosomes. The chromosome cycle did not differ, in general, from that found in other snails, C. chinensis malleata has 18 diploid chromosomes and they were identified and classified into 2 groups. The mitotic chromosome complement of this species consists of 2 pairs of metacentric and 7 pairs of submetacentric chromosomes, Spermatogonial metaphase chromosomes range in length from 4.10 ${\mu}{\textrm}{m}$ for the largest pair to 2.20 ${\mu}{\textrm}{m}$ for the smallest pair.

The Effect of Abraxane on Cell Kinetic Parameters of HeLa Cells

  • Abraxane (nab-paclitaxel) is a member of the group of nano chemotherapeutics. It is approved for metastatic breast cancer and non small cell lung cancer. Trials for several cancer types including gynecological cancers, head and neck, and prostatic cancer are being studied. In this study, the antiproliferative and apoptotic effect of abraxane was evaluated on HeLa cell line originated from human cervix carcinoma. Three different doses ($D_1$=10 nM, $D_2$=50 nM, $D_3$=100 nM) were administered to HeLa cells for 24, 48 and 72 h. The 50 nM dose of abraxane decreased DNA synthesis from 4.62-0.08%, mitosis from 3.36-1.89% and increased apoptosis from 10.6-30% at 72 h. Additionally, tripolar metaphase plates were seen in mitosis preparations. In this study, abraxane effected cell kinetic parameters significantly. This results are consistent with other studies in the literature.

  • To obtain baseline data required for ploidy induction in Silurus asotus, a species with enormous aquacultural potential, histological events related to early embryonic development at 24$\pm$0.5$^{\circ}C$ are described. The histological location of the one-celled embryos indicated that metaphase stage of the first cleavage occurs at 31 min after insemination, when the highest yield can be obtained for the chromosome set manipulation.

  • The elucidation of the genetic changes of cholangiocarcinoma is very important for understanding the molecular mechanism of carcinogenesis and progression of cholangiocarcinoma. In order to identify the gains or losses of the copy number of DNA sequence in cholangiocarcinoma, we used comparative genomic hybridization to study 33 cases of cholangiocarcinoma. The whole DNAs from each tumor tissue were labeled with different fluorochromes and then simultaneously hybridized to normal metaphase spread chromosomes.(omitted)

A Chromosome Aberration Test of HMC05 on Cultured Chinese Hamster Lung Cells (HMC05의 배양 Chinese Hamster Lung 세포를 이용한 염색체이상 시험)

  • Objectives : We investigated genetic toxicity of HMCO5 in relation to chromosome aberration test on Cultured Chinese Hamster Lung (CHL) in the presence and absence of S-9 mix. Methods : Experimental groups were divided into two groups: with S-9mix (+S) or without S-9 mix (-S). -S group was also divided 2 series by treatment hours (6 hr: 6-S; or 24 hr; 24-S). Each group treated with vehicle only (complete culture medium), HMCO5 (1,250, 2,500, $5,000\;{\mu}g/ml$), and cyclophosphamide monohydrate (CPA) and ethylmethanesulfonate (EMS), respectively. Results : HMC05 did not show any aberrant metaphase. However, there were significant (p < 0.01) aberrant metaphases with CPA in S+ and with EMS in S-. Conclusions : These results indicate that HMC05 formula does not show any toxicity in chromosome aberration test.

Karyological Analysis of Somaclonal Variation in Callus Cells of Allium victorialis var. platyphyllum (산마늘의 캘러스세포에서 체세포군 변이의 핵학적 분석)

  • Calli obtained from basal disc explants of Allium victorialis var.platyphyllum wre grown in three kinds of nutrient media (MS, BDS, and B5), and the frequencies of mitotic index and the chromosomal aberrations were analysed. The mitotic index varied from 0.55% to 1.01% with respect to culture media and ages. The mitotic irregularities like micro-, bi- and multi-nuclei, chromosome bridge and laggards were noted in each types of calli. The chromosome number variations observed in metaphase stage were identified aneuploid and tetraploid. Structural variations such as dicentric chromosomes, centromere breakage and small chromosomes were observed. Relationship between basal medium and chromosomal variability was not observed in this study. But, in BDS medium, NAA and BA had a more effect on number variation than kinetin.

Chromosome Variation in Suspension Cells Derived from Cultured Immature Embryo of Triticum spp. (밀(Triticum spp.)의 미성숙배로부터의 유도한 현탁 배양세포에서의 염색체 변이)

  • Suspension cell lines have been newly established from the calli derived from the immuature embryo culture of hexapolid (Triticum aestivum var. sicco), tetrapolid (T. durum) and diploid (T. tauchii or Aegilops squarrosa) wheat species. The chromosomal variation in suspension cultured cell lines was examined and old cell line, C82d, established from T. aestivum var. copain was also used. New method using 1-bromonaphthalene for metaphase rapping of suspension cells was developed. Variation in chromosome number was observed among all the suspension lines. Cells with doubled chromosome number and deleted chromosome were also observed. Extensive structural changes in chromosome were found in C82d line. Chromosome aberrations showed loss of chromosome arms and chromosome segment. The mean chromosome number in suspension cells of T. aestivum var. sicco was 40, in C82d line 33, in T. durum 28 and in T. tauchii 14. The stability of chromosome in suspension cells of diploid and tetrapolid wheats was higher than that of hexaploid wheat.
